XCCNAU-92生产黄原胶的工业发酵培养基成份是:蔗糖、玉米淀粉、氮源X、鱼粉、CaCO3、MgSO4、K2HPO4。适宜的C/N是:蔗糖(玉米淀粉)/氮源X=60.0/1.0,蔗糖(玉米淀粉)/鱼粉=60.0/10.0。CaCO3、MgSO4对XCCNAU-92合成黄原胶有明显促进作用,K2HPO4在发酵过程中使pH保持稳定,Mn2+、Zn2+、Fe3+、柠檬酸和谷氨酸对生产黄原胶无促进作用。  相似文献

应用抗人淋巴细胞单克隆抗体检测32例鼻咽癌患者放疗前、后外周血T淋巴细胞亚群,并与12例非癌人群(对照组)进行对比。结果:鼻咽癌患者OKT3、OKT4及OKT4/OKT8值与对照组相比明显降低,OKT8显著上升(P<0.05);放疗后OKT3、OKT4及OKT4/OKT8值进一步下降,OKT8进一步升高(P<0.01)。提示鼻咽癌患者细胞免疫功能低下,放射治疗可进一步抑制鼻咽癌患者的细胞免疫功能。  相似文献

本文研究了用吸附交联技术共固定化蔗糖酶和葡萄糖氧化酶(GOD)的方法,考查了共固定化酶的动力学性质。试验结果表明:与溶液酶相比较,固定化蔗糖酶和GOD的响应滞迟期分别为3分钟和2分钟,4态响应时间增加6分钟和4分钟,Km值增大,pH─活力曲线变宽,最适pH值分别增大0.7和0.64,最适温度则降低7.3℃和16℃。以活性氧化铝作载体,戊二醛作交联剂制备的共固定化蔗糖酶和GOD,其蛋白质固定化率为92.9%,分解葡萄糖的总速度为441.6IU,当蔗糖浓度在0.2%以内时其反应速度与蔗糖浓度呈正相关(r=0.996),使用半衰期1623次,在4℃下保存120天活力残存为83.7%。  相似文献

本文测定了粘虫核多角体病毒的一个长3336bp的大片段,该片段包括了3个完整的开放阅读框(ORF2、ORF3、ORF4)和两个不完整的开放阅读框(ORF1、ORF5)。与AcNPV比较发现,ORF1编码重蛋白与P94同源,ORF3、ORF4、ORF5分别与AcNPV的ORF60、ORF59、ORF57同源。且转录方向一致。ORF2则是LsNPV的特有基因。分析结果表明,LsNPV基因组上基因的组织  相似文献

为探讨花榈木体胚发生过程中不同碳氮源处理对胚性愈伤组织诱导、发育和有机物积累的影响,并筛选出有利于花榈木体胚发生的碳氮源,优化体胚发生体系,该研究以成熟胚为外植体,通过单因素试验分析3种碳源、4种蔗糖浓度和6种氮源处理下胚性愈伤组织诱导、发育和有机物积累的差异。结果表明:(1)蔗糖中胚性愈伤组织诱导率显著高于葡萄糖和麦芽糖,但其体胚诱导率、体胚分化率、胚性愈伤组织可溶性糖、淀粉和可溶性蛋白含量差异不显著。(2)随着蔗糖浓度的升高,胚性愈伤组织、体细胞胚(体胚)诱导率、体胚分化率、胚性愈伤组织重量和可溶性蛋白含量呈先升高后降低的趋势,均以添加30 g·L-1蔗糖最高,而胚性愈伤组织可溶性糖和淀粉含量呈增加的趋势。(3)在6种氮源处理中,胚性愈伤组织诱导率以添加500 mg·L-1谷氨酰胺的处理最高,体胚诱导率则以添加谷氨酰胺和水解酪蛋白的处理较高,但不同氮源处理间体胚分化率无差异; 添加有机氮源的处理其胚性愈伤组织可溶性蛋白含量显著高于无氮源处理。总之,不同的碳氮源通过影响花榈木胚性愈伤组织的诱导、发育和有机物的积累,从而影响其体胚诱导率,但对体胚分化率影响不显著。初步认为30 g·L-1蔗糖和500 mg·L-1谷氨酰胺作为碳氮源可促进花榈木体胚发生诱导。  相似文献

采用花粉发育双核期的矮牵牛花药,研究不同浓度植物生长调节剂配比及不同浓度蔗糖和麦芽糖对花药诱导率的影响。结果表明,采用6-BA和IBA组合诱导效果较好;NH+6-BA 1.5mg/L+IBA 1.5mg/L花药诱导率较高;麦芽糖诱导效果明显比蔗糖好。  相似文献

Sucrose has been the carbohydrate traditionally used for potatomicrotuber production. Added to nutrient media, sucrose canact solely as a carbon source, or as an osmoticum, or both.Preliminary tests showed that the osmolarity of sucrose solutionswas increased by autoclaving, indicating some breakdown of thesugar. This was taken into consideration in experiments whichinvolved supplementing 4% sucrose media with sucrose, maltose,glucose or fructose, while keeping the osmotic potential ofthe media constant. A medium concentration of about 400 mM withonly sucrose was more suitable for microtuber production thanmedia supplemented with maltose, glucose or fructose. However,a better microtuber yield was obtained when hexoses were addedthan with unsupplemented 4% sucrose media. When glucose wassupplied at concentrations which had the same number of carbonatoms as 8% sucrose, the high osmolarity inhibited microtuberisation.Sugar movement in the tubering plantlet was followed using radio-labelledsucrose, glucose and fructose. The sucrose was translocatedand used at a faster rate than the other sugars, which tendedto remain in the roots of the plantlets. Furthermore, therewas no difference in microtuber production on media to whichthe sucrose was added before or after autoclaving, indicatingthat levels of breakdown were not severe enough to affect theprocess. The influences of carbon sources, fructose, glucose, sorbitol and sucrose on shoot proliferation and in vitro rooting of cork oak (Quercus suber L.) were compared at a wide range of concentrations (1–6%, w/v). The highest number of shoots occurred on glucose-containing medium. Nevertheless, we have chosen 3% sucrose which induced a similar rate of proliferation but favoured shoot elongation, permitting an effectively higher number of shoots during transfers. Sorbitol and autoclaved fructose did not stimulate shoot proliferation. Adventitious root formation was strongly dependent on carbohydrate supply. Sorbitol and autoclaved fructose were completely ineffectively on rooting induction. Glucose was the most effective carbon source on rooting promotion followed by sucrose and filter-sterilized fructose. The rooting response induced by fructose was dependent on the sterilizing procedure. The number of adventitious roots produced per shoot increased with increasing glucose and sucrose concentration. The content of reducing sugars in leaves of proliferation cultures and in leaves and roots of rooted plantlets was more dependent on carbon concentration than on glucose or sucrose supplement. The results presented here show that carbohydrate requirements during cork oak micropropagation depend upon the phase of culture. Sucrose (3%) and glucose (4%) were the best carbon sources respectively during proliferation and rooting phases.  相似文献

Three carbohydrate sources, maltose, sucrose and cellobiose, were compared for the respective effects on regeneration from internodal sections of flax. It was found that different patterns of morphogenesis were attributable to the type of carbohydrate and its concentration. Mean shoot regeneration was highest on a medium containing 87 mM maltose with a rate of 2.4 shoots per explant and root formation was stimulated to the greatest degree on a medium with 87 mM sucrose. This medium also resulted in the highest tissue dry weight.  相似文献

Different carbon sources, sterilized by autoclaving or filter-sterilization, were tested during induction, maintenance, and plant regeneration of embryogenic Miscanthus x ogiformis Honda `Giganteus' callus, derived from various explant types. Explants from small immature inflorescences, between 2.5 and 8 mm, produced more embryogenic callus than explants from shorter or longer inflorescences, shoot apices or leaf explants. On medium containing mannitol or sorbitol, only small amounts of callus were induced and no embryogenic callus was formed. Callus induction and embryogenic callus formation on shoot apices and immature inflorescences did not differ significantly between media containing sucrose, glucose, fructose, maltose or a mixture of glucose and fructose. However, callus induction and embryogenic callus formation from leaf explants were best on glucose. A higher percentage of leaf explants formed callus on autoclaved sucrose, as opposed to the other carbon sources where filter-sterilization in general resulted in a higher callus percentage. The growth rate of embryogenic callus was influenced both by carbon source and sterilization method when less than 1 g of callus was inoculated. None of the tested carbon sources could considerably improve plant regeneration from M. `Giganteus' callus, but a higher number of plants tended to be regenerated per callus piece from filter-sterilized carbon sources. Summary Ten cultivars of tomato were screened for their ability to produce shoots and shoot primordia on media containing a range of 6-benzyladenine (BA) concentrations (0.0–4.0 mgl−1; 0.0–17.7 μM) in combination with indole-3-acetic acid (IAA; 0.0, 0.1, 0.2 mgl−1; 0.0, 0.57, 1.14 μM, respectively). Both genotypes and growth regulator levels showed significant differences at α=0.05. However, their interaction was not significantly different. A comparison of the mean number of shoots produced six cultivar classes, with UC82, UC97-3, Castlerock, Red Rock, and Peto86 producing the highest means. There were four cultiver frequency classes, the highest including cultivars Peto Rock (93.7%), Peto86 (92.3%), and Strain B (90.7%). Growth regulator mean comparisons produced eight classes, the highest included three different BA/IAA combinations, with media containing 2.0 mgl−1 (8.87 μM) BA and 0.2 mgl−1 (1.14. μM) IAA giving the highest mean and frequency (94.1%). The effect of carbon source was studied using glucose, fructose, maltose, and sucrose at 3.0% concentration. Three high regenerating genotypes and four media were used in combination with each sugar. Sugars were significantly different with maltose giving the highest number of shoots (31.6) followed by glucose (20.9). Growth regulator means, sugar-growth regulator interaction, and cultivar-sugar interaction were significant. A three-way interaction was nonsignificant. The effect of maltose and sucrose concentration (1.5–9.0%) showed the positive effect of maltose over sucrose in inducing shoots and in reducing loss rates of shoots on the regeneration media. Maltose at 1.5, 3.0, and 4.5% was the most effective. Six pH values (4.0–6.5) were used to test their effect on shoot induction in three cultivars (Peto86, UC97-3 and Castlerock). pH effects and cultivar-pH interaction were not significant.  相似文献

本文研究了培养基中碳源和氮源变化对悬浮培养玫瑰茄细胞生长和花青素合成的影响。在8种不同的碳源中,麦芽糖有利于花青素的积累,而蔗糖和葡萄糖适合细胞生长,并有较高的花青素产率。在1%~10%蔗糖浓度范围内,4%浓度下细胞生长和花青素产率最高,而6%浓度下细胞花青素含量最高,高渗环境较有利于细胞花青素的积累。135mM的氮源总量已足够维持玫瑰茄细胞生长和花青素合成,氮源总量增加对细胞代谢有抑制作用。NH+4对细胞有显著抑制作用。总量135mM,NO-3与NH+4比例25∶2和23∶4时细胞生长和花青素合成最佳。  相似文献

Sprouts of potato tubers were excised from the three potato cultivars Agria, Hermes, and Spunta, sterilized and subjected to shoot formation and propagation on Murashige and Skoog (MS) medium supplemented with 1 mg dm-3 6-benzylaminopurine (BAP) + 0.5 mg dm-3 gibberellic acid. Shoots were rooted on MS medium supplemented with 1 mg dm-3 indole-3-butyric acid. To increase shoot vigour prior tuber formation, shoots were subcultured on MS medium supplemented with 0.56 mg dm-3 BAP, 0.11 mg dm-3 2,4-dichlorophenoxyacetic acid, and 0.96 mg dm-3 naphthaleneacetic acid. Under dark, microtuberization on MS media supplemented with 4 mg dm-3 of both BAP and kinetin was better than 4 mg dm-3 BAP alone, where they induced higher number of microtubers per shoot and/or the percentage of shoots that formed microtubers. The highest frequency of microtuber formation was achieved when sucrose at high concentration (8 %) was used as carbon source in culture media. Glucose ranked at the second position whereas fructose reduced the microtuber formation frequency when it was used alone or in combination with glucose. Under the applied culture conditions, cvs. Agria and Hermes showed better micropropagation and microtuberization in comparison to cv. Spunta. In addition, isozyme and RAPD techniques revealed that Agria and Hermes are closer to each other when compared with the third cultivar.  相似文献
