Soils are an important source of N2O, which can be produced both in the nitrification and the denitrification processes. Grassland soils in particular have a high potential for mineralization and subsequent nitrification and denitrification. When ploughing long term grassland soils, the resulting high supply of mineral N may provide a high potential for N2O losses. In this work, the short-term effect of ploughing a permanent grassland soil on gaseous N production was studied at different soil depths. Fertiliser and irrigation were applied in order to observe the effect of ploughing under a range of conditions. The relative proportions of N2O produced from nitrification and denitrification and the proportion of N2 gas produced from denitrification were determined using the methyl fluoride and acetylene specific inhibitors. Irrespectively to ploughing, fertiliser application increased the rates of N2O production, N2O production from nitrification, N2O production from denitrification and total denitrification (N2O + N2). Application of fertiliser also increased the denitrification N2O/N2 ratio both in the denitrification potential and in the gaseous N productions by denitrification. Ploughing promoted soil organic N mineralization which led to an increase in the rates of N2O production, N2O production from nitrification, N2O production from denitrification and total denitrification (N2O + N2). In both the ploughed and unploughed treatments the 0–10 cm soil layer was the major contributing layer to gaseous N production by all the above processes. However, the contribution of this layer decreased by ploughing, gaseous N productions from the 10 to 30 cm layer being significantly increased with respect to the unploughed treatment. Ploughing promoted both nitrification and denitrification derived N2O production, although a higher proportion of N2O lost by denitrification was observed as WFPS increased. Recently ploughed plots showed lower denitrification derived N2O percentages than those ploughed before as a result of the lower soil water content in the former plots. Similarly, a lower mean nitrification derived N2O percentage was found in the 10–30 cm layer compared with the 0–10 cm.

N2O emissions from two slightly alkaline sandy soils, from arable land and a woodland, were determined in a laboratory experiment in which the soils were incubated with different sources of nitrogen, with or without glucose, and with 0, 1 and 100 mL C2H2 L-1. Large differences in the rate of N2O production were observed between the two soils and between the different N treatments. The arable soil showed very low N2O emissions derived from reduced forms of N as compared with the N2O which was produced when the soil was provided with NO 2 - or NO 3 - and a C source, suggesting a very active denitrifier population. In contrast, the woodland soil showed a very low denitrification activity and a much higher N2O production derived from the oxidation of NH 4 + and reduction of NO 2 - by some processes probably mediated by autotrophic or heterotrophic nitrifiers or dissimilatory NO 2 - reducers. In both soils, the highest N2O emissions were induced by NO 2 - addition. Those emissions were demonstrated to have a biological origin, as no significant N2O emissions were measured when the soil was autoclaved.

Production and sources of N2O were determined in soil columns amended with autoclaved yeast cells either mixed into or added as 0.5 cm3 lumps to the soil in combination with no or 200 g NO3 --N g-1. At four occasions over a two-week study period, subsets of cores were measured for N2O production during 4-hour incubations under atmospheres of ambient air, 10 Pa of C2H2, and N2, respectively. Denitrification enzyme activity (DEA) was assessed in subsamples of cores that had been incubated continuously under air.Autoclaved yeast provided a C-source readily available for denitrifying bacteria in the soil. Nitrous oxide production was negligible in unamended columns whereas accumulated N2O losses in the presence of yeast material were substantial, varying between 15 to 49 ng N2O-N g-1 h-1. Mixing yeast into the soil caused the highest production of N2O followed by the yeast lump and no yeast treatments. Incubation in the presence of 10 Pa C2H2 indicated that denitrification was the sole source of N2O, in accordance with an increase in DEA. Nitrous oxide production and DEA peaked after 4–7 days of incubation, and both were unaffected by additional NO3 -. Two-to four-fold responses to anaerobiosis and accumulation of NO3 - and NH4 + in proximity of the lumps indicated that N2O production here was limited by relatively low C-availability. In contrast, 10- to 12-fold responses to anaerobiosis and no accumulation of inorganic N suggested a higher C-availability where yeast was mixed into the soil.

Background and aims

The direct measurement of denitrification dynamics and its product fractions is important for parameterizing process-oriented model(s) for nitrogen cycling in various soils. The aims of this study are to a) directly measure the denitrification potential and the fractions of nitrogenous gases as products of the process in laboratory, b) investigate the effects of the nitrate (NO 3 ? ) concentration on emissions of denitrification gases, and c) test the hypothesis that denitrification can be a major pathway of nitrous oxide (N2O) and nitric oxide (NO) production in calcic cambisols under conditions of simultaneously sufficient supplies of carbon and nitrogen substrates and anaerobiosis as to be found to occur commonly in agricultural lands.

Methods

Using the helium atmosphere (with or without oxygen) gas-flow-soil-core technique in laboratory, we directly measured the denitrification potential of a silt clay calcic cambisol and the production of nitrogen gas (N2), N2O and NO during denitrification under the conditions of seven levels of NO 3 ? concentrations (ranging from 10 to 250 mg N kg?1 dry soil) and an almost constant initial dissolved organic carbon concentration (300 mg C kg?1 dry soil).

Results

Almost all the soil NO 3 ? was consumed during anaerobic incubation, with 80–88 % of the consumed NO 3 ? recovered by measuring nitrogenous gases. The results showed that the increases in initial NO 3 ? concentrations significantly enhanced the denitrification potential and the emissions of N2 and N2O as products of this process. Despite the wide range of initial NO 3 ? concentrations, the ratios of N2, N2O and NO products to denitrification potential showed much narrower ranges of 51–78 % for N2, 14–36 % for N2O and 5–22 % for NO.

Conclusions

These results well support the above hypothesis and provide some parameters for simulating effects of variable soil NO 3 ? concentrations on denitrification process as needed for biogeochemical models.

We examined the effects of growth carbon dioxide (CO2)concentration and soil nutrient availability on nitrogen (N)transformations and N trace gas fluxes in California grasslandmicrocosms during early-season wet-up, a time when rates of Ntransformation and N trace gas flux are high. After plant senescenceand summer drought, we simulated the first fall rains and examined Ncycling. Growth at elevated CO2 increased root productionand root carbon:nitrogen ratio. Under nutrient enrichment, elevatedCO2 increased microbial N immobilization during wet-up,leading to a 43% reduction in gross nitrification anda 55% reduction in NO emission from soil. ElevatedCO2 increased microbial N immobilization at ambientnutrients, but did not alter nitrification or NO emission. ElevatedCO2 did not alter soil emission of N2O ateither nutrient level. Addition of NPK fertilizer (1:1:1) stimulatedN mineralization and nitrification, leading to increased N2Oand NO emission from soil. The results of our study support a mechanisticmodel in which elevated CO2 alters soil N cycling and NOemission: increased root production and increased C:N ratio in elevatedCO2 stimulate N immobilization, thereby decreasingnitrification and associated NO emission when nutrients are abundant. This model is consistent with our basic understanding of how C availabilityinfluences soil N cycling and thus may apply to many terrestrial ecosystems.

Summary Total denitrification (N2O+N2) and nitrous oxide emission were measured on intact soil cores using the acetylene inhibition technique.Total denitrification from the depth 0–8 cm during the growth period from April to August was 7 kg N/ha from plots supplied with 30 kg N/ha and 19 kg N/ha from plots supplied with 120 kg N/ha. The amounts of precipitation, plant growth, and N application were found to affect the denitrification rate. These factors also affected the ratio (N2O+N2)/N2O, which varied from 1.0 to 7.2. Plant growth and precipitation increased the proportion of N2 produced, whereas a high nitrate content increased the proportion of N2O.

N2O production from denitrification in soils contributes to the enhanced greenhouse effect and the destruction of the stratospheric ozone. Ungulate grazing affects denitrification and the production of N2O. The short-term effect of grazing on denitrification and N2O production has been examined in several grassland ecosystems. However, the effects of long-term grazing have rarely been studied. We measured denitrification and N2O production during the 2005 and 2006 growing seasons in a long-term (17 years) experiment that had five grazing intensities (GI; 0.00, 1.33, 2.67, 4.00 and 5.33 sheep ha−1). We found that denitrification and N2O production rates were seasonally variable during the measurement period, with higher values observed in summer and lower values found in spring and autumn. The grazed treatments resulted in decreased denitrification and N2O production, primarily due to the reduced soil nitrate concentration and organic N content under the long-term grazing. This supported our hypothesis that long-term over-grazing suppresses denitrification and N2O production. Although significant differences in denitrification and N2O production were not found between the four GI, there was a general trend that cumulative denitrification and N2O production decreased as grazing intensity increased, especially in 2006. Lower N losses via denitrification and N2O production in the grazed plots, to some extent, may contribute to the mitigation of greenhouse gas emission and help to preserve soil N and ameliorate the negative impacts of grazing on plant growth, productivity, and ecological restoration processes in the temperate steppe in northern China.

The effects of grassland conversion to forest vegetation and of individual tree species on microbial activity in Siberia are largely unstudied. Here, we examined the effects of the six most commonly dominant tree species in Siberian forests (Scots pine, spruce, Arolla pine, larch, aspen and birch) on soil C and N mineralization, N2O-reduction and N2O production during denitrification 30 years after planting. We also documented the effect of grassland conversion to different tree species on microbial activities at different soil depths and their relationships to soil chemical properties. The effects of tree species and grassland conversion were more pronounced on N than on C transformations. Tree species and grassland conversion did significantly alter substrate-induced respiration (SIR) and basal respiration, but the differences were not as large as those observed for N transformations. Variances in SIR and basal respiration within species were markedly lower than those in N transformations. Net N mineralization, net nitrification, and denitrification potential were highest under Arolla pine and larch, intermediate under deciduous aspen and birch, and lowest beneath spruce and Scots pine. Tree species caused similar effects on denitrification potential, net N mineralization, and net nitrification, but effects on N2O reduction rate were idiosyncratic, indicating a decoupling of N2O production and reduction. We predict that deciduous species should produce more N2O in the field than conifers, and that Siberian forests will produce more N2O if global climate change alters tree species composition. Basal respiration and SIR showed inverse responses to tree species: when basal respiration increased in response to a given tree species, SIR declined. SIR may have been controlled by NH4 + availability and related therefore to N mineralization, which was negatively affected by grassland conversion. Basal respiration appeared to be less limited by NH4 + and controlled mostly by readily available organic C (DOC), which was higher in concentration under forests than in grassland and therefore basal respiration was higher in forested soils. We conclude that in the Siberian artificial afforestation experiment, soil C mineralization was not limited by N.

N2O gas is involved in global warming and ozone depletion. The major sources of N2O are soil microbial processes. Anthropogenic inputs into the nitrogen cycle have exacerbated these microbial processes, including nitrification. Ammonia-oxidizing archaea (AOA) are major members of the pool of soil ammonia-oxidizing microorganisms. This study investigated the isotopic signatures of N2O produced by soil AOA and associated N2O production processes. All five AOA strains (I.1a, I.1a-associated and I.1b clades of Thaumarchaeota) from soil produced N2O and their yields were comparable to those of ammonia-oxidizing bacteria (AOB). The levels of site preference (SP), δ15Nbulk and δ18O -N2O of soil AOA strains were 13–30%, −13 to −35% and 22–36%, respectively, and strains MY1–3 and other soil AOA strains had distinct isotopic signatures. A 15N-NH4+-labeling experiment indicated that N2O originated from two different production pathways (that is, ammonia oxidation and nitrifier denitrification), which suggests that the isotopic signatures of N2O from AOA may be attributable to the relative contributions of these two processes. The highest N2O production yield and lowest site preference of acidophilic strain CS may be related to enhanced nitrifier denitrification for detoxifying nitrite. Previously, it was not possible to detect N2O from soil AOA because of similarities between its isotopic signatures and those from AOB. Given the predominance of AOA over AOB in most soils, a significant proportion of the total N2O emissions from soil nitrification may be attributable to AOA.

试验共设4个处理:CK(对照)、C1(低量生物炭4.5 t hm~(-2)a~(-1))、C2(高量生物炭9.0 t hm~(-2)a~(-1))和SR(秸秆还田straw return)。

During two intensive field campaigns in summer and autumn 2004 nitrogen (N2O, NO/NO2) and carbon (CO2, CH4) trace gas exchange between soil and the atmosphere was measured in a sessile oak (Quercus petraea (Matt.) Liebl.) forest in Hungary. The climate can be described as continental temperate. Fluxes were measured with a fully automatic measuring system allowing for high temporal resolution. Mean N2O emission rates were 1.5 μg N m−2 h−1 in summer and 3.4 μg N m−2 h−1 in autumn, respectively. Also mean NO emission rates were higher in autumn (8.4 μg N m−2 h−1) as compared to summer (6.0 μg N m−2 h−1). However, as NO2 deposition rates continuously exceeded NO emission rates (−9.7 μg N m−2 h−1 in summer and −18.3 μg N m−2 h−1 in autumn), the forest soil always acted as a net NO x sink. The mean value of CO2 fluxes showed only little seasonal differences between summer (81.1 mg C m−2 h−1) and autumn (74.2 mg C m−2 h−1) measurements, likewise CH4uptake (summer: −52.6 μg C m−2 h−1; autumn: −56.5 μg C m−2 h−1). In addition, the microbial soil processes net/gross N mineralization, net/gross nitrification and heterotrophic soil respiration as well as inorganic soil nitrogen concentrations and N2O/CH4 soil air concentrations in different soil depths were determined. The respiratory quotient (ΔCO2 resp ΔO2 resp−1) for the uppermost mineral soil, which is needed for the calculation of gross nitrification via the Barometric Process Separation (BaPS) technique, was 0.8978 ± 0.008. The mean value of gross nitrification rates showed only little seasonal differences between summer (0.99 μg N kg−1 SDW d−1) and autumn measurements (0.89 μg N kg−1 SDW d−1). Gross rates of N mineralization were highest in the organic layer (20.1–137.9 μg N kg−1 SDW d−1) and significantly lower in the uppermost mineral layer (1.3–2.9 μg N kg−1 SDW d−1). Only for the organic layer seasonality in gross N mineralization rates could be demonstrated, with highest mean values in autumn, most likely caused by fresh litter decomposition. Gross mineralization rates of the organic layer were positively correlated with N2O emissions and negatively correlated with CH4 uptake, whereas soil CO2 emissions were positively correlated with heterotrophic respiration in the uppermost mineral soil layer. The most important abiotic factor influencing C and N trace gas fluxes was soil moisture, while the influence of soil temperature on trace gas exchange rates was high only in autumn.

The effects of elevated concentrations of atmospheric CO2 on CH4 and N2O emissions from rice soil were investigated in controlled-environment chambers using rice plants growing in pots. Elevated CO2 significantly increased CH4 emission by 58% compared with ambient CO2. The CH4 emitted by plant-mediated transport and ebullition–diffusion accounted for 86.7 and 13.3% of total emissions during the flooding period under ambient level, respectively; and for 88.1 and 11.9% of total emissions during the flooding period under elevated CO2 level, respectively. No CH4 was emitted from plant-free pots, suggesting that the main source of emitted CH4 was root exudates or autolysis products. Most N2O was emitted during the first 3 weeks after flooding and rice transplanting, probably through denitrification of NO3 contained in the experimental soil, and was not affected by the CO2 concentration. Pre-harvest drainage suppressed CH4 emission but did not cause much N2O emission (< 10 μg N m−2 h−1) from the rice-plant pots at both CO2 concentrations.

In order to quantify N2-emissions from a spruce and a beech site at the Höglwald Forest, a new measuring system was developed, that allowed simultaneous, direct determination of N2- and N2O-emission with high accuracy (detection limit approx. 10 g N m–2 h–1 for N2 and <1 g for N2O) using a gas-flow core method. This method requires exchange of the soil atmosphere with an artificial atmosphere, that differs only in that N2 is substituted by He. The measuring system, the methodology of measurements and validation experiments are described in detail. Due to the huge heterogeneity of denitrification activity in different soil cores taken from our forest sites, no general trends of N2 and N2O production in relation to soil moisture and temperature could be demonstrated. Based on reasonable number of measurements, this work gives for the first time an estimate of the magnitude of N2-losses from temperate forest soils. Both the magnitude of N2-emissions (spruce: 7.2±0.7 kg N2-N ha–1 yr–1; beech: 12.4±3.1 kg N2-N ha–1 yr–1), as well as the N2O–N2 ratio (spruce: 0.136±0.04; beech: 0.52±0.19) were significantly higher for soils from the beech sites as compared to soils from the spruce site. The results suggests that N2-emissions from N-saturated forest soils, still receiving high loads of atmospheric N-deposition, are approx. 30% of atmospheric N-input at the spruce site, and approx. 50% at the beech site. Our results demonstrate that losses of nitrogen in the form of N2 cannot be neglected in the context of calculating N-balances for given forest sites.

Nitrous oxide (N2O) is a potent greenhouse gas with a high contribution from agricultural soils and emissions that depend on soil type, climate, crops and management practices. The N2O emissions therefore need to be included as an integral part of environmental assessments of agricultural production systems. An algorithm for N2O production and emission from agricultural soils was developed and included in the FASSET whole-farm model. The model simulated carbon and nitrogen (N) turnover on a daily basis. Both nitrification and denitrification was included in the model as sources for N2O production, and the N2O emissions depended on soil microbial and physical conditions. The model was tested on experimental data of N2O emissions from grasslands in UK, Finland and Denmark, differing in climatic conditions, soil properties and management. The model simulated the general time course of N2O emissions and captured the observed effects of fertiliser and manure management on emissions. Scenario analyses for grazed and cut grasslands were conducted to evaluate the effects of soil texture, climatic conditions, grassland management and N fertilisation on N2O emissions. The soils varied from coarse sand to sandy loam and the climatic variation was taken to represent the climatic variation within Denmark. N fertiliser rates were varied from 0 to 500 kg N ha−1. The simulated N2O emissions showed a non-linear response to increasing N rates with increasing emission factors at higher N rates. The simulated emissions increased with increasing soil clay contents. N2O emissions were slightly increased at higher temperatures, whereas increasing annual rainfall generally lead to decreasing emissions. Emissions were slightly higher from grazed grasslands compared with cut grasslands at similar rates of total N input (fertiliser and animal excreta). The results indicate higher emission factors and thus higher potentials for reducing N2O emissions for intensively grazed grasslands on fine textured soils than for extensive cut-based grasslands on sandy soils.

The effect of photosynthesis on N2O emission from soil was investigated by shading soybean (Gycline max. L) plant at flowering, pod-setting and grain-filling stages. The results showed that by stopping photosynthesis through shading the plants stimulated N2O emission significantly at flowering stage and pod-setting stage, and suppressed N2O emission dramatically at grain-filling stage. At flowering stage, soybean species seem to rely mainly on fertilizer N and shaded plants decreased the N uptake. Interaction between the relative increase in available N for N2O production by shading and the presence of root exudates promoted N transformation (nitrification/denitrification) and N2O emission. At pod-setting stage, the available soil nitrogen seems to be a critical limiting factor and without substantial release of symbiotically fixed N through plant roots, resulted in a weak effect of shading on N2O emission. At grain-filling stage, available N for N2O production was derived from symbiotically fixed N and was greatly affected by photosynthesis. These results indicated that the effect of soybean growth on N2O emission from soil varies with plant growth stages as available N for N2O production is mainly from fertilizer N and organic mineralization during the early growth of soybean plants, while N2O emission is controlled by the quantity and perhaps also the quality of root exudates, which is closely related with plant photosynthesis in the late season of soybean growth.
