Characterization of Cre recombinase mouse lines enabling cell type-specific targeting of postnatal intervertebral discs

Cre/loxP technology is an important tool for studying cell type-specific gene functions. Cre recombinase mouse lines, including Agc1-CreER^T2, Col2a1-Cre; Col2a1-CreER^T2, Shh-Cre, Shh-CreER^T2, and Osx-Cre, have been proven to be valuable tools to elucidate the biology of long bones, yet the information for their activity in postnatal intervertebral disc (IVD) tissues was very limited. In this study, we used R26-mTmG fluorescent reporter to systematically analyze cell specificity and targeting efficiency of these six mouse lines in IVD tissues at postnatal growing and adult stages. We found that Agc1-CreER^T2 is effective to direct recombination in all components of IVDs, including annulus fibrosus (AF), nucleus pulposus (NP), and cartilaginous endplate (CEP), upon tamoxifen induction at either 2 weeks or 2 months of ages. Moreover, Col2a1-Cre targets most of the cells in IVDs, except for some cells in the outer AF (OAF) and NP. In contrast, the activity of Col2a1-CreER^T2 is mainly limited to the IAF of IVD tissues at either stage of tamoxifen injection. Similarly, Shh-Cre directs recombination specifically in all NP cells, whereas Shh-CreER^T2 is active only in a few NP cells when tamoxifen is administered at either stage. Finally, Osx-Cre targets cells in the CEP, but not in the NP or AF of IVDs tissues at these two stages. Thus, our data demonstrated that all these Cre lines can direct recombination in IVD tissues at postnatal stages with different cell type specificity and/or targeting efficiency, and can, therefore, serve as valuable tools to dissect cell type-specific gene functions in IVD development and homeostasis.     

山岳型乡村旅游地“三生”空间演变及优化——德庆金林水乡的案例实证

以德庆县金林水乡为例,采用参与式观察和空间统计法,选取2000、2008年和2018年3个时段分析旅游发展对山岳型乡村旅游地"三生"空间的影响,并探讨了金林水乡"三生"空间的发展瓶颈及优化路径。研究发现:(1)旅游开发前,金林水乡土地结构与用地功能单一且呈片状分布;村落呈现传统乡村风貌,基础设施不健全;空间形态变化稳定,扩张缓慢。(2)旅游开发后,土地利用类型多样化,出现新型复合用地;土地功能利用复杂化,以服务旅游业为主;村庄景观风貌现代化,生活空间更加宜居。(3)旅游开发前后对比可得,土地平面占地规模化,空间用地以居民点为核心,呈圈层状向外围扩张;生产-生活-生态空间相互转化,乡村聚落重构特征较为显著;村落景观风貌的变化较大,呈现城镇化趋势。(4)金林水乡"三生"空间演化与旅游发展存在的问题表现在生产用地效率不高,生活用地质量较低,生态空间不断萎缩,在旅游产业发展上表现为旅游产品单一且缺乏创新,旅游服务功能不完善等。为此从生活空间的提质、生产空间增效、生态空间保护、旅游产业创新以及土地利用五方面提出优化建议。     

Cytoplasmic RNA complementary to pre-albumin intron V

We have detected an RNA species, containing sequences complementary to pre-albumin intron V in poly(A)+ RNA from rat liver cytoplasm (but not in nuclear RNA). Its relative abundance roughly parallels that of mature albumin mRNA, when comparing control with acute phase preparations.     

Initial Commitment to Pre-Exposure Prophylaxis and Circumcision for HIV Prevention amongst Indian Truck Drivers

Studies of HIV prevention interventions such as pre-exposure prophylaxis (PREP) and circumcision in India are limited. The present study sought to investigate Indian truck-drivers initial commitment to PREP and circumcision utilizing the AIDS Risk Reduction Model. Ninety truck-drivers completed an in-depth qualitative interview and provided a blood sample for HIV and HSV-2 testing. Truck-drivers exhibited low levels of initial commitment towards PREP and even lower for circumcision. However, potential leverage points for increasing commitment were realized in fear of infecting family rather than self, self-perceptions of risk, and for PREP focusing on cultural beliefs towards medication and physicians. Cost was a major barrier to both HIV prevention interventions. Despite these barriers, our findings suggest that the ARRM may be useful in identifying several leverage points that may be used by peers, health care providers and public health field workers to enhance initial commitment to novel HIV prevention interventions in India.     

Inhibition of soil urease activity by amido derivatives of phosphoric and thiophosphoric acids

Summary A laboratory experiment was conducted to study inhibition of soil urease activity by amido derivatives of phosphoric and thiophosphoric acids. Results showed that derivatives with higher amido substitutions have greater inhibitory effect on urea hydrolysis in the soils used in our study. Triamides of phosphoric and thiophosphoric acids were found to be very effective inhibitors of soil urease. These compounds seem to have potential as fertilizer amendments for inhibiting soil urease activity and for improving the efficiency of nitrogen use from urea.
