酵母是一种真核模式生物同时也是一种耐盐微生物,其基因表达和信号传导系统的调节机制及离子运输机制与高等真核生物类似。酵母耐盐机制的研究有助于阐明真核生物的耐盐机制。本文综述了酵母在盐胁迫下的信号传导途径和分子应答机制,以及在酵母耐盐机制研究中主要的研究方法。 Abstract:Yeast is a model eukoryotic organism and salt-tolerant microorganism.The regulative mechanism of gene expression and signal transduction and ion transport of yeast is similar to that of higher eukoryotic organism.The research on salt-tolerant mechanism of yeast will be helpful to the illustrate the salt-tolerant mechanism of higher eukoryotic organism.This review summarized the signal transduction pathway and molercular responses of yeast under salt stress and the major research methods in the research on the salt-tolerant mechenism in yeast.  相似文献

利用F1花药培养、EMS诱变和耐盐性反复筛选后已稳定9代的小麦耐盐突变体RH8706-49、H8706-34、H8706-44、H8706-48、H8706-57及其亲本濮农3665、百农3039为材料,用生化标记(醇溶蛋白)及分子标记(RAPD)分析了各材料间的差异,发现突变体与亲本相比,不仅发生了蛋白质水平的变异,而且也在DNA水平上证明了突变的发生,从而为耐盐突变体的真实性提供了有力的证据,排除了盐适应的可能性; 经用218 个引物对5个突变体之间的多态性进行RAPD分析,结果表明,它们之间的差异很小,其遗传背景相似,因而它们是一系列耐盐性不同的近似等位基因系。 Abstract: In this paper, 5 wheat salt tolerant mutants(H8706-34、H8706-44、H8706-48、RH8706-49、H8706-57)derived from anther culture、EMS induction and salt tolerance selection and their parents (Punong3665、Bainong3039)were used as materials, all the mutants have inherited stably for 9 generations. Differences were revealed between the mutants and their parents using chemical marker(gliadin)and molecular marker(RAPD), the results showed that compared with the parents, the mutants varied not only on the protein level,but also on the DNA level,which supplied hard evidence of the truth of the mutants and ruled out the possibility of salt adaptation. RAPD analysis were conducted among the 5 mutants by 218 primers,which proved they were a series of near isogenic lines of different salt tolerance because of their little difference and similar genetic background.  相似文献

In an attempt to isolate and identify the target genes relevant to salt tolerance in a mangrove plant (Sesuvium portulacastrum L.), a subtracted cDNA library was constructed via suppressive subtractive hybridization (SSH), in which the poly(A)+RNA isolated from salt-tolerant S. portulacastrum leaves was used as a tester, whereas the driver was poly(A)+RNA, derived from salt-sensitive S. portulacastrum leaves. Screening of this subtracted cDNA library revealed five clones, of which the expression levels in the salt-tolerant plant were markedly higher than those observed in the salt-sensitive plant, indicating that these candidate clones may be involved in salt-tolerance pathways. Among the clones isolated, P66, P175, and P233 are novel because no significant similarity was obtained upon alignment with the GenBank database. Clone P89 demonstrated high homology with NADPH of Arabidopsis thaliana, whereas clone P152 was highly homologous with the gene encoding late embryogenesis abundant (LEA) protein of A. thaliana. The full-length gene of clone P152, with a predicated 344 amino acid residues, was shown to bear LEA-2 domains, a signature motif for proteins that have been enriched under salty and drought conditions. It is thus implied that clone P152 would be a salt-tolerance gene of S. portulacastrum. In addition, we have also developed a strategy for the extraction of total RNA from mangrove plants.  相似文献

The pearl oyster Pinctada martensii is an important economic shellfish species and mainly cultured in the southern provinces of China. The species is cultured for marine pearl production. However, pearl quality has recently decreased because the cultured stocks had slow growth and mass mortalities caused by inbreeding depression. We have initiated a selective breeding program to improve the cultivated stocks since 2002. In 2004, a base population was developed by collecting the breeders from Beibuwan and Liushagang stocks. During the period of 2005–2007, a two successive generation selection for shell length in the base population was carried out to produce the second generation selected and unselected lines. In May of 2008, three types of lines were produced by selecting the breeders from the second generation selected and unselected lines. The three types of lines were designated as SS (Selected for three generations), SC (selected for the two generations) and CC (unselected for three generations). Realized heritability for the third generation, cumulative (over three generation) and current (for the third generation) genetic gains were evaluated by comparing the growth performance of three types of lines at days 8, 16, 45, 75, 180 and 360. It was found that the SS lines had significant larger mean shell length and shell height than the SC and CC lines at all sampling ages (P < 0.05). The realized heritability estimate for adult shell length for the third generation was 0.41, similar with those detected for the first and second generation. The cumulative and current genetic gains for adult shell length were 20.94% and 13.27%, respectively. The present results indicate that there exists a high genetic variation in the population and mass selection is potential to improve pearl oyster stocks.  相似文献

Common wheat (Triticum aestivum L.) is one of the most important crops,and intra-specific wheat hybrids have obvious heterosis in yield and protein quality.Therefore,utilization of hybrid wheat varieties offers an effective way to increase yield and nutrition.Cytoplasmic male sterility (CMS) systems are a useful genetic tool for hybrid crop breeding,and are ideal models for studying the genetic interaction and cooperative function of mitochondrial and nuclear genomes in plants (Schnable and Wise,1998;Hanson and Bentolila,2004).The breeding of hybrid wheat using male sterility caused by the cytoplasm of T.timopheevii has been studied since the early 1960's.But it is unsuccessful because there are some deficiencies in the practical application of this cytoplasm,including limited restoration resources,thin seeds,pre-harvest sprouting and lower germination rate (Wilson and Ross,1962).The Sv type of cytoplasmic male sterility (CMS-Sv) in wheat is general accessions for four types of CMS lines that were derived from four Aegilops species:Ae.kotschyi,Ae.variabilis,Ae.ventricosa,and Ae.bicornis.Based on the observation of alloplasmic lines produced in all possible combinations between 12 wheat nuclear genotypes and 47 cytoplasms of two related genera,Triticum (wheat) and Aegilops,Ogihara and Tsunewaki (1988) concluded that the D2-cytoplasm of Ae.crassa and its relatives,N-cytoplasm of Ae.uniaristata,and SV-cytoplasm of Ae.kotschyi and its relatives might be used as the alternative male sterile cytoplasm to replace the T.timopheevii cytoplasm for hybrid wheat breeding.Ikeguchi et al.(1999) proposed that spring-type hybrid wheat may be bred by combination of the 1BL-1RS chromosome and Ae.kotschyi cytoplasm with a new fertility-restorer gene discovered in a wheat variety Kitamiharu 48.Zhang et al.(1996) also proposed the use of CMS-Sv lines as the most effective male sterile cytoplasm.  相似文献

Thinopyrum elongatum(2n = 2x = 14,EE),a wild relative of wheat,has been suggested as a potentially novel source of resistance to several major wheat diseases including Fusarium Head Blight(FHB).In this study,a series of wheat(cv.Chinese Spring,CS) substitution and ditelosomic lines,including Th.elongatum additions,were assessed for TypeⅡresistance to FHB.Results indicated that the lines containing chromosome 7E of Th.elongatum gave a high level of resistance to FHB,wherein the infection did not spread beyond the inoculated floret.Furthermore,it was determined that the novel resistance gene(s) of 7E was located on the short-arm(7ES) based on sharp difference in FHB resistance between the two 7E ditelosomic lines for each arm.On the other hand,Th.elongatum chromosomes 5E and 6E likely contain gene(s) for susceptibility to FHB because the disease spreads rapidly within the inoculated spikes of these lines. Genomic in situ hybridization(GISH) analysis revealed that the alien chromosomes in the addition and substitution lines were intact,and the lines did not contain discernible genomic aberrations.GISH and multicolor-GISH analyses were further performed on three translocation lines that also showed high levels of resistance to FHB.Lines TA3499 and TA3695 were shown to contain one pair of wheat-Th. elongatum translocated chromosomes involving fragments of 7D plus a segment of the 7E,while line TA3493 was found to contain one pair of wheat-Th.elongatum translocated chromosomes involving the D- and A-genome chromosomes of wheat.Thus,this study has established that the short-arm of chromosome 7E of Th.elongatum harbors gene(s) highly resistant to the spreading of FHB,and chromatin of 7E introgressed into wheat chromosomes largely retained the resistance,implicating the feasibility of using these lines as novel material for breeding FHB-resistant wheat cultivars.  相似文献

Defects in mitochondrial energy metabolism have been implicated in the pathology of several neurodegenerative disorders. In addition, the reactive metabolites generated from the metabolism and oxidation of the neurotransmitter dopamine (DA) are thought to contribute to the damage to neurons of the basal ganglia. We have previously demonstrated that infusions of the metabolic inhibitor malonate into the striata of mice or rats produce degeneration of DA nerve terminals. In the present studies, we demonstrate that an intrastriatal infusion of malonate induces a substantial increase in DA efflux in awake, behaving mice as measured by in vivo microdialysis. Furthermore, pretreatment of mice with tetrabenazine (TBZ) or the TBZ analogue Ro 4-1284 (Ro-4), compounds that reversibly inhibit the vesicular storage of DA, attenuates the malonate-induced DA efflux as well as the damage to DA nerve terminals. Consistent with these findings, the damage to both DA and GABA neurons in mesencephalic cultures by malonate exposure was attenuated by pretreatment with TBZ or Ro-4. Treatment with these compounds did not affect the formation of free radicals or the inhibition of oxidative phosphorylation resulting from malonate exposure alone. Our data suggest that DA plays an important role in the neurotoxicity produced by malonate. These findings provide direct evidence that inhibition of succinate dehydrogenase causes an increase in extracellular DA levels and indicate that bioenergetic defects may contribute to the pathogenesis of chronic neurodegenerative diseases through a mechanism involving DA.  相似文献

Somatostatin (SST) peptide is a potent inhibitor of insulin secretion and its effect is mediated via somatostatin receptor 5 (SSTR5) in the endocrine pancreas. To investigate the consequences of gene ablation of SSTR5 in the mouse pancreas, we have generated a mouse model in which the SSTR5 gene was specifically knocked down in the pancreatic beta cells (betaSSTR5Kd) using the Cre-lox system. Immunohistochemistry analysis showed that SSTR5 gene expression was absent in beta cells at three months of age. At the time of gene ablation, betaSSTR5Kd mice demonstrated glucose intolerance with lack of insulin response and significantly reduced serum insulin levels. Insulin tolerance test demonstrated a significant increase of insulin clearance in vivo at the same age. In vitro studies demonstrated an absence of response to SST-28 stimulation in the betaSSTR5Kd mouse islet, which was associated with a significantly reduced SST expression level in betaSSTR5Kd mice pancreata. In addition, betaSSTR5Kd mice had significantly reduced serum glucose levels and increased serum insulin levels at 12 months of age. Glucose tolerance test at an older age also indicated a persistently higher insulin level in betaSSTR5Kd mice. Further studies of betaSSTR5Kd mice had revealed elevated serum C-peptide levels at both 3 and 12 months of age, suggesting that these mice are capable of producing and releasing insulin to the periphery. These results support the hypothesis that SSTR5 plays a pivotal role in the regulation of insulin secretion in the mouse pancreas.  相似文献

Abstract. Changes are described in aquatic vegetation in oligotrophic, groundwater-fed Rhine floodplain streams in Alsace (eastern France), resulting from disturbance. Disturbance factors include changes in nutrients, either permanent ones - effluent from a waste water treatment plant or trout hatcheries - or periodic ones: flooding. Regular inputs of high levels of phosphate and ammonia modified the macrophyte vegetation in these streams. The floristic composition, which was characteristic of oligotrophic waters upstream of the eutrophicated sector, changed to that of a eutrophic situation as originally found downstream. Periodic disturbance by floods which normally occur once a year, irregularly eutrophicates the small streams, causing the development of a mixture of eutrophic and oligotrophic species. Six macrophyte communities are distinguished, indicating different trophic levels. The aquatic vegetation is adapted to the variations of phosphate and ammonia levels. Hence, aquatic macrophytes can be used as bio-indicators of fluctuations in water nutrient levels in relation to the type of disturbance.  相似文献

We report the appearance of apoptotic cells in experimental myocardial infarction (rabbit heart) in in situ and in vitro preparations. Apoptosis was recognized by intravital staining with Hoechst 33342 (Ho342), by nick-end labeling (TUNEL) and by DNA laddering. A steady rise in the relative number of apoptotic cardiomyocytes (apoptotic index) was noted in in situ preparations. Apoptosis was first noted 6 h after the onset of ischemia with its highest value occurring after 72 h. Apoptotic nuclei were absent in remote areas of the left and right ventricles. Apoptotic nuclei within the infarcted area showed diminished intensity of Ho342 fluorescence. Three days after ischemia, a border zone adjacent to the infarcted area consisting of apoptotic macrophages was recognized. A novel finding was the appearance of apoptotic cardiomyocytes in the isolated perfused ischemic heart. Occurring as early as 50 min after the onset of ischemia, a high apoptotic index was present adjacent to the ligature placed around the coronary artery. This observation provides the opportunity to selectively examine factors leading to apoptosis in the ischemic heart under controlled experimental conditions.  相似文献

The detailed mechanisms underlying morphine-signaling pathways in platelets remain obscure. Therefore, we systematically examined the influence of morphine on washed human platelets. In this study, washed human platelet suspensions were used for in vitro studies. Furthermore, platelet thrombus formation induced by irradiation of mesenteric venules with filtered light in mice pretreated with fluorescein sodium was used for an in vivo thrombotic study. Morphine concentration dependently (0.6, 1, and 5 microM) potentiated platelet aggregation and the ATP release reaction stimulated by agonists (i.e., collagen and U46619) in washed human platelets. Yohimbine (0.1 microM), a specific alpha(2)-adrenoceptor antagonist, markedly abolished the potentiation of morphine in platelet aggregation stimulated by agonists. Morphine also potentiated phosphoinositide breakdown and intracellular Ca(2+) mobilization in human platelets stimulated by collagen (1 microg/ml). Moreover, morphine (0.6-5 microM) markedly inhibited prostaglandin E(1) (10 microM)-induced cyclic AMP formation in human platelets, while yohimbine (0.1 microM) significantly reversed the inhibition of cyclic AMP by morphine (0.6 and 1 microM) in this study. The thrombin-evoked increase in pH(i) was markedly potentiated in the presence of morphine (1 and 5 microM). Morphine (2 and 5 mg/g) significantly shortened the time require to induce platelet plug formation in mesenteric venules. We concluded that morphine may exert its potentiation in platelet aggregation by binding to alpha(2)-adrenoceptors in human platelets, with a resulting inhibition of adenylate cyclase, thereby reducing intracellular cyclic AMP formation followed by increased activation of phospholipase C and the Na(+)/H(+) exchanger. This leads to increased intracellular Ca(2+) mobilization, and finally potentiation of platelet aggregation and of the ATP release reaction.  相似文献
