Grass pea (Lathyrus sativus L.) is a legume crop known from its tolerance to various abiotic stresses, especially drought. In this study, we investigated: (1) the response of grass pea seedlings to osmotic stress generated in vitro by polyethylene glycol (PEG); (2) potential drought acclimatization mechanisms of two polish grass pea cultivars. Grass pea seeds of two cultivars were sown on media containing different PEG concentrations (0, 5.5, 11.0 mM) and cultivated for 14 days in controlled conditions. Plants’ dry matter increased under osmotic stress (regardless of PEG concentration). In turn, the highest dose of PEG caused a reduction in seedling growth in both cultivars. Furthermore, PEG caused the peroxidase activity increase in whole seedlings and catalase (CAT) activity in roots. However, differences between cultivars were noted in: CAT activity in shoots; while phenols and anthocyanin content as well as electrolyte leakage in shoots and roots. In turn, in both tested genotypes, accumulation of proline increased in shoots under osmotic stress. Obtained results indicate that the examined plants, although belonging to the same species, differ in acclimatization processes leading to elevated tolerance to osmotic stress.  相似文献

以酿酒葡萄‘雷司令’(Riesling)一年生营养袋扦插苗为材料,采用人工气候室水培试验,考察在聚乙二醇6000(PEG)模拟干旱条件下,不同浓度(0.05、0.10和0.20mg/L)24-表油菜素内酯(EBR)预处理对‘雷司令’幼苗活性氧、抗氧化物质、渗透调节物质含量和抗氧化酶活性的影响,以揭示EBR预处理对干旱胁迫下葡萄幼苗的抗旱机理。结果显示:(1)与正常生长(对照)相比,干旱胁迫显著提高葡萄幼苗叶片中超氧阴离子自由基(■)、过氧化氢(H_2O_2)和丙二醛(MDA)含量;与干旱胁迫处理(PEG)相比,不同浓度EBR预处理均可降低叶片中■、H_2O_2和MDA的含量。(2)与对照相比,PEG处理显著降低葡萄幼苗叶片的抗坏血酸(AsA)和还原型谷胱甘肽(GSH)含量;与PEG处理相比,各浓度EBR预处理均可显著提高葡萄叶片AsA与GSH的含量,且以0.10mg/LEBR处理效果最好。(3)随着干旱胁迫时间的延长,葡萄幼苗叶片中的超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化物酶(POD)与抗坏血酸过氧化物酶(APX)活性均呈先上升后下降的变化趋势,而在正常生长条件下酶活性基本保持不变;EBR预处理的葡萄叶片SOD、CAT、POD和APX活性均始终高于同期PEG处理。(4)PEG处理条件下,渗透调节物质脯氨酸和可溶性蛋白的含量整体高于对照;与PEG处理相比,不同浓度EBR预处理在干旱胁迫中后期均能显著提高葡萄叶片中脯氨酸和可溶性蛋白含量。研究表明,在干旱胁迫下,外源EBR预处理能够提高葡萄叶片抗氧化系统酶活性和渗透调节物质含量,有效降低干旱胁迫诱导的活性氧过度积累及膜脂过氧化程度,提高葡萄幼苗的抗旱能力,且以0.10mg/L EBR处理效果最佳。  相似文献

Flooding stress constrains crop growth and yield because most agricultural species are flood-sensitive. However, many of the plant species that live in permanently or temporarily flooded habitats have evolved specific traits to cope with these harsh conditions. Grass pea (Lathyrus sativus L.) is a legume that tolerates stresses such as drought, diseases, and pests; however, it is unclear whether grass pea has a tolerance mechanism for flooding stress. To understand if grass pea tolerates hypoxia and how it deals with hypoxic stress, the effects of hypoxia on root tip death, physiological, and morpho-anatomical alterations in grass pea and pea (Pisum sativum), which is sensitive to hypoxia, were compared. The results showed that activities of antioxidant enzymes, namely superoxide dismutase, catalase, ascorbate peroxidase, and glutathione content in grass pea were greater than in pea during hypoxia, which protected the root tip from oxidative damage and reduced ion leakage, which helped maintain membrane integrity. Furthermore, aerenchyma and lateral root development accompanied by ethylene production, moderate ROS accumulation-mediated cell death, and Ca2+ spatial-temporal heterogeneity developed well in grass pea compared to pea, which may not only facilitate internal gas diffusion but also promote removal of toxic by-products under hypoxic conditions. These results demonstrate that grass pea is more tolerant to hypoxic stress induced by flooding than garden pea seedlings. This discovery not only provides significant information for understanding the hypoxia-tolerant mechanisms in plants, but also promotes the usability of grass pea in flood-prone areas.  相似文献

Riboflavin is the precursor of the coenzymes flavin monophosphate (FMN) and flavin adenine dinucleotide (FAD), which serve as indispensable redox cofactors in all plants. Numerous data indicate that riboflavin is involved in pathogen resistance but less data are available on abiotic stress tolerance. In this experiment, the overexpression of the riboflavin-binding protein resulted in an enhancement of vegetative growth and net photosynthetic rate, and an acceleration of floral transition in transgenic Arabidopsis thaliana REAT11 (containing less than half the normal levels of free riboflavin, FMN, and FAD) compared to wild-type Col-0 under nonstressed conditions. The effect of drought stress on the antioxidant response of Col-0 and REAT11 was compared, where 20- and 40-day-old grown plants were subjected to 10 % PEG 6000 treatment for 2 days. Stress conditions caused a significant increase in H2O2 accumulation, lipid peroxidation, and membrane permeability in Col-0 over that in REAT11. Greater activity levels of superoxide dismutase, ascorbate peroxidase, and glutathione reductase were observed in the leaves of REAT11 compared to those of Col-0. Significant increases in total ascorbate and glutathione content and higher ratios of ASC/DHA: (ASC and DHA are reduced and oxidized ascorbate, respectively) and GSH/GSSG: (GSH and GSSG are reduced and oxidized glutathione, respectively) were observed in the leaves of REAT11 compared to those in Col-0 under drought conditions. In addition, enhancement of free proline and soluble sugar accumulation was observed in REAT11 compared to Col-0 under stress. Our results suggest that a slight deficiency in free riboflavin can paradoxically induce both a higher vegetative growth rate and an enhanced tolerance to drought in transgenic plants. The “stress escape” hypothesis is proposed here to explain this interesting phenomenon.  相似文献

Drought stress is a major threat to plant production in semi-arid and arid areas of the world. This research was laid out to asses the effects of sodium nitroprusside (SNP) as a nitric oxide donor on growth, physiological and biochemical changes of in vitro-cultured Allium hirtifolium under polyethylene glycol (PEG) induced drought stress. Basal plate explants of A. hirtifolium were cultured on MS medium containing different levels of PEG (0, 2, 4, 8 and 16 mM) and SNP (0, 10, 40 and 70 µM). After prolonged drought, growth responses, oxidative stress indicators, and phytochemical variations of regenerated plantlets with or without PEG and/or SNP treatments were recorded. Water limitation reduced regeneration potential of explants and consequently number of shoots per explant. Relative water content, total chlorophyll and carotenoid contents of regenerated A. hirtifolium plantlets decreased, but accumulation of malondialdehyde, H2O2 and proline and the activities of superoxide dismutase, ascorbate peroxidase, catalase and peroxidase enzymes increased with decreasing water availability. Total phenol and allicin contents were also increased in response to drought stress. Exogenous SNP in 10 and particularly in 40 µM was effective in enhancing regeneration rate and relative water content as well as protecting photosynthetic pigments under different levels of water availability. SNP also inhibited the hydrogen peroxide (H2O2) accumulation and lipid peroxidation in cell membranes via increasing the activities of superoxide dismutase and ascorbate peroxidase enzymes and accumulating proline and allicin. In general, these results suggest that exogenous SNP at 40 µM not only could somewhat protect A. hirtifolium from drought stress, but also can help to improve the propagation and allicin production of that plant under in vitro condition.  相似文献

Caper (Capparis ovata Desf.) is a perennial shrub (xerophyte) and drought resistant plant which is well adapted to Mediterranean Ecosystem. In the present study we investigated the plant growth, relative water content (RWC), chlorophyll fluorescence (FV/FM), lipid peroxidation (TBA-reactive substances content) as parameters indicative of oxidative stress and antioxidant enzymes such as superoxide dismutase (SOD), ascorbate peroxidase (APX), peroxidase (POX), catalase (CAT) and glutathione reductase (GR) in relation to the tolerance to polyethylene glycol mediated drought stress in C. ovata seedlings. For induction of drought stress, the 35 days seedlings were subjected to PEG 6000 of osmotic potential −0.81 MPa for 14 days. Lipid peroxidation increased in PEG stressed seedlings as compared to non-stressed seedlings of C. ovata during the experimental period. With regard to vegetative growth, PEG treatment caused decrease in shoot fresh and dry weights, RWC and FV/FM but decline was more prominent on day 14 of PEG treatment. Total activity of antioxidative enzymes SOD, APX, POX, CAT and GR were investigated in C. ovata seedlings under PEG mediated drought. Induced activities of SOD, CAT and POX enzymes were high and the rate of increment was higher in stressed seedling. APX activity increased on both days of PEG treatment, however, increase in GR activity was highest on day 14 of drought stress. We concluded that increased drought tolerance of C. ovata is correlated with diminishing oxidative injury by functioning of antioxidant system at higher rates under drought stress.  相似文献

Interrelationship among abscisic acid (ABA) content, accumulation of free polyamines and biosynthesis of beta-N-oxalyl-l-alpha,beta-diaminopropionic acid (ODAP) was studied in grass pea (Lathyrus sativus L.) seedlings under drought stress induced by 10% polyethylene glycol (PEG6000). Increase of ABA content occurred prior to that of ODAP and polyamine contents, and was found significantly positive correlation between ABA content and ODAP content. Addition of exogenous ABA increased ODAP content in leaves. On the other hand, pretreatment with alpha-difluoromethylarginine (DFMA), a polyamine biosynthesis inhibitor, significantly suppressed the accumulation of free putrescine (Put), free spermidine (Spd) and free spermine (Spm), which in turn inhibited biosynthesis of ODAP in well-watered leaves. Meanwhile, addition of exogenous Put alleviated DFMA-induced inhibition on the biosynthesis of Put and Spd, but did not affect the biosynthesis of Spm and ODAP in well-watered leaves. Same result was also achieved in drought-stressed leaves. Increasing accumulation of ODAP was significantly correlated with increasing Spm content (R=0.7957**) but not with that of Spd and Put. Therefore, it can be argued that ABA stimulated the biosynthesis of ODAP simultaneously with increasing the level of free Spm under drought stress condition.  相似文献

Drought stress usually causes a serious yield reduction in wheat production. Silicon (Si) has been reported to be able to alleviate drought stress damage; however, the mechanism is still poorly understood. In this article, the effects of Si (as sodium silicate) on some parameters related to oxidative damage, proline, soluble sugar, and inorganic ions in the leaves of wheat under 20% (w/v) polyethylene glycol (PEG-6000) simulative drought stress are investigated. PEG stress depressed the growth of shoot and root and decreased leaf water potential and chlorophyll concentration. Addition of 1.0 mM Si could partially improve the growth of shoot (but not root) and increase the leaf chlorophyll concentrations of stressed plants. Inclusion of Si in culture solution also maintained leaf water potential of stressed plants at the same level as that of the control plants. PEG stress induced significant accumulation of leaf hydrogen peroxide (H2O2) and malondialdehyde (MDA) as well as an increase in electrolyte leakage, which were all decreased by added silicon. These results suggest that stress-induced membrane lipid peroxidation could be partly alleviated by added silicon. Moreover, the results were also supported by the observation that PEG stress-induced decrease in glutathione concentration in the leaves was reversed by added silicon. The proline concentration in the leaves was markedly increased under PEG stress, whereas added silicon partially reversed this. PEG stress decreased the leaf soluble sugar concentration. There were significant negative regressions between proline concentration and both shoot dry weight and leaf chlorophyll concentrations, whereas there were positive regressions between the proline concentration and both H2O2 and MDA concentrations in the leaves, supporting the view that proline accumulation is a symptom of stress damage rather than stress tolerance. Addition of Si obviously increased Si accumulation in the shoot. Analyses of Na, Mg, K, and Ca showed no accumulation of these ions in the shoot (on the basis of per tissue dry weight) under water stress, and added Si even decreased their concentrations. These results suggest that under short-term PEG-induced water stress conditions (1 week), antioxidant defense, rather than osmotic adjustment, contributed to the improved wheat growth by Si.  相似文献

为了解干旱对五唇兰(Phalaenopsis pulcherrima)生长的影响,以聚乙二醇(PEG)溶液模拟干旱胁迫,对其叶片的光合色素、渗透调节物质和非结构碳水化合物(NSC)含量变化进行研究。结果表明,随着PEG浓度增加,五唇兰植株含水量和鲜质量逐渐下降,以PEG为13.75%~14.84%时最显著。PEG处理显著降低叶片的叶绿素a和b含量。随着植株含水量的降低,叶片可溶性蛋白、淀粉(St)含量均呈下降趋势,可溶性糖(SS)含量、NSC和SS/St均呈先升后降的趋势。因此,干旱胁迫会影响五唇兰植株的含水量和光合产物的积累;在较低程度干旱胁迫下,可溶性糖在抗旱响应中发挥主要作用;随着干旱胁迫程度加深,五唇兰的生理代谢受到严重影响。  相似文献

The responses of the antioxidant defense system in plant species to drought stress are still relatively unknown. In order to further understand how the system responds to drought stress, the leaves of Fargesia denudata seedlings were investigated. Antioxidant enzyme activities, antioxidant contents, hydrogen peroxide (H2O2), superoxide anion (O 2 ·? ) and MDA contents in the seedling leaves were measured under well-watered (WW), moderate drought-stressed (MD), and severe drought-stressed (SD) treatments. Although drought stress significantly increased H2O2 and O 2 ·? levels in F. denudata leaves, only weak lipid peroxidation was observed. This is attributed to the higher superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), glutathione reductase (GR), monodehydroascorbate reductase (MDHAR), and dehydroascorbate reductase (DHAR) activities in F. denudata leaves during the entire drought period. Reduced and oxidized ascorbate (AsA and DHA) contents were almost not affected by drought except that DHA under SD showed an obvious increase on day 30. Furthermore, reduced glutathione (GSH) content under drought stress significantly decreased, while oxidized glutathione (GSSG) markedly increased under SD on days 30 and 45 as well as under MD on day 30; as a result, the ratio GSH/GSSG declined considerably. These results indicated that GSH was involved in scavenging H2O2 and O 2 ·? under drought stress and it was more sensitive to drought stress in scavenging H2O2 and O 2 ·? than AsA. As a result, a highly efficient antioxidant defense system in drought-stressed F. denudate leaves operated mainly through the synergistic functioning of SOD, CAT, APX, MDHAR, DHAR, GR, and GSH against oxidative damage.  相似文献

Seedling stage is a critical period for survival and growth under drought stress. In the current study, we determined effects of drought stress on physiological and biochemical parameters of leaves and roots of Lycium ruthenicum Murr. seedling. The variables measured were lipid peroxidation (in terms of malondialdehyde (MDA) content), osmotic substances (free proline, soluble protein, and soluble sugar), and antioxidative enzymes (peroxidase (POD), superoxide dismutase (SOD), and catalase (CAT)). Free proline, soluble sugar, and MDA of leaves and roots increased with increasing stress level. Leaves displayed higher accumulations of free proline and MDA than roots. However, roots showed higher total soluble sugar than leaves. Under drought stress, soluble proteins in leaves and roots decreased initially and then increased. Meanwhile, measured proteins were higher in leaves. Under drought stress, SOD, POD, and CAT activities in leaves increased initially and then decreased but increased with increasing drought stress level in roots. Under drought the level of accumulation of osmotics was higher in the leaves than in the roots, while increased activity of antioxidant enzymes persisted in the stressed roots longer that in the leaves.  相似文献

A factorial experiment based on RCB design with three replicates was conducted to investigate changes in some physiological responses of two chickpea (Cicer arietinum L.) cultivars (Pirouz from Desi type and ILC482 from Kabuli type) to arbuscular mycorrhiza (Glomus etunicatum Becker and Gerdman) under different irrigation treatments. The experiment was carried out in the greenhouse of the Agricultural Faculty of Kurdistan University from April to August 2009. The results showed that leaf chlorophyll content of chickpea cultivars was significantly increased by arbuscular mycorrhiza (AM) under both well and limited irrigation conditions. Proline accumulation in chickpea leaves under moderate and severe drought stresses was significantly stronger than that under optimum irrigation. Inoculation of chickpea with mycorrhizal fungi caused an increase in the activities of polyphenol oxidase and peroxidase, but a decrease in the activity of catalase. Comparisons among different irrigation levels showed that chickpea plants under drought stress had the most active lipid peroxidation. Non-AM plants showed stronger lipid peroxidation under moderate and severe water stresses than AM plants. Lipid peroxidation was more active in Pirouz leaves than in ILC482 leaves. It seems that Kabuli-type cultivar responded better to mycorrhizal symbiosis under drought stress than Desitype cultivar.  相似文献

This study evaluates the role of exogenous foliar application of 5-aminolevulinic acid (ALA) on water relations, gas exchange, chlorophyll fluorescence, and the activities and gene expression patterns of antioxidant enzymes in leaves of oilseed rape under drought stress and recovery conditions. Seedlings at four-leaf stage were imposed to well-watered condition (80 % of water-holding capacity) or drought stress (40 % of water-holding capacity) and subsequently foliar sprayed with water or ALA (30 mg l?1). Drought suppressed the accumulation of plant biomass and decreased chlorophyll content and leaf water status (relative water content and water potential). The actual quantum yield of photosystem II and electron transport rates were hampered in parallel to net photosynthetic rate. However, drought stress induced the accumulation of malondialdehyde (MDA) and hydrogen peroxide, enhanced the activities of catalase (CAT), peroxidase (POD), ascorbate peroxidase (APX), glutathione reductase (GR) and superoxide dismutase and up-regulated the expression of APX and GR. After rehydration for 4 days, the growth of drought-treated seedlings was restored to normal level for most of the physiological parameters. Foliar application of ALA maintained relatively higher leaf water status and enhanced chlorophyll content, net photosynthetic rate, actual quantum yield of photosystem II, photochemical quenching, non-photochemical quenching and electron transport rates in stressed leaves. Exogenous ALA also alleviated the accumulation of MDA and hydrogen peroxide, increased the activities of antioxidant enzymes and enhanced the expression of CAT and POD in drought-treated plants. These results indicate that ALA may effectively protect rapeseed seedlings from damage induced by drought stress.  相似文献

Seedlings of grass pea (Lathyrus sativus L.) were treated with polyethylene glycol (PEG), in addition with putrescine (Put) and α-difluoromethylarginine (DFMA) in order to study the relation between polyamine metabolism and accumulation of β-N-oxalyl-L-α, β-diaminopropionic acid (ODAP) under water stress. The results indicated that Put, spermidine (Spd) and spermine (Spm) contents of leaves gradually increased along with the extension of PEG-treatment time. Especially, the increase in Spm contents was significant together with a gradual increase of ODAP contents. Put and Spd contents of leaves markedly increased along with the extension of PEG+Put treatment time. There was little effect on Spm contents and ODAP contents in leaves with PEG+Put treatment. The accumulation of Put, Spd, Spm and ODAP in leaves was inhibited by PEG+DFMA treatment. The inhibition of DFMA for Put and Spd biosynthesis was partially overcome by adding exogenous Put, but little effect for the inhibition of DFMA on Spm biosynthesis by adding exogenous Put. The accumulation of ODAP was inhibited with PEG+DFMA+Put treatment in leaves. This indicated that there was a relationship between the increase of Spm contents and the accumulation of ODAP in leaves under water stress.  相似文献

To ascertain the effect of exogenously applied hydrogen peroxide (H2O2) on drought stress, we examined whether the spraying of soybean leaves with H2O2 would alleviate the symptoms of drought stress. Pre-treatment by spraying leaves with H2O2 delayed foliar wilting caused by drought stress compared to leaves sprayed with distilled water (DW). Additionally, the relative water content of drought-stressed leaves pre-treated with H2O2 was higher than that of leaves pre-treated with DW. Therefore, we analyzed the effect of H2O2 spraying on photosynthetic parameters and on the biosynthesis of oligosaccharides related to water retention in leaves during drought stress. Under conditions of drought stress, the net photosynthetic rate and stomatal conductance of leaves pre-treated with H2O2 were higher than those of leaves pre-treated with DW. In contrast to DW spraying, H2O2 spraying immediately caused an increase in the mRNA levels of d-myo-inositol 3-phosphate synthase 2 (GmMIPS2) and galactinol synthase (GolS), which encode key enzymes for the biosynthesis of oligosaccharides known to help plants tolerate drought stress. In addition, the levels of myo-inositol and galactinol were higher in H2O2-treated leaves than in DW-treated leaves. These results indicated that H2O2 spraying enabled the soybean plant to avoid drought stress through the maintenance of leaf water content, and that this water retention was caused by the promotion of oligosaccharide biosynthesis rather than by rapid stomatal closure.  相似文献

A drought stress-responsive Cys2/His2-type zinc finger protein gene DgZFP3 was previously isolated (Liu et al., Afr J Biotechnol 11:7781–7788, 2012b) from chrysanthemum. To assess roles of DgZFP3 in plant drought stress responses, we performed gain-of-function experiment. The DgZFP3-overexpression tobacco plants showed significant drought tolerance over the wild type (WT). The transgenic lines exhibited less accumulation of H2O2 under drought stress, more accumulation of proline and greater activities of peroxidase (POD) and superoxide dismutase than the WT under both control conditions and drought stress. In addition, there was greater up-regulation of the ROS-related enzyme genes (NtSOD and NtPOD) and stress-related genes (NtLEA5 and NtDREB) in transgenic lines under normal or drought conditons. Thus DgZFP3 probably plays a positive regulatory role in drought stress response and has the potential to be utilized in transgenic breeding to improve drought stress tolerance in plants.  相似文献

The study on Stevia callus has the potential to advance the knowledge of antioxidant mechanisms involved in unorganized cells response to drought stress. The effects of polyethylene glycol (PEG; 0 and 4% w/v) in combination with paclobutrazol (PBZ; 0 and 2 mg l?1) and gibberellin (GA; 0 and 2 mg l?1) were studied on Stevia rebaudiana callus. PEG treatment led to an oxidative stress, as indicated by increased H2O2 content whose accumulation was prevented with PBZ and GA treatments. All treatments of PEG, PBZ and GA increased the total antioxidant capacity, with the highest antioxidant power in PBZ and GA treatments without PEG. The activity of superoxide dismutase, catalase and ascorbate peroxidase significantly increased in PEG treatment alone or in combination with PBZ and GA. All treatments of PEG, PBZ and GA significantly increased proteins, amino acids and proline contents, with the highest increase in presence of PBZ in medium culture. In contrary to proline, the activity of pyrroline-5-carboxylate synthetase and proline dehydrogenase did not change in response to any of the treatments. Collectively, our results demonstrated that PBZ and GA increased reactive oxygen species scavenging and osmolytes in PEG-treated calli more than PEG treatment alone to alleviate negative effects of PEG on Stevia calli. These findings will enable us to design effective genetic engineering strategies in callus culture to generate some somaclonal variation that may be useful in enhancing drought resistance in Stevia.  相似文献

Drought resistance is increased in plants by the absence of the hormone gibberellic acid (GA) or by a lack of GA sensitivity. We studied the effects of tissue-specific reduction in GA levels on drought tolerance, on recovery from drought stress, and on primary and secondary growth using transgenic tobacco plants expressing the GA-inactivating gene PtGA2ox 1 (GA 2-oxidase) specifically in leaves, stems, or roots. Localized reduction of bioactive GA1 levels was achieved by tissue-specific expression of the PtGA2ox 1 gene in leaves using the rbcs promoter (LD plants), in roots using the TobRB7 promoter (RD plants), and in stems using the LMX5 promoter (SD plants). In response to drought stress, all transgenic tobacco plants exhibited reduced primary and secondary growth and increased drought tolerance with a corresponding reduction in malondialdehyde levels, higher relative water content, increased proline and sugar content, and elevated peroxidase, superoxide dismutase, and catalase activities relative to wild-type plants. The highest level of drought tolerance and the most rapid recovery from stress was achieved by localized reduction of GA1 in the roots of the RD transgenic plants. In addition, although the total bioactive GA1 content in RD and LD plants was essentially identical, the heights of LD plants were significantly greater and drought tolerance was significantly less than in RD plants.
