微藻具有极高的应用价值,近些年来针对于微藻的研究也越来越受到重视。抗生素的使用是微藻进行无菌化及突变体筛选等微藻研究中常用的手段之一,抗生素的作用会对微藻的生长代谢、生理活性等产生影响。现将近年来抗生素在微藻工程中的应用研究进行综述,包括抗生素在微藻无菌化培养及微藻突变株筛选中的应用及抗生素对微藻生长和生理上的影响,并对今后抗生素在微藻工程中的研究前景进行展望。  相似文献

培养基琼脂浓度及抗生素对3种海洋微藻生长的影响   总被引:1,自引:0,他引:1  
目的:确定用于分离、纯化及保种所用的固体培养基中优化的琼脂浓度和利于微藻生长并抑菌的抗生素浓度。方法:设计固体培养基中的琼脂浓度以及培养液中抗生素的种类和浓度,定时测定培养体系中三种微藻的生物量及细菌菌落随培养时间的变化。结果:利于固体培养基上三种微藻生长的琼脂浓度均为0.6%~0.8%;牟氏角毛藻用200IU/ml的庆大霉素和200IU/ml的青霉素联合作用,球等鞭金藻8701和盐藻分别用300IU/ml和400IU/ml的青霉素时可保证微藻较好生长。结论:不同微藻生长所需的抗生素浓度不同,抗生素对微藻生长的影响不完全取决于抑菌作用,青霉素促进球等鞭金藻8701生长主要是产生促生长因子,庆大霉素抑制生长可能是产生抑制因子。  相似文献

微藻作为地球上最古老的物种之一,其诞生可追溯到35亿多年前。微藻的种类十分丰富,形态也多种多样。微藻一般都含有叶绿体,因此可进行光合作用,有研究表明微藻固定CO_2的能力是陆地植物的10倍。微藻以其丰富的代谢产物及独特的生理特性在可再生能源、生物医药、食品工业和环境监测等方面有着广泛的应用。然而如何在控制成本的前提下对微藻进行规模化培养成为困扰微藻应用行业的一大难题。为此,本文将从微藻的生化特点及其在各领域中的应用、微藻的规模化培养和微藻的采收3个方面对微藻的规模化培养近十年的研究进展进行综述,旨在为微藻高效培养、低成本采收的研究开发提供参考。  相似文献

污水资源化、二氧化碳减排及微藻生物柴油是当前能源与环境领域的前沿课题。以下围绕污水及烟道气资源化培养产油微藻的培养体系,就藻种、营养条件、培养方式、培养环境及微藻生物反应器等影响产油微藻培养的因素研究进展进行了综述。在综述的基础上提出:由于微藻具有特殊营养方式,通过藻种筛选、微藻营养条件和培养环境的优化以及高效光生物反应器和生产工艺等的创新,可利用污水进行产油微藻生产,以获得生物柴油等高附加值产品,实现微藻生物能源、污水资源化处理和CO2减排三者高度耦合的产油微藻生产体系,从而减少微藻培养费用及污水处理费用,因此,该体系具有重要的环境、社会、经济价值和商业化应用前景。  相似文献

城市生活废水用于产油微藻培养   总被引:10,自引:2,他引:8  
将废水与产油微藻培养结合起来,可以实现废水的无害化处理,还可为微藻的培养提供营养组分和大量水源。利用高产油栅藻,以城市生活废水为水源,在气泡柱式光反应器中,考察了添加不同营养组分对栅藻细胞的生长、生物质产量、总脂含量以及氮磷的去除情况的影响。结果表明:生活废水非常适合于产油微藻的培养,利用生活废水进行微藻培养中,仅需补充添加无机氮、无机磷、柠檬酸铁铵以及微量元素。但这些营养组分的加入量对藻细胞的生长、生物量和油脂积累有重要影响。在优化的废水培养基中微藻细胞浓度可达8.0 g/L左右,远高于标准BG11培养基5.0 g/L的水平。微藻细胞对于无机氮与磷有着高的吸收能力,在废水中加入185.25 mg/L以下无机氮,16.1 mg/L以下无机磷的条件下培养3~4 d后,培养液水体中未检测到有氮磷残留。由此表明利用城市生活废水培养含油微藻可以在获得微藻油脂产品的同时实现水体的无害化处理。  相似文献

微藻细胞富含油脂、淀粉及其他高值代谢物,可用于食品、饲料、化学品和能源的生产。但在规模化培养中,微藻的高生长速率和高产物含量难以兼得,制约了其商业化应用。通过微藻的两阶段培养技术可以将生长和产物积累的时期分离,从而同时获得较高的微藻生物量和产物含量。该技术具有产品得率高、节能减排、适用范围广的优点,是推进微藻商业化的关键之一。本综述总结了现有微藻两阶段培养技术的优势和产品类型,解析了目前微藻两阶段培养技术的限制因素及发展前景,并提出微藻两阶段培养中存在阶段转换时间尚不明确、中间采收步骤成本高这两个限制该技术应用的关键瓶颈,从而为未来微藻两阶段培养技术规模化生产方案的科学决策与实施提供参考。  相似文献

利用TD-700型叶绿素荧光仪测定了6种不同海洋和淡水微藻,即锥状斯氏藻(Scrippsiella trochoidea)、球形棕囊藻(Phaeocystis globosa)、三角褐指藻(Phaeodactylum tricornutum)、眼点拟微绿球藻(Nannochloropsis oculata)、四列藻(Tetraselmis tetrethele)和雨生红球藻(Haematococcus pluvialis)的叶绿素荧光值,同时采用经典方法同步测定其细胞密度和叶绿素a含量。统计分析结果显示,微藻细胞密度、叶绿素a含量、叶绿素荧光值三者之间具有显著或极显著的正相关关系。因此,利用叶绿素荧光仪测定微藻生物量的方法是切实可行的,具有快捷方便、灵敏度高的优点。  相似文献

The kinetics of biomass and antibiotic formation in batch and dialysis culture of Streptomyces baarnensis at various initial concentrations limiting the substrate growth (glucose) has been studied. The antibiotic substances were synthesized by actively growing culture, its concentration in the cultural media was maximum in the log-phase. In continuous dialysis culture on the background of biomass lianer growth in the course of time the constant antibiotic concentration in the media proportional to the glucose input concentration has been established. The inactivation (decomposition) of antibiotic was immediately initiated after discontinuation of substrate supply and followed first kinetics order. Observed features were used for construction of kinetical model of antibiotic biosynthesis. A conclusion has been made that the dialysis culture gives opportunity for more effective antibiotic synthesis as compared with the batch one.  相似文献

The production of iturin A by Bacillus subtilis was studied with respect to the composition of the culture medium. Increasing phosphate concentrations did not modify the antibiotic yield. Fructose, sucrose and mannitol were better carbon sources than glucose for antibiotic production. The nature of the nitrogen source was an important factor in the production of antibiotic. Among the amino acids which are components of iturin A, L-asparagine was the best substrate for the biosynthesis of iturin A; L-glutamine and L-serine were rather poor substrates while L-proline and D-tyrosine gave no antibiotic. Ammonium salts permitted good synthesis of antibiotic but the addition of calcium ions to the culture medium inhibited the excretion of antibiotic from the cells.  相似文献

The stability of the tobramycin-producing organism was studied by the property of the antibiotic production. The organism was stored under conditions of different exposures to light and subculture on slants. The culture was also subjected to long-term storage with various methods. The organism was stable in preserving its antibiotic activity for 1.5 months when stored on the Gauze organic agar No. 2. Subcultures of the organism on this medium provided preservation of the antibiotic activity throughout 4 passages. The culture storage on millet provided preservation of the antibiotic activity for 6 months.  相似文献

Glycerol as the sole carbon source was added to the medium or biosynthesis of heliomycin by Streptomyces olivocinereus and the effect of its concentration on the culture growth and antibiotic production was studied. The culture growth and the amount of the antibiotic synthesized per 1 unit of the fermentation broth were limited by glycerol added in quantities of 0.05 to 1 per cent. Further increasing of the glycerol concentration had no significant effect on the culture growth and antibiotic biosynthesis. The amount of the antibiotic synthesized per 1 unit of the mycelial mass relatively slightly depended on the glycerol concentration. The rate of glycerol consumption by the young 24-hour culture in batch fermentations markedly exceeded that of glycerol consumption by the 48-hour culture. The younger mycelium significantly increased its rate of glycerol consumption when the initial concentration was increased whereas the rate of glycerol consumption by the more mature mycelium did not depend on the initial concentration of the carbon source (within 0.5-2 per cent). The rate of heliomycin biosynthesis practically slightly depended on the initial concentration of glycerol.  相似文献

An actinomycetous culture 2562 inhibiting the growth of gramnegative bacteria was isolated from a soil sample. The culture was classified as Streptomyces griseovariabilis. It was found that culture 2562 produced an antibiotic belonging to the group of novobiocin. It consists of 2 components. One of them is identical to chlorobiocin and the other is a minor component of this group. Some parameters of the antibiotic complex production by strain 2562 under submerged conditions were studied. Nutrient media providing the predominant biosynthesis of the first (main) or the second component of the antibiotic were developed.  相似文献

研究不同溶剂的浒苔干粉末组织提取液对两种赤潮藻—东海原甲藻和中肋骨条藻生长的克生效应。结果表明, 浒苔提取物中确实含有可以影响赤潮藻类生长的克生物质,克生作用具有较明显的浓度效应,添加浓度低时可能会表现为一定的促进生长的作用,添加浓度较高时表现为抑制作用,添加浓度越大,抑制作用越强,即“低促高抑”的特点,这与浒苔提取物对其他微藻的作用相似。其中,蒸馏水提取物对这两种赤潮藻的克生作用小于有机溶剂提取物,有机溶剂中,甲醇和乙酸乙酯提取物对这两种赤潮藻的克生效果最好,正己烷相对较差。根据相似相溶原理,可以初步推断最有效的克生物质应为具有相对较高的极性的有机物。两种赤潮藻对克生物质的敏感程度不同。东海原甲藻对克生物质的敏感性高于中肋骨条藻。蒸馏水、甲醇、乙酸乙酯、正己烷的浒苔干粉末提取物影响东海原甲藻生长的致死作用阈值浓度分别为5.00 g/L、0.50 g/L、0.50 g/L、0.63 g/L(相当于浒苔新鲜藻体浓度为22.00 g-wet/L、2.20 g-wet/L、2.20 g-wet/L、2.75 g-wet/L);影响中肋骨条藻生长的致死作用阈值浓度则分别为20.00 g/L、1.25 g/L、1.25 g/L、2.50 g/L(相当于新鲜藻体浓度为88.00 g-wet/L、5.50 g-wet/L、5.50 g-wet/L、11.00 g-wet/L)。  相似文献

Mitomycin C, a DNA-tropic antibiotic, was shown to have a lethal effect on spore sprouts of two strains of Streptomyces antibioticus, an organism producing oleandomycin. When the time of exposure to the antibiotic increased there was an almost equal decrease in the survival rate. The mutagen action on the morphological variation and antibiotic production of the two closely related strains were diverse due to their genetic differences. The strain isolated after the culture treatment with a chemical mutagen and subjected to a more prolonged maintaining selection showed lower variation with respect to its colony morphology. The other strain isolated after treatment of the culture with high concentrations of its own antibiotic showed lower variation with respect to its antibiotic production property. The shift in the antibiotic production in the direction of the low active variants was characteristic of the both highly productive strains.  相似文献

Kinetic parameters of Streptomyces olivocinereus 11-98 growth and biosynthesis of heliomycin were studied. It was shown that carbon sources such as glycerol, mannitol and ramnose were the most favourable for the antibiotic biosynthesis. These carbon sources belonged to the group of substances providing high growth rates of the culture. Ranging of the culture growth rates and antibiotic production levels revealed a set of carbon sources providing a converse relationship between the growth rate and antibiotic biosynthesis i.e. L-arabinose, potassium gluconate, raffinose and sucrose. It was suggested that these compounds were catabolic type regulators of heliomycin biosynthesis.  相似文献
