Clearance and ingestion rates of Brachionus plicatilis were measured using 14C-labeled Isochrysis galbana Tahiti. Experiments were conducted at 20–22 °C, 20 ppt salinity, and algal concentrations ranging from 0.13–64 mg C 1–1. Clearance rates were constant and maximal at concentrations <2 mg C 1–1, with maximum rates ranging from 3.4–6.9 µl ind.–1 hr–1. The ingestion rate varied with food concentration, and was described by a rectilinear model. The maximum ingestion rate varied considerably, and was dependent on the growth rate of the rotifers. Depending on the pre-conditions, B. plicatilis ingested about 0.5 to 2 times its body carbon per day at saturating food concentrations.  相似文献

We studied the effect of different concentrations (0, 3, 6, 9 and 12 g l–1) of sodium chloride at one food level of Chlorella (1×106 cells ml–1) on competition between the rotifers B. rotundiformis and H. jenkinae, both of which were isolated from a saline lake. The population growth experiments were conducted for 3 weeks. Both the rotifer species did not survive beyond one week at a salinity of 0 g l–1. Regardless of salt concentration and the presence of a competitor, H. jenkinae reached higher densities than B. rotundiformis. When grown alone, both B. rotundiformis and H. jenkinae showed optimal peak population densities at the salinity of 6 and 9 g l–1. Since biomass wise, B. rotundiformis was larger than H. jenkinae, it showed a lower numerical abundance. Thus, the maximum peak population densities of B. rotundiformis and H. jenkinae recorded in this study were 107±3 and 203±28 ind. ml–1. The maximal rates of population increase for B. rotundiformis and H, jenkinae when grown alone were 0.264±0.003 and 0.274±0.004, respectively. Our results also indicated that B. rotundiformis and H. jenkinae coexisted better at a salinity of 6 and 9 g l–1 of sodium chloride while a salinity of 3 g l–1 favoured Hexarthra over B. rotundiformis. At 12 g l–1, both the rotifer species grown alone or together showed lower growth rates compared to those at lower salinity levels. Except 0 g l–1, in all other salinity treatments, H. jenkinae was a superior competitor to B. rotundiformis.  相似文献

Evidence for the influence of food type and heavy metals on shell growth and fertility is presented for a freshwater population of the snail P. jenkinsi. When fed an excess of lettuce or lamb heart (protein source), growth rates were higher for lettuce. Highest growth rates occurred at a diet of lettuce plus lamb heart. Fertility was favoured by a diet of lamb heart. When fed an excess of lettuce, the EC50 growth values were 16 µg Cd l–1, 13 µg Cu l–1, and 103 µg Zn l–1 in lake water; snail fertility was inhibited at 25 µg Cd l–1 and 30 µg Cu l–1. A diet of lake detritus spiked with Cd or Cu resulted in a decrease of approximately 50% in growth rates, when compared with growth on non-spiked detritus. Spiked detritus lost metals into lake water. Food type positively interacted with metal stress, both for growth rate and fertility. The assessment of inhibitory effects of detritus contaminated either in the field or, notably, by spiking, and serving as food source for deposit feeders is hampered by sampling problems in the field and by redistribution processes of pollutants between particles and water in laboratory-scale experiments.  相似文献

Two serine carboxypeptidases, MpiCP-1 and MpiCP-2, were purified to homogeneity from Monascus pilosus IFO 4480. MpiCP-1 is a homodimer with a native molecular mass of 125 kDa composed of two identical subunits of 61 kDa, while MpiCP-2 is a high mass homooligomer with a native molecular mass of 2,263 kDa composed of about 38 identical subunits of 59 kDa. This is unique among carboxypeptidases and distinguishes MpiCP-2 as the largest known carboxypeptidase. The two purified enzymes were both acidic glycoproteins. MpiCP-1 has an isoelectric point of 3.7 and a carbohydrate content of 11%, while for MpiCP-2 these values were 4.0 and 33%, respectively. The optimum pH and temperature were around 4.0 and 50°C for MpiCP-1, and 3.5 and 50°C for MpiCP-2. MpiCP-1 was stable over a broad range of pH between 2.0 and 8.0 at 37°C for 1 h, and up to 55°C for 15 min at pH 6.0, but MpiCP-2 was stable in a narrow range of pH between 5.5 and 6.5, and up to 50°C for 15 min at pH 6.0. Phenylmethylsulfonylfluoride strongly inhibited MpiCP-1 and completely inhibited MpiCP-2, suggesting that they are both serine carboxypeptidases. Of the substrates tested, benzyloxycarbonyl-l-tyrosyl-l-glutamic acid (Z-Tyr-Glu) was the best for both enzymes. The Km, Vmax, Kcat and Kcat/Km values of MpiCP-1 for Z-Tyr-Glu at pH 4.0 and 37°C were 1.33 mM, 1.49 mM min–1, 723 s–1 and 545 mM–1 s–1, and those of MpiCP-2 at pH 3.5 and 37°C were 1.55 mM, 1.54 mM min–1, 2,039 s–1 and 1,318 mM–1 s–1, respectively.  相似文献

Degradation of polyaromatic hydrocarbons by Burkholderia cepacia 2A-12   总被引:1,自引:1,他引:0  
A new strain of bacterium degrading polyaromatic hydrocarbons (PAHs), Burkholderia cepacia 2A-12, was isolated from oil-contaminated soil. Of three PAHs, the isolated strain could utilize naphthalene (Nap) and phenanthrene (Phe) as a sole carbon source but not pyrene (Pyr). However, the strain could degrade Pyr when a cosubstrate such as yeast extract (YE) was supplemented. The PAH degradation rate of the strain was enhanced by the addition of other organic materials such as YE, peptone, glucose, and sucrose. YE was a particularly effective additive in stimulating cell growth as well as PAH degradation. When 1 g YE l–1, an optimum concentration, was supplemented into the basal salt medium (BSM) with 215 mg Phe l–1, the specific growth rate (0.30 h–1) and Phe-degrading rate (29.6 mol l–1 h–1) were enhanced approximately ten and three times more than those obtained in the BSM with 215 mg Phe l–1, respectively. Both cell growth and PAH degradation rates were increased with increasing Phe and Pyr concentrations, and B. cepacia 2A-12 had a tolerance against Phe and Pyr toxicity at the high concentration of 730–760 mg l–1. Through kinetic analysis, the maximum specific growth rate ( max) and PAH degrading rate ( max) for Phe were obtained as 0.39 h–1 and 300 mol l–1 h–1, respectively. Also, max and max for Pyr were 0.27 h–1 and 52 mol l–1 h–1, respectively. B. cepacia 2A-12 could simultaneously degrade crude oil as well as PAHs, indicating that this bacterium is very useful for the removal of oils and PAHs contaminants.  相似文献

The best culture medium composition for the production of bikaverin by Gibberella fujikuroi in shake-flasks, i.e. 100 g glucose l–1; 1 g NH4Cl l–1; 2 g rice flour l–1; 5 g KH2PO4 l–1 and 2.5 g MgSO4 l–1, was obtained through a fractional factorial design and then scaled-up to a fluidized bioreactor. The effects of carbon and nitrogen concentrations, inoculum size, aeration, flow rate and bead sizes on batch bikaverin production using immobilized G. fujikuroi in a fluidized bioreactor were determined by an orthogonal experimental design. Concentrations of up to 6.83 g bikaverin l–1 were obtained when the medium contained 100 g glucose l–1 and 1 g NH4Cl l–1 with an inoculum ratio of 10% v/v, an aeration rate of 3 volumes of air per volume of medium min–1, and a bead size of 3 mm. Based on dry weight, the bikaverin production was 30–100 times larger than found in submerged culture and approximately three times larger than reported for solid substrate fermentation.  相似文献

Carboxypeptidase produced by Monascus purpureus IFO 4478 was purified to homogeneity. The purified enzyme is a heterodimer with a molecular mass of 132 kDa and consists of two subunits of 64 and 67 kDa. It is an acidic glycoprotein with an isoelectric point of 3.67 and 17.0% carbohydrate content. The optimum pH and temperature were 4.0 and 40 °C, respectively. The enzyme was stable between pH 2.0 and 8.0 at 37 °C for 1 h, and up to 50 °C at pH 5.0 for 15 min. The enzyme was strongly inhibited by piperastatin A, diisopropylfluoride phosphate (DFP), phenylmethylsulfonylfluoride (PMSF), and chymostatin, suggesting that it is a chymotrypsin-like serine carboxypeptidase. Monascus purpureus carboxypeptidase was also strongly inhibited by p-chloromercuribenzoic acid (PCMB) but not by ethylenediaminetetraacetic acid (EDTA) and 1,10-phenanthroline, indicating that it requires cysteine residue but not metal ions for activity. Benzyloxycarbonyl-l-tyrosyl-l-glutamic acid (Z-Tyr-Glu), among the substrates tested, was the best substrate of the enzyme. The Km, Vmax, Kcat, and Kcat/Km values of the enzyme for Z-Tyr-Glu at pH 4.0 and 37 °C were 0.86 mM, 0.917 mM min–1, 291 s–1, and 339 mM–1 s–1, respectively.  相似文献

Feeding of Euchlanis dilatata lucksiana, a brachionid rotifer isolated from the Loosdrecht Lakes (The Netherlands), was examined in the laboratory using 14C-labelled food. The gut-filling time at a food concentration of 9.6 µg C ml–1 was about 15 minutes. Animals which were fed on 3 size fractions of the lake seston (< 7 µm, 7–15 µm, and 15–33 µm) showed a clear preference (Jacobs' selectivity index) for the largest size fraction. This fraction was composed predominantly of filamentous cyanobacteria. For animals weighing 0.37–0.49 µm C ind.–1 the daily ration (daily food consumption per unit body weight × 100) ranged from 50 to 100% at food levels of 2 mg C l–1 and below, but increased to 150–250% at food concentrations of 5 mg C l–1 and above. The assimilation efficiency was 100% up to 5 mg C l–1 of food, but decreased to about 80% at higher food levels.  相似文献

This paper reports on a two-year analysis of the wind climateand its effect on phytoplankton primary production in ashallow (mean depth = 1.9 m), hypertrophic South Africancoastal lake, Zeekoevlei. The lake is subject to continuousmixing of the euphotic zone (Z eu = 0.8 m), andcomplete mixing of the water column to the mean depth on adaily basis. Median annual wind speeds, prevailing fromeither the north or the south, were 6.4 m s–1. There wasan almost total absence of calms, measured as hourly meanwind speeds of <1 m s–1. Notwithstanding the highfrequency of mixing, the lake supports a dense population ofphytoplankton, dominated by Cyanophyte and Chlorophytespecies. Mean concentrations of chlorophyll-a were240 g l–1. The attenuation of photosyntheticallyavailable radiation, PAR, was high, with mean K dvalues of 6.4 m–1 and water transparencies of <0.5 m.Levels of primary productivity, determined using the lightand dark bottle oxygen method, were very high, comparable toor exceeding that of the most productive systems yet studied.Maximum volumetric productivity ranged from 525 to 1524 mg Cm–3 h–1, and was confined to the upper 0.5 m of thewater column. Daily areal productivity, P d,varied between 1.2 and 4.3 g C m–2 d–1, and that ofthe maximum chlorophyll-a specific photosynthetic rate,P B max, between 1.6 and 7.9 mg C (mgChl-a)–1 h–1. Primary production was limited bywater temperature and the attenuation of PAR. The highfrequency of wind-induced mixing resulted in regular mixingof the phytoplankton through the euphotic zone, and reducedthe overall importance of P max at a single layer inthe depth profile. AxenicTrentepohlia odorata was cultured at three different NH4Cl levels (3.5 × 10–2, 3.5 × 10–3, 3.5 × 10–4 M) and three different light intensities (48, 76, 122 µmol m–2 s–1). Chloride had no effect on growth over this range of concentration. High light intensity and high NH4Cl concentration enhanced the specific growth rate. The carotenoid content increased under a combination of high light intensity and low N concentration. WhenD. bardawil was exposed to the same combination of growth conditions, there was an increase in its carotenoid content. The light saturation and the light inhibition constants (K s andK i, respectively) for growth, and the saturation constant (K m) for NH4Cl were determined. TheK s andK i values were higher inT. odorata (66.7 and> 122 mol m–2 s–1, respectively) than inD. bardawil (5.1 and 14.7 µmol m–2 s–1, respectively). Summary Microbial conversion of glycerol into dihydroxyacetone (DHA) byGluconobacter oxydans was subjected to inhibition by excess substrate. Comparison of cultures containing increasing initial DHA contents (0 to 100 g l–1) demonstrated that DHA also inhibited this fermentation process. The first effect was on bacterial growth (cellular development stopped when DHA concentration reached 67 gl–1), and then on oxidation of glycerol (DHA synthesis only occurred when the DHA concentration in the culture medium was lower than 85 g l–1). Productivity, specific rates and, to a lesser extent, conversion yields decreased as initial concentrations of DHA increased. The changes in the specific parameters according to increasing initial DHA contents were described by general equations. Spirulina platensis (= Arthrospira fusiformis) was isolated from Lake Chitu, a saline, alkaline lake in Ethiopia, where it forms an almost unialgal population. Optimum growth conditions were studied in a turbidostat. Cultures grown in modified Zarrouk's medium and exposed to a range of light intensities (20–500 µmol photons m–2s–1) showed a maximum specific growth rate (µmax) of 1.78 d–1. Quantum yield for growth (µ) was 3.8% at the optimum light for growth of 330 µmol photons m–2s–1, and ranged from 2.8 to 9.4%. With increase in irradiance, the chlorophyll a concentration decreased, and the carotenoids/chlorophyll a ratio increased by a factor of 2.4. The phosphorus to carbon ratio (P/C) showed some variation, while the nitrogen to carbon ratio (N/C) remained relatively constant, thus causing fluctuations in the N:P ratio (7–11) of cells. An optimum N:P ratio of about 7 was attained in cells growing at the optimum light for growth. Results from the continuous culture experiments agreed well with maximum values of photosynthetic efficiency given in the literature for natural populations of S. platensis in the soda lakes of East Africa, Lake Arenguade (Ethiopia), and Lake Simbi (Kenya).  相似文献

A flow-through method was employed to study the algal part of the diet spectrum of the calanoid Arctodiaptomus salinus in the surface layer of the lake and in the phytoplankton biomass maximum zone (10 m deep). Daily consumption rates of this calanoid differed markedly in these layers, being 1 and 11 g ind –1 d–1, respectively. The cyanobacterial taxa, Lyngbya contorta and Microcystis sp., accounted for more than a half of the ration. Nevertheless, the negative Ivlev electivity coefficient indicated that Arctodiaptomus prefers none of these two taxa. The low measured uptakes suggest that the energy expenditure is compensated by other resources, presumably microzooplankton and detritus.  相似文献

During four cruises in continental shelf waters of the northern Gulf of Mexico in the winters of 1981–83, we performed quantitative studies on the grazing of the copepods Acartia tonsa, Centropages velificatus, and Eucalanus pileatus, on phytoplankton using natural particulate assemblages as food. Stations were in, or adjacent to the plume of the Mississippi River, thereby providing wide spectra of phytoplankton and suspended riverine particulate concentrations. Phytoplankton cell volume was converted to carbon, and this, coupled with carbon content measurements of these three copepod species, allowed comparisons of daily ingestion effort even though the copepods were of different sizes. Data were expressed in the same units (% of copepod body carbon ingested copepod –1 d–1) for each species. Over similar ranges of phytoplankton carbon concentrations (0.21–92.06 gCl–1), Acartia tonsa had higher carbon-specific ingestion rates (x = 22.31%, range = 0.08–152.37%) than C. velificatus (x = 2.8%, range = 0.00–31.09 %) or E. pileatus (x = 1.27%, range = 0.10–2.80%). Carbon-specific ingestion rates increased with increasing phytoplankton carbon concentration for A. tonsa (R2 = 0.81) and there was no evidence of saturated feeding on the carbon concentrations offered. A similar, but weaker trend was evident for E. pileatus (R2 = 0.71), but not C. velificatus (R2 = 0.49). Over a wide range of suspended particulate concentrations (10.6–95.2 mg l–1), there was no systematic effect of particulates on carbon-specific ingestion rate for any of the three copepod species. However, A. tonsa appeared more adept at grazing in highly turbid water than C. velificatus or E. pileatus.  相似文献

The uptake of ammonium, nitrate and phosphate by laboratory-grown young sporophytes of Laminaria abyssalis was measured in a perturbed system (batch mode) at 18 °C and 35 ± 5 µE m–2 s–1 photon flux density. Uptake of all appeared to follow saturation-type nutrient uptake kinetics. The NO inf3 sup– (K s = 14.0 µM, V max = 5.0 µmol h–1 g–1 dry wt) and NH inf4 sup+ (K s = 4.6 µM, V max= 2.0 µmol h–1 g–1 dry wt) were taken up simultaneously, although NH inf4 sup+ was taken up more rapidly. Values of K 3 and V max for phosphate were, respectively, 2.21 µM and 0.83 µmol h–1 g–1 dry wt. Nitrate and phosphate were both consumed in similar rates (V max /Ks 0.37) at low concentrations. NH inf4 sup+ , thus, might be a more efficient form of N fertilizer if artificial enrichment of seawater is used.  相似文献

A detailed study of the life history of the rotifer Brachionus plicatilis was done at 20 °C, 20 ppt salinity, and 90 mg C 1–1 food concentration. Rotifers were grown individually in culture plate wells (150 µl culture volume) and fed Isochrysis galbana Tahiti, Tetraselmis sp., Nannochloris atomus, or a l : 1 mixture (weight) of two of the algae. Observations were made every 2–8 hr and rotifers were sized and transferred to new food daily. A total of 19 different parameters were compared. Rotifers fed Isochrysis averaged 21 offspring per female, a 6.7 day reproductive period, a lifespan of 10.5 days and a mean length of 234 µm. After Isochrysis, the foods giving the highest growth, survival, and reproduction in decreasing order were Isochrysis + Nannochloris, Nannochloris, Isochrysis + Tetraselmis, Tetraselmis + Nannochloris, and Tetraselmis. Although the small volume culture system used in this study seems appropriate for studying life history of B. plicatilis, the results cannot always be directly applied to larger cultures.  相似文献

Experiments conducted on samples collected from a large oligotrophic lake revealed the following: (1) excretion rates of PO inf4 sup3– by single Daphnia thorata were below detection (5 pmol animal–1 min–1) in 20 ml of oligotrophic lake water over a period of 10 min, (2) experimental addition of D. thorata to 20 ml aliquots of lake water decreased community-wide microbial uptake of PO inf4 sup3– on two occasions (as measured by 32PO inf4 sup3– incorporation), and (3) the presence of D. thorata increased uptake by organisms smaller than 1µm, and decreased uptake by large phytoplankton. The specific mechanism for these responses remains unclear, but the results imply that when phytoplankton larger than 1µm encounter cm scale patches of water recently occupied by Daphnia they may experience decreased PO inf4 sup3– availability rather than elevated concentrations of PO inf4 sup3– caused by excretion. We show that 32P uptake experiments using natural plankton assemblages can be influenced by the presence or absence of large zooplankton, and that neither grazing, turbulence, nor PO inf4 sup3– excretion can account for this influence.  相似文献

A fourth order Runge–Kutta approximation was used to determine the Monod kinetics of Candida rugopelliculosa by using unsteady state data from only one continuous unsteady state operation at a fixed dilution rate. The maximum microbial growth rates, max, and half saturation coefficient, K s, were 0.82 ± 0.22 h–1 and 690 ± 220 mg soluble chemical oxygen demand (SCOD) l–1, respectively. The microbial yield coefficient, Y, and microbial decay rate coefficient, k d, were 1.39 ± 0.22 × 104 cells mg–1 SCOD and 0.06 ± 0.01 h–1, respectively.  相似文献

The kinetic properties of a microsomal gill (Na+,K+)-ATPase from the blue crab Callinectes danae were analyzed using the substrate p-nitrophenylphosphate. The (Na+,K+)-ATPase hydrolyzed PNPP obeying cooperative kinetics (n=1.5) at a rate of V=125.4±7.5 U mg−1 with K0.5=1.2±0.1 mmol l−1; stimulation by potassium (V=121.0±6.1 U mg−1; K0.5=2.1±0.1 mmol l−1) and magnesium ions (V=125.3±6.3 U mg−1; K0.5=1.0±0.1 mmol l−1) was cooperative. Ammonium ions also stimulated the enzyme through site–site interactions (nH=2.7) to a rate of V=126.1±4.8 U mg−1 with K0.5=13.7±0.5 mmol l−1. However, K+-phosphatase activity was not stimulated further by K+ plus NH4+ ions. Sodium ions (KI=36.7±1.7 mmol l−1), ouabain (KI=830.3±42.5 μmol l−1) and orthovanadate (KI=34.0±1.4 nmol l−1) completely inhibited K+-phosphatase activity. The competitive inhibition by ATP (KI=57.2±2.6 μmol l−1) of PNPPase activity suggests that both substrates are hydrolyzed at the same site on the enzyme. These data reveal that the K+-phosphatase activity corresponds strictly to a (Na+,K+)-ATPase in C. danae gill tissue. This is the first known kinetic characterization of K+-phosphatase activity in the portunid crab C. danae and should provide a useful tool for comparative studies.  相似文献

The optimal growth of Cistanche deserticola callus and formation of phenylethanoid glycosides (PeG) was at 25°C with light irradiation intensity of 24 mol m–2 s–1 on solidified B5 media supplemented with 0.5 mg 6-benzylaminopurine l–1, 10 mg gibberellin l–1, 800 mg casein hydrolysate l–1 and 20 g sucrose l–1. After 30 d culture, the biomass reached 15.5 g dry wt callus l–1 medium and its PEG content was 10.7% (w/w). The PeG content was 42%–127% higher than those in explants.  相似文献
