激光诱变青霉PT95原生质体选育类胡萝卜素高产菌株   总被引:5,自引:0,他引:5  
采用激光对青霉PT95菌株的原生质体进行了诱变处理,选育到一株菌核生物量和类胡萝卜素含量均有显提高的突变菌株L05。与出发菌株相比,L05菌株的菌核生物量提高了98.6%,菌核中的类胡萝卜素含量提高了28.3%,在查氏平板上的类胡萝卜素产率提高了154.0%。所选育的L05菌株经3次传代培养,菌落没有发生扇形变异,菌核生物量和类胡萝卜素含量没有明显改变,说明该菌株具有良好的遗传稳定性。  相似文献

初步研究了外源β-胡萝卜素和光照对青霉PT5菌株菌核分化和类胡萝卜素产率的影响。结果表明,在培养基中加入外源β-胡萝卜素后,PT5菌株渗出液出现的时间、菌核出现的时间延迟了,但菌核成熟的时间没变。培养基中的外源β-胡萝卜素浓度越大,其渗出液、菌核出现的时间越迟。外源β-胡萝卜素亦能降低PT5菌株的脂质过氧化水平和菌核中的类胡萝卜素含量。高氧胁迫的光照培养条件有利于PT95菌株的菌核分化和色素在菌核中的积累;与低氧胁迫的黑暗培养条件相比,其菌核生物量和类胡萝卜素产率分别增加了18.7%和101%。以上实验结果表明,若想获得高的菌核生物量和类胡萝卜素产率,应该尽可能在高氧胁迫、无抗氧化剂存在的条件下培养PT5菌株。  相似文献

AIMS: To determine the effect of oxidative stress and exogenous ascorbic acid on sclerotial biomass and carotenoid yield of Penicillium sp. PT95. METHODS: In this experiment, high oxidative stress was applied by the inclusion of FeSO(4) in the growth medium and exposure to light. Low oxidative stress was applied by omitting iron from the growth medium and by incubation in the dark. Supplementation of exogenous ascorbic acid (as antioxidant) to the basal medium caused a concentration-dependent delay of sclerotial differentiation (up to 48 h), decrease of sclerotial biomass (up to 40%) and reduction of carotenoid yield (up to 91%). On the contrary, the exogenous ascorbic acid also caused a concentration-dependent decrease of lipid peroxidation in colonies of this fungus. CONCLUSIONS: Under high oxidative stress growth condition, the sclerotial biomass and carotenoid yield of PT95 strain in each plate culture reached 305 mg and 32.94 microg, which were 1.23 and 3.71 times higher, respectively, than those at low oxidative stress growth condition. These data prompted us to consider that in order to attain higher sclerotial biomass and pigment yield, the strain PT95 should be grown under high oxidative stress and in the absence of antioxidants. SIGNIFICANCE AND IMPACT OF THE STUDY: These results suggest that strain PT95 may be used for solid-state fermentation of carotenoid production under high oxidative stress growth conditions.  相似文献

以紫外(UV)与亚硝基胍诱变的竹黄菌(Shiraia bambusicola)菌株NU12和UV4为出发菌株,60℃处理5 min、紫外(距离30 cm,30 W)照射90s进行双亲原生质体灭活,通过30%聚乙二醇PEG6000介导原生质体融合20 min.结合平板初筛和高效液相色谱( HPLC)分析进行复筛,通过3轮重组融合操作,筛选出6株高产竹红菌甲素的融合株.其中融合菌株FIII - 21的竹红菌甲素产量达到80.4 mg/L,比原始出发菌株提高了58.9%~167.1%,且遗传稳定,具有较高的医药及工业应用价值.  相似文献

Various inocula and grains were evaluated for carotenoid production by solid-state fermentation using Penicillium sp. PT95. Millet medium was more effective in both sclerotia growth and carotenoid production than other grain media. An inoculum in the form of sclerotia yielded higher sclerotia biomass compared to either a spore inoculum or a mycelial pellet inoculum. Adding wheat bran to grain medium favored the formation of sclerotia. However, neither the inoculum type nor addition of wheat bran resulted in a significant change in the carotenoid content of sclerotia. Among grain media supplemented with wheat bran (wheat bran:grain =1:4 w/w, dry basis), a medium consisting of rice and wheat bran gave the highest sclerotia biomass (15.10 g/100 g grain), a medium consisting of buckwheat and wheat bran gave the highest content of carotenoid in sclerotia (0.826 mg/g dry sclerotia), and a medium consisting of millet and wheat bran gave the highest carotenoid yield (11.457 mg/100 g grain).  相似文献

Using corn meal as fermentation substrate, the effect of some factors, fermentation time and supplementation of saccharide and nitrogen sources as well as vegetable oil, on the sclerotia growth and carotenoid production of Penicillium sp PT95 during solid state fermentation were studied. When PT95 strain was grown on the amended medium by supplementing of 3g NaNO3, 10g maltose and 2.5g soybean oil per liter of salt solution to basal medium for 20 days, the dry sclerotia weight and carotenoid yield reached 9.70 g and 5260 g / 100 g of substrate, respectively. AIMS: To determine the effect of oxidative stress and exogenous beta-carotene on sclerotial differentiation and carotenoid yield of Penicillium sp. PT95. METHODS AND RESULTS: In this experiment, high oxidative stress was applied by inclusion of FeCl(3) (10 micromol l(-1)) in the growth medium and by light exposure. Low oxidative stress was applied by omitting iron from the growth medium and by incubation in the dark. Supplementation of exogenous beta-carotene (as antioxidant) to the basal medium caused a concentration-dependent delay of sclerotial differentiation (up to 72 h), decrease of sclerotial biomass (up to 43%) and reduction of carotenoid yield (up to 92%). On the contrary, the exogenous beta-carotene also caused a concentration-dependent decrease of lipid peroxidation in colonies of this fungus. CONCLUSIONS: Under high oxidative stress growth condition, the sclerotial biomass and carotenoid yield of PT95 strain in each plate culture reached 141 mg and 30.03 microg, which were 1.53 and 3.51 times higher respectively, than that at low oxidative stress growth condition. SIGNIFICANCE AND IMPACT OF THE STUDY: These data prompted us to consider that in order to attain higher sclerotial biomass and pigment yield, the strain PT95 should be grown under high oxidative stress and in the absence of antioxidants.  相似文献

本文对青霉Penicilliumsp.PT95菌株在固态发酵条件下菌核内产生类胡萝卜素进行了初步研究。结果表明,在3种固态发酵培养基中,玉米粉培养基(SMA)比麸皮2基和棉籽壳培养基更适合于PT95菌株固态发酵产生类胡萝卜素。为了增加菌核干重和提高类胡萝卜素产率,SMA中需要添加氮源、碳源和植物油。在所度的各种氮、碳源中,以硝酸钠和麦芽糖效果最佳。通过我试验确定了在培养基盐溶液中添加硝酸钠3g/L  相似文献

精氨酸脱亚胺酶(arginine deiminase,EC 3.5.3.6,ADI)因其可作为精氨酸营养缺陷型肿瘤细胞的靶向治疗药物而受到广泛关注. 目前,支原体来源的重组ADI处于肝癌和黑素瘤的三期临床研究阶段. 作为药用酶,当前报道的ADI在体内生理条件下普遍存在酶活低、半衰期短、底物亲和性弱等局限性.本研究结合随机突变及基于理性设计的定点突变两种方法,对研究室前期自主筛选得到的变形假单胞菌Pseudomonas plecoglossicida来源的ADI经一轮定向进化后所获优势突变株M314(A128T/H404R/I410L)进行分子改造.通过对随机突变法获得的1480个突变株进行96孔板高通量筛选,得到优良突变株M173(A128T/H404R/I410L/K272R);同时,基于同源序列比对及ADI蛋白三维结构同源建模,采用PyMOL软件理性预测和分析其活性中心及附近保守区域氨基酸位点对蛋白功能的影响,选择了6个位点D78E、L223I、P230I、S245D、A275N、R400M分别在M314的基础上进行定点突变,最终获得优势突变株M04(A128T/H404R/I410L/S245D). 通过对突变株的酶学性质以及动力学参数分析发现:生理pH值下,突变株M173的酶比活(12.32 U/mg)在M314(9.02 U/mg)的基础上提升3659%,Kcat/Km提高5236%;而突变株M04的最适pH由6.5升高至7.0,更接近体内生理pH,其比酶活(14.66 U/mg)较M314提升62.53 %,Kcat/Km提高了37.12%. 综上结果,本研究结合两种分子改造方法成功地对该ADI在生理pH条件下的酶活和酶学性质进行了改良,并为蛋白质的分子改造策略提供了理论基础和实验依据.  相似文献

Candida tropicalis isolated from acclimated activated sludge was used in this study. Cell suspensions with 5 x 10(7) cells ml(-1) were irradiated by using a He-Ne laser. After mutagenesis, the irradiated cell suspension was diluted and plated on yeast extract-peptone-dextrose (YEPD) medium. Plates with approximately 20 individual colonies were selected, and all individual colonies were harvested for phenol biodegradation. The phenol biodegradation stabilities for 70 phenol biodegradation-positive mutants, mutant strains CTM 1 to 70, ranked according to their original phenol biodegradation potentials, were tested continuously during transfers. Finally, mutant strain CTM 2, which degraded 2,600 mg liter(-1) phenol within 70.5 h, was obtained on the basis of its capacity and hereditary stability for phenol biodegradation. The phenol hydroxylase gene sequences were cloned in wild and mutant strains. The results showed that four amino acids were mutated by irradiation with a laser. In order to compare the activity of phenol hydroxylase in wild and mutant strains, their genes were expressed in Escherichia coli BL21(DE3) and enzyme activities were spectrophotometrically determined. It was clear that the activity of phenol hydroxylase was promoted after irradiation with a He-Ne laser. In addition, the cell growth and intrinsic phenol biodegradation kinetics of mutant strain CTM 2 in batch cultures were also described by Haldane's kinetic equation with a wide range of initial phenol concentrations from 0 to 2,600 mg liter(-1). The specific growth and degradation rates further demonstrated that the CTM 2 mutant strain possessed a higher capacity to resist phenol toxicity than wild C. tropicalis did.  相似文献

Amerindian Mapuche (Araucanians) are now living in Chile and Argentina at both sides of Andean Mountains. They are anthropologically and genetically different from southernmost South America Patagonian Amerindians. Most of the HLA alleles found in our Mapuche sample are frequent or very frequent in North and South America Amerindians: (1) Class I: A*02:01, A*03:01, A*68:01, B*39:09, B*51:01, (2) Class II: DRB1*03:01, DRB1*04:03, DRB1*07:01, DRB1*08:02, DRB1*14:02, DRB1*16:02. One of the nine most frequent extended haplotypes seems to be from European origin, suggesting the existence of a degree of admixture with Europeans in our Mapuche sample. It has been calculated of about 11 % admixture. Three of the extended haplotypes are also found in other Amerindians and five of them are newly found in Mapuche Amerindians: A*68:01-B*39:09-DRB1*08:02-DQB1*04:02; A*68:01-B*51:01-DRB1*04:03-DQB1*03:02; A*29:01-B*08:01-DRB1*03:01-DQB1*02:01; A*02:01-B*15:01-DRB1*04:03-DQB1*03:02; A*33:01-B*14:02-DRB1*07:01-DQB1*03:03. The medical importance of calculating HLA profile is discussed on the diagnostic (HLA and disease) and therapeutical bases of HLA pharmacogenomics and on the construction of a virtual transplantation HLA list profile. Summary A protoplast fusion experiment was designed in which the selectable marker, nitrate reductase (NR), also served as a biochemical marker to provide direct evidence for intergeneric specific gene transfer. NR-deficient tobacco (Nicotiana tabacum) mutant Nia30 protoplasts were the recipients for the attempted transfer of the NR structural gene from 50 krad -irradiated barley (Hordeum vulgare L.) protoplasts. Barley protoplasts did not form colonies and Nia30 protoplasts could not grow on nitrate medium; therefore, selection was for correction of NR deficiency allowing tobacco colonies to grow on nitrate medium. Colonies were selected from protoplast fusion treatments at an approximate frequency of 10-5. This frequency was similar to the Nia30 reversion frequency, and thus provided little evidence for transfer of the barley NR gene to tobacco. Plants regenerated from colonies had NR activity and were analyzed by western blotting using barley NR antiserum to determine the characteristics of the NR conferring growth on nitrate. Ten plants exhibited tobacco NR indicating reversion of a Nia30 mutant NR locus. Twelve of 26 regenerated tobacco plants analyzed had NR subunits with the electrophoretic mobility and antigenic properties of barley NR. These included plants regenerated from colonies selected from 1) co-culturing a mixture of Nia30 protoplasts with irradiated barley protoplasts without a fusion treatment, 2) a protoplast fusion treatment of Nia30 and barley protoplasts, and 3) a fusion treatment of Nia30 protoplasts with irradiated barley protoplasts. No barley-like NR was detected in plants regenerated from a colony that grew on nitrate following selfed fusion of Nia30 protoplasts. Because tobacco plants expressing barley-like NR were recovered from mixture controls as well as fusion treatments, explanations for these results other than protoplast fusionmediated gene transfer are discussed.  相似文献

The aim was to assess heterosis in a set of 16 summer-squash hybrids, and evaluate the combining capacity of the respective parental lines, which differed as to the degree of parthenocarpy and resistance to PRSV-W (Papaya Ringspot Virus-Watermelon strain). The hybrids were obtained using a partial diallel cross design (4 × 4). The lines of parental group I were 1 = ABX-037G-77-03-05-01-01-bulk, 2 = ABX-037G-77-03-05-03-10-bulk, 3 = ABX-037G-77-03-05-01-04-bulk and 4 = ABX-037G-77-03-05-05-01-bulk, and of group II, 1' = ABX-037G-77-03-05-04-08-bulk, 2' = ABX-037G-77-03-05-02-11-bulk, 3' = Clarice and 4' = Caserta. The 16 hybrids and eight parental lines were evaluated for PRSV-W resistance, parthenocarpic expression and yield in randomized complete-block designs, with three replications. Parthenocarpy and the resistance to PRSV-W were rated by means of a scale from 1 to 5, where 1 = non-parthenocarpic or high resistance to PRSV-W, and 5 = parthenocarpic or high susceptibility to PRSV-W. Both additive and non-additive gene effects were important in the expression of parthenocarpy and resistance to PRSV-W. Whereas estimates of heterosis in parthenocarpy usually tended towards a higher degree, resistance to PRSV-W was towards higher susceptibility. At least one F(1) hybrid was identified with a satisfactory degree of parthenocarpy, resistance to PRSV-W and high fruit-yield.  相似文献

A large amount of genetic mapping information has been obtained in the chicken from the East Lansing, Compton and Wageningen reference populations. Physical mapping information has however, been more limited. We have mapped 14 new clones, both genetically and physically, and all 14 have been assigned to macrochromosomes. The orientation of linkage groups E01C01C11W01 (Chr 1), E06C02W02 (Chr 2), E02C03W03 (Chr 3), E05C04W04 (Chr 4), E07E34C05W05 (Chr 5), E11C10W06 (Chr 6), E45C07W07 (Chr 7) and E43C12W11 (Chr 8) has been established. Here we present integrated maps of the eight macrochromosomes and the Z chromosome of the chicken and correlate genetic with physical distances for chromosomes 1-3 and the Z sex chromosome.  相似文献

研究武汉亮斑扁角水虻幼虫肠道微生物对武汉亮斑水虻成虫产卵行为的影响。首先使用LB(Luria-Bertani)培养基对武汉亮斑水虻幼虫肠道微生物进行分离纯培养,筛选得到23株肠道微生物。以灭菌的LB液体培养基为阴性对照,使用已证实引诱产卵效果好的来自武汉亮斑水虻卵表的属于戈登氏菌属的细菌(Gordonia sihwensis)FE06为阳性对照,利用筛选到的武汉亮斑水虻幼虫肠道微生物摇瓶发酵液对武汉亮斑水虻成虫进行产卵行为实验。发现有5株武汉亮斑水虻幼虫肠道微生物(BSFLG01、BSFLG03、BSFLG04、BSFLG05、BSFLG06)对武汉亮斑水虻产卵有显著的积极促进作用,其中3株(BSFLG03、BSFLG05、BSFLG06)对水虻成虫产卵的引诱作用明显强于所设立的阳性对照FE06;菌株BSFLG07为一种毕赤酵母菌(Pichia kudriavzevii),对武汉亮斑水虻产卵有消极影响。经菌落培养特征、菌体形态比较、16SrDNA或18SrDNA序列测定和发育树比对分析,鉴定出这5种具有显著引诱武汉亮斑水虻产卵作用的微生物分别为:枯草芽胞杆菌(Bacillus subtilis)BSFLG01、一种沙雷氏菌(Serratiasp.)BSFLG03、库德毕赤酵母(Pichia kudriavzevii)BSFLG04、皱落假丝酵母(Candida rugosas)BSFLG05、粘质沙雷氏菌(Serratia marcescens)BSFLG06。  相似文献

A mutant strain EMS-1 ofSclerotium rolfsii lacking the ability to develop mature sclerotia was isolated following chemical mutagenesis of macerated sclerotia with ethyl methane-sulfonate. The mutant failed to form sclerotia even in the presence of lactose, threonine or iodo-acetic acid which promoted sclerotial development in the wild strain and the UV-8 mutant. EMS-1 exhibited higher (1.5 – 3.0 times) cellulase and hemicellulase activity compared to the wild strain. Possible correlation between sclerotial morphogenesis and cellulase and/or oxalic acid production is discussed.  相似文献
