Endogenous adventitious buds develop in situ from dedifferentiated parenchyma cells of the trunk-bark in the temperate trees Tilia platyphyllos, Acer pseudoplatanus, and Fraxinus excelsior, as well as in the tropical cauliflorous trees Artocarpus integrifolia, Swartzia schomburgkii, and Couroupita guianensis. On aerial roots of Clusia rosea endogenous adventitious buds originate within the proliferated phelloderm beneath lenticels. In Salix alba, Fraxinus excelsior, and Terminalia arjuna, exogenous dormant buds are overgrown during secondary growth and engulfed within the bark tissue, so that they give the impression of apparent endogeny. In the leaf axils of young shoots of Araucaria angustifolia, superficial tissue layers divide and form axillary protrusions, which soon become parenchymatic and partly suberized. A few cells at their bases stay meristematic, however, and develop as rudimentary endogenous bud primordia, which persist in the bark for many years.  相似文献

Primordial shoot explants excised from buds of one Larix decidua tree, about 30 years old, produced more adventitious buds, elongating into shoots, when grown on half strength Litvay medium than when grown on other basal media. Thidiazuron and N6-benzyladenine (BA) were equally effective in adventitious bud induction. In a comparative study of 30-year-old L. decidua, L. leptolepis, L. eurolepis, and L. laricina trees, explants from L. eurolepis and L. decidua produced a high number of cultures with adventitious buds that elongated into shoots; those from L. leptolepis were less productive, and those from L. laricina failed to form adventitious buds. The highest response was obtained with material collected in August and September, and in March and April; the lowest response occurred in explants from the October collection.  相似文献

The effects of fruit on bud sprouting and vegetative growth were compared on fruiting and defruited loquat trees from fruit set onward. Carbohydrate and nitrogen content in leaves and bark tissues and hormone concentrations were studied during the fruit development and vegetative growth periods. On defruited trees, a significant proportion of buds sprouted in winter, whereas buds from fruiting trees sprouted only in the spring when fruit reached its final size. Furthermore, when panicles were completely removed in autumn, the buds also sprouted. In addition, fruit directly affected vegetative growth by reducing shoot length. An effect of sink removal (flower or fruit) promoting bud sprouting, regardless of the season, was then demonstrated. Neither soluble sugar concentration nor nitrogen fraction concentration in leaves or bark tissues was related to bud sprouting, but a certain nutritional imbalance was observed during the most active period of fruit development. Moreover, fruit sink activity significantly modified hormone content by increasing indole-3-acetic acid (IAA) and reducing zeatin concentrations, resulting in a higher IAA/zeatin ratio parallel to the lower bud sprouting intensity. Therefore, these changes caused by fruit removal are all related to vegetative growth, but there is no evidence that they are responsible for bud burst.  相似文献

Resting vegetative buds of Norway spruce, Picea abies (L.) Karst., were induced to form adventitious bud primordia when cultured on medium -containing cytokinin. After transfer of the induced buds to medium lacking cytokinin, adventitious buds developed. The adventitious buds arose from meristems formed de novo in the needle primordia. No differences were found in the ability to form adventitious buds among buds collected from trees ranging from 5–50 years old.  相似文献

A method was developed for the establishment of shoot cultures from Douglas-fir trees selected for outstanding growth and form in a 12-year-old genetic test. Vegetative buds from the lower crown were sterilized and grafted in vitro onto juvenile clonal rootstock. The rootstocks were produced from adventitious buds induced on cotyledons, and were maintained through micropropagation. Buds that established grafts slowly elongated into shoots, which were harvested and multiplied through micropropagation. Grafts often grew several new shoots which in turn could be harvested. In 1987, 2830 buds were grafted from 18 superior trees. Twenty nine grafts (1%) produced shoots which established 11 of the 18 trees in culture. Their appearance and behavior in vitro became more juvenile over 1–3 years, as indicated by shoot and needle morphology, disappearance of episodic growth pattern, increase in multiplication rates, and ability of needles to produce adventitious buds.The five most prolific of the 11 clones were given a pre-rooting treatment and planted in soil under fog. The success of rooting and subsequent establishment in soil varied from 5 to 17% depending on clone. In contrast, trees multiplied in vitro for 1–2 years longer showed soil establishment rates from 8–60%. This technique allows establishment, multiplication, and maintenance in vitro of cultures from high value Douglas-fir genotypes. Such cultures may serve as a starting point for further research on rejuvenation and cloning.  相似文献

研究不同生态环境条件下胡杨和灰叶胡杨横走侧根的空间分布及其形态特征、不定芽及克隆分株的数量格局特征及其与土壤水分的关系。研究结果表明:(1)16团3个生境中灰叶胡杨克隆分株所在横走侧根均分布在10—20cm的土层;阿瓦提混交林内灰叶胡杨和胡杨克隆分株所在横走侧根分布在20—40cm的土层;轮台县河道边胡杨克隆分株所在横走侧根分布在10—20cm的土层,林内胡杨克隆分株所在横走侧根分布在20—40cm的土层。不定芽、克隆分株所在部位横走侧根最粗,向两端逐渐变细,且远端根始终比近端根要粗;(2)不定芽具有以前期克隆分株为中心向两端延伸发生分布的空间格局特征,横走侧根上不定芽数与出土克隆分株数呈极显著正相关,未出土克隆分株数与出土克隆分株数呈显著正相关,说明不定芽、未出土克隆分株的数量都直接影响出土克隆分株的数量。(3)不定芽、未出土克隆分株和出土克隆分株出现频率、空间分布范围在同一生境同一根段均表现为不定芽未出土克隆分株出土克隆分株,在不同研究区表现为16团(地下水位1.08m)轮台县(地下水位2.56m)阿瓦提县(地下水位3.34m);相关分析显示土壤水分含量与不定芽和出土克隆分株数量均呈极显著正相关。综合分析认为,地下水位和土壤含水量均对不定芽发生以及不定芽向克隆分株转化有显著的影响。  相似文献

Summary Explants derived from adventitious buds, rhizomes, stems, and leaves of a medicinal plant, Polygonatum cyrtonema, were studied for plantlet regeneration, and only adventitious bud explants were able to be regenerated into plantlets. Regeneration was also accompanied by the formation of rhizome-like tissue, the medicinal portion of the plant. The optimum hormone combination for plantlet regenertion was 4.44 μM benzyladenine plus 2.26 μM 2,4-dichlorophenoxyacetic acid, at which new adventitious buds were obtained from 96.6% of the adventitious bud explants, with an average of 5.2 buds per explant. The best medium for root induction was half-strength Murashige and Skoog medium with 4.57 μM α-naphthaleneacetic acid, as 92% of regenerated buds rooted. Regenerated plantlets were successfully transferred to a greenhouse with 86% survival. Histological observation indicated that new adventitious buds originated from the superficial meristematic cell cluster of the granular callus induced from adventitious bud explants via organogenesis.  相似文献

Pinus contorta Douglas ex Loudon var. yukonensis W. L Strong (Yukon pine), a new variety, is described from south–central Yukon, Canada. It is a stout tree with long lower branches, often with paired lower and forked upper stems, and weakly platy to smooth gray bark on the lower stem, when < 70 years old; with bark on the lower stem becoming furrowed and platy on older trees. This contrasts with the narrow columnar habit, simple stem, and scaly bark associated with the more common inland var. latifolia. In several respects, var. yukonensis is more similar to var. contorta (a coastal variant), although occupying a different ecological regime and spatially disjunct (i.e., a high latitude and high elevation variant). Variety yukonensis may represent the taxon alluded to in previous leaf oil terpene and genetic studies, and a possible Beringian survivor.  相似文献

Spatial and temporal aspects of overwintering in the holocyclic sycamore aphid Drepanosiphum platanoidis (Schrank) (Homoptera: Callaphididae) were investigated. Eggs of D. platanoidis were principally laid on trunks and branches of Acer pseudoplatanusL., at considerable distances from the buds. Trunks with rough bark, followed by branches with moderate roughness supported more overwintering eggs than the relatively exposed smooth surfaces of twigs and terminal buds. Eggs were aggregated on rough bark. Aspect had no effect on the number of eggs laid. Experimental manipulations varying ovipara densities showed intra-specific competition among oviparae for optimal oviposition sites. Overwintering egg mortality was a function of time, with the greatest mortality rate occurring in late winter. Egg mortality varied from 80.4% to 76.9% on trunks and 83.0% to 65.9% on branches in 1996 and 1997.  相似文献

Resting vegetative buds of Picea abies collected from phytotron-grown rooted cuttings of 24-year-old trees or a 12-year-old hedge were tested for their capacity to form adventitious buds after various cytokinin treatments. The most effective method for obtaining a high yield of adventitious buds within 8 weeks was to pulse treat the buds in 250 M BA for 3 h and then culture them on medium containing 5 M each of BA and kinetin for 1 week. The developmental pattern for adventitious bud production, with the formation of 10 to 20 adventitious buds per bud, was similar for all tested genotypes, although the number of buds giving rise to adventitious buds varied significantly. The capability of some clones to form adventitious buds was correlated to endogenous cytokinin content. The clone which contained most endogenous cytokinin in its resting bud had the highest potential for adventitious bud formation.  相似文献

Fast-growing willows are cultivated as coppice in short rotation biomass plantations. The production and sustainability of the system is based on the ability of trees to resprout after repeated harvesting. The large variation in coppicing ability is due to plant genotypic differences in structure and physiology as well as environmental factors. Morphological and structural prerequisites for resprouting were compared in two shrubby willows with high coppicing ability, S. viminalis and S. eriocephala, and one tree-formed species, S. amygdaloides, with low coppicing ability. The initiation and development of buds and the resprouting pattern of coppiced stools were compared. All buds were axillary in origin and showed the same principal structure consisting of one main shoot primordium and two lateral primordia. In S. viminalis and S. eriocephala the lateral buds contained several leaf primordia and sprouted shortly after the main bud. In S. amygdaloides further development of lateral buds was inhibited after formation of two budscales, and leaf primordia were not formed until the buds were forced to sprout. The number of sprouts developing after coppicing were correlated to the structure and number of buds and their position on the stools. Self-thinning rate was high and many shoots originating from lateral buds died. Most buds were located above ground on the remaining basal portions of harvested stems. No adventitious buds were found on the stools. Significantly different bud differentiation pattern and frequent sylleptic sprouting resulted in lower coppice response in S. amygdaloides compared to S. viminalis and S. eriocephala.  相似文献

为探究小檗科植物八角莲组织培养的器官发生方式,该研究以八角莲离体叶片、叶柄在MS培养基上诱导产生的愈伤组织、不定芽、不定根为对象,用连续石蜡切片技术分析八角莲组织培养的器官发生途径。结果表明:八角莲愈伤组织形成的解剖学特征是靠近表皮的薄壁细胞经激素刺激恢复分裂能力,继续培养形成拟分生组织。拟分生组织可形成许多分化中心。通过对八角莲组织培养产生的不定芽细胞组织学观察发现芽原基起源于愈伤组织外侧的几层薄壁细胞,芽原基背离愈伤组织中央生长形成不定芽,故八角莲脱分化形成的芽起源方式为外起源。而八角莲的根原基起源于组织深处髓部薄壁细胞和部分维管形成层细胞,进而形成类似球形或楔形并朝韧皮部突起的根原基轮廓,根原基继续发育会突破表皮生成不定根,起源方式为内起源。八角莲离体再生途径为器官发生型,在组培苗生长过程中先诱导形成不定芽,再诱导形成不定根,在愈伤组织上形成维管组织将不定芽和不定根连接成完整植株。  相似文献

This study describes the origin and early development of a distinct organ of clonal regeneration in Ginkgo biloba, the basal chichi. These aggregates of suppressed shoot buds originate from superficial meristems located in the cotyledonary axils of all Ginkgo seedlings as part of their normal ontogeny. Within 6 wk of germination these buds become embedded in the cortex of the stem, and their subsequent growth and development occurs below the surface of the bark. When stimulated by some traumatic event that damages the seedling axis, one of these embedded cotyledonary buds usually grows down from the trunk to form a woody, rhizomelike basal chichi which, under appropriate conditions, is capable of generating both aerial shoots and adventitious roots. Vegetative regeneration by means of basal chichi has not only contributed to the long-term persistence of G. biloba in the forests of China, but may also have played a role in the remarkable survival of the genus since the Cretaceous.  相似文献

The adventitious bud forming ability of Sitka spruce (Picea sitchensis (Bong.) Carr.) buds in vitro was found to be dependent upon the age of tree from which the explants were taken. Bud formation declined exponentially with increasing tree age when 1.0 and 10 μM 6-benzylaminopurine (BA) were used to induce adventitious buds. When less BA was used (0.1 μM) bud production was much lower with all ages of tree and no mathematical relationship between declining bud production and tree age was found. By a tree age of 6 years bud-forming ability had declined severely. Even the few buds that developed on older tree tissues failed to elongate into shoots, became necrotic and eventually died, indicating that adventitious bud induction in this species is not a rejuvenative process. Callusing of bud explants also declined with increasing tree age when 0.1 μM BA was used whilst very little callusing occurred at the higher cytokinin concentrations (1.0 and 10 μM BA). Tissue necrosis in vitro increased with tree age, whilst the ability of BA to retard necrosis declined with increasing tree age. Buds from epicormic shoots, formed on the lower trunks of 20-year-old trees when these were exposed to light, were not significantly better at forming adventitious buds in vitro than buds taken from the lower branches of the crown.  相似文献

采用室内生测法研究了雷公藤愈伤组织、悬浮细胞、不定根等组培产物对小菜蛾毒杀、生长发育影响以及不定根提取物对番茄灰霉病等植物病原真菌菌丝生长抑制作用,以明确雷公藤组培产物的生物活性及其应用前景。结果表明,不同雷公藤组培产物对小菜蛾3龄幼虫都具有明显的毒杀作用,悬浮细胞以及不定根的LC50均超过了根皮粉的效果,其中的不定根提取物对小菜蛾3龄幼虫毒杀作用的LC50为根皮粉的1.95倍;不同组培产物提取物对小菜蛾幼虫的生长均有明显的抑制作用,其中不定根提取物处理后每天的小菜蛾体重显著下降,72h后70%左右已经死亡,存活的试虫体重比试验前下降了18.33%;雷公藤不定根提取物对供试的11种植物病原真菌菌丝生长均有一定的抑制作用,有5种的抑制率超过60%,并对番茄灰霉病菌的抑制作用最强,其EC50为10.074mg/mL,且不定根的抑制效果均超过了根皮粉的抑制效果。  相似文献

The adventitious bud development was induced in epicotyl segments of Valencia sweet orange (Citrus sinensis L. Osbeck). Seeds were cultured in vitro for three weeks in the dark, followed by one week at a 16-h photoperiod. Epicotyl segments were cultured horizontally for the induction of organogenesis in Murashige and Tucker (1969, MT) culture medium supplemented with 1.0 mg dm−3 benzylaminopurine. Samples were observed by light and scanning electron microscopy from day zero to day 25, when buds were well grown. It was shown that the adventitious buds originated directly from the cambial region on the cut ends of the explants.  相似文献

Axillary bud development and adventitious bud formation was obtained with decapitated shoot explants of Aloe barbadensis Mill. Maximal bud growth and rooting of shoots was obtained on a modified medium of Murashige and Skoog supplemented with 5 M IBA. More adventitious and axillary buds developed on nutrient media supplemented with IBA than with NAA. Axillary buds but not adventitious buds developed with IAA in the medium. Morphogenesis was inhibited by 2,4-D. Kinetin, benzyladenine and thidiazuron were toxic to the explants and did not stimulate the development of axillary of adventitious buds. The optimal temperature for bud growth and development was 25°C. Axillary bud growth and the formation of adventitious buds was slowed down at 10°C and totally inhibited by 30°C. The optimal sucrose concentration was 3% with the inhibition of bud growth and development by higher sucrose levels.  相似文献

Summary Treatment with gibberellic acid (GA3) enhances the number of adventitious roots and the number and length of sprouted buds on stem cuttings of Ipomoea fistulosa. Ultrastructure of the cortical cells of living bark, leaf buds, and flower buds of apple trees (Malus pumila Mill. var. domestica Schneid, cv. McIntosh) was studied. The ultrastructural changes in preexisting plastids, leading to formation of the plastid initials and subsequent intermediates in the formation of plastids, started in mid-January or early February and continued to occur through late March. The plastid initials of apple trees resemble that of poplar, and prolamellar bodies were abundant in the developing intermediates in the cells of leaf buds and flower buds. Association of the endoplasmic reticulum and vesicles with developing plastid initials suggests that the former two organelles may possibly participate in the development of the plastid initial. Thus, formation of plastids from plastid initials takes place seasonally at this stage.  相似文献
