Kocide 101 (77% cupric hydroxide) and Ridomil plus (15% metalaxyl and 35% copper oxychloride) were used in the treatment of tomato plants in the field. Generally, the two fungicides exhibited an inhibitive effect on the total and individual counts of glucophilic fungi after different periods of treatment except in some cases, the counts were not affected or promoted compared with the control. Fungal genera and species associated with roots and shoots of tomato plants were isolated from untreated plants as control and then compared with those isolated from plants treated with different doses of fungicides. The most common fungal species recovered from soil and different parts of plants were Alternaria alternata, Aspergillus flavus, A. fumigatus, A. niger, A. terreus, Cochliobolus spicifer, Emericella nidulans, E. nidulans var. lata, Fusarium oxysporum, Gibberella fujikuroi, Mycosphaerella tassiana, Nectria haematococca, Penicillium chrysogenum, P. funiculosum, P. oxalicum, Stachybotrys chartarum and Rhizopus stolonifer.  相似文献

Argemone mexicana L. (Papaveraceae), a tropical annual weed, is known to be phytotoxic to many crop species. This study was designed to examine the possible impact of A. mexicana on root‐infecting fungi, changes in fungal community structure and the growth of tomato. A. mexicana decaying shoots in soil provided a marked decrease in the infectivity of Fusarium solani and Rhizoctonia solani but Macrophomina phaseolina remained unaffected. Plant height and shoot growth of tomato plants increased markedly though high concentration of A. mexicana (5% w/w) was deleterious to tomato plants. General species diversity of soil fungal communities increased in the amended soils over the controls and greater increase in diversity occurred at higher concentrations of decaying A. mexicana. Likewise, equitability and richness components of diversity increased in treatments compared to controls but declined with increasing sampling period. Aspergillus nidulans, Cephaliophora irregularis, Drechslera halodes, Paecilomyces lilacinus and Trichoderma viride were isolated exclusively from the amended soils. Aqueous extract of A. mexicana when applied in soil greatly suppressed all three of the above root‐infecting fungi, and at lower concentration actually enhanced plant growth. The influence of different levels of N‐fertilization with NH4NO3 on the modification of the effect of decaying A. mexicana on root‐infecting fungi was also investigated. N‐fertilization to some extent alleviated the phytotoxicity to tomato plants while suppressing the root‐infecting fungi. A. mexicana in conjunction with Pseudomonas aeruginosa, a plant growth‐promoting rhizobacterium, significantly suppressed root‐infecting fungi with concomitant increase in plant growth. Whereas P. aeruginosa was reisolated from the rhizosphere and inner root tissues of tomato, its population slightly declined in the amended soil but not to an extent that could reduce the biocontrol and growth promoting potential of the bacterium.  相似文献

Summary Hippophaë rhamnoides seedlings were grown in sterilized and unsterilized soil from a decliningH. rhamnoides scrub, to which different numbers ofLongidorus sp. andTylenchorhynchus microphasmis were added. In sterilized and unsterilized soil, retardation of growth, content of dry matter in the shoots, and incidence of deformed short lateral roots of test plants were positively correlated with counts of both nematode species. Nitrogen content in the shoots, nodulation on the roots of test plants and increase increase in nematodes were negatively correlated with the initial number of both nematode species in sterilized soil. In unsterilized soil, an unknown biotic factor was present that reduces growth ofH. rhamnoides, nodulation and multiplication of the nematodes. A very large number of plant species are capable of forming symbiotic associations with arbuscular mycorrhizal (AM) fungi. The roots of these plants are potentially capable of absorbing P from the soil solution both directly through root epidermis and root hairs, and via the AM fungal pathway that delivers P to the root cortex. A large number of phosphate (P) transporters have been identified in plants; tissue expression patterns and kinetic information supports the roles of some of these in the direct root uptake pathways. Recent work has identified additional P transporters in several unrelated species that are strongly induced, sometimes specifically, in AM roots. The primary aim of the work described in this paper was to determine how mycorrhizal colonisation by different species of AM fungi influenced the expression of members of the Pht1 gene families in the cereals Hordeum vulgare (barley), Triticum aestivum (wheat) and Zea mays (maize). RT-PCR and in-situ hybridisation, showed that the transporters HORvu;Pht1;8 (AY187023), TRIae;Pht1;myc (AJ830009) and ZEAma;Pht1;6 (AJ830010), had increased expression in roots colonised by the AM fungi Glomus intraradices,Glomus sp. WFVAM23 and Scutellospora calospora. These findings add to the increasing body of evidence indicating that plants that form AM associations with members of the Glomeromycota have evolved phosphate transporters that are either specifically or preferentially involved in scavenging phosphate from the apoplast between intracellular AM structures and root cortical cells. Operation of mycorrhiza-inducible P transporters in the AM P uptake pathway appears, at least partially, to replace uptake via different P transporters located in root epidermis and root hairs. In the years 1985–1989, the occurrence of arbuscular fungi and mycorrhizae on the Hel Peninsula (Poland) was investigated with the help of 45 soil and root samples collected under 20 plant species of eight families. Except for Zea mays, the other plant species were from uncultivated sites. All soil samples contained spores of arbuscular fungi, of which about 45% were of the genus Glomus. Acaulospora spp. preferred members of the Cupressaceae. Spores of Gigaspora occurred rarely and only in two plant families. Glomus spp. were most frequently associated with plants of the Rosaceae, and species of Scutellospora were found at markedly higher frequencies among roots of plants of the Gramineae and Cupressaceae. A total of 29 spore-forming species and Glomus tenue (a fungus recognizable by its distinctive infections) were found. The most frequently recovered fungus, Glomus tenue, was present in roots of 56.8% of examined plants. Of the spore-forming fungi, the most frequently isolated spores were those of Scutellospora dipurpurascens, then Glomus constrictum, Acaulospora 61, and Glomus microcarpum. The overall spore density in examined samples averaged 99.8 in 100 g dry soil in the range 1 to 547, and was highest in a sample taken from around roots of Festuca arundinacea. The dominant fungi forming spores in sampled soils were Glomus constrictum, Glomus microcarpum, and Scutellospora dipurpurascens. The average species density was 3.9 in 100 g dry soil in the range 1 to 10, and was highest in Corynephorus canescens, Rosa canina, and Thuja occidentalis. Levels of colonization by arbuscular fungi ranged from 0.0 to94.0% (mean 23.3%) of the root length and were highest in Festuca arundinaceae and Zea mays.  相似文献

Zn uptake by maize plants may be affected by the presence of arbuscular mycorrhizal fungi (AMF). Collembola often play an important controlling role in the inter-relationship between AMF and host plants. The objective of this experiment was to examine whether the presence of Collembola at different densities (0.4 and 1 individuals g−1 dry soil) and their activity have any effect on Zn uptake by maize through the plant–AMF system. The presence of the AMF (Glomus intraradices) and of the Collembola species Folsomia candida was studied in a laboratory microcosm experiment, applying a Zn exposure level of 250 mg kg−1 dry soil. Biomass and water content of the plants were no different when only AMF or when both AMF and Collembola were present. In the presence of AMF the Zn content of the plant shoots and roots was significantly higher than without AMF. This effect was reduced by Collembola at both low and high density. High densities of Collembola reduced the extent of AMF colonization of the plant roots and hyphal length in the soil, but low densities had no effect on either. The results of this experiment reveal that the F. candidaG. intraradices interaction affects Zn uptake by maize, but the mechanisms are still unknown.  相似文献

Communities of indigenous arbusuclar mycorrhizal (AM) fungi are expected to alter phosphorus uptake and biomass productivity of plants according to characteristics of the life cycles of the fungi present and the way they interact with each other inside roots and with host plants. Differences in the relative abundance of AM fungi inside roots could influence P uptake if the fungi present differ in effectiveness at accessing P and transferring it to the plant. However, it is difficult to assess the contribution of AM fungi under field conditions. We investigated P uptake, from point sources of P placed 2, 4 and 6 cm from roots, by plants colonised by a community of AM fungi in jarrah forest soil. Roots were retained within a mesh bag to prevent them from growing towards the point source of P. The relative abundance of morphotypes of fungi inside roots and the P status of plants were assessed after 12 and 16 weeks. First, a bioassay was carried out in undisturbed forest soil cores using two host plants, a forest understorey plant Phyllanthus calycinus Labill and the annual pasture species subterranean clover (Trifolium subterraneaumL.), to assess the infectivity of the indigenous community of AM fungi. Roots of both bioassay host plants were colonised in similar proportions by morphotypes of AM fungi resembling Glomus, Acaulospora, Scutellospora and fine endophytes. In this bioassay, there were positive correlations between the proportion of root length colonised and plant biomass and P uptake for P. calycinus, but not for subterranean clover. In the experiment assessing the capacity of P. calycinus to access P placed at increasing distances from the root, shoot P content and concentration in P. calycinus were greater when P was placed 2 cm compared with 4 and 6 cm from roots. The length of hyphae in the vicinity of the point source of P decreased with increasing distance from the plant. The extent to which the individual AM fungi were involved in P uptake is not known. The Glomus morphotype was dominant at both times of sampling.  相似文献

Low-input agricultural systems that do not rely on fertilizers may be more dependent on vesicular-arbuscular mycorrhizal [VAM] fungi than conventionally managed systems. We studied populations of spores of VAM fungi, mycorrhiza formation and nutrient utilization of maize (Zea mays L.) grown in moldboard plowed, chisel-disked or no-tilled soil under conventional and low-input agricultural systems. Maize shoots and roots were collected at four growth stages. Soils under low-input management had higher VAM fungus spore populations than soils under conventional management. Spore populations and colonization of maize roots by VAM fungi were higher in no-tilled than in moldboard plowed or chisel-disked soil. The inoculum potential of soil collected in the autumn was greater for no-till and chisel-disked soils than for moldboard plowed soils and greater for low-input than conventionally farmed soil. The effects of tillage and farming system on N uptake and utilization varied with growth stage of the maize plants. The effect of farming system on P use efficiency was significant at the vegetative stages only, with higher efficiencies in plants under low-input management. The effect of tillage was consistent through all growth stages, with higher P use efficiencies in plants under moldboard plow and chisel-disk than under no-till. Plants grown in no-tilled soils had the highest shoot P concentrations throughout the experiment. This benefit of enhanced VAM fungus colonization, particularly in the low-input system in the absence of effective weed control and with likely lower soil temperatures, did not translate into enhanced growth and yield.  相似文献

Arbuscular mycorrhizal (AM) fungi can form symbiosis with 90% of the vascular plants and play important roles in ecosystem. To realize the AM fungal colonization at different succession stages in saline-alkali land and screen AM fungi species with great functions, roots and soil samples were collected from the three succession stages of Songnen saline-alkali grassland. The soil properties and AM fungal colonization were measured, and the fungus distributed extensively in three stages was annotated by sequencing for AML1/AML2 target, subsequently, maize was selected as the host to verify its colonization. The results showed that the soil properties improved with the succession of saline-alkali grassland. The plants’ communities of the three stages could be colonized by AM fungi, and the colonization rate of Leymus chinensis (the third stage) ranged from 66.67% to 100%, Puccinellia tenuiflora (the second stage) ranged from 50% to 80%, while the Suaeda glauca (the first stage) was only 35%–60%. Glomeraceae sp1 was identified as the dominant AM fungi species which occurred frequently in the succession of saline-alkali land with the isolation frequency, relative abundance, and importance value of 100%, 18.1%, and 59.1%, respectively. The colonization rate of Glomeraceae sp1 in maize ranged from 80% to 87% and similar mycorrhizal characteristics were detected in the roots of P. tenuiflora, S. glauca, and L. chinensis, indicating that Glomeraceae sp1 colonized the samples in the field. The correlation matrix indicated that colonization rate, colonization intensity, and vesicle abundance were closely related to soil conditions most, and they were related significantly to all the soil properties except cellulase activity. Besides, redundancy analysis (RDA) showed that soil properties drove the changes of AM fungal colonization and sporulation. These results will provide theoretical support for realizing the relationship between AM fungal colonization and soil conditions, and also for the exploration of AM fungi species with great functions.

Pot culture experiments were established to determine the effects of colonization by arbuscular mycorrhizal fungi (AMF) (Glomus mosseae and G. sp) on maize (Zea mays L.) grown in Pb, Zn, and Cd complex contaminated soils. AMF and non-AMF inoculated maize were grown in sterilized substrates and subjected to different soil heavy metal (Pb, Zn, Cd) concentrations. The root and shoot biomasses of inoculated maize were significantly higher than those of non-inoculated maize. Pb, Zn, and Cd concentrations in roots were significantly higher than those in shoots in both the inoculated and non-inoculated maize, indicating the heavy metals mostly accumulated in the roots of maize. The translocation rates of Pb, Zn, and Cd from roots to shoots were not significantly difference between inoculated and non-inoculated maize. However, at high soil heavy metal concentrations, Pb, Zn, and Cd in the shoots and Pb in the roots of inoculated maize were significantly reduced by about 50% compared to the non-inoculated maize. These results indicated that AMF could promote maize growth and decrease the uptake of these heavy metals at higher soil concentrations, thus protecting their hosts from the toxicity of heavy metals in Pb, Zn, and Cd complex contaminated soils.  相似文献

The impact of different defoliation intensities on the ability of Lotus tenuis plants to regrowth, mobilise nutrients and to associate with native AM fungi and Rhizobium in a saline‐sodic soil was investigated. After 70 days, plants were subjected to 0, 25, 50, 75 and 100% defoliation and shoot regrowth was assessed at the end of subsequent 35 days. Compared to non‐defoliated plants, low or moderate defoliation up to 75% did not affect shoot regrowth. However, 100% treatment affected shoot regrowth and the clipped plants were not able to compensate the growth attained by non‐defoliated plants. Root growth was more affected by defoliation than shoot growth. P and N concentrations in shoots and roots increased with increasing defoliation while Na+ concentration in shoots of non‐defoliated and moderately defoliated plants was similar. Non‐defoliated and moderately defoliated plants prevented increases of Na+ concentration in shoots through both reducing Na+ uptake and Na+ transport to shoots by accumulating Na+ in roots. At high defoliation, the salinity tolerance mechanism is altered and Na+ concentration in shoots was higher than in roots. Reduction in the photosynthetic capacity induced by defoliation neither changed the root length colonised by AM fungi nor arbuscular colonisation but decreased the vesicular colonisation. Spore density did not change, but hyphal density and Rhizobium nodules increased with defoliation. The strategy of the AM symbiont consists in investing most of the C resources to preferentially retain arbuscular colonisation as well as inoculum density in the soil.  相似文献

Three arbuscular mycorrhizal (AM) fungi (Glomus mosseae, Glomus claroideum, and Glomus intraradices) were compared for their root colonizing ability and activity in the root of Astragalus sinicus L. under salt-stressed soil conditions. Mycorrhizal formation, activity of fungal succinate dehydrogenase, and alkaline phosphatase, as well as plant biomass, were evaluated after 7 weeks of plant growth. Increasing the concentration of NaCl in soil generally decreased the dry weight of shoots and roots. Inoculation with AM fungi significantly alleviated inhibitory effect of salt stress. G. intraradices was the most efficient AM fungus compared with the other two fungi in terms of root colonization and enzyme activity. Nested PCR revealed that in root system of plants inoculated with a mix of the three AM fungi and grown under salt stress, the majority of mycorrhizal root fragments were colonized by one or two AM fungi, and some roots were colonized by all the three. Compared to inoculation alone, the frequency of G. mosseae in roots increased in the presence of the other two fungal species and highest level of NaCl, suggesting a synergistic interaction between these fungi under salt stress.  相似文献

Two distinct populations of Arnica montana, an endangered medicinal plant, were studied under field conditions. The material was investigated using microscopic and molecular methods. The analyzed plants were always found to be mycorrhizal. Nineteen arbuscular mycorrhizal fungal DNA sequences were obtained from the roots. They were related to Glomus Group A, but most did not match any known species. Some showed a degree of similarity to fungi colonizing liverworts. Conventional analysis of spores isolated from soil samples allowed to identify different fungal taxa: Glomus macrocarpum, Glomus mosseae, Acaulospora lacunosa, and Scutellospora dipurpurescens.  The ability of arbuscular mycorrhizal (AM) fungi from a metal-tolerant plant (Viola calaminaria, violet) to colonise and reduce metal uptake by a non-tolerant plant (Trifolium subterraneum, subterranean clover) in comparison to a metal-tolerant AM fungus isolated from a non-tolerant plant was studied. AM spores from the violet rhizosphere and from violet roots were characterised by polymerase chain reaction (PCR) amplification of the SSU rDNA, and sequencing. Subterranean clover was grown in pots containing a soil supplemented with Cd and Zn salts and inoculated either with a mixture of spores extracted from the violet rhizosphere or with spores of a Cd-tolerant Glomus mosseae P2 (BEG 69), or non-inoculated. The diversity of fungi, including AM fungi, colonising clover roots was assessed and analysed using terminal-restriction fragment length polymorphism. At least four different Glomus species were found in the violet rhizosphere. After 8 weeks in a growth chamber, colonisation of clover roots with spores from the violet rhizosphere increased Cd and Zn concentrations in clover roots without significantly affecting the concentrations of metals in the shoot and plant growth. G. mosseae P2 reduced plant growth and slightly increased the Cd concentration. Only one AM fungus (Glomus b) from the violet rhizosphere colonised clover roots, but other fungi were present. AM fungi from heavy metal-contaminated soils and associated with metal-tolerant plants may be effective in accumulating heavy metals in roots in a non-toxic form. Accepted: 7 July 2000  相似文献

Maize roots were rapidly infected by soil inhabiting fungi as soon as they differentiated. The first tissues to develop were the seminal roots and mesocotyls. As their function became superseded by that of the adventitious roots, they rapidly became, completely rotted. Adventitious root rot developed slower and the roots did not become as severely rotted. Numerous fungi were isolated from the roots. The most frequently isolated were Helminthosporium pedicellatum and Fusarium moniliforme. Trichoderma sp. was the next most frequently isolated fungus; it was, however, significantly less frequently isolated than the above. Young and Kucharek (1977) found that the fungi isolated from maize roots occur in communities associated with certain plant growth stages. This was not clearly evident in this study. Numbers of root lesion nematodes (Pratylenchus species) correlated non-significantly with fungus frequencies and root rot.  相似文献

The seed germination of orchids under natural conditions requires association with mycorrhizal fungi. Dendrobium nobile and Dendrobium chrysanthum are threatened orchid species in China where they are considered medicinal plants. For conservation and application of Dendrobium using symbiosis technology, we isolated culturable endophytic and mycorrhizal fungi colonized in the protocorms and adult roots of two species plants and identified them by morphological and molecular analyses (5.8S and nrLSU). Of the 127 endophytic fungi isolated, 11 Rhizoctonia-like strains were identified as Tulasnellales (three strains from protocorms of D. nobile), Sebacinales (three strains from roots of D. nobile and two strains from protocorms of D. chrysanthum) and Cantharellales (three strains from roots of D. nobile), respectively. In addition, species of Xylaria, Fusarium, Trichoderma, Colletotrichum, Pestalotiopsis, and Phomopsis were the predominant non-mycorrhizal fungi isolated, and their probable ecological roles in the Dendrobium plants are discussed. These fungal resources will be of great importance for the large-scale cultivation of Dendrobium plants using symbiotic germination technology and for the screening of bioactive metabolites from them in the future.  相似文献

Diversity in phosphorus (P) acquisition strategies was assessed among eight isolates of arbuscular mycorrhizal fungi (AMF) belonging to three Glomus species, all obtained from the same field site. Maize (Zea mays L. cv. Corso) was used as a test plant. Compartmented cultivation containers coupled with 33P radioisotope labeling of soil P were employed to estimate (1) the distance from the roots that AMF were able to acquire soil P from, (2) the rate of soil colonization, (3) the efficiency of uptake of soil P by AMF, (4) benefits provided to maize in terms of P acquisition and growth. Glomus mosseae and G. intraradices took up P 10 cm from roots, whereas G. claroideum only up to 6 cm from the roots. G. mosseae most rapidly colonized the available soil volume and transported significant amounts of P to maize from a distance, but provided no net P uptake benefit to the plants. On the other hand, both G. intraradices and three out of four G. claroideum isolates significantly improved net P uptake by maize. These effects seem to be related to variability between and to a limited extent also within AMF species, in mycelium development, efficiency of hyphal P uptake and effects on plant P acquisition via the root pathway. In spite of absence of maize growth responses to inoculation with any of the AMF isolates, this study indicates remarkable functional diversity in the underground component of the studied field site.  相似文献

为了解喀斯特典型物种-小蓬竹根际土壤微生物及不同部位内生真菌多样性,采用沿等高线等距离取样法采集小蓬竹根际土壤及健康植株,通过可培养对根际土微生物及内生菌进行分离,利用分子技术对其进行鉴定,根据鉴定结果构建系统发育树,并计算小蓬竹根际土壤微生物和根茎叶内生真菌多样性。结果如下:(1)共从根际土壤、根、茎、叶分离得到139个真菌菌株,隶属于27属,其中根际土壤分离得到34个真菌菌株隶属于12属,根部分离得到的63个内生真菌菌株隶属于17个属,茎部分离得到的14个内生真菌菌株隶属于8个属,叶部分离得到28个内生真菌菌株隶属于9个属;(2)根际土壤共分离得到41株细菌菌株,隶属于7个属26个种,20株放线菌菌株,隶属于1属15种;从Shannon-Wiener多样性指数、均匀度指数、Simpson指数排序来看,真菌主要表现为根 > 根际土壤 > 茎 > 叶,细菌和放线菌多样性均较低。(3)按层次聚类分析可分别将真菌、细菌、放线菌聚为3支。小蓬竹根际土壤、根、茎和叶具有丰富的微生物多样性,不同部位菌群组成存在差异性(P<0.05),且存在以假单胞菌属、芽孢杆菌属等为优势属的抗盐耐旱菌群,这有助于揭示小蓬竹对喀斯特生境的适应性,以及为微生物-植物群落之间相互关系提供一定基础数据,为后期寻找小蓬竹相关耐性功能菌奠定基础。  相似文献

Effects of arbuscular mycorrhizal fungus (Glomus mosseae) on the accumulation and speciation of selenium (Se) in alfalfa, maize, and soybean were investigated by using Se(IV)-spiked soil. Mycorrhizal inoculation decreased Se accumulation in roots and shoots of all the plants at Se spiked level of 0 or 2 mg kg−1, while an increased Se accumulation was observed in alfalfa shoots and maize roots and shoots at the spiked level of 20 mg kg−1. Concentration of inorganic Se (especially Se(VI)) in roots and shoots of the three plants was much higher in mycorrhizal than non-mycorrhizal treatment. Mycorrhizal inoculation decreased the portion of total organic Se in plant tissues with the exception of alfalfa and maize shoots at Se spiked level of 20 mg kg−1, in which organic Se portion did not reduced greatly (<5%) for mycorrhizal treatment. Mycorrhizal effects on alfalfa and maize were more obvious than on soybean in terms of root colonization rate, biomass, and Se accumulation.  相似文献
