本文对佛波醇酶诱导人早幼粒白血病细胞系HL-60细胞分化为巨噬细胞样细胞对蛋白激酶c活力及其在亚细胞分布的变化进行了研究。蛋白激酶c活力在TPA处理1小时即明显降低,此低水平的酶活力持续整个实验时期。酶的亚细胞分布研究提示TPA处理细胞胞质组分酶活力剧烈降低,而颗粒组分存在一高盐浓度洗脱的酶活力峰。蛋白激酶c抑制剂三氟过(口了)嗪单独处理HL-60细胞导致胞质和颗粒组分酶活力升高,但并不诱导细胞分化;若与TPA合并处理细胞,酶活力又降低,此时细胞又分化为巨噬细胞样细胞。对上述结果的可能机理进行了讨论。  相似文献

目的:通过,IPA诱导K562细胞分化过程中干预细胞铁代谢探讨白血病细胞铁与细胞分化的关系及对EGR1mRNA表达的影响。方法:应用体外细胞培养技术通过细胞形态,细胞化学染色观察细胞生长分化情况;用FCM、RT—PCR等技术检细胞周期、细胞表面分化抗原CD33、CD14及EGR1mRNA的表达。结果:在,IPA诱导K562细胞分化过程中铁剥夺可明显抑制K562细胞生长,并可阻止,IPA诱导K562细胞分化,使K562细胞停止在S期。铁剥夺可降低,TPA诱导K562细胞分化过程中EGR1mRNA的表达。讨论:铁剥夺明显抑制K562细胞生长、阻止TPA诱导K562细胞分化,故铁剥夺剂(DFO)可能作为一种辅助抗癌药用于白血病的化疗,但由于它能阻止白血病细胞的分化,故不宜用于白血病的诱导分化治疗。铁剥夺使K562细胞分化过程中E—GR1mRNA表达降低可能参与了阻止TPA诱导K562细胞的分化过程。  相似文献

目的:对树突状细胞体外大量扩增及树突状细胞的临床应用提供理论基础.方法:以重组小鼠粒细胞-巨噬细胞集落刺激因子(rmGM-CSF)、重组小鼠白介素-4(rmIL-4)和重组小鼠肿瘤坏死因子-α(rm TNF-α)体外诱导小鼠骨髓细胞分化为DC,倒置显微镜动态观察细胞形态学变化,流式细胞术分析细胞表面分子,并应用混合淋巴细胞反应检测其刺激T淋巴细胞的增殖能力.结果:经体外诱导培养第2天即可见大量细胞集落形成;培养至第9天,DC成熟,具有典型的形态,同时DC可以显著刺激同种异体混合淋巴细胞增殖.结论:体外诱导培养可以获得大量小鼠骨髓来源的DC,可广泛应用于临床实验及实验研究.  相似文献

目的:观察全反式维甲酸(ATRA)处理和ATRA与十四烷酰佛波醇乙酸酯(TPA)序贯处理(ATRA/TPA)对人类神经母细胞瘤细胞系SH-SY5Y细胞增殖抑制和形态分化的影响。方法:应用10μM ATRA处理6天和10μM ATRA处理3天继以80 nMTPA处理3天这两种方法使SH-SY5Y细胞分化;用倒置光学显微镜动态观察SH-SY5Y细胞形态学变化;并用MTT比色法比较两种分化方法对SH-SY5Y细胞的体外抗增殖作用。结果:ATRA处理和ATRA与TPA序贯处理对SH-SY5Y细胞都有抗增值和诱导细胞分化作用,细胞形态发生明显的变化,分化成神经元表型,前者主要表现为两端带有长突起的纺锤体样细胞形态,而后者主要是由细胞体延伸出多个突起的多边形的细胞。ATRA分化6天的细胞的存活率下降为78.7%±2.0%。当去除ATRA后,继续培养1天的细胞存活率上升为89%±0.2%,而继续培养2天的细胞存活率为86.3%±1.4%;ATRA与TPA序贯分化6天细胞存活率下降为75.9±0.4%。当去除TPA后,继续培养一天的细胞存活率为75.5±0.7%,继续培养2天的细胞存活率为74.9±1.0%。结论:维甲酸(ATRA)处理和ATRA与十四烷酰佛波醇乙酸酯(TPA)序贯处理(ATRA/TPA)均能明显诱导SH-SY5Y细胞分化。这两种分化细胞为神经科学的研究提供了优良的体外培养模型细胞,尤其是ATRA与TPA序贯处理能获得分化完全而稳定的神经元样细胞。  相似文献

为了观察正常人骨髓成纤维样细胞系HFCL对急性单核细胞白血病U937细胞促分化作用,及其对经典诱导分化剂TPA诱导分化作用的影响,先建立U937细胞和HFCL细胞共培养体系,以细胞形态学改变、硝基四氦唑蓝(NBT)、流式细胞仪检测细胞周期和CD11b、CD13、CD14、CD33细胞表面抗原作为诱导分化指标;Western印迹检测P38蛋白的表达变化。结果发现,与HFCL细胞共培养后,U937细胞出现分化成熟的形态学改变,且与HFCL细胞直接接触组的诱导分化作用大于用transwell组。同时发现U937细胞与HFCL细胞共培养后,G1期细胞增高,S期细胞减少;CD11b、CD13、CD14和CD33表达增高;且NBT阳性细胞增高至46、3%。Western印迹检测结果显示,直接接触组总P38蛋白表达增加。而且HFCL细胞还能增强TPA对U937的诱导分化作用。  相似文献

用微量细胞病变抑制法测定了53例急性白血病患者外周血白细胞诱生干扰素的水平,以及18例急性淋巴细胞性白血病患者的淋巴细胞转化功能。发现前者体外诱生干扰素的水平显著下降,并与病程转归有密切关系;后者淋巴细胞转化指数低于正常,但与干扰素水平之间无平行关系。本文还对急性白血病患者白细胞体外诱生干扰素水平下降的可能原因进行了讨论。  相似文献

白细胞介素-12(interleukin-12,IL-12)具有明显的抗肿瘤作用,但其作用机制较复杂。前期形态学和功能的研究发现,IL-12可诱导单核细胞白血病细胞分化。该研究从巨噬细胞表面分化标志、细胞增殖能力、周期以及凋亡四个方面探讨了IL-12诱导急性单核细胞白血病细胞分化和凋亡的相关作用机理。结果发现,以人源性重组IL-12 p70处理的THP-1细胞,巨噬细胞表面标志CD68和CD11b的表达量明显增加并呈时间依赖性,CD68+和CD11b+阳性细胞数也显著增多;IL-12诱导分化过程中伴有THP-1细胞生长缓慢、G1期或G1/S期细胞周期阻滞现象;IL-12处理后的THP-1细胞凋亡率也明显增加,以早期凋亡细胞为主,并伴有抗凋亡蛋白Bcl-2表达下调及促凋亡蛋白Fas表达增加。上述实验结果提示,IL-12对于急性单核细胞白血病可通过诱导肿瘤细胞向成熟巨噬细胞分化,抑制细胞增殖以及增加细胞凋亡来发挥抗肿瘤作用。  相似文献

利用佛波酯体外分化人急性单核白血病细胞形成巨噬细胞,并用低温超速离心法从巨噬细胞培养上清收集Exosomes,并分析其生物学特征。Exosomes经透射电显微镜分析形态,并用Western blot方法分析Exosomes膜上特异性蛋白CD80、ICAM-3、HSP-70的表达对其生物学特征进行鉴定。研究结果表明佛波酯成功诱导人急性单核白血病细胞分化成人巨噬细胞。获得的巨噬细胞分泌微小囊泡经透射电镜分析后,微小囊泡的直径在30~100 nm,并且其含有Exosomes的特征性蛋白CD80、ICAM-3、HSP-70等蛋白,表明其为Exosomes。使用低温超速离心法成功收集到Exosomes,为后续开展Exosomes的成份及功能分析提供了保障。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献

Microbial resistance to antibiotics is an unresolved global concern, which needs urgent and coordinated action. One of the guidelines of the Centers for Disease Control and Preventions (CDC) to combat antibiotic resistance is the development of new antibiotics to treat drug-resistant bacteria. In our effort to find new antibiotics, we report the synthesis and antimicrobial studies of 30 new pyrazole derivatives. These novel molecules have been synthesized by using readily available starting materials and benign reaction conditions. Some of these molecules have shown activity with MIC values as low as 0.78?µg/mL against four bacterial strains; Staphylococcus aureus, methicillin-resistant S. aureus, Bacillus subtilis, and Acinetobacter baumannii. Furthermore, active molecules are non-toxic to mammalian cell line.
