荞麦柱头、花柱的结构及ATP酶的超微细胞化学定位   总被引:1,自引:0,他引:1  
利用超薄切片结合ATP酶定位技术,研究了荞科柱头及花柱的结构。结果表明,成熟的荞麦柱头表面无乳突细胞,柱头细胞的细胞壁存在发达的壁内突,花柱为实心型无引导组织,柱头表面的细胞壁根据电子密度的不同可划分为3层,最外一层电子密度最大,最内一层电子近透明,并形成壁内突的结构,柱头表面和花柱细胞的质膜上ATP酶的活性较高,表明成熟的柱头和花柱细胞物质代谢和运输十分活跃,成熟的柱头和花柱细胞中有丰富的蛋白质和多糖类物质。  相似文献

通过Western印迹法、免疫组织化学和超微细胞化学等技术,研究了烟草柱头和花柱中阿拉伯半乳糖蛋白(arabinogalactan-proteins,AGPs)的分布。结果表明烟草柱头和花柱组织中含有大量的AGPs,主要分布于柱头表皮细胞的细胞质和分泌层细胞的胞外基质中,且授粉前后AGPs的分布情况差异不明显;而花柱中的AGPs主要分布于表皮细胞的外层细胞壁、维管组织周围细胞的细胞质及引导组织的胞外基质中;花粉管通过后,引导组织胞外基质中AGPs减少,而花粉管细胞质和花粉管壁中检测到大量AGPs。  相似文献

向日葵柱头,花柱和珠孔中钙分布的超微细胞化学定位   总被引:16,自引:0,他引:16  
用焦锑酸盐沉淀法对向日葵(HelianthusannuusL.)授粉前后柱头、花柱和珠孔中的钙进行了超微细胞化学定位。同时还运用X射线能谱(EDX)和波谱(WDX)两种方法进行了X射线定性分析,证明了前法所得沉淀确系焦锑酸钙。观察表明,花粉萌发和花粉管生长所经的柱头接受面,花柱引导组织和珠孔引导组织中含钙较柱头、花柱和珠孔的其它部位明显地多。柱头乳突细胞的表面和花柱引导组织的胞间基质中、尤其胞间基质与细胞壁外层相接之处钙很密集。在珠孔外端引导组织中,以角质层为界,钙主要分布于其近珠柄侧。花粉管壁果胶质层中有相当多的钙。结合向日葵中已有的研究和其他文献,讨论了钙的分布与花粉管生长的关系  相似文献

烟草柱头表面有两层覆盖物,其中含有少量细小的钙颗粒.花粉落到柱头上后,储存在花粉外壁中的钙被释放到覆盖层中.当花粉管穿过覆盖层长入柱头细胞之间时,花粉管顶端的细胞壁中出现了大量的细小钙颗粒.开花后22 h观察时,在花柱引导组织中形成了钙的梯度分布:花柱上部引导组织中的钙较少,而下部连接子房处的花柱引导组织中含有较多的钙颗粒.去雄花开花后1 d时,花柱上部引导组织中的钙明显增多;3 d时,连柱头细胞中也出现了较多的钙颗粒.讨论了烟草花柱引导组织中钙梯度分布和花粉管生长的关系.  相似文献

山茱萸成熟雌蕊的柱头为干型,具表皮起源的单细胞乳突。在开花后授粉前乳突细胞的顶部发生凹陷,授粉后乳突细胞迅速萎缩。花柱在发育初期为开放型,发育后期花柱道消失,成为一种具特殊结构的闭合型。成熟花柱由表皮、皮层和特殊的引导组织等部分构成,在皮层中贯穿有3条维管束,引导组织的中央则有二列类似腺质的内表皮细胞,该细胞富含蛋白质、核酸和多糖类物质。  相似文献

应用扫描电镜和荧光显微镜对软枣猕猴桃同种花粉在柱头上原位萌发及花粉管生长情况进行了观察,并用焦锑酸盐沉淀法对其授粉前后柱头及花柱中Ca2+进行超微细胞化学定位.结果显示:(1)猕猴桃柱头属于干性柱头,具有一道裂沟,乳突呈长圆柱形,授粉前后形态差异不明显.(2)授粉后3 h,花粉管生长穿过柱头表面,授粉后7 h,花粉管生长到达花柱底部;(3)授粉前后,柱头接受面靠近柱头外围细胞的角质层一侧细胞器内含有丰富的钙,而柱头非接受表面钙颗粒分布很少;(4)授粉前和授粉后3 h,花柱顶端钙颗粒较少,基部钙颗粒较多;授粉后7 h,花柱顶端和基部钙分布密度无明显差别;(5)授粉前后花柱顶端钙主要均匀分布在细胞质膜位置;在花柱基部授粉前钙主要分布在引导组织胞间隙中,授粉后3 h主要分布在细胞质内,授粉后7 h主要存在于细胞质、内质网上.研究表明,猕猴桃授粉前后,柱头和花柱组织中均含有钙,授粉前和授粉后3 h花柱中的钙呈现出梯度分布,授粉后7 h钙的梯度分布现象减弱甚至消失.  相似文献

为了深入了解植物捕获花粉的机制和生殖策略,我们对3种耧斗菜属Aquilegia L.植物华北耧斗菜A. yabeana Kitagawa、无距耧斗菜A. ecalcarata Maxim.和秦岭耧斗菜A. incurvata P. K. Hsiao的雌蕊进行了扫描电镜和荧光显微镜观察,发现其柱头发育的特殊式样。其柱头位于花柱的腹缝面,由花柱腹缝两侧细胞发育成柱头乳突,在花期中从顶端朝花柱基部渐次成熟。伴随花柱向外弯曲,成熟的柱头表面相继呈现,逐渐增加了花粉落置的空间。这种柱头发育的式样通过延长柱头的可授期,增加了植物在不可预测的传粉环境下的繁殖机会。  相似文献

An ultrastructural investigation of the entire transmitting tract in Trimezia fosteriana (Iridaceae) was undertaken. The transmitting tissue is secretory but transfer cells do not occur at any level. With exception for the stigma papillae, the cells are covered with large amounts of secretory products. The papillae have a thick and ridged cuticle. The cuticle in the rest of the transmitting tract is thin and detached from the cell wall by the secretory products. It is more or less ruptured in the secretory parts of the stigma and ovary. In the stylar canal the major part of the cuticle is continuous and covers the secretory products. The occurence of a large amount of vesicles in the stigma transmitting tissue cells is interpreted as a result of high dictyosome activity. An electron opaque material is produced in the dictyosomes and appears in vesicles and vacuoles but also between the plasma membrane and the cell walls in the stigma. A small amount of such material is present in the cell walls. Corresponding material is also present in the style and the ovary but declines basipetally. Plastids with strongly electron opaque plastoglobules are present at all levels in the transmitting tract.  相似文献

利用透射电镜技术研究了黄瓜(Cucumis sativus L.)雌花柱头发育过程中传递组织、分泌组织和乳突细胞的超微结构。在整个发育过程中,乳突细胞和分泌组织细胞的细胞质内密布很多管状及槽库膨大的内质网,产生很多分泌囊泡;在成熟柱头的传递组织和分泌组织细胞间观察到大量的胞间连丝;乳突细胞和分泌细胞高度液泡化,质膜内折;在柱头发育过程中分泌组织细胞的核周腔扩大形成裂瓣状核,到柱头成熟阶段裂瓣状核更加明显。进一步的研究显示,在成熟柱头的不同组织细胞中, ATPase的活性呈现在质膜和液泡膜上,随着柱头的发育,PM-H+-ATPase的比活性明显增强。结果表明,黄瓜雌花柱头的腺特征随发育进程而趋于显著。  相似文献

The detailed ontogeny of postgenital fusions within the gynoecium of Catharanthus roseus was investigated. The basal margins of the young carpel primordia infold and fuse together to seal shut the loculi. Independently, the opposing distal tips of the two carpels also unite, with the fusion region subsequently developing into the stigma, style, and a small distal region of the compound ovary. The basal ovary regions of the two opposing carpels remain unfused, thus leaving the tip fusion spatially restricted. In the region of contact, cells with distinctively epidermal features progressively lose their epidermal character after their participation in the fusion. In the fused stigma these former epidermal cells redifferentiate into transmitting and secretory tissues; in the fused style these cells undergo a tremendous expansion in length while forming stylar transmitting tissue; but in the compound ovary region corresponding cells experience little expansion or redifferentiation. It is concluded that the loss of epidermal features or the occurrence of periclinal cell divisions in the epidermis is a definitive indication that cells have fused postgenitally. However, studies with the transmission electron microscope are necessary to detect the first indications of a postgenital fusion. The compound ovary region within the gynoecium of C. roseus is a tissue appropriate for a high resolution ultrastructural study of the cytological events accompanying postgential tissue fusion because the fusion occurs quickly and little subsequent cell expansion takes place within this region.  相似文献

Soybeans have a wet stigma overtopped by a pellicle that originates from the cuticle. There are numerous exudate-filled, axially oriented channels between cells of the transmitting tissue in the stigma and style. Pollen tubes grow in these channels and receive nutrition and mechanical guidance. Transmitting-tissue cells of the obturator are secretory also, but the obturator in soybean does not appear to control direction of pollen tube growth mechanically. The significant function of transmitting tissue in soybeans is to provide nutrition and to control direction of pollen tube growth.  相似文献

The gynoecium of Phaseolus acutifolius var. latifolius, a self-compatible legume, is characterized by a wet non-papillate stigma, an intermeditae hollow/solid style type, and secretory cells on the ventral surface of the ovary which direct pollen tube growth. The stigma is initially receptive 5–6 days prior to anthesis. Production of stigmatic secretions, composed primarily of carbohydrates and lipids, fragment the cuticle covering epidermal cells of the stigma early in ontogeny; the lipidic aspect of the copious secretions apparently serves to inhibit desiccation after the cuticle is ruptured. Stylar canal development occurs as a combination of elongation of a basal canal present early in development, and dissolution of part of a solid transmitting tract tissue just below the stigma. Anthers dehisce and the tricolporate pollen is released onto the receptive stigma one day before anthesis. Following initial growth in intercellular spaces in the transmitting tract of the stigma, pollen tubes adhere to epidermal secretory cells along the ventral side of the stylar canal and upper ovary; here the transmitting tract is apparently limited in the number of tubes it can accommodate, providing a possible site of selection of male gametes.  相似文献

The bilobed papillate stigma of Nicotiana sylvestris Speg. andComes, is covered at maturity with a copious exudate containinglipid, protein and carbohydrate. The stigma is receptive fromthe very early stage of development and it also stains positivelyfor esterase activity. The stigma has three distinct zones:an epidermis with papillae; a subepidermal secretory zone; anda parenchymatous ground tissue. The behaviour of the cells ofthese three zones has been followed from 6 d before anthesisto one day after anthesis and pollination. The cells of theepidermis and the secretory zone stain intensively for lipids,proteins and carbohydrates in the initial stages. The secretoryzone develops large intercellular spaces containing heterogenoussecretory products which also stain positively for the aforesaidthree compounds. At maturity the secretory products are releasedto the surface through gaps formed in the epidermis by cellseparation. The main secretion of the stigma is produced bythe cells of the secretory zone. Less secretion is derived fromthe stigmatic papillae. Some amount of secretion is also releasedfrom the stylar transmitting tissue adjoining the stigma. Theglandular cells of the stigma contain numerous plastids, mitochondria,ribosomes, ER, cytoplasmic lipid droplets and some dictyosomes.The plastids and the vacuoles in the secretory cells of thestigma have a lot of electron dense (osmiophilic) inclusionsrespectively in the initial and later stages of development.The former are probably involved in the production of thesematerials. It is suggested that the proteins are directly secretedby rough ER compartments whereas smooth ER is involved in thesynthesis of lipidic materials. The carbohydrate moiety of theexudate is released by the eccrine mode (sugar mono- and dimers)with some addition of polymers by disintegration of the middlelamellae. Pistil development was studied in transgenic tobacco plants in which the stigma is ablated by expression of a stigma-specific cytotoxic gene. These plants offer a tool to investigate the process of differentiation of the secretory zone, in that cell death caused by barnase activity provides a marker to follow cell fate at high resolution. After fusion of the carpel walls in the region most distal from the ovary, the epidermal cells begin to divide in both wild-type and stigmaless plants. Divisions of the L1 layer of the pistil are immediately followed by the morphogenetic events that lead to three different cell types: rounded-angular cells showing an equal number of anti- and periclinal divisions, cells that are more oblong forming the transition zone, and the square cells of the transmitting tissue dividing mostly anticlinally with respect to the original carpel wall. In the stigmaless plants, cell death caused by the expression ofSTIG 1-barnase begins at stage –1 and proceeds gradually, but is always associated with round epidermal cells and with angular-rounded cells underneath them. Studies at the ultrastructural level show that cell death caused by barnase activity occurs first in solitary cells and gradually extends to groups of cells.In situ hybridizations using the STIG 1 RNA probe in wild-type pistils confirm these results. Most likely, the cells in whichSTIG 1 is expressed are those that have just differentiated into the secretory cell type. Our results indicate that the transition zone or neck is autonomously differentiated from the secretory zone and the transmitting tissue. Furthermore, our results indicate that in both wild-type and stigmaless pistils secretion of lipids most likely occurs through the plasmodesmata. This observation suggests that bulk transport can occur via plasmodesmata.  相似文献
