Complementary DNAs encoding two types of acetylcholinesterase (ACHE) were isolated from the silkworm, Bombyx mori. The type 1 (Bmacel) and type 2 (Bmace2) ORFs are 2052 and 1917 bp in length, respectively. Both the complete ORFs of the Bmaces and C- terminal truncated forms were recombined into the Bacmid baculovirus vector under the control of the polyhedrin promoter and expressed in Trichoplusia ni (Tn-5B 1-4) cells. The resulting products exhibited ACHE activity and glycosylation of the expressed proteins. An inhibition assay indicated that the ace2-type enzyme was more sensitive than the acel-type enzyme to inhibition by eserine and paraoxon.  相似文献

【目的】非编码RNA(non-coding RNA,ncRNA)在家蚕Bombyx mori发育过程中具有重要调控作用。本研究旨在探索ncRNA参与家蚕神经系统发育的分子机理。【方法】采用实时荧光定量PCR技术对22个中等长度的ncRNA及3个ncRNA的邻近编码基因在家蚕幼虫神经系统中的表达水平进行检测。【结果】8个ncRNA(包括1个C/D box snoRNA,4个H/ACA box snoRNA和3个不能归类的ncRNA)在家蚕5龄幼虫神经组织和非神经组织中均有表达,且在胸腹神经中的表达量明显高于头部神经中的表达量。其中,snoRNA Bm-51,Bm-18和Bm-86在胸腹神经中的表达量分别是头部神经中的23,5和4.7倍。进一步研究发现,这3个内含子起源的ncRNA与其宿主基因在家蚕神经系统中的表达趋势一致,宿主基因在胸腹神经中的表达量也明显高于头部神经中的表达量。【结论】本研究筛选到的在家蚕不同神经部位存在差异表达的ncRNA,特别是在胸腹神经中高表达的ncRNA可能协同其邻近编码基因,参与家蚕神经发育或神经活动过程。该结果为研究ncRNA参与家蚕神经系统发育提供了分子依据,为从非编码RNA角度探索鳞翅目害虫防治提供了新的思路。  相似文献

The domesticated silkworm (Bombyx mori) was domesticated from wild silkworm (Bombyx mandarina) more than 5,000 years ago. During domestication, body color between B. mandarina and B. mori changed dramatically. However, the molecular mechanism of the silkworm body color transition is not known. In the present study, we examined within- and between-species nucleotide diversity for eight silkworm melanin synthesis pathway genes, which play a key role in cuticular pigmentation of insects. Our results showed that the genetic diversity of B. mori was significantly lower than that of B. mandarina and 40.7% of the genetic diversity of wild silkworm was lost in domesticated silkworm. We also examined whether position effect exists among melanin synthesis pathway genes in B. mandarina and B. mori. We found that the upstream genes have significantly lower levels of genetic diversity than the downstream genes, supporting a functional constraint hypothesis (FCH) of metabolic pathway, that is, upstream enzymes are under greater selective constraint than downstream enzymes because upstream enzymes participate in biosynthesis of a number of metabolites. We also investigated whether some of the melanin synthesis pathway genes experienced selection during domestication. Neutrality test, coalescent simulation, as well as network and phylogenetic analyses showed that tyrosine hydroxylase (TH) gene was a domestication locus. Sequence analysis further suggested that a putative expression enhancer (Abd-B-binding site) in the intron of TH gene might be disrupted during domestication. TH is the rate-limiting enzyme of melanin synthesis pathway in insects. Real-time polymerase chain reaction assay did show that the relative expression levels of TH gene in B. mori were significantly lower than that in B. mandarina at three different developmental stages, which is consistent with light body color of domesticated silkworm relative to wild silkworm. Therefore, we speculated that expression change of TH gene may contribute to the body color transition from B. mandarina to B. mori. Our results emphasize the exceptional role of gene expression regulation in morphological transition of domesticated animals.  相似文献

Mulberry latex contains extremely high concentrations of alkaloidal sugar mimic glycosidase inhibitors, such as 1,4-dideoxy-1,4-imino-D-arabinitol (D-AB1) and 1-deoxynojirimycin (DNJ). Although these compounds do not harm the silkworm, Bombyx mori, a mulberry specialist, they are highly toxic to insects that do not normally feed on mulberry leaves. D-AB1 and DNJ are strong inhibitors of alpha-glucosidases (EC 3.2.1.20); however, they do not affect the activity of beta-fructofuranosidases (EC 3.2.1.26). Although alpha-glucosidase genes are found in a wide range of organisms, beta-fructofuranosidase genes have not been identified in any animals so far. In this study, we report the identification and characterization of beta-fructofuranosidase genes (BmSuc1 and BmSuc2) from B. mori. The BmSuc1 gene was highly expressed in the midgut and silk gland, whereas the expression of BmSuc2 gene was not detected. BmSuc1 encodes a functional beta-fructofuranosidase, whose enzymatic activity was not inhibited by DNJ or D-AB1. We also showed that BmSUC1 protein localized within the midgut goblet cell cavities. Collectively, our data clearly demonstrated that BmSuc1 serves as a sugar-digesting enzyme in the silkworm physiology. This anomalous presence of the beta-fructofuranosidase gene in the B. mori genome may partly explain why the silkworm can circumvent the mulberry's defense system.  相似文献

The VAP-peptide (BmACP-6.7) is a hydrophobic peptide localized in adult cuticle of the silkworm, Bombyx mori. We isolated and characterized the VAP-peptide gene as a useful marker gene to analyze molecular mechanisms of terminal differentiation processes in the adult. The gene is composed of two exons interrupted by one intron. The 5' upstream promoter region is shown to bear a nucleotide sequence similar to the cis-element that is recognized and bound by the Bombyx mori FTZ-F1 protein (BmFTZ-F1). Expression of the BmFTZ-F1 gene preceded expression of the VAP-peptide gene and injection of 20-hydroxyecdysone suppressed the expression of both genes. An in vitro binding assay indicated direct interaction of BmFTZ-F1 with the VAP-peptide gene promoter sequence. Therefore, BmFTZ-F1 is proposed to be a possible factor regulating the stage-specific expression of the VAP-peptide gene towards adult life.  相似文献

明确农药对家蚕的影响对综合评价农药的生态效应与合理使用农药具有理论和实践意义.从生长发育、经济性状和食物利用方面综述了农药对家蚕的亚致死效应;从生理代谢、组织器官亚显微结构和培养细胞方面介绍了亚致死效应产生的机制,并结合杀虫剂对其它鳞翅目昆虫亚致死影响方面的研究结果对其进行了讨论,提出了今后加强研究的重点.总的来说,亚致死剂量农药的胁迫可使家蚕幼虫眠性不齐,眠蚕体重和蛹重减轻,发育历期延长,严重者结茧(上蔟)时间推迟,且吐丝营茧功能失常,最终导致蚕茧质量下降.桑叶上残留的农药在一定程度上还会对家蚕的取食行为以及食物摄入后的消化吸收产生影响.农药对家蚕亚致死效应产生的机制目前主要有3个:家蚕体内正常生理水平和代谢平衡受到扰动、家蚕的一些重要组织器官如中肠和后丝腺等微形态结构受到损伤和家蚕卵巢细胞出现凋亡现象.农药对家蚕的亚致死效应因农药种类、试验剂量和家蚕受药时间而异,产生的机理也十分复杂.今后应着重开展新农药对家蚕亚致死效应研究,特别是对生化和分子机制方面的研究以及农药复合污染对家蚕影响的研究.  相似文献

【目的】明确基于电穿孔的基因功能分析方法在家蚕Bombyx mori活体内的应用实效。【方法】针对调控家蚕幼虫体表斑纹黑色素合成的靶基因Wnt1 (Wingless),人工合成特异性siRNA,向4龄第3天家蚕幼虫注射Wnt1 siRNA并进行电穿孔作为处理组(ERFA-RNAi),以注射Wnt1 siRNA但未进行电穿孔的幼虫作为阴性对照组(negative control, NC),解剖5龄幼虫斑纹区的表皮,用实时定量RT-PCR测定表皮中Wnt1的相对表达量,验证电穿孔介导的RNAi效果。带有增强型绿色荧光蛋白报告基因EGFP和红色荧光蛋白报告基因DsRed2表达元件的转座子载体pPIG-A3GR,通过电穿孔导入家蚕2龄幼虫;正常饲养72 h后,用荧光体视显微镜观察幼虫中EGFP和DsRed2的表达,验证当世代家蚕的嵌合体转基因。【结果】家蚕4龄第3天幼虫中导入Wnt1的特异性siRNA后,5龄幼虫体表特定部位斑纹的形成明显受到抑制,实时定量RT PCR分析表明5龄幼虫表皮中Wnt1的表达水平显著降低。嵌合体转基因家蚕的阳性率达56.60%,并且两个荧光报告基因EGFP和DsRed2在幼虫、蛹及成虫期持续表达。【结论】电穿孔技术可快捷、高效地向家蚕活体内导入外源RNA或DNA,是家蚕乃至其他昆虫基因功能解析的有效方法。  相似文献

A genome-wide analysis of innate immunity-related genes and gene families was conducted using the silkworm, Bombyx mori. We identified orthologs for a large number of genes involved in insect immunity that have been reported from Drosophila melanogaster (Diptera), Anopheles gambiae (Diptera), Apis mellifera (Hymenoptera) and Tribolium castaneum (Coleoptera). B. mori has a unique recognition gene and antimicrobial peptide genes that are not present in the Drosophila, Anopheles, Apis and Tribolium genomes, suggesting a lineage-specific gene evolution for lepidopteran insects. The comparative analysis of the insect immune repertoires indicated a dynamic and flexible gene expansion in recognition, modulation and effector mechanisms due to different selection pressures. Differential gene regulation by different bacterial species was found in PGRP and Serpin genes, suggesting that Bombyx has a highly selective gene regulation system depending on bacterial species.  相似文献

Signal inactivation is a crucial step in the dynamic of olfactory process and involves various Odorant-Degrading Enzymes. In the silkworm Bombyx mori, one of the best models for studying olfaction in insects, the involvement of an antennal-specific aldehyde oxidase in the degradation of the sex pheromone component bombykal has been demonstrated over the three past decades by biochemical studies. However, the corresponding enzyme has never been characterized at the molecular level. Bioinformatic screening of B. mori genome and molecular approaches have been used to isolate several candidate sequences of aldehyde oxidases. Two interesting antennal-expressed genes have been further characterized and their putative functions are discussed in regard to their respective expression pattern and to our knowledge on aldehyde oxidase properties. Interestingly, one gene appeared as specifically expressed in the antennae of B. mori and associated in males with the bombykal-sensitive sensilla, strongly suggesting that it could encode for the previously biochemically characterized enzyme.  相似文献

采用RT-PCR技术,利用简并引物从棉蚜(Aphis gossypii Glover)中克隆出2个乙酰胆碱酯酶基因的cDNA片段,Ag,ace 1l和Ag.ace2.Ag.ace1基因的cDNA片段为282bp,编码94个氨基酸;Ag.ace2基因的cDNA片段为264bp,编码88个氨基酸。扩增获得的2个乙酰胆碱酯酶基因cDNA片段所编码的氨基酸序列均与其他昆虫的乙酰胆碱酯酶基因有很高的同源性。首次从一种昆虫中克隆出2个乙酰胆碱酯酶基因片段,为同一种昆虫中存在多个乙酰胆碱酯酶基因的假设提供了直接的分子生物学证据。  相似文献

Acetylcholinesterase is a key enzyme of the animal nerve system. The enzyme is the primary target of organophosphorous (OP) and carbamate (CB) insecticides. The insect AChE is being extensively used in development of new insecticides or in vitro selection of the new designed insecticides, and in pharmacological and toxicological field. Rapid assays using AChE-based methods have been proposed as an efficient and rapid method for the detection of pesticides, especially in many Asian markets. In this study, the acetylcholinesterase gene was cloned from housefly (Musca domestica) susceptible to organophosphate (OP) and carbamate (CB) insecticides, and expressed in baculovirus-insect cells system using a bioreactor with oxygen supplementation. The recombinant housefly AChE was purified using ammonium sulfate precipitation and procainamide affinity chromatography, and approximately 0.42 mg of the purified AChE with high biological activity (118.9 U/mg) was obtained from 100 ml of culture solution. The purified AChE was highly sensitive to OP and CBs insecticides. In conclusion, an efficient expression and purification system has been developed for large-scale production of recombinant housefly AChE. The recombinant enzyme is potential to be used for the detection of pesticide residues.  相似文献

本研究以优良杂交品种"两广二号"家蚕为试材,克隆了该杂交品种家蚕两个抗家蚕核型多角体病毒(BmNPV)基因:脂肪酶基因Bmlipase-1和丝氨酸蛋白酶基因BmSP-2,测序并分别与不同品种蚕的同源基因序列进行比较。结果显示,"两广二号"家蚕Bmlipase-1基因ORF长度为885bp,编码294个氨基酸,BmSP-2扩增长度为855bp,编码284个氨基酸;它们的核苷酸和推导氨基酸序列同源性皆达92%以上,Bmlipase-1更保守,同源性大于99%";两广二号"家蚕的Bmlipase-1基因脂肪酶活化部位和BmSP-2基因酶催化三联体位点的氨基酸残基与不同品种蚕的完全相同。以上结果说明这两个抗病毒基因在蚕的遗传进化过程中高度保守,提示其可能在机体消化或者免疫防御方面起着重要生理作用。将这两个抗病毒基因在大肠杆菌BL21中进行融合表达,获得的融合Bmlipase-1和BmSP-2蛋白分子量分别为47kD和42kD左右。  相似文献

Ecdysis-triggering hormone (ETH) is an integration factor in the ecdysis process of most insects, including Bombyx mori (silkworm). To understand the function of the ETH gene in silkworm, we developed an effective approach to knockdown the expression of ETH in vivo based on RNA interference (RNAi) and a binary UAS/GAL4 expression system that has been successfully used in other insect species. Two kinds of transgenic silkworm were established with this method: the effector strain with the ETH RNAi sequence under the control of UAS and the activator strain with the GAL4 coding sequence under the control of Bombyx mori cytoplasmic actin3. By crossing the two strains, double-positive transgenic silkworm was obtained, and their ETH expression was found to be dramatically lower than that of each single positive transgenic parent. Severe ecdysis deficiency proved lethal to the double-positive transgenic silkworm at the stage of pharate second instar larvae, while the single positive transgenic or wild-type silkworm had normal ecdysis. This UAS/GAL4 RNAi approach provides a way to study the function of endogenous silkworm genes at different development stages.  相似文献

转基因家蚕Bombyx mori是指利用分子生物学手段,将外源基因转移到家蚕染色体中, 使之出现先前不具有的性状和产物,并且可以保持传代,在个体水平可以体现外源基因的功能,使外源基因获得大量表达。目前转基因家蚕研究主要以piggyBac转座系统为常用载体, 绿色荧光蛋白(green fluorescent protein, GFP)基因为常用报告基因,经显微注射法获得转基因家蚕的成功率可达40%。通过转基因家蚕技术已经探明了家蚕外源导入核受体基因BmFtz-F1,调控家蚕体壁半透明的BmBLOS2基因、蜕皮启动激素(ecdysis-triggering hormone, ETH)基因以及家蚕抗菌肽CecB(cecropin B)基因的功能;获得了具有高品质、高细纤度、高拉伸强度和高弹性丝品种,能吐带绿色荧光或粉红色荧光的蚕丝品种, 抗家蚕核型多角体病毒(Bombyx mori nucleopolyhedrovirus, BmNPV)品种及抗藤黄微球菌的品种;成功表达纯化了人的Ⅲ型前胶原蛋白、 人碱性纤维生长因子、人血清蛋白、人脑源性神经营养因子、人胰岛素生长因子Ⅰ、猫干扰素、单克隆抗体等生物活性蛋白、疫苗及特殊的生物材料。随着家蚕转基因技术的深入研究, 转基因家蚕产物将在国防、 军工、 航天、 医药等方面有着更为广阔的应用前景。  相似文献

To identify genes involved in the innate immunity of the silkworm Bombyx mori, we constructed a cDNA library from the fat body of Escherichia coli-challenged B. mori larvae. Based on the expressed sequence tag (EST) data and whole genome shotgun sequence analysis, we found four Gloverin-like genes, BmGlov1-4, in the Bombyx genome. Northern blot and RT-PCR analysis showed that BmGlov1-4 were induced in the larval fat body after an immune challenge by the injection of E. coli; however, less induction was observed after the injection of a yeast Candida albicans. In silico sequence analysis revealed the presence of a motif homologous to NF-kappaB binding site in the upstream region of each BmGlov gene. Moreover, we expressed recombinant BmGlov1-4 proteins using the baculovirus expression system, and found that all the recombinant BmGlov1-4 significantly inhibited the growth of E. coli.  相似文献

In order to establish a transient expression system for genes introduced into early embryos of the silkworm, Bombyx mori, we tested various promoters ligated with CAT reporter genes. The embryos into which we injected supercoiled plasmid DNA of pFb(-860/+10)CAT containing the Bombyx fibroin promoter region ligated to the CAT gene showed a reasonably high CAT activity beginning around 30 h after oviposition. This high activity was observed only when the plasmid was injected before termination of the early nuclear cleavage stage, which was about 8 h after oviposition, but not after this stage. This means that the expression of injected DNA is closely related to the presence of cleavage nuclei in early embryos. Promoters originating from insect genes, like the Bombyx sericin-1 gene, Drosophila hsp70 and Drosophila copia LTR, functioned as strong promoters in the embryos. On the contrary, promoters from mammalian virus genes, such as the SV40 early and Moloney murine leukemia virus LTR genes, functioned as weak promoters. Moreover, linearized DNAs showed no or weak activity of expression in embryos. From these results, we conclude that the silkworm embryo transient expression system is a useful tool for studying the mechanism of regulation of insect genes.  相似文献

对昆虫乙酰胆碱酯酶(acetylcholinesterase, AChE,EC 3.1.1.7)的基因结构和表达等方面的研究进展进行了综述。分析了昆虫乙酰胆碱酯酶基因的结构,包括10个外显子的特征。对已经报道的昆虫AChE基因进行了系统归纳,并基于已知全序列的昆虫AChE基因,进行了昆虫AChE基因的分子进化分析。对昆虫AChE基因的结构特点及其功能,以及昆虫AChE基因的活性位点、AChE的变构与昆虫抗药性的关系进行了探讨。最后对昆虫AChE基因研究中存在的问题和前景进行了分析和展望。  相似文献
