在体外培养瘤细胞的实验中,胰RNA提取物不仅对艾氏腹水癌细胞而且对其他瘤细胞(S180、P388、H22)也具有杀伤作用。在胰RNA提取物中,对瘤细胞具有杀伤作用的活性物质不是外源性凝集素样多糖类或能透过半透膜的低分子化合物,而是具有一定二级结构的低分子RNA。  相似文献

用液体闪烁计数法测定离体再灌注鼠心肌肌质网(SR)和线粒体(Mit)内 45Ca2+放射性强度(cpm),比较能量制剂ATP-MgCl2,活性氧自由基清除剂SOD和钙阻滞剂Verapamil对离体缺血再灌注鼠心肌细胞SR和Mit钙离子浓度的影响.结果表明,SR内ATp-MgCl2,SOD和Verapamil组 45Ca2+的cpm均高于对照组(P<0.0l或P<0.05),而Mit内均低于对照组(P<0.01).此三种药均能提高离体大鼠心肌细胞内SR 45Ca2+和降低Mit 45Ca2+积聚,从而保护了心肌细胞,防止缺血再灌注损伤.  相似文献

用快速微量等电聚焦技术对190名北京地区汉族健康人血清Gc蛋白亚型、Pi蛋白亚型进行分型鉴定和基因频率调查.上样量为1.5μl,电泳和染色各0.5h.Gc1F=0.4891,Gc1S=0.2432,Gc2=0.2678.观察值与期望值吻合良好.(∑X2=1.404,0.7<P<0.8).PiM1=0.7542,PiM2=0.1808,PiM3=0.0650,观察值与期望值吻合也良好,(∑X2=1.1233,0.7<P<0.8).  相似文献

植物最大净光合速率的季节变异性及其氮调控机制是近年来植物生理生态领域研究热点,对荒漠植物光合最大净光合速率季节动态及其叶氮影响,特别是叶氮分配对最大净光合速率调控机制的了解仍非常有限。2018年5-10月在宁夏盐池毛乌素沙地,对当地主要建群种油蒿(Artemisia ordosica)进行生长季原位观测,测定其叶光合光响应曲线(A-PAR)、CO2响应曲线(A-Ci)和叶氮含量,结合环境观测数据,分析A-PAR关键参数最大净光合速率(Amax)的季节变异和叶片氮分配相关参数对Amax的调控。结果表明,油蒿叶片Amax在生长期季节变异系数(Cv)为14%,在完全展叶中期,光合氮利用效率(PNUE)有最大值11.82 μmolCO2 gN-1 s-1,此时叶片氮素在光合系统中的分配比例最大,Amax有最大值29.48 μmol CO2 m-2s-1,油蒿光合能力较强。在完全展叶末期,油蒿叶片氮在光合系统中的分配比例(Pp)有最小值0.21,此时叶氮分配更倾向于非光合系统,Amax较小,光合能力较弱,有利于油蒿延长叶片寿命。Amax与单位干重叶片氮含量(Nmass)、氮在捕光系统中分配比例(PL)、氮在生物力能系统中分配比例(Pb)正相关(P<0.05),其中,PLAmax产生了最大的直接正向影响,氮在羧化系统中分配比例(Pc)主要通过PbAmax的产生了间接的正向影响,PbAmax季节动态的解释度最大(R2=0.49,P<0.01)。Amax与比叶重(LMA)负相关(P<0.05),即当有较高LMA时油蒿具有较低的光合速率,此时叶片光合能力较弱,叶片机械韧性较强。不同物候时期,油蒿叶片氮素分配的变化及叶片光合能力与结构性状之间的权衡关系,标志着荒漠植物对其生境的适应性调节。研究结果不但可提高荒漠灌木油蒿生态系统生产力预测的准确性,也是对植物光合适应机理认识的补充和深入。  相似文献

The calculated maximum net photosynthetic rate (P N) at saturation irradiance (I m) of 1 314.13 μmol m−2 s−1 was 25.49 μmol(CO2) m−2 s−1, and intrinsic quantum yield at zero irradiance was 0.103. The results fitted by nonrectangular hyperbolic model, rectangular hyperbolic method, binomial regression method, and the new model were compared. The maximum P N values calculated by nonrectangular hyperbolic model and rectangular hyperbolic model were higher than the measured values, and the I m calculated by nonrectangular hyperbolic model and rectangular hyperbolic model were less than measured values. Results fitted by new model showed that the response curve of P N to I was nonlinear at low I for Oryza sativa, P N increased nonlinearly with I below saturation value. Above this value, P N decreased nonlinearly with I.  相似文献

为寻求一种可替代人肝细胞研究载脂蛋白(apolipoprotein, apo)CⅢ受体的生理模型,并探讨apoCⅢ受体的生理功能及其在内源性高甘油三酯血症发病机制中的作用,首先以125I标记的人apoCⅢ为配体,利用放射性配体结合分析法观察了人肝癌细胞系HepG2细胞是否有apoCⅢ受体存在.结果证实HepG2细胞上存在高亲和力的、可饱和的、特异的apoCⅢ受体结合位点,其受体的亲和力(Kd)和apoCⅢ受体结合容量(Bmax)分别为(9.53±1.03)×10-9 mol/L和(3.28±0.31) μg/g.随后又分别研究了胰岛素及胰高血糖素对HepG2细胞apoCⅢ受体功能的影响,结果表明:胰岛素可使HepG2细胞apoCⅢ受体结合容量(Bmax)显著增加,但对受体的亲和力(Kd)无影响;胰高血糖素可使HepG2细胞apoCⅢ受体亲和力显著下降(即Kd值升高),对受体的结合容量无影响.提示人肝apoCⅢ受体的功能可能受胰岛素及胰高血糖素的不同调节.  相似文献

We determined the interactive effects of irradiance, elevated CO2 concentration (EC), and temperature in carrot (Daucus carota var. sativus). Plants of the cv. Red Core Chantenay (RCC) were grown in a controlled environmental plant growth room and exposed to 3 levels of photosynthetically active radiation (PAR) (400, 800, 1 200 μmol m−2 s−1), 3 leaf chamber temperatures (15, 20, 30 °C), and 2 external CO2 concentrations (C a), AC and EC (350 and 750 μmol mol−1, respectively). Rates of net photosynthesis (P N) and transpiration (E) and stomatal conductance (g s ) were measured, along with water use efficiency (WUE) and ratio of internal and external CO2 concentrations (C i/C a). P N revealed an interactive effect between PAR and C a. As PAR increased so did P N under both C a regimes. The g s showed no interactive effects between the three parameters but had singular effects of temperature and PAR. E was strongly influenced by the combination of PAR and temperature. WUE was interactively affected by all three parameters. Maximum WUE occurred at 15 °C and 1 200 μmol m−2 s− 1 PAR under EC. The C i /C a was influenced independently by temperature and C a. Hence photosynthetic responses are interactively affected by changes in irradiance, external CO2 concentration, and temperature. EC significantly compensates the inhibitory effects of high temperature and irradiance on P N and WUE.  相似文献

The relationships between dark respiration rate (R D) and net photosynthetic rate (P N) in Quercus ilex L. shrubs growing at the Botanical Garden in Rome were analysed. Correlation analysis of the data sets collected in the year 2006 confirmed the dependence among the considered leaf traits, in particular, R D was significantly (p<0.05) correlated with P N (r = 0.40). R D and P N increased from March to May [1.40±0.10 and 10.1±1.8 μmol(CO2) m−2 s−1 mean values of the period, respectively], when air temperature was in the range 14.8–25.2 °C, underlining the highest metabolic activity in the period of the maximum vegetative activity that favoured biomass accumulation. On the contrary, the highest R D [1.60±0.02 μmol(CO2) m−2 s−1], associated to the lowest P N rates (44 % of the maximum) and carbon use efficiency (CUE) in July underlined the mobilization of stored material during drought stress by a higher air temperature (32.7 °C).  相似文献

高胆固醇饲料喂养造成的动脉粥样硬化(As)模型家兔通过静脉注射人血浆HDL制剂,观察HDL对As家兔肝细胞膜LDL受体活性的影响.结果发现,摄取高胆固醇饲料的As家兔,其肝细胞膜LDL受体Kd值虽无明显变化但Bmax值显著减小(P<0.01,与正常对照组比较);注射HDL制剂后,As家兔肝细胞膜LDL受体Kd值仍无明显改变,但Bmax值却显著回升(P<0.01,与高脂组比较).表明人血浆HDL具有增加As家兔肝细胞膜LDL受体活性的作用.  相似文献

为深入了解Cr6+胁迫对人工湿地植物薏苡光合特性和微量元素吸收的影响,该文通过构筑微型垂直流薏苡(Coix lacryma-jobi)人工湿地,采用1/2 Hoagland''s营养液配制的含0、5、20、40 mg·L-1 Cr6+废水为灌溉用水,研究铬胁迫对人工湿地植物生长、光合特性、抗氧化酶活性和微量元素吸收等的影响。结果表明:(1)低浓度(5 mg·L-1)Cr6+对薏苡的株高、茎粗和分蘖影响不显著,高浓度(20、40 mg·L-1)Cr6+则显著抑制薏苡的生长。(2)低浓度Cr6+处理下,薏苡叶片净光合速率(Pn)、气孔导度(Gs)和蒸腾速率(Tr)有不同程度的提高,提高幅度分别为6.8%~54.8%、13.0%~40.3%和9.1%~36.4%。高浓度Cr6+处理下,叶片PnGsTr等指标显著下降,但叶片胞间CO2浓度(Ci)则显著提高。(3)薏苡叶片超氧化物歧化酶(SOD)和过氧化物酶(POD)活性均随着处理时间的延长而提高; 低浓度Cr6+处理下,薏苡叶片SOD活性与对照差异不大,高浓度Cr6+处理下,SOD活性受到显著抑制。薏苡叶片POD活性和丙二醛(MDA)含量均随Cr6+处理浓度的提高而提高。(4)高浓度Cr6+处理显著抑制根系、茎、叶对Cu 、Fe、Mn和Zn的吸收。(5)5、20 mg·L-1 Cr6+处理下,人工湿地对Cr6+去除率最高可达99%,40 mg·L-1 Cr6+处理下最高则为86%。综上认为,Cr6+胁迫导致Fe、Mn、Zn和Cu等元素吸收量显著下降,光合受阻,抗氧化系统受损,植物生长受到抑制,最终导致人工湿地处理含Cr6+废水的能力下降。  相似文献

【目的】探索研究反刍动物胃肠道微生物合成维生素B12的方法,并评估植物乳酸菌或博落回提取物对断奶山羊回肠食靡微生物合成维生素B12的影响。【方法】选取体重相近年龄相仿的断奶黑山羊20只,随机分为对照组(CON, n=7)、乳酸菌组(LAC, n=7)和博落回组(MAC, n=6)。CON组饲喂普通的日粮,LAC组饲喂基础日粮+10 g/d的植物乳酸菌(Lactobacillus plantarum P-8 strains, 4.0×109 CFU/g),MAC组饲喂基础日粮+0.3 g/d的博落回提取物(Macleaya cordata 3.75%)。试验结束后,采集回肠中段食靡样品。利用宏基因组测序技术,比对最新功能基因数据库VB12Path和公共数据库KEGG,分析植物乳酸菌和博落回提取物对山羊回肠食靡微生物合成维生素B12的影响。【结果】结果显示,比对VB12Path数据库共注释到55个与维生素B12合成相关的基因。与CON组相比,LAC组和MAC组中合成维生素B12基因的丰富度和均匀度降低(P<0.05)。3组间基因的β多样性也有显著的差异(P<0.05);比对KEGG数据库共注释到49个与维生素B12合成相关的基因,LAC组的多样性与CON组没有差异,但MAC组的α多样性显著降低(P<0.05)。值得注意的是,比对VB12Path数据库和KEGG数据库均发现LAC组和MAC组中参与前咕啉2合成途径、参与无氧合成途径、有氧合成途径、参与重排转换途径以及腺苷钴胺素合成途径的部分基因(gltXcbiTcobTbtuD等)的丰度均显著地高于CON组(P<0.05)。【结论】2个数据库比对后的相似结果表明博落回提取物在对断奶山羊回肠微生物合成维生素B12相关代谢上与植物乳酸菌的作用相似,均可以通过改变其多样性和提高部分关键基因的丰度,从而影响微生物合成维生素B12的潜能,为后期博落回提取物和植物乳酸菌在畜牧养殖中的运用提供一定的理论支撑。此外,2个数据库比对的差异提示未来研究胃肠道微生物维生素B12相关代谢时,应用多个数据库比对,能更全面精确地进行评价,为后期分析过程奠定研究基础和提供新的思路。  相似文献

以黄土丘陵区退耕草地群落典型优势种白羊草、长芒草和达乌里胡枝子为研究对象,测定不同氮磷(主区N:0,50,100 kg N hm-2 a-1;副区P:0,40,80 kg P2O5 hm-2 a-1)添加处理下叶片光合气体交换参数、叶绿素荧光参数、叶片氮磷含量、光合氮利用效率(PNUE)和光合磷利用效率(PPUE),探究不同优势种光合生理特征对外源氮磷添加的响应特征与物种差异。结果表明:单独氮添加下,3个优势种的净光合速率(Pn)、蒸腾速率、水分利用效率(WUEi)、气孔限制值(Ls)和光系统Ⅱ(PSⅡ)活性高于或显著高于未施肥处理,白羊草和达乌里胡枝子的PNUE和PPUE以及长芒草的PPUE显著增加。单独磷添加下,仅达乌里胡枝子的Pn、WUEiLs和PNUE相比未施肥处理显著增加。氮磷共同添加下,50 kg N hm-2 a-1处理下,施磷后达乌里胡枝子的Pn以及三个优势种的PNUE显著增加,白羊草的PPUE显著降低;100 kg N hm-2 a-1处理下,施磷后达乌里胡枝子的Pn和PNUE均显著增加,白羊草PPUE和长芒草的PNUE的仅在40 kg P2O5 hm-2 a-1处理下显著增加。3个优势种的Pn均与PSⅡ最大光化学效率和叶片氮含量呈显著正相关关系,长芒草和达乌里胡枝子的Pn与PSⅡ潜在活性和叶片磷含量呈显著正相关关系。总体表明,氮磷对黄土丘陵区退耕草地优势种光合生理具有一定的交互作用,物种属性和施肥水平影响其光合生理特征响应程度。  相似文献

Six months old in vitro-grown Anoectochilus formosanus plantlets were transferred to ex-vitro acclimation under low irradiance, LI [60 μmol(photon) m−2 s−1], intermediate irradiance, II [180 μmol(photon) m−2 s−1], and high irradiance, HI [300 μmol(photon) m−2 s−1] for 30 d. Imposition of II led to a significant increase of chlorophyll (Chl) b content, rates of net photosynthesis (P N) and transpiration (E), stomatal conductance (g s), electron transfer rate (ETR), quantum yield of electron transport from water through photosystem 2 (ΦPS2), and activity of ribulose-1,5-bisphosphate carboxylase/ oxygenase (RuBPCO, EC 4.1.1.39). This indicates that Anoectochilus was better acclimated at II compared to LI treatment. On the other hand, HI acclimation led to a significant reduction of Chl a and b, P N, E, g s, photochemical quenching, dark-adapted quantum efficiency of open PS2 centres (Fv/Fm), probability of an absorbed photon reaching an open PS2 reaction centre (Fv′/Fm′), ETR, ΦPS2, and energy efficiency of CO2 fixation (ΦCO2PS2). This indicates that HI treatment considerably exceeded the photo-protective capacity and Anoectochilus suffered HI induced damage to the photosynthetic apparatus. Imposition of HI significantly increased the contents of antheraxanthin and zeaxanthin (ZEA), non-photochemical quenching, and conversion of violaxanthin to ZEA. Thus Anoectochilus modifies its system to dissipate excess excitation energy and to protect the photosynthetic machinery.  相似文献

Kandelia candel (L.) Druce is the dominant mangrove species on the west coast of northern Taiwan. We have measured the net photosynthetic rate (P N) and chlorophyll (Chl) a fluorescence of seedlings grown at combinations of two nitrogen (0.01 and 0.1 mM) and two NaCl (250 and 430 mM NaCl) controls. With the same nitrogen level, seedlings grown at higher salinity (HS) had a significantly lower P N and stomatal conductance (g s) than those at lower salinity (LS). An increase in nitrogen availability significantly elevated P N and g s of the LS-grown seedlings. Compared to dark adapted leaves, the maximum quantum yield of photosystem 2 (PS2) (Fv/Fm) of leaves exposed to PFDs of 1200 and 1600 μmol m-2 s-1 for 2 h was significantly reduced. The degree of Fv/Fm reduction differed among leaves of the four types of treated plants. Chl fluorescence quenching analysis revealed differences among the examined plants in coefficients of non-photochemical and photochemical quenching. 植物资源(光、氮、水分)利用效率是反映生态功能适应气候变化的关键指示,然而鲜有研究综合考虑植物资源利用效率间的相对变化及其调控机制。选取宁夏盐池毛乌素沙地优势物种油蒿(Artemisia ordosica)为研究对象,于2017-2019年生长季对油蒿光合生理参数和环境因子进行原位监测,实验室测定叶片比叶面积(SLA)和叶氮含量(LNC),分析叶片光利用效率(LUE)、水分利用效率(WUE)、氮利用效率(NUE)的相对变化特征及其生物和非生物影响因子,探讨油蒿叶片资源利用效率对环境的响应。结果显示:油蒿LUE和NUE的季节变化趋势基本一致,两者呈正相关(R2=0.17;P<0.01),且皆与WUE季节变化不同,无显著关系(P>0.05),WUE波动幅度最高(CV=48%),NUE最低(CV=39%);研究期间LUE、WUE和NUE月均值基本在夏季出现高峰值,分别为0.12 mol/mol,104.02 μmol/mol和11.49 μmol g-1 s-1。土壤含水量(SWC)>0.09 m3/m3,叶片资源利用效率不受其影响,而SWC<0.09 m3/m3,WUE和SWC关系为二次函数;SWC调节土壤氮含量(Nsoil)和光合有效辐射(PAR)对叶片资源利用效率的影响。叶片资源利用效率与LNC无显著相关性;SLA与LUE显著负相关(P<0.01),与NUE显著正相关(P<0.01),与WUE相关性不显著(P>0.05)。LUE主要受SLA和Nsoil影响,NUE主要受SLA和SWC影响,SWC和Nsoil还可通过SLA和LNC间接影响LUE和NUE。结果表明水分和土壤氮含量是限制油蒿叶片资源利用效率的主要非生物因子,比叶面积则是调控其资源利用效率的关键生物因子,是深入探究荒漠植物群落对环境响应策略的重要补充。  相似文献

为探讨叶片斑纹的光合适应意义,该文选择有斑和无斑两种叶型的维西堇菜(Viola monbeigii)作为研究材料,采用石蜡切片制片和显微镜观察叶片结构及GFS 3000便携式光合测量仪测量光合参数并进行比较。结果表明:(1)两种叶型维西堇菜气孔均为不等型气孔,有斑叶片上、下表皮气孔密度、栅栏组织(PT)厚度、栅栏组织与海绵组织的比值(PT/ST)均显著低于无斑叶片; 而上表皮气孔较大,表现出更适应弱光环境的结构特征。(2)两种叶型维西堇菜暗呼吸速率(Rd)、初始荧光(Fo)、最大荧光产量(Fm)、光化学猝灭系数(qP)、非光化学猝灭系数(qN)、PSⅡ最大光化学量子产量(Fv/Fm)、实际光化学反应量子效率(Yield)均无显著差异,有斑植株叶片叶绿素含量、最大净光合速率(Pmax)显著低于无斑植株; 有斑叶片表观光合电子传递速率(ETR)在光合有效辐射(PAR)处于400~2 000 μmol·m-2·s-1之间时显著低于无斑叶片,无斑植株的光饱和点(LSP)高于有斑植株,而补偿点(LCP)较低。综上认为,无斑植株PAR利用范围较宽,光合适应能力更强,有利于维西堇菜充分利用环境中有限的资源,保障物种生存; 有斑植株具有较适应弱光胁迫的特征,表明叶片斑纹的出现可能是维西堇菜适应林缘弱光环境的一种策略。  相似文献

根据天然超氧化物歧化酶(SOD)活性部位结构合成了含苯并咪唑的5种配体及其32种分别含Cu(Ⅱ)、Fe(Ⅲ)、Mn(Ⅱ)、Co(Ⅱ)的模拟化合物.经光谱、电化学测试证明这些化合物具有拟S3D活性,其50%抑制率浓度(IC50)为10-6~10-8mol·L-1,催化超氧离子自由基(O-·2)歧化的反应速率常数kq为106~108mol-1·L·s-1.同时观察到几种模拟化合物有抗肿瘤活性或增强水稻抗寒性.  相似文献

双须叶须鱼(Ptychobarbus dipogon Regan)隶属裂腹鱼亚科(Schizothoracinae)叶须鱼属(Ptychobarbus),是西藏特有经济鱼类,2016年被列入中国脊椎动物红色名录,必须加快推动该鱼的养护工作。2013年2~3月份及2014年2~6月份,在雅鲁藏布江谢通门段与支流拉萨河上游段采集1 030尾双须叶须鱼样本,以脊椎骨作为年龄鉴定材料进行年龄与生长特点研究,以期分析和评价该类群鱼类资源情况。研究结果如下:双须叶须鱼样本中最大年龄为49龄,最小年龄为4龄;体重与体长关系是样本总体W=4.4×10-5?SL2.7688、雌鱼W=5.0×10-4?SL2.3474、雄鱼W=2.8×10-5?SL2.8414;体长生长方程为雌鱼Lt(♀)=431.8[1-e-0.19t+1.19)],渐进体长L(♀)=431.8 mm,拐点年龄为3.3龄,雄鱼Lt(♂)=367.6[1-e-0.42t+3.37)],渐进体长L(♂)=367.7 mm;体重生长方程为雌鱼Wt(♀)=767.40[1-e-0.19t+1.19)]2.3474,雄鱼Wt(♂)=545.02[1-e-0.42t+3.37)]2.8414;群体异速生长指数b=2.768 8与匀速生长指数3存在显著性差异,即P<0.05。研究表明双须叶须鱼体趋向低龄化,应当予以充分关注。  相似文献

The seasonal changes of photosynthesis of cones of Japanese larch (Larix kaempferi Carr.) trees showed that gross photosynthetic rate of young cones (P G) was 2–3 μmol m−2 s−1 at surface area unit and P G/R D (dark respiration of cones) peaked about 0.7 in the same period, indicating that 70 % of respiratory CO2 was re-fixed. With maturation, P G and P G/R D sharply decreased. Chlorophyll content in cones was 3–20 % of that in leaves, which made it a limiting factor for photosynthesis and its content was closely correlated with photosynthetic capacity. Although sunken and linearly arranged stomatal organs were found on the scale of young cones, differently from the significant regulation of leaf photosynthesis, these stomata tended to be non-functional since CO2 is not limiting factor for cone photosynthesis. Thus photosynthesis of larch cones is an additional contribution to their development.  相似文献
