Growth, photosynthesis, and Na+, K+, Ca2+, and Mg2+ distributions were examined in two-year-old hydroponically cultured Populus nigra and Populus alba cuttings exposed to salt stress (0, 50, or 100 mM NaCl) for four or six weeks and to nonaeration stress for one or three weeks, followed by a three-week aeration period in 2/5 Hoagland solution. Salt stress with 100 mM NaCl totally inhibited height increase in P. nigra cuttings. Combined salinity and nonaeration inhibited height increase to a greater degree than either stress alone in both species. Simple salt stress did not affect diameter increase in P. alba, whereas combined high salinity (100 mM NaCl) and nonaeration inhibited diameter increase. Growth and biomass accumulation were more sensitive to salt stress in P. nigra cuttings than in P. alba, although P. alba showed a more rapid decrease in photosynthesis in response to nonaeration stress. Ion distributions in the leaves and roots differed between species. P. alba was superior to P. nigra in terms of Na+ exclusion capacity, such that most of the absorbed Na+ was confined to the root system, with little reaching the leaves. The distributions of K+, Ca2+, and Mg2+ in the leaves and roots of each species under the two stressors were also analyzed. The lower Na+/K+ ratio in leaves indicated that P. alba was more tolerant to salt stress than P. nigra.  相似文献

Nitraria retusa and Atriplex halimus (xero-halophytes) plants were grown in the range 0–800 mM NaCl while Medicago arborea (glycophyte) in 0–300 mM NaCl. Salt stress caused a marked decrease in osmotic potential and a significant accumulation of Na+ and Cl in leaves of both species. Moderate salinity had a stimulating effect on growth rate, net CO2 assimilation, transpiration and stomatal conductance for the xero-halophytic species. At higher salinities, these physiological parameters decreased significantly, and their percentages of reduction were higher in A. halimus than in N. retusa whereas, in M. arborea they decreased linearly with salinity. Nitraria retusa PSII photochemistry and carotenoid content were unaffected by salinity, but a reduction in chlorophyll content was observed at 800 mM NaCl. Similar results were found in A. halimus, but with a decrease in the efficiency of PSII (F′v/F′m) occurred at 800 mM. Conversely, in M. arborea plants we observed a significant reduction in pigment concentrations and chlorophyll fluorescence parameters. The marked toxic effect of Na+ and/or Cl observed in M. arborea indicates that salt damage effect could be attributed to ions’ toxicity, and that the reduction in photosynthesis is most probably due to damages in the photosynthetic apparatus rather than factors affecting stomatal closure. For the two halophyte species, it appears that there is occurrence of co-limitation of photosynthesis by stomatal and non-stomatal factors. Our results suggest that both N. retusa and A. halimus show high tolerance to both high salinity and photoinhibition while M. arborea was considered as a slightly salt tolerant species.  相似文献

Thirty bacterial strains were isolated from the rhizosphere of plants collected from Egypt and screened for production of chitinase enzymes. Bacillus thuringiensis NM101-19 and Bacillus licheniformis NM120-17 had the highest chitinolytic activities amongst those investigated. The production of chitinase by B. thuringiensis and B. licheniformis was optimized using colloidal chitin medium amended with 1.5% colloidal chitin, with casein as a nitrogen source, at 30°C after five days of incubation. An enhancement of chitinase production by the two species was observed by addition of sugar substances and dried fungal mats to the colloidal chitin media. The optimal conditions for chitinase activity by B. thuringiensis and B. licheniformis were at 40°C, pH 7.0 and pH 8.0, respectively. Na+, Mg2+, Cu2+, and Ca2+ caused enhancement of enzyme activities whereas they were markedly inhibited by Zn2+, Hg2+, and Ag+. In vitro, B. thuringiensis and B. licheniformis chitinases had potential for cell wall lysis of many phytopathogenic fungi tested. The addition of B. thuringiensis chitinase was more effective than that of B. licheniformis in increasing the germination of soybean seeds infected with various phytopathogenic fungi.  相似文献

Drought and salinity are major abiotic stresses affecting rice production. To improve plant tolerance to salinity and drought, we overexpressed rice Na+/H+ exchangers (OsNHX1) and H+-pyrophosphatase in tonoplasts (OsVP1) in a japonica elite rice cultivar, Zhonghua 11. Compared with our wild-type control, transgenic plants overexpressing both genes incurred less damage when exposed to long-term treatment with 100 mM NaCl or water deprivation. Under high-saline conditions, the transformants accumulated less Na+ and malondialdehyde in the leaves, thereby allowing the plants to maintain a low level of leaf water potential and reduce stress-induced damage. Those transgenics also had higher photosynthetic activity during the stress period. Under those conditions, they also showed an increase in root biomass, which enabled more water uptake. These results suggest that OsVP1 and OsNHX1 improve the tolerance of rice crops against drought and salt by employing multiple strategies in addition to osmotic regulation.  相似文献

Salinity adversely affects plant growth and development. Halotolerant plant-growth-promoting rhizobacteria (PGPR) alleviate salt stress and help plants to maintain better growth. In the present study, six PGPR strains were analyzed for their involvement in salt-stress tolerance in Arachis hypogaea. Different growth parameters, electrolyte leakage, water content, biochemical properties, and ion content were analyzed in the PGPR-inoculated plants under 100 mM NaCl. Three bacterial strains, namely, Brachybacterium saurashtrense (JG-06), Brevibacterium casei (JG-08), and Haererohalobacter (JG-11), showed the best growth of A. hypogaea seedlings under salt stress. Plant length, shoot length, root length, shoot dry weight, root dry weight, and total biomass were significantly higher in inoculated plants compared to uninoculated plants. The PGPR-inoculated plants were quite healthy and hydrated, whereas the uninoculated plant leaves were desiccated in the presence of 100 mM NaCl. The percentage water content (PWC) in the shoots and roots was also significantly higher in inoculated plants compared to uninoculated plants. Proline content and soluble sugars were significantly low, whereas amino acids were higher than in uninoculated plants. The MDA content was higher in uninoculated plants than in inoculated plants at 100 mM NaCl. The inoculated plants also had a higher K+/Na+ ratio and higher Ca2+, phosphorus, and nitrogen content. The auxin concentration was higher in both shoot and root explants in the inoculated plants. Therefore, it could be predicted that all these parameters cumulatively improve plant growth under saline conditions in the presence of PGPR. This study shows that PGPR play an important role in inducing salinity tolerance in plants and can be used to grow salt-sensitive crops in saline areas.  相似文献

A pot-culture experiment was carried out to investigate the effect of arbuscular mycorrhizal (AM) fungus (Glomus macrocarpum Tul. and Tul.) on plant growth and Cd2+uptake by Apium graveolens L. in soil with different levels of Cd2+. Mycorrhizal (M) and non-mycorrhizal (NM) plants were grown in soil with 0, 5, 10, 40 and 80 Cd2+ mg kg−1soil. The infectivity of the fungus was not affected by the presence of Cd2+ in the soil. M plants showed better growth and less Cd2+ toxicity symptoms. Cd2+ root : shoot ratio was higher in M plants than in NM plants. These differences were more evident at highest Cd2+ level (80 mg kg−1 soil). Chlorophyll a and chlorophyll b concentrations were significantly higher in AM-inoculated celery leaves. The dilution effect due to increased biomass, immobilization of Cd2+ in root and enhanced P-uptake in M plants may be related to attenuation of Cd2+toxicity in celery.  相似文献

Three types of transgenic tobacco plants were acquired by separate transformation or co-transformation of a vacuolar Na+/H+ antiporter gene, SeNHX1, and a betaine synthesis gene, BADH. When exposed to 200 mM NaCl, the dual gene-transformed plants displayed greater accumulation of betaine and Na+ than their wild-type counterparts. Photosynthetic rate and photosystem II activity in the transgenic plants were less affected by salt stress than wild-type plants. Transgenic plants exhibited a greater increase in osmotic pressure than wild-type plants when exposed to NaCl. More importantly, the dual gene transformed plants accumulated higher biomass than either of the single transgenic plants under salt stress. Taken together, these findings indicate that simultaneous transformation of BADH and SeNHX1 genes into tobacco plants can enable plants to accumulate betaine and Na+, thus conferring them more tolerance to salinity than either of the single gene transformed plants or wild-type tobacco plants. The effects of treatment with NaCl (3, 100 and 300 mM) for 1, 2, 3 and 7 d on plant growth and ion accumulation were analyzed in 2-week and 8-week-old Annona muricata and A. squamosa plants. Fresh mass and root growth inhibition were directly related to the increase in salinity, particularly for A. squamosa. Two-weeks old seedlings were sensitive to 100 and 300 mM NaCl particularly after 7 d, whereas 8-week-old plants were shown to be more resistant to NaCl even at 300 mM NaCl. Na+ and Cl mostly accumulated in young leaves. Our results suggest that A. squamosa is more sensitive than A. muricata to salt stress and that older seedlings of both species are more tolerant than younger seedlings.  相似文献

This study describes two phenotypes of Arabidopsis thaliana (ecotype Columbia) developed in vitro under salt stress (75 mM NaCl). The phenotypes 01 and 02 appeared visibly distinguishable by rosette morphology and competence to produce flowers. Phenotype 01, sensible to salt stress, accumulated high quantities of Na+, showed a slight reduction in dry mass, and high protein and chlorophyll contents. Moreover, its anatomy exhibited some xeromorphic traits. Phenotype 02, clearly salt tolerant, showed a morphology similar to control plants, displaying typical phyllotactic rosette and flowering stalk production. Accumulation of Na+, protein and chlorophyll contents were close to control plants. Reversion experiments on NaCl free MS medium, showed a partially recovered phenotype 01. A threshold salt stress concentration that permits the simultaneous development of two phenotypes, was found.  相似文献

Soil salinity is a major factor limiting apple production in some areas. Tonoplast Na+/H+ antiporters play a critical role in salt tolerance. Here, we isolated MdNHX1, a vacuolar Na+/H+ antiporter from Luo-2, a salt-tolerant rootstock of apple (Malus × domestica Borkh.), and introduced it into apple rootstock M.26 by Agrobacterium-mediated transformation. PCR and DNA gel blot analyses confirmed successful integration of MdNHX1. RT-PCR analysis indicated that the gene was highly expressed in transgenic plants, but the degree of this expression varied among lines. Its overexpression conferred high tolerance to salt stress. Analysis of ion contents showed that, when exposed to salinity stress, the transgenics compartmentalized more Na+ in the roots and also maintained a relatively high K+/Na+ ratio in the leaves compared with non-transformed plants. Under normal conditions, however, amounts of potassium and sodium did not differ significantly between transgenic and control plants.  相似文献

The salinity tolerance and ion transport of 2-month-old seedlings of stone pine (Pinus pinea L.) grown in hydroponic solution containing various concentrations of NaCl (0–100 mM) were studied. The presence of salt of up to 100 mM did not significantly reduce growth. Seedling hydration was insensitive to salinity. High salt concentrations reduced K+ and Ca2+ uptake, root accumulation, and export to shoots. Na+ and Cl ions, representing the major part of the ionic uptake, were effectively compartmentalized in vacuoles. We concluded that seedlings of stone pine cultivated hydroponically were highly tolerant to salt concentrations of up to 100 mM for a culture period of 38 days. This tolerance was associated with the accumulation of Na+ and Cl ions in the shoots.  相似文献

Total ion (Na+, K+, Ca2+, SO4 2? and Cl?) accumulation by plants, ion contents in plant tissues and ion secretion by salt glands on the surface of shoots of Tamarix ramosissima adapted to different soil salinity, namely low (0.06 mmol Na+/g soil), moderate (3.14–4.85 mmol Na+/g soil) and strong (7.56 mmol Na+/g soil) were analyzed. There are two stages of interrelated and complementary regulation of ion homeostasis in whole T. ramosissima plants: (1) regulation of ion influx into the plant from the soil and (2) changing the secretion efficiency of salt glands on shoots. The secretion efficiency of salt glands was appraised by the ratio of ion secretion to tissue ion content. Independent of soil salinity, the accumulation of K+ and Ca2+ was higher than the contents of these ions in the soil. Furthermore, the accumulation of K+, Ca2+ and SO4 2? ions by plants was maintained within a narrow range of values. Under low soil salinity, Na+ was accumulated, whereas under moderate and strong salinity, the influxes of Na+ were limited. However, under strong salinity, the accumulation of Na+ was threefold higher than that under low soil salinity. This led to a change in the Na+/K+ ratio (tenfold), an increase in the activity of salt glands (tenfold) and a reduction in plant growth (fivefold). An apparently high Na+/K+ ratio was the main factor determining over-active functioning of salt glands under strong salinity. Principal component analysis showed that K+ ions played a key role in ion homeostasis at all levels of salinity. Ca2+ played a significant role at low salinity, whereas Cl? and interrelated regulatory components (K+ and proline) played a role under strong salinity. Proline, despite its low concentration under strong salinity, was involved in the regulation of secretion by salt glands. Different stages and mechanisms of ion homeostasis were dominant in T. ramosissima plants adapted to different levels of salinity. These mechanisms facilitated the accumulation of Na+ in plants under low soil salinity, the limitation of Na+ under moderate salinity and the over-activation of Na+ secretion by salt glands under strong salinity, which are all necessary for maintaining ion homeostasis and water potential in the whole plant.  相似文献

On the basis of the results of gene chip analysis of the salt-tolerant wheat mutant RH8706-49 under conditions of salt stress, we identified and cloned an unknown salt-induced gene TaST (Triticum aestivum salt-tolerant). Real-time quantitative PCR analysis showed that the expression of the gene was induced by salt stress. Transgenic Arabidopsis plants overexpressing the TaST gene showed higher salt tolerance than the wild-type controls. Subcellular localization studies revealed that the protein encoded by this gene was in the nucleus. In comparison with wild-type controls, transgenic Arabidopsis plants accumulated more Ca2+, soluble sugar, and proline and less Na+ under salt stress. Real-time quantitative PCR analysis showed that Arabidopsis plants overexpressing TaST also showed increased expression of many stress-related genes. All these findings indicated that TaST can enhance the salt tolerance of transgenic Arabidopsis plants.  相似文献

The effects of NaCl and mannitol iso-osmotic stresses on calli issued from sugarcane cultivars (cvs.) R570, CP59-73 and NCo310 were investigated in relation to callus growth, water content, ion and proline concentrations. Callus growth and water content decreased under both stresses with the highest reduction under mannitol-induced osmotic stress. The ion concentration was drastically affected after exposure to NaCl and mannitol. Salt stress induced an increase in Na+ and Cl accumulation and a decrease in K+ and Ca2+ concentrations. Under mannitol-induced osmotic stress, K+ and Ca2+ concentrations decreased significantly while Na+ and Cl concentrations remained unchanged. Free proline accumulation occurred under both stresses and was more marked in stress-sensitive cv. than in stress-resistant one. Our results indicated that the physiological mechanisms operating at the plant cell level in response to salt- and osmotic-induced stress in sugarcane cvs. are different. Among the cvs., we concluded that the stress resistance is closely related to the maintain of an adequate water status and a high level of K+ and Ca2+ under both stresses and a low level of Na+ concentration in the presence of NaCl. Thus, sugarcane (Saccharum sp.) can be regarded as a Na+ excluder. We also provided evidence that proline accumulation is a stress-sensitive trait rather than a stress resistance marker.  相似文献

Tibetan wild barley is rich in genetic diversity with potential allelic variation useful for salinity-tolerant improvement of the crop. The objectives of this study were to evaluate salinity tolerance and analysis of the allelic function of HvHKT1 and HvHKT2 in Tibetan wild barley. Salinity tolerance of 189 Tibetan wild barley accessions was evaluated in terms of reduced dry biomass under salinity stress. In addition, Na+ and K+ concentrations of 48 representative accessions differing in salinity tolerance were determined. Furthermore, the allelic and functional diversity of HvHKT1 and HvHKT2 was determined by association analysis as well as gene expression assay. There was a wide variation among wild barley genotypes in salt tolerance, with some accessions being higher in tolerance than cultivated barley CM 72, and salinity tolerance was significantly associated with K+/Na+ ratio. Association analysis revealed that HvHKT1 and HvHKT2 mainly control Na+ and K+ transporting under salinity stress, respectively, which was validated by further analysis of gene expression. The present results indicated that Tibetan wild barley offers elite alleles of HvHKT1 and HvHKT2 conferring salinity tolerance.  相似文献

To elucidate the osmotic adjustment characteristics of mangrove plants, inorganic ion and organic solute contents of intermediate leaves were investigated in 3-month-old Kandelia candel (L.) Druce seedlings during 45 days of NaCl treatments (0, 200, and 500 mM NaCl). The contents of Na+, Cl, total free amino acids, proline, total soluble sugars, pinitol and mannitol increased to different degree by salinity, whereas, K+ content decreased by salinity compared with control. NaCl treatment induced an increase of inorganic ion contribution while a decrease of organic solute contribution. It was concluded that accumulating a large amount of inorganic ions was used as the main osmotic adjustment mechanism under salinity treatment. However, accumulation of organic osmolytes might be considered to play much more important role in osmoregulation under severe salinity (500 mM NaCl) than under moderate salinity (200 mM NaCl), thus the damage caused by high toxic ions (Na+ and Cl) concentration in K. candel leaves could be avoided.  相似文献

Rice yield is severely affected by high-salt concentration in the vicinity of the plant. In an effort to engineer rice for improved salt tolerance Agrobacterium-mediated transformation of rice cv. Binnatoa was accomplished with the Pennisetum glaucum vacuolar Na+/H+ antiporter gene (PgNHX1) under the constitutive CaMV35S promoter. For the molecular analysis of putative transgenic plants, PCR and RT-PCR were performed. Transgenic rice plants expressing PgNHX1 showed better physiological status and completed their life cycle by setting flowers and seeds in salt stress, while wild-type plants exhibited rapid chlorosis and growth inhibition. Moreover, transgenic rice plants produced higher grain yields than wild-type plants under salt stress. Assessment of the salinity tolerance of the transgenic plants at seedling and reproductive stages demonstrated the potential of PgNHX1 for imparting enhanced salt tolerance capabilities and improved yield.  相似文献

Increased salinization of arable land is expected to have devastating global effects in the coming years. Arbuscular mycorrhizal fungi (AMF) have been shown to improve plant tolerance to abiotic environmental factors such as salinity, but they can be themselves negatively affected by salinity. In this study, the first in vitro experiment analyzed the effects of 0, 50, or 100 mM NaCl on the development and sporulation of Glomus intraradices. In the second experiment, the effects of mycorrhization on the expression of key plant genes expected to be affected by salinity was evaluated. Results showed that the assayed isolate G. intraradices DAOM 197198 can be regarded as a moderately salt-tolerant AMF because it did not significantly decrease hyphal development or formation of branching absorbing structures at 50 mM NaCl. Results also showed that plants colonized by G. intraradices grew more than nonmycorrhizal plants. This effect was concomitant with a higher relative water content in AM plants, lower proline content, and expression of Lsp5cs gene (mainly at 50 mM NaCl), lower expression of the stress marker gene Lslea gene, and lower content of abscisic acid in roots of mycorrhizal plants as compared to nonmycorrhizal plants, which suggest that the AM fungus decreased salt stress injury. In addition, under salinity, AM symbiosis enhanced the expression of LsPIP1. Such enhanced gene expression could contribute to regulating root water permeability to better tolerate the osmotic stress generated by salinity.  相似文献

High salinity is an environmental factor that inhibits plant growth and development, leading to large losses in crop yields. We report here that mutations in SIZ1 or PHO2, which cause more accumulation of phosphate compared with the wild type, enhance tolerance to salt stress. The siz1 and pho2 mutations reduce the uptake and accumulation of Na+. These mutations are also able to suppress the Na+ hypersensitivity of the sos3-1 mutant, and genetic analyses suggest that SIZ1 and SOS3 or PHO2 and SOS3 have an additive effect on the response to salt stress. Furthermore, the siz1 mutation cannot suppress the Li+ hypersensitivity of the sos3-1 mutant. These results indicate that the phosphate-accumulating mutants siz1 and pho2 reduce the uptake and accumulation of Na+, leading to enhanced salt tolerance, and that, genetically, SIZ1 and PHO2 are likely independent of SOS3-dependent salt signaling.  相似文献
