中国春小麦株高、育性近等基因系的建立及应用   总被引:1,自引:0,他引:1

以矮败小麦和中国春小麦为材料,经过杂交和连续回交,得到了中国春小麦遗传背景的分别表现矮秆不育、矮秆可育、高秆不育、高秆可育的近等基因系。根据近等基因系各成员系的株高表现,计算出矮秆基因Rht10的降秆强度是69.8%。借助于赤霉酸处理,在幼芽期就可分出矮败中国春小麦后代的不育株与可育株。 Abstract: Use Dwarfing Male-sterile Wheat and cv. Chinese Spring as parents, after cross and continuously back cross, the isogenic lines with Chinese Spring background were developed. These lines include dwarfing male-sterile line, dwarfing fertile line, tall male-sterile line and tall fertile line. The dwarfing intensity of gene Rht10 was calculated to be 69.8% according the differences between the isogenic lines. Treated with GA3solution, the male-sterile and fertile plants in Chinese Spring Dwarfing Male-sterile Wheat can be identified clearly when they are seedlings.  相似文献

利用蓝粒太谷核不育硬粒小麦89-2343[AABB 4D(MS2)/4E]与普通小麦7739-3(2n=42)杂交、回交所产生的蓝粒可育株与白粒矮败材料杂交、回交,育成了一份矮败蓝粒小麦.选用13份遗传背景不同的白粒普通小麦与之杂交、回交,育成了13份矮败蓝粒小麦.对后代的粒色和育性分离进行分析,蓝粒矮败不育株占22.1%,白粒非矮秆可育株占77.7%,表明蓝粒基因、Ms2和Rht10均位于附加染色体上,且连锁紧密;但不同轮回亲本,矮败蓝粒的传递率有差异,477A的传递率最高,接近50%.细胞学分析表明矮败蓝粒小麦仍为单体附加系;探讨了矮败蓝粒小麦在群体改良和杂种小麦生产中的应用.  相似文献

根据大白菜及其近缘芸薹属作物核不育材料育成的纯合两型系和杂合两型系可育株间互交F~1|可育株自交,其子代可能出现无育性分离情况或者产生13(可育株)∶3(不育株)两种表型的育性比资料,认为前者宜用复等位基因假说解释,而后者用抑制作用解释为妥。抑制作用的内涵有两种可能,一是由一对决定育性表现的育性基因与另一对不决定育性表现的抑制基因互作表现抑制作用,即抑制作用假说;二是由性质相同、作用相反且可育基因起上位作用的两对育性基因彼此互作产生抑制效应,权称之为上位抑制假说解释其育性遗传现象。 Abstract:The heading Chinese cabbage-pe-tsai and related crops genic male sterile materials can breed up homozygous two-type line and hyterozygous two-type line,intercrossing between fertile plants of this two lines,its F1 fertile plant selfed,generation show two possibles,one is being without fertility segregation,another is fertility segregation rate of 13 (fertile) ∶3(sterile).According literature above,its considerd that the former is proper to be interpreted by means of multiple alleles hypothesis,and the fertility heredity of the latter is appropriate to be interpreted using inhibition.The implecation of the inhibition has two possibilities,one is that a pair of fertility genes controlling fertility and another pair of inhibition genes not controlling fertility interact showing inhibition,i.e.,inhibiting effect hypothesis,and the other is that two pairs of fertility genes with identical property and contrary action and its fertile genes acting epistatically interact demonstrating inhibiting effect.It was temporarily here defined as epistatic inhibition hypothesis interpreting its fertility inheritance phenomenon.  相似文献

研究表明,本院获得的一种紫色水稻的植株色遗传受控于C、 A、Pl3个独立基因座位上显性基因的互补作用,另有一独立的显性基因对Pl基因的表达起抑制作用。由于该抑制基因在籼稻中的高频率存在,因而,紫稻与一般绿稻品种杂交F1多表现为绿株。紫稻光敏不育(株)系的不育性表达和配合力表现,均可达到与普通光敏不育系相似的水平。本文还讨论了选育籼型紫稻光敏不育系设想的可行性。 Abstract:The inheritance of a purple rice in crosses to green rices was investigated.A group of dominant and interactive genes,C,A and Pl,was found to control the expression of the trait and the other independent inhibitor I-Pl-1 to inhibit the effect of the gene Pl.Because of the wide existence of the gene I-Pl-1 in indica rice,most of F1 plants of the crosses between purple rice and green cultivars were green.The primary study indicated that,for the purple photoperiod-sensitive genic male-sterile lines,the degrees of the sterility and its stability in the sterile stage,and of the fertility in the fertile stage,and the combining ability levels of the purple rice were as high as the degrees and levels for the green rice.A tentative idea on breeding purple photoperiod-sensitive male-sterile lines of indica rice was suggested and its feasibility and advantage was discussed.  相似文献

从轮选可育株衍生的后代中发现一个雄性不育株,即P740不育材料。在P7 40不育材料第1、2、3次姊妹交后代中,可育株与不育株的分离比分别符合4:1、 7:5和15:13。在第2次姊妹交后代中可育株产生的40个株系中,7个株系的育性分离比是15:1,33个株系是3:1,二者约分别占1/7和6/7。以上结果表明,P740是一个双隐性基因控制的核雄性不育材料。本文提出了计算隐性核不育材料姊妹交后代育性分离比率的公式。 Abstract:A male sterile material was found in the progeny derived from a fertile plant in the population of recurrent selection.The ratios of fertile to sterile plants in three sib-mating of the male sterile material with fertile plants were to 4:1,7:5 and 15:13,respcctively.In the 40 plant lines derived from the fertile plants of second sib-mating population,33 lines approximated to the segregation ratio of 3(fertile):a double recessive nuclear male sterile material.A genetic pattern for calculation of fertile segregation in sib-mating progenies of recessive nuclear male sterile material was advanced in this paper.  相似文献

通过连续回交,将抗除草剂基因EPSPS-G6转育花粉败育彻底(无微量花粉,不育度达100%)的棉花单基因隐性控制的核不育系Yu98-8A,进而培育成抗除草剂核不育系Yu98-8A1。对该转育不育系花冠表型测量观察表明,与同质系正常可育株比较,不育株花冠较小, 不育株子房直径略大于可育株,花柱长和花柱外露长度均明显高于同质系正常可育株,花柱头外露为其最显著的表型特征;显微观察显示,不育系Yu98-8A1小孢子败育主要是在四分体形成后的小孢子发育期。小孢子败育特征表现为花粉粒无内含物、无刺突产生,最后解体、退化。PCR分子鉴定表明,抗除草剂基因EPSPS-G6转育入Yu98-8A1,除草剂抗性试验表明,该转育不育系可抗质量百分比浓度达0.3%的草甘膦。该抗除草剂核不育系的培育在棉花杂种优势利用方面有重大利用价值。  相似文献

Chromosome 2C from Aegilops cylindrica has the ability to induce chromosome breakage in common wheat (Tritivum aestivum). In the BC1F3 generation of the T. aestivum cv. Chinese Spring and a hybrid between T. aestivum-Leymus racemosus Lr.7 addition line and T. aestivum-Ae, cylindrica 2C addition line, three disomic translocation addition lines (2n = 44) were selected by mitotic chromosome C-banding and genomic in situ hybridization. We further characterized these T. aestivum-L, racemosus translocation addition lines, NAU636, NAU637 and NAU638, by chromosome C-banding, in situ hybridization using the A- and D-genome-specific bacterial artificial chromosome (BAC) clones 676D4 and 9M13; plasmids pAsl and pSc119.2, and 45S rDNA; as well as genomic DNA of L. racemosus as probes, in combination with double ditelosomic test cross and SSR marker analysis. The translocation chromosomes were designated as T3AS-Lr7S, T6BS-Lr7S, and T5DS-Lr7L. The translocation line T3AS-Lr7S was highly resistant to Fusarium head blight and will be useful germplasm for resistance breeding.  相似文献

对由离子束介导大豆DNA导入豫麦52获得的第四代两个转化株系9004和9002中的可育株籽粒和矮秆不育株杂交籽粒以及受体豫麦52籽粒,进行酯酶和可溶性蛋白的聚丙烯酰胺凝胶电泳分析。结果表明,可育株籽粒可溶性蛋白电泳在谱带上与受体存在差异,矮秆不育株材料的酯酶和可溶性蛋白电泳图谱谱带差异更明显。由此推测,外源大豆DNA导入受体后某些片段有可能插入受体基因组,从而导致基因突变或受体基因表达发生改变。  相似文献

对萝卜(RaphanussativusL.)2种雄性不育系及相应保持系花药发育过程进行组织化学研究,研究结果表明,2种不育花药组织化学变化与小孢子败育变化皆类似,不育系475A的花药发育受阻于孢原细胞分化之前,此时花药时含少量蛋白质,核酸的较多淀粉粒。随着花粉的长大,不再含有蛋白质,核酸和淀粉粒,不育系春红A的花药在四分体以前与可育系类似,含丰富蛋白质,核酸和多糖。在单核期绒毡层异常引起小孢子败育  相似文献

Dwarfing and sensitivity to the duration of a single inductive dark period for flowering ofPharbitis nil in F2 progeny of a cross between the tall strain Tendan, and the dwarf, Kidachi appear to be controlled by the alleles at two independent loci. Progeny of a similar cross between the tall strain Violet and the dwarf Kidachi at F2 and F3 also showed single locus segregation for tall: dwarf plants. In this cross, differences in photoperiodic response could be identified in F3 families but they were not simply inherited. There was some evidence of difficulties with classification of the F2 plants, but also, the flowering of the F1 between the two less sensitive strains Tendan and Violet indicated complex inheritance of their photoperiodic response. Complementary dominant alleles at three independent loci may be necessary for flowering in even shorter dark periods with the sensitive strain Kidachi. The dwarf strain Kidachi has a reduced gibberellin (GA) content (Barendse and Lang 1972), it flowers in a short dark period without terminal flowering, and it responds positively to GA application both for flowering and growth. However, since control of dwarfing and photoperiodic sensitivity can be separated genetically, there is no strick link between the gibberellin responsiveness of Kidachi for its growth and flowering. Despite the complexity of flowering genetics in Violet×Kidachi, a short-dark-period-sensitive, terminal flowering and tall F7 line was obtained in a pedigree previously held heterozygous for the dwarf: tall character but not selected for flowering time. Thus, flowering in a short dark period can also be obtained in the presence of the non-dwarfing allele from strain Violet, again demonstrating genetic independence.  相似文献

In order to understand the molecular characteristics of the Chinese radish, the mitochondrial DNA structure and sequence were analyzed in Chinese wild radish and cultivated varieties. A total of four male-sterile lines, four maintainer lines, 17 cultivars, and a single Chinese wild radish were used, along with 25 male-sterile individuals and 159 fertile plants. We found that the cytoplasm of Chinese radishes could be classified into two types: the normal type and the Ogura type. The Ogura-type cytoplasm was detected in 25 male-sterile plants. The 159 fertile plants had normal cytoplasm. Both the Ogura cytoplasm and the normal cytoplasm were detected in the male-sterile ??RA??. The orf138 gene in mitochondrial DNA was detected in cultivated Chinese radish cultivars but not in the wild radish. The Chinese radish orf138 nucleotide sequence was determined in four male-sterile lines and displayed complete homology to the known orf138 type A nucleotide sequence. Three types of mitochondrial orfB (type 1, type 2 and type 3) were found in Chinese radishes. Type 1 was only present in the male-sterile lines. Chinese cultivated radishes were divided into type 2 and type 3, while the Chinese wild radish only had type 3 cytoplasm.  相似文献

The male sterility gene from a male-sterile, green,Gelidium vagum line was introduced to a wild-type line through repeated backcrossing and selection for five generations. The plants from the recurrent parent, the male-sterile green, the backcross-5 fertile and the backcross-5 male-sterile lines were compared for their growth performance. The backcross-5, red, male-sterile plants grew at a significantly higher rate than the plants from other lines, suggesting that male-sterile gametophytes would likely be better candidates for aquaculture than normal wild-type plants of this species. This study also provides evidence that the growth rate ofG. vagum decreases as a consequence of reproduction.  相似文献

The soluble proteins of germinated embryos and young shoots in Wheat were studied by means of isoelectrofocusing (IEF) electrophoresis. The materials used were three species of cytoplasmic male-sterile (CMS) lines (Type E, T and A) and their maintainers. The protein quantity of pI 4. 9 in male-fertile materials is higher than the corresponding male-sterile ones; the protein of pI 6.85 is probably the result of gene expression in Timopheevi cytoplasm; the protein of pI 7.6 is a typical band of Jinfeng male-sterile line. Evident differences on soluble protein chromatograms of IEF electrophoresis were observed both in germinated embryos and young shoots which were from different kinds of CMS line, these differences could be used as indexes identifying various CMS lines.  相似文献

A number of S male-sterile plants from several shrunken-2 inbred lines were crossed initially with an R138-TR inbred line pollinator carrying the nonrestoring genotype for S sterile cytoplasm. One such cross, involving a male-sterile female parent from inbred line M825, produced, unexpectedly, a number of male-fertile F1 progeny, along with the expected male-sterile off-spring. Pollen records of plants in F2, F3 and F4 progenies in the exceptional pedigree, and of a variety of testcross and backcross progenies from these male-fertile exceptions, indicate that the exceptional male fertility is not attributable to the action of either dominant or recessive nuclear restorer genes. They are, however, consistent with the hypothesis that the event responsible for the appearance of exceptional male-fertile offspring among progeny of the original cross involved a change from male-sterile to male-fertile condition in the cytoplasm of the male-sterile M825 plant involved as the female parent in this cross. It appears that this plant bore an ear in which there was a relatively early mutational event at the cytoplasmic level resulting in a chimera involving some kernels which carried S male-sterile cytoplasm, and others which carried the mutated fertile cytoplasmic condition. The finding of a number of additional ear chimeras supports this contention.—The evidence suggests that the change from sterile to fertile cytoplasm has occurred in a number of other instances. The male-sterile line M825 is especially prone to this change. These findings are of particular interest because it has heretofore been considered that both S and T types of male-sterile cytoplasm are highly stable.—The data presented here are not sufficient to support the notion that the exceptional event involves a qualitative change, analogous to gene mutation, in a cytoplasmic entity governing the expression of male fertility. It is equally plausible that the exceptional male fertility is the result of occasional transfer of normal cytoplasm through the male germ cells of maintainer parents.  相似文献
