A greenhouse experiment was conducted to examine the changes in antioxidant enzyme activities of arbuscular mycorrhizal (AM) fungus Glomus intraradices Schenck and Smith inoculated (M+) and non-inoculated (M−) maize (Zea mays L.) plants (variety COHM5) under varying levels of zinc (0, 1.25, 2.5, 3.75 and 5.0 mg kg−1). Roots and shoots sampled at 45 days after sowing (DAS) were estimated for its antioxidant enzymes (superoxide dismutase, peroxidase) IAA oxidase, polyphenol oxidase, acid phosphatase and nutritional status especially P and Zn concentrations. Mycorrhizal inoculation significantly (P ≤ 0.01) increased all the four antioxidant enzymes in both roots and shoots at 45 DAS regardless of Zn levels. All enzyme activities except SOD increased progressively with increasing levels of Zn under M+ and M− conditions. The SOD activity got decreased in roots and shoots at 2.5 and 3.75 mg Zn kg−1. Acid phosphatase activity in M+ roots and shoots were higher in all levels of Zn but the values decreased with increasing levels of Zn particularly in roots. Mycorrhizal fungus inoculated plants had higher P and Zn concentrations in both stages in comparison to non-inoculated plants. Our overall data suggest that mycorrhizal symbiosis plays a vital role in enhancing activities of antioxidant enzymes and nutritional status that enables the host plant to sustain zinc deficient conditions.  相似文献

Effects of inoculation with the arbuscular mycorrhizal (AM) fungus Glomus mosseae on the behavior of Hg in soil–plant system were investigated using an artificially contaminated soil at the concentrations of 0, 1.0, 2.0, and 4.0 mg Hg kg−1. Mercury accumulation was lower in mycorrhizal roots than in nonmycorrhizal roots when Hg was added at the rates of 2.0 and 4.0 mg kg−1, while no obvious difference in shoot Hg concentration was found between mycorrhizal and nonmycorrhizal treatments. Mycorrhizal inoculation significantly decreased the total and extractable Hg concentrations in soil as well as the ratio of extractable to total Hg in soil. Equilibration sorption of Hg by soil was investigated, and the results indicated that mycorrhizal treatment enhanced Hg sorption on soil. The uptake of Hg was lower by mycorrhizal roots than by nonmycorrhizal roots. These experiments provide further evidence for the role of mycorrhizal inoculation in increasing immobilization of Hg in soil and reducing the uptake of Hg by roots. Calculation on mass balance of Hg in soil suggests the presence of Hg loss from soil presumably through evaporation, and AM inoculation enhanced Hg evaporation. This was evidenced by a chamber study to detect the Hg evaporated from soil.  相似文献

Effects of selenium (Se) on growth and some physiological traits of roots in wheat (Triticum aestivum L. cv Han NO.7086) seedlings exposed to enhanced ultraviolet-B (UV-B) stress are reported. Responses of roots were different depending on the Se concentration. Compared with the control, root weight of wheat seedlings treated with 1.0 and 2.0 mg Se kg−1 soil increased by 39.47% and 16.28%, respectively. The lower amount Se (0.5 mg kg−1) and the higher amount Se treatments (3.0 mg kg−1) did not significantly affect on root weight. Se treatments significantly increased root activity, flavonoids and proline content, and activities of peroxidase and superoxide dimutase in wheat roots exposed to enhanced UV-B. In addition, the treatments with 0.5, 1.0, and 2.0 mg Se kg−1 significantly reduced malondialdehyde content and the rate of superoxide radical (O2) production of roots, whereas the higher amount Se treatment only induced a decrease in the rate of O2 production. The results of this study demonstrated that optimal Se supply promoted roots growth of wheat seedlings, and that optimal Se supply could reduce oxidative stress in wheat roots under enhanced UV-B radiation.  相似文献

Responses of Wheat Seedlings to Exogenous Selenium Supply Under Cold Stress   总被引:2,自引:0,他引:2
Dose-dependent effects of selenium on growth and physiological trait of wheat seedlings (Triticum aestivum L. cv Han NO.7086) exposed to cold stress are reported. Responses of seedlings were different depending on the Se concentration. The treatments with 0.5 and 1.0 mg Se kg−1 significantly increased biomass and chlorophyll content of seedlings. However, the treatments at 2.0 and 3.0 mg Se kg−1 only induced an evident increase in chlorophyll content and did not promote biomass accumulation of seedlings. Antioxidant compounds content (anthocyanins, flavonoids, and phenolic compounds) and antioxidant enzymes’ activities (peroxidase and catalase) increased by different Se treatments, while only the treatment with 1.0 mg Se kg−1 induced a significant reduce in malondialdehyde content and the rate of superoxide radical production of wheat seedlings. The results of this study demonstrated that Se supply could increase antioxidant capacity of seedlings, and optimal Se supply reduced production of free radicals, membrane lipid peroxidation, and promoted biomass accumulation.  相似文献

An efficient micropropagation system for mining ecotype Sedum alfredii Hance, a newly identified Zn/Cd hyperaccumulator, was developed. Frequency of callus induction reached up to 70% from leaves incubated on Murashige and Skoog (MS) medium supplemented with 1.0 mg l−1 2,4-dichlorophenoxy acetic acid (2,4-D) and 0.5 mg l−1 6-benzyladenine (BA), and 83% from internodal stem segments grown on MS medium with 0.1 mg l−1 2,4-D and 0.1 mg l−1 BA. Callus proliferated rapidly on MS medium containing 0.2 mg l−1 2,4-D and 0.05 mg l−1 thidiazuron. The highest number of adventitious buds per callus (17.3) and frequency of shoot regeneration (93%) were obtained when calli were grown on MS medium supplemented with 2.0 mg l−1 BA and 0.3 mg l−1 α-naphthalene acetic acid (NAA). Elongation of shoots was achieved when these were incubated on MS medium containing 3.0 mg l−1 gibberellic acid. Induction of roots was highest (21.4 roots per shoot) when shoots were transferred to MS medium containing 2.0 mg l−1 indole 3-butyric acid rather than either indole 3-acetic acid or NAA. When these in vitro plants were acclimatized and transferred to the greenhouse, and grown in hydroponic solutions containing 200 μM cadmium (Cd), they exhibited high efficiency of Cd transport, from roots to shoots, and hyperaccumulation of Cd.  相似文献

In order to characterize uptake of vanadium in alfalfa grown in vanadium–cadmium (Cd)-contaminated soil, 104 soil samples and 94 plant samples were collected from pot experiment. The results showed alfalfa had strong metal adaptability (up to 400 mg kg−1) and high accumulation (up to 3,440.14 mg kg−1) of vanadium. Root had higher contents and better absorption to vanadium than overground part. Moreover, both root and overground part had direct correlation with vanadium in soil, especially with the sum of first three fractions and reducible fraction. With the increasing of vanadium, higher concentration of Cd may inhibit the absorption of vanadium in alfalfa.  相似文献

A pot experiment was conducted to investigate the uptake of Zn from experimentally contaminated calcareous soil of low nutrient status by maize inoculated with the arbuscular mycorrhizal (AM) fungus Glomus caledonium. EDTA was applied to the soil to mobilize Zn and thus maximize plant Zn uptake. The highest plant dry matter (DM) yields were obtained with a moderate Zn addition level of 300 mg kg?1. Plant growth was enhanced by mycorrhizal colonization when no Zn was added and under the highest Zn addition level of 600 mg kg?1, while application of EDTA to the soil generally inhibited plant growth. EDTA application also increased plant Zn concentration, and Zn accumulation in the roots increased with increasing EDTA addition level. The effects of inoculation with Gcaledonium on plant Zn uptake varied with Zn addition level. When no Zn was added, Zn translocation from roots to shoots was enhanced by mycorrhizal colonization. In contrast, when Zn was added to the soil, mycorrhizal colonization resulted in lower shoot Zn concentrations in mycorrhizal plants. The P nutrition of the maize was greatly affected by AM inoculation, with mycorrhizal plants showing higher P concentrations and P uptake. The results indicate that application of EDTA mobilized soil Zn, leading to increased Zn accumulation by the roots and subsequent plant toxicity and growth inhibition. Mycorrhizal colonization alleviated both Zn deficiency and Zn contamination, and also increased host plant growth by influencing mineral nutrition. However, neither EDTA application nor arbuscular mycorrhiza stimulated Zn translocation from roots to shoots or metal phytoextraction under the experimental conditions. The results are discussed in relation to the environmental risk associated with chelate-enhanced phytoextraction and the potential role of arbuscular mycorrhiza in soil remediation.  相似文献

Wild barley (Hordeum spontaneum), the progenitor of cultivated barley, is an important genetic resource for cereal improvement. Selenium (Se) is an essential trace mineral for humans and animals with antioxidant, anticancer, antiarthropathy, and antiviral effects. In the current study, the grain Se concentration (GSeC) of 92 H. spontaneum genotypes collected from nine populations representing different habitats in Israel was investigated in the central area of Guizhou Province, China. Remarkable variations in GSeC were found between and within populations, ranging from 0 to 0.387 mg kg−1 among the 92 genotypes with an average of 0.047 mg kg−1. Genotype 20_C from the Sede Boqer population had the highest GSeC, while genotype 25_1 from the Atlit population had the lowest. The mean value of GSeC in each population varied from 0.010 to 0.105 mg kg−1. The coefficient of variation for each population ranged from 12% to 163%. Significant correlations were found between GSeC and 12 ecogeographical factors out of 14 studied. Habitat soil type also significantly affected GSeC. The wild barley exhibited wider GSeC ranges and greater diversity than its cultivated counterparts. The higher Se grain concentrations found in H. spontaneum populations suggest that wild barley germplasm confer higher abilities for Se uptake and accumulation, which can be used for genetic studies of barley nutritional value and for further improvement of domesticated cereals.  相似文献

The paper reports the effects of selenium (Se) supply on growth and antioxidant traits of wheat (Triticum aestivum L. cv Han NO.7086) seedlings exposed to enhanced ultraviolet-B (UV-B) stress. Antioxidant responses of seedlings were different depending on the Se concentration. Compared with the control, the lower amount used (0.5 mg Se kg−1 soil) had no significant effect on biomass accumulation. The treatments with 1.0, 2.0, and 3.0 mg Se kg−1 promoted biomass accumulation of wheat seedlings, and the increased amount in biomass was the most at 1.0 mg Se kg−1 treatment. Se treatments with 1.0, 2.0, and 3.0 mg kg−1 also significantly increased activities of peroxidase (POD) and superoxide dismutase (SOD) and reduced the rate of superoxide radical (O2) production and malondialdehyde (MDA) content of wheat seedlings. In addition, anthocyanins and phenolic compounds content in wheat seedlings evidently increased by the treatments with 1.0 and 2.0 mg Se kg−1. The lower Se treatment had no significant effect on MDA content, although it increased activities of antioxidant enzymes (POD, SOD, and catalase activities) and reduced the rate of O2 production in wheat seedlings. These results suggest that optimal Se supply is favorable for the growth of wheat seedlings and that optimal Se supply can reduce oxidative stress of seedlings under enhanced UV-B radiation.  相似文献

Silicate (Si) can enhance plant resistance or tolerance to the toxicity of heavy metals. However, it remains unclear whether Si can ameliorate lead (Pb) toxicity in banana (Musa xparadisiaca) roots. In this study, treatment with 800 mg kg−1 Pb decreased both the shoot and root weight of banana seedlings. The amendment of 800 mg kg−1 Si (sodium metasilicate, Na2SiO3·9H2O) to the Pb-contaminated soil enhanced banana biomass at two growth stages significantly. The amendment of 800 mg kg−1 Si significantly increased soil pH and decreased exchangeable Pb, thus reducing soil Pb availability, while Si addition of 100 mg kg−1 did not influence soil pH. Results from Pb fractionation analysis indicated that more Pb were in the form of carbonate and residual-bound fractions in the Si-amended Pb-contaminated soils. The ratio of Pb-bound carbonate to the total Pb tended to increase with increasing growth stages. Treatment with 100 mg kg−1 Si had smaller effects on Pb forms in the Si-amended soils than that of 800 mg kg−1 Si. Pb treatment decreased the xylem sap greatly, but the addition of Si at both levels increased xylem sap and reduced Pb concentration in xylem sap significantly in the Si-amended Pb treatments. The addition of Si increased the activities of POD, SOD, and CAT in banana roots by 14.2% to 72.1% in the Si-amended Pb treatments. The results suggested that Si-enhanced tolerance to Pb toxicity in banana seedlings was associated with Pb immobilization in the soils, the decrease of Pb transport from roots to shoots, and Si-mediated detoxification of Pb in the plants.  相似文献

Sustainability of soil-plant systems requires, among other things, good development and function of mycorrhizal symbioses. The effects of P and micronutrient levels on development of an arbuscular mycorrhizal fungus (AMF) and uptake of Zn, Cu, Mn and Fe by maize (Zea mays L.) were studied. A pot experiment with maize either inoculated or not with Glomus intraradices was conducted in a sand:soil (3 :1) mix (pH 6.5) in a greenhouse. Our goal was to evaluate the contribution of mycorrhizae to uptake of Cu, Zn, Mn and Fe by maize as influenced by soil P and micronutrient levels. Two levels of P (10 and 40 mg kg−1 soil) and three levels of a micronutrient mixture: 0, 1X and 2X (1X contained, in mg kg−1 soil, 4.2 Fe, 1.2 Mn, 0.24 Zn, 0.06 Cu, 0.78 B and 0.036 Mo), were applied to pots. There were more extraradical hyphae at the low P level than at the high P level when no micronutrients were added to the soil. Root inoculation with mycorrhiza and application of micronutrients increased shoot biomass. Total Zn content in shoots was higher in mycorrhizal than non-mycorrhizal plants grown in soils with low P and low or no micronutrient addition. Total Cu content in shoots was increased by mycorrhizal colonization when no micronutrients were added. Mycorrhizal plants had lower Mn contents than non-mycorrhizal plants only at the highest soil micronutrient level. AMF increased total shoot Fe content when no micronutrients were added, but decreased shoot Fe when plants were grown at the high level of micronutrient addition. The effects of G. intraradices on Zn, Cu, Mn, and Fe uptake varied with micronutrient and P levels added to soil. Accepted: 27 December 1999  相似文献

Some plants can tolerate and even detoxify soils contaminated with heavy metals. This detoxification ability may depend on what chemical forms of metals are taken up by plants and how the plants distribute the toxins in their tissues. This, in turn, may have an important impact on phytoremediation. We investigated the impact of arbuscular mycorrhizal (AM) fungus, Glomus intraradices, on the subcellular distribution and chemical forms of cadmium (Cd) in alfalfa (Medicago sativa L.) that were grown in Cd-added soils. The fungus significantly colonized alfalfa roots by day 25 after planting. Colonization of alfalfa by G. intraradices in soils contaminated with Cd ranged from 17% to 69% after 25–60 days and then decreased to 43%. The biomass of plant shoots with AM fungi showed significant 1.7-fold increases compared to no AM fungi addition under the treatment of 20 mg·kg−1 Cd. Concentrations of Cd in the shoots of alfalfa under 0.5, 5, and 20 mg·kg−1 Cd without AM fungal inoculation are 1.87, 2.92, and 2.38 times higher, respectively, than those of fungi-inoculated plants. Fungal inoculation increased Cd (37.2–80.5%) in the cell walls of roots and shoots and decreased in membranes after 80 days of incubation compared to untreated plants. The proportion of the inactive forms of Cd in roots was higher in fungi-treated plants than in controls. Furthermore, although fungi-treated plants had less overall Cd in subcellular fragments in shoots, they had more inactive Cd in shoots than did control plants. These results provide a basis for further research on plant-microbe symbioses in soils contaminated with heavy metals, which may potentially help us develop management regimes for phytoremediation.  相似文献

Dorema ammoniacum D. Don. (Apiaceae), a native medicinal plant in Iran, is classified as a vulnerable species. Root, hypocotyl, and cotyledon segments were cultured on Murashige and Skoog (MS) (1962) medium supplemented with either 2,4-dichlorophenyoxyacetic acid (2,4-D) or naphathalene acetic acid (NAA), at 0–2 mg l−1, alone or in combination with either benzyladenine (BA) or kinetin (KN), at 0–2 mg l−1 for callus induction. The best response (100%) was observed from root segments on MS medium containing 1 mg l−1 NAA and 2 mg l−1 BA. The calli derived from various explants were subcultured on MS medium supplemented with BA (1–4 mg l−1) alone or in combination with NAA or indole-3-butyric acid (IBA), at 0.2 or 0.5 mg l−1 for shoot induction. Calli derived from hypocotyl segments showed significantly higher frequency of plantlet regeneration and number of plantlets than the calli derived from root and cotyledon segments. Therefore, MS medium supplemented with 2 mg l−1 BA and 0.2 mg l−1 IBA produced the highest frequency of shoot regeneration (87.3%) in hypocotyl-derived callus. The optimal medium for rooting contained 2.5 mg l−1 IBA on which 87.03% of the regenerated shoots developed roots with an average number of 5.2 roots per shoots within 30 days. These plantlets were hardened and transferred to the soil. The described method can be successfully employed for the large-scale multiplication and conservation of germplasm this plant.  相似文献

Morinda citrifolia adventitious roots were cultured in shake flasks using Murashige and Skoog medium with different types and concentrations of auxin and cytokinin. Root (fresh weight and dry weight) accumulation was enhanced at 5 mg l−1 indole butyric acid (IBA) and at 7 and 9 mg l−1 naphthalene acetic acid (NAA). On the other hand, 9 mg l−1 NAA decreased the anthraquinone, phenolic and flavonoid contents more severely than 9 mg l−1 IBA. When adventitious roots were treated with kinetin (0.1, 0.3 and 0.5 mg l−1) and thidiazuron (TDZ; 0.1, 0.3 and 0.5 mg l−1) in combination with 5 mg l−1 IBA, fresh weight and dry weight decreased but secondary metabolite content increased. The secondary metabolite content (including 1,1-diphenyl-2-picrylhydrazyl activity) increased more in TDZ-treated than in kinetin-treated roots. Antioxidative enzymes such as catalase (CAT) and guaiacol peroxidase (G-POD), which play important roles in plant defense, also increased. A strong decrease in ascorbate peroxidase activity resulted in a high accumulation of hydrogen peroxide. This indicates that adventitious roots can grow under stress conditions with induced CAT and G-POD activities and higher accumulations of secondary metabolites. These results suggest that 5 mg l−1 IBA supplementation is useful for growth and secondary metabolite production in adventitious roots of M. citrifolia.  相似文献

This greenhouse study aimed to examine the contribution of arbuscular mycorrhizal (AM) colonization on the uptake of and tolerance to nickel (Ni) in sunflower (Helianthus annuus L.). We hypothesized that AM colonization increases Ni content and tolerance in sunflower grown under varying soil Ni concentrations. The combined effect of AM colonization and soil Ni input on the assimilation of nitrogen, in particular the activity of glutamine synthetase (GS), in sunflower plants was also investigated. A factorial experimental design was performed with sunflower cv. Lemon Queen, with or without the AM fungus, Glomus intraradices Schenck & Smith, and treated with 0, 100, 200, or 400 mg Ni kg−1 dry soil (DS). The AM colonization significantly enhanced plant growth and Ni content, especially at the lower soil Ni treatments. Furthermore, the AM plants exposed to the highest soil Ni level of 400 mg Ni kg−1 DS had a significantly higher shoot Ni extracted percentage than non-AM plants, suggesting that the AM symbiosis contributed to Ni uptake, then its translocation from roots to shoots. The AM colonization also significantly increased the GS activity in roots, this being likely an indicator of an enhanced Ni tolerance. These findings support the hypothesis that AM symbiosis contributes to an enhanced Ni plant uptake and tolerance and should be considered as part of phytoremediation strategies.  相似文献

The objective of the present work was selection of cultivar and suitable medium for regenerating shoots from leaf segments of non-heading Chinese cabbage. We evaluated six types of supplemented media with 2.0, 5.0 and 10.0 mg l−1 6-BA; 1.0 and 2.0 mg l−1 TDZ; 0.1, 0.3, 0.5, 0.8 and 1.0 mg l−1NAA; 3.0, 5.0 and 7.5 mg l−1AgNO3; 0.01 mg l−1 2–4, D and 4.0 mg l−1 KT for shoot regeneration and six cultivars “Sanchidaye”, “Liuchuandasuomian”, “Qingyou 4”, “Liangbaiye”, “AiKang 5” and “Hanxiao F3”, furthermore for root formation three types of supplemented media with 0.2, 0.3, 0.5 mg l−1 NAA, and for survival rate two types of base media: turf + vermiculite + manure (1:2:0.2) and soil + vermiculite (1:2). Culturing leaf segments on MS medium supplemented with 2 mg l−1 TDZ; 0.5 mg l−1 NAA and 7.5 mg l−1 AgNO3 gave the highest number of shoots per leaf segment (66) while roots were best formed on the medium supplemented with 0.2 mg l−1 NAA. Survival rate was highest (61.6%) in the turf: vermiculite: manure (1:2:0.2) medium. The highest percentage of responding leaf segments, number of shoots per leaf segment, rooting percentage and survival rate were observed in “Liuchuandasuomian”. The plantlets were transferred to the soil and grown into mature plants in pots. These results could be used for preliminary selections of cultivars to transfer disease resistance (Bt) gene through agrobacterium in non-heading Chinese cabbage.  相似文献

This study demonstrates the morphogenic potential of pulvinus, an important organ situated at the base of the petiole or rachis of leguminous plants. Plant regeneration via pulvinus-derived calli of Caesalpinia bonduc has been achieved. Organogenic calli have been derived from the explant 45 days after culture on Murashige and Skoog (MS) medium supplemented with 2,4-dichlorophenoxyacetic acid (2,4-D) alone or in combination with 6-benzylaminopurine (BA). Optimum callus induction (100%) occurred when the pulvini were cultured on MS medium fortified with 6 mg l−1 2,4-D and 1 mg l−1 BA. The highest shoot induction was obtained when the calli were transferred to MS medium supplemented with 5 mg l−1 BA and 1 mg l−1 indole-3-acetic acid (IAA). On this medium, 87% cultures responded with an average number of 4.2 shoots per culture. The maximum root induction from the regenerated shoots was observed on half strength MS medium containing 6 mg l−1 indole-3-butyric acid (IBA). Here 100% shoots rooted with a mean number of 6.3 roots per shoot. The regenerated plantlets were acclimatized and subsequently showed normal growth. This efficient protocol will be helpful for propagating elite clones on a mass scale and could be utilized for genetic transformation study.  相似文献

The present study describes the potential of in vitro grown adventitious roots of Hypericum perforatum L. commonly known as St. John’s wort at low nutrient and auxin levels in the liquid medium for micropropagation. Roots were regenerated from shoot-derived callus on MS medium containing 4.0 mg l−1 Indole-3 acetic acid (IAA). IAA and Indole-3 butyric acid (IBA) were equally effective for the induction of roots from shoot cultures. Half strength MS medium containing 1.0 mg l−1 IAA was most found suitable for culturing roots in liquid medium. A total biomass of 4.13 ± 0.67 g comprising 226 ± 34.4 shoots and shoot buds along with roots was obtained per culture starting with 200 mg roots inoculum. Pretreatment with kinetin (2.0 mg l−1) enhanced the shoot multiplication. Shoots proliferated profusely from excised roots in static liquid medium supported with glass bead matrix. Growtek vessel was found suitable and cost effective system for high throughput plantlet production. In vitro grown roots regardless of their source of origin were an excellent and easy to handle source of explant for aseptic production of plantlets without loosing the morphogenetic potential over the generations. The plants exhibited 84–99% similarity among themselves through RAPD. The in vitro shoots produced can either be multiplied or rooted perpetually, and alternatively they can also be explored for the in vitro production of hypericin and hyperforin.  相似文献

We have developed a system for the in vitro regeneration of pasqueflowers (Pulsatilla koreana Nakai). The system was based on somatic embryogenesis and shoot organogenesis. Over a growth period of 6 weeks, multiple shoots were initiated from leaf, petiole, and pedicel explants on Murashige and Skoog (MS) medium containing 0.5 mg l−1 indole-3-acetic acid (IAA) and zeatin (Zn), kinetin (Kin), or 6-benzyladenine (BA). We achieved 100% of adventitious shoot induced when petiole and pedicel explants were cultured on MS, 0.5–2.0 mg l−1 Zn, and 0.5 mg l−1 IAA. Somatic embryos developed from the explants and generated shoots on MS medium containing 0.25 mg l−1 Zn and 0.5 mg l−1 IAA. Globular and heart-shaped stages of somatic embryos were observed. Histological studies have revealed the stages of development of somatic embryos. For propagation and growth, the regenerated shoots from organogenic or embryogenic calluses were transferred to MS medium containing either (1) 1.5 mg l−1 Zn and 0.05 mg l−1 IAA or (2) 1.0 mg l−1 BA and 0.05 mg l−1 IAA. After the length of the shoots reached 3 cm, the shoots initiated by organogenesis as well as those initiated by somatic embryogenesis were transferred to the root induction medium. After 2 months of culture in half-strength MS with 1.5 mg l−1 α-naphthalene acetic acid (NAA), the rooting ratio was 93%. Finally, the rooted plantlets were acclimatized in a mixture of mountain soil and perlite.  相似文献
