Two-month-old healthy seedlings of a true mangrove, Bruguiera parviflora, raised from propagules in normal nursery conditions were subjected to varying concentrations of NaCl for 45 d under hydroponic culture conditions to investigate the defence potentials of antioxidative enzymes against NaCl stress imposed oxidative stress. Changes in the activities of the antioxidative enzymes catalase (CAT), ascorbate peroxidase (APX), guaiacol peroxidase (POX), glutathione reductase (GR) and superoxide dismutase (SOD) were assayed in leaves to monitor the temporal regulation. Among the oxidative stress triggered chemicals, the level of H2O2 was significantly increased while total ascorbate and total glutathione content decreased. The ratio of reduced to oxidized glutathiones, however, increased due to decreased levels of oxidized glutathione in the leaf tissue. Among the five antioxidative enzymes monitored, the APX, POX, GR and SOD specific activities were significantly enhanced at high concentration (400 mM NaCl), while the catalase activities declined, suggesting both up and downregulations of antioxidative enzymes occurred due to NaCl imposed osmotic and ionic stress. Analysis of the stress induced alterations in the isoforms of CAT, APX, POX, GR and SOD revealed differential regulations of the isoforms of these enzymes. In B. parviflora one isoform of each of Mn-SOD and Cu/Zn-SOD while three isoforms of Fe-SOD were observed by activity staining gel. Of these, only Mn-SOD and Fe-SOD2 content was preferentially elevated by NaCl treatment, whereas isoforms of Cu/Zn-SOD, Fe-SOD1 and Fe-SOD3 remained unchanged. Similarly, out of the six isoforms of POX, the POX-1,-2,-3 and -6 were enhanced due to salt stress but the levels of POX-4 and -5 remained same as in control plants suggesting preferential upregulation of selective POX isoforms. Activity staining gel revealed only one prominent band of APX and this band increased with increased salt concentration. Similarly, two isoforms of GR (GR1 and GR2) were visualized on activity staining gel and both these isoforms increased upon salt stress. In this mangrove four CAT-isoforms were identified, among which the prominent CAT-2 isoform level was maximally reduced again suggesting differential downregulation of CAT isoforms by NaCl stress. The results presented in this communication are the first report on the resolutions of isoforms APX, POX and GR out of five antioxidative enzymes studied in the leaf tissue of a true mangrove. The differential changes in the levels of the isoforms due to NaCl stress may be useful as markers for recognizing salt tolerance in mangroves. Further, detailed analysis of the isoforms of these antioxidative enzymes is required for using the various isoforms as salt stress markers. Our results indicate that the overproduction of H2O2 by NaCl treatment functions as a signal of salt stress and causes upregulation of APX, POX, GR and deactivations of CAT in B. parviflora. The concentrations of malondialdehyde, a product of lipid peroxidation and lipoxygenase activity remained unchanged in leaves treated with different concentrations of NaCl, which again suggests that the elevated levels of the antioxidant enzymes protect the plants against the activated oxygen species thus avoiding lipid peroxidation during salt stress.  相似文献

Chickpea plants were subjected to salt stress for 48 h with 100 mM NaCl, after 50 days of growth. Other batches of plants were simultaneously treated with 0.2 mM sodium nitroprusside (NO donor) or 0.5 mM putrescine (polyamine) to examine their antioxidant effects. Sodium chloride stress adversely affected the relative water content (RWC), electrolyte leakage and lipid peroxidation in leaves. Sodium nitroprusside and putrescine could completely ameliorate the toxic effects of salt stress on electrolyte leakage and lipid peroxidation and partially on RWC. No significant decline in chlorophyll content under salt stress as well as with other treatments was observed. Sodium chloride stress activated the antioxidant defense system by increasing the activities of peroxidase (POX), catalase (CAT) superoxide dismutase (SOD) and ascorbate peroxidase (APX). However no significant effect was observed on glutathione reductase (GR) and dehydro ascorbate reductase (DHAR) activities. Both putrescine and NO had a positive effect on antioxidant enzymes under salt stress. Putrescine was more effective in scavenging superoxide radical as it increased the SOD activity under salt stress whereas nitric oxide was effective in hydrolyzing H2O2 by increasing the activities of CAT, POX and APX under salt stress.  相似文献

When rice seedlings grown for 10 and 20 days were subjected to in vitro drought stress of −0.5 and −2.0 MPa for 24 h, an increase in the concentration of superoxide anion (O2.−), increased level of lipid peroxidation and a decrease in the concentration of total soluble protein and thiols was observed in stressed seedlings compared to controls. The concentration of H2O2 as well as ascorbic acid declined with imposition of drought stress, however glutathione (GSH) concentration declined only under severe drought stress. The activities of total superoxide dismutases (SODs) as well as ascorbate peroxidase (APX) showed consistent increases with increasing levels of drought stress, however catalase activity declined. Mild drought stressed plants had higher guaiacol peroxidase (GPX) and chloroplastic ascorbate peroxidase (c-APX) activity than control grown plants but the activity declined at the higher level of drought stress. The activities of enzymes involved in regeneration of ascorbate i.e. monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR) and glutathione reductase (GR) were higher in drought stressed plants compared to controls. Results suggest that drought stress induces oxidative stress in rice plants and that besides SOD, the enzymes of ascorbate-glutathione cycle, which have not been studied in detail earlier under stressful conditions, appear to function as important component of antioxidative defense system under drought stress.  相似文献

This study investigated oxidative stress and the antioxidant response to boron (B) of chickpea cultivars differing in their tolerance to drought. Three‐week‐old chickpea seedlings were subjected to 0.05 (control), 1.6 or 6.4 mm B in the form of boric acid (H3BO3) for 7 days. At the end of the treatment period, shoot length, dry weight, chlorophyll fluorescence, B concentration, malondialdehyte content and the antioxidant enzymes superoxide dismutase (SOD), peroxidase (POX), catalase (CAT), ascorbate peroxidase (APX) and glutathione reductase (GR) were measured. The 1.6 mm B treatment did not cause significant changes in shoot length of cultivars, although shoot length increased in the drought‐tolerant Gökce and decreased in the drought‐sensitive Küsmen after 6.4 mm B treatment. Dry weights of both cultivars decreased with 6.4 mm B treatment. Chlorophyll fluorescence (Fv/Fm) did not change in Gökce at either B level. Nor did it change in Küsmen with 1.6 mm B but Fv/Fm decreased with 6.4 mm B. Boron concentration in the shoots of both cultivars increased significantly with increasing levels of applied B. Significant increases in total SOD activity were observed in shoots of both cultivars given 1.6 and 6.4 mm B. Shoot extracts exhibited five activity bands, two of which were identified as MnSOD and Cu/ZnSOD. In comparison to the control group, all enzyme activities (except APX and SOD) decreased with 1.6 mm B stress. GR activity decreased, while activities of CAT, POX and APX did not change with 6.4 mm B in Küsmen. On the other hand, activities of CAT, APX and SOD increased in Gökce at both B levels. In addition, lipid peroxidation was higher in Küsmen than in Gökce, indicating more damage by B to membrane lipids in the former cultivar. These results suggest that (i) Gökce is tolerant and Küsmen is sensitive to B, and (ii) B tolerance of Gökce might be closely related to increased capacity of the antioxidative system (total SOD, CAT and APX) to scavenge reactive oxygen species and thus suppress lipid peroxidation under B stress. To the best of our knowledge, this is the first report on the antioxidant response of chickpea seedlings to B toxicity.  相似文献

The combined effects of drought and low light on biomass partition, foliar nitrogen concentration, membrane stability and active oxygen species (AOS) and antioxidant system were investigated in dragon spruce (Picea asperata Mast.) seedlings grown at two watering regimes (well-watered, 100% of field capacity and drought, 30% of field capacity) and light availabilities (HL, 100% of full sunlight and low light, 15% of full sunlight). Under high light condition drought not only reduced foliar nitrogen concentration (Nmass) and membrane stability index (MSI) but also significantly increased biomass partitioning to roots, AOS, ascorbic acid (AsA) content and antioxidant enzyme activities including superoxide dismutase (SOD, EC 1.15.1.1), peroxidase (POD, EC 1.11.1.7), catalase (CAT, EC 1.11.1.6), ascorbate peroxidase (APX, EC 1.11.1.11) and glutathione reductase(GR, EC 1.6.4.2). However, no prominently drought-induced differences in biomass partitioning to root, SOD, GR activities, hydrogen peroxide (H2O2) and MSI were observed in low light seedlings. On the other hand, significant interaction of drought and low light was found on MSI, the antioxidant enzymes activities (SOD, POD, CAT, APX, GR), H2O2 and superoxide radical (O2 ). Two cultivars of peanut (Arachis hypogaea L.) which were designated as resistant (Florispan) and sensitive (Gazipasa) according to their growth retardation under drought stress conditions were compared for their oxidative damage and antioxidant responses. Sixteen days-old peanut seedlings were subjected to PEG-6000 solutions of two different osmotic potentials; −0.4 and −0.8 MPa, and various growth parameters, photosystem II activity, changes in malondialdehyde (MDA), hydrogen peroxide (H2O2) and proline levels, activities of ascorbate peroxidase (APX), catalase (CAT), peroxidase (POX) and gluthatione reductase (GR) enzymes were determined. Both cultivars exhibited water deficit at −0.8 MPa osmotic potential of PEG-6000 and H2O2 levels significantly increased during exposure to −0.4 MPa osmotic potential. However, H2O2 levels were under control in both cultivars at exposure to −0.8 MPa osmotic potential. Significant proline accumulation was observed in the tissues of cv. Florispan at −0.8 MPa osmotic potential, whereas proline accumulation did not appear to be an essential part of the protection mechanism against drought in cv. Gazipasa. No significant variation in chlorophyll fluorescence values were detected in neither of the cultivars. Enzyme activity measurements revealed that Gazipasa copes well with lesser magnitudes of drought stress by increasing the activity of mainly APX, and during harsh stress conditions, only APX maintains its activity in the tissues. In cultivar Florispan, GR activity appears to take role in lesser magnitudes of drought stress, whereas CAT and APX activities appear to be very crucial antioxidative defenses during intense stress conditions. The results indicate that, the level of proline and activities of the enzymes CAT and APX are important mechanisms for the maintenance of drought tolerance in peanut plants.  相似文献

用20%聚乙二醇(PEG 6000)研究了渗透胁迫对黑麦(Secale cereale L.)幼苗活性氧(reactive oxygen species, ROS)和主要抗氧化酶—— 超氧化物歧化酶(superoxide dismutase, SOD)、过氧化氢酶(catalase, CAT)、抗坏血酸过氧化物酶(ascorbate peroxidase, APX)和谷胱甘肽还原酶(glutathione reductase, GR)活性的影响。结果表明, 与对照相比, PEG处理明显提高了叶子和根中丙二醛(malondialdehyde, MDA)的含量、ROS的水平和以上4种抗氧化酶的活性。渗透胁迫下,叶子和根中MDA和ROS水平变化的规律基本相似, 但抗氧化酶活性在2种器官中表现不完全相同, 叶子中CAT的活性在对照和处理中无显著差异, 但在根中差异明显, 表明叶子中SOD、APX和GR在植物应答渗透胁迫中起重要作用, 而根中这4种抗氧化酶都参与植物对胁迫的反应。GR活性随PEG处理变化幅度显著高于其它抗氧化酶, 表明GR在黑麦应答渗透胁迫中所起作用可能强于其它抗氧化酶。  相似文献

Antioxidant enzymes are related to the resistance to various abiotic stresses including salinity. Barley is relatively tolerant to saline stress among crop plants, but little information is available on barley antioxidant enzymes under salinity stress. We investigated temporal and spatial responses of activities and isoform profiles of superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), non-specific peroxidase (POX), and glutathione reductase (GR) to saline stress in barley seedlings treated with 200 mM NaCl for 0, 1, 2, 5 days, respectively. In the control plant, hydrogen peroxide content was about 2-fold higher in the root than in the shoot. Under saline stress, hydrogen peroxide content was decreased drastically by 70% at 2 d after NaCl treatment (DAT) in the root. In the leaf, however, the content was remained unchanged by 2 DAT and increased about 14 % at 5 DAT. In general, the activities of antioxidant enzymes were increased in the root and shoot under saline stress. But the increase was more significant and consistent in the root. The activities of SOD, CAT, APX, POX, and GR were increased significantly in the root within 1 DAT, and various elevated levels were maintained by 5 DAT. Among the antioxidant enzymes, CAT activity was increased the most drastically. The significant increase in the activities of SOD, CAT, APX, POX, and GR in the NaCl-stressed barley root was highly correlated with the increased expression of the constitutive isoforms as well as the induced ones. In order to observe the possible regulatory role of selenium (Se) in relation to the changes in ascorbate (AsA) glutathione (GSH) levels and to the activities of antioxidant and glyoxalase pathway enzymes, rapeseed (Brassica napus) seedlings were grown in Petri dishes. A set of 10-day-old seedlings was pretreated with 25 μM Se (Sodium selenate) for 48 h. Two levels of drought stress (10% and 20% PEG) were imposed separately as well as on Se-pretreated seedlings, which were grown for another 48 h. Drought stress, at any level, caused a significant increase in GSH and glutathione disulfide (GSSG) content; however, the AsA content increased only under mild stress. The activity of ascorbate peroxidase (APX) was not affected by drought stress. The monodehydroascorbate reductase (MDHAR) and glutathione reductase (GR) activity increased only under mild stress (10% PEG). The activity of dehydroascorbate reductase (DHAR), glutathione S-transferase (GST), glutathione peroxidase (GPX), and glyoxalase I (Gly I) activity significantly increased under any level of drought stress, while catalase (CAT) and glyoxalase II (Gly II) activity decreased. A sharp increase in hydrogen peroxide (H2O2) and lipid peroxidation (MDA content) was induced by drought stress. On the other hand, Se-pretreated seedlings exposed to drought stress showed a rise in AsA and GSH content, maintained a high GSH/GSSG ratio, and evidenced increased activities of APX, DHAR, MDHAR, GR, GST, GPX, CAT, Gly I, and Gly II as compared with the drought-stressed plants without Se. These seedlings showed a concomitant decrease in GSSG content, H2O2, and the level of lipid peroxidation. The results indicate that the exogenous application of Se increased the tolerance of the plants to drought-induced oxidative damage by enhancing their antioxidant defense and methylglyoxal detoxification systems.  相似文献

Rice ( Oryza sativa L.) cv. Tulsi is recommended for Eastern India, for upland ecological cultivation systems where a crop experiences natural cycles of water deficit and water sufficiency, depending upon the monsoon rains. In this experiment, this cultivar was subjected to three cycles of water stress of increasing stress intensity. Each stress cycle was terminated by rewatering the plants for a 48-h period. The level of stress was measured by quantification of H2O2. The response of antioxidant metabolites such as ascorbate and glutathione, and enzymes such as superoxide dismutase (SOD, EC 1.15.1.1), catalase (CAT, EC 1.11.1.6), ascorbate peroxidase (APX, EC 1.11.1.11), glutathione reductase (GR, EC 1.6.4.2) and guaiacol peroxidase (POX, EC 1.11.1.7) was analysed in terms of activity and isozyme pattern for each cycle of stress and recovery. The differential response of the antioxidant enzymes with increasing stress intensity followed by recovery, highlight the different role of each in the drought acclimation process of upland rice. SOD and POX activity in stressed plants was higher than the controls in all the three cycles. The second level of stress saw an increase in all the enzymes with APX and GR showing its maximum activity and there was a better management of H2O2 levels. There was an induction of a new CAT isoform in stressed plants in the third cycle of water stress. The co-ordinated defense helped the plants to recover in terms of growth on rewatering after stress cycles.  相似文献

In order to elucidate the response of the ascorbate-glutathione (ASC-GSH) cycle to drought stress, the activities of antioxidant enzymes and the levels of molecules involved in the ASC-GSH metabolism were studied in Trifolium repens L. seedlings subjected to PEG-induced water deficit. Compared to the control, the contents of H2O2, thiobarbituric acid reactive substances (TBARS), ascorbate (ASC), dehydroascorbate (DHA), and glutathione disulfide (GSSG) increased in PEG-treated seedlings, whereas the glutathione (GSH) content kept constant during the drought period. Further more, the ASC/DHA and GSH/GSSG ratios decreased in the presence of PEG. Except for that of monodehydroascorbate reductase (MDHAR), the activities of ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR), and glutathione reductase (GR) were up-regulated during water deficit, and the increases in APX and DHAR activities were much higher than those in GR activity. These data indicate that fluctuations in the ASC-GSH metabolism resulted from PEG treatment may have a positive effect on drought stress mitigation in T. repens.  相似文献

以酿酒葡萄‘雷司令’(Riesling)一年生营养袋扦插苗为材料,采用人工气候室水培试验,考察在聚乙二醇6000(PEG)模拟干旱条件下,不同浓度(0.05、0.10和0.20mg/L)24-表油菜素内酯(EBR)预处理对‘雷司令’幼苗活性氧、抗氧化物质、渗透调节物质含量和抗氧化酶活性的影响,以揭示EBR预处理对干旱胁迫下葡萄幼苗的抗旱机理。结果显示:(1)与正常生长(对照)相比,干旱胁迫显著提高葡萄幼苗叶片中超氧阴离子自由基(■)、过氧化氢(H_2O_2)和丙二醛(MDA)含量;与干旱胁迫处理(PEG)相比,不同浓度EBR预处理均可降低叶片中■、H_2O_2和MDA的含量。(2)与对照相比,PEG处理显著降低葡萄幼苗叶片的抗坏血酸(AsA)和还原型谷胱甘肽(GSH)含量;与PEG处理相比,各浓度EBR预处理均可显著提高葡萄叶片AsA与GSH的含量,且以0.10mg/LEBR处理效果最好。(3)随着干旱胁迫时间的延长,葡萄幼苗叶片中的超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化物酶(POD)与抗坏血酸过氧化物酶(APX)活性均呈先上升后下降的变化趋势,而在正常生长条件下酶活性基本保持不变;EBR预处理的葡萄叶片SOD、CAT、POD和APX活性均始终高于同期PEG处理。(4)PEG处理条件下,渗透调节物质脯氨酸和可溶性蛋白的含量整体高于对照;与PEG处理相比,不同浓度EBR预处理在干旱胁迫中后期均能显著提高葡萄叶片中脯氨酸和可溶性蛋白含量。研究表明,在干旱胁迫下,外源EBR预处理能够提高葡萄叶片抗氧化系统酶活性和渗透调节物质含量,有效降低干旱胁迫诱导的活性氧过度积累及膜脂过氧化程度,提高葡萄幼苗的抗旱能力,且以0.10mg/L EBR处理效果最佳。  相似文献

水分胁迫下,柚树苗叶片RWC、ψw、Pn及可溶性蛋白质,叶绿素含量下降,游离脯氨酸、MDA含量及SOD活性升高,CAT活性先上升后下降,APX活性、AsA和GSH含量明显下降,抗旱性强的品种具有较高的活性氧清除能力。CPPU处理提高柚树苗叶片RWC、Pn、叶绿素、游离脯氨酸、AsA、GSH含量,并增加SOD、APX活性,降低MDA水平,从而提高柚树苗抗旱性。实验结果表明,CPPU处理提高叶片对膜脂过氧化作用的保护能力可能是其提高柚树苗抗旱性的原因。  相似文献

To understand the functions of antioxidant enzymes during leaf development in sweetpotato, we investigated the activities of several antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POX), ascorbate peroxidase (APX) and catalase (CAT). Significant increases were observed in the activities of SOD, POX and APX during the late stage of leaf development, whereas CAT activity increased during the early developmental stage. By RT-PCR analysis, various POX and APX genes showed differential expression patterns during leaf development. Four POX genes swpa3, swpa4, swpa6, swpb4 and one APX gene swAPX1 exhibited high levels of gene expression during the senescence stage of leaf development, but two POX genes, swpa1 and swpa7 were preferentially expressed at both the mature green and the late senescence stages of leaf development. These results indicate that hydrogen peroxide (H2O2)-related antioxidant enzymes are differentially regulated in the process of leaf development of sweetpotato.  相似文献

Effects of the antioxidant system and chlorophyll fluorescence on drought tolerance of four common bean (Phaseolus vulgaris L.) cultivars were studied. The cultivars were positioned in the order of a decrease in their drought tolerance: Yakutiye, Pinto Villa, Ozayse, and Zulbiye on the basis of changes in the water potential, stomatal conductance, photosynthetic pigment content, and lipid peroxidation. Under drought conditions, the level of H2O2 was not changed in cv. Pinto Villa but decreased in other cultivars. Antioxidant enzymes (superothide dismutase (SOD), guaiacol peroxidase (GPX), catalase (CAT), ascorbate peroxidase (APX), and glutathione reductase (GR)) were generally activated in all cultivars. Interestingly, CAT, APX, and GR activities were not changed in cv. Pinto Villa, APX activity decreased in cv. Yakutiye, and CAT activity was not changed in cv. Zulbiye. The increases in SOD and GPX activities in cv. Ozayse were higher than in other cultivars. Drought stress reduced the effective quantum yield of PS2 (ΦPS2) and the photochemical quenching (qp), while it increased nonphotochemical quenching (NPQ) in all cultivars. The reduction or increase was more pronounced in cv. Zulbiye. There were generally significant correlations between qp, NPQ, and ROS scavenging by SOD and APX. Also, there were significant correlations between SOD and qp in tolerant cultivars and APX and qp in sensitive ones. The results indicate that activation of SOD and APX was closely related to the efficiency of PS2 in common bean cultivars. This interaction was essential for protection of photosystems and plant survival under drought.  相似文献

We studied the effect of water stress imposed at anthesis and pre-anthesis stages on oxidative stress and antioxidant activity in four wheat cultivars, two hexaploid Triticum aestivum cultivars, drought resistant cv. C 306 and drought susceptible cv. Hira, and two tetraploid cultivars, T. durum cv. A 9-30-1 and T. dicoccum cv. HW 24. Water stress decreased relative water content (RWC), membrane stability index (MSI), and increased H2O2 and malondialdehyde (MDA) contents as well as activity of superoxide dismutase (SOD), catalase (Cat) and peroxidase (POX) in all the genotypes at all the stages. Both the tetraploid cultivars showed higher RWC, MSI and SOD activity, and lower H2O2 and MDA contents under water stress than hexaploid ones. Cat and POX activities were highest in C 306.  相似文献

The effect of exogenously applied glycinebetaine (GB) on the alleviation of damaging effects of NaCl treatment was studied in view of relative water content (RWC), malondialdehyde content, and the activity of some antioxidant enzymes in two rice (Oryza sativa L.) cultivars differing in salt tolerance (salt-tolerant Pokkali and--sensitive IR-28), comparatively. Both cultivars took up exogenously applied GB through their roots and accumulated it to considerable levels. Leaf RWC of both cultivars under salt treatment showed an increase with GB application. The activities of superoxide dismutase (SOD), ascorbate peroxidase (AP), catalase (CAT), and glutathione reductase (GR) increased in leaves of Pokkali, but peroxidase (POX) activity decreased under salinity. In IR-28, the activities of SOD, AP and POX increased, whereas CAT and GR decreased upon exposure to salt treatment. When compared to the salt-treated group alone, GB application decreased the activities of SOD, AP, CAT, and GR in Pokkali, whereas it increased the activities of CAT and AP in IR-28 under salinity. However, the activity of POX in IR-28 under salinity showed a decrease with GB application compared to the NaCl group. In addition, lipid peroxidation levels of both cvs. under salt treatment showed a decrease with GB treatment. Therefore, we conclude that GB protects both rice seedlings from salinity-induced oxidative stress.  相似文献
