DNA依赖性蛋白激酶(DNA-dependent protein kinase,DNA-PK)是由3个亚基组成的丝/苏氨酸蛋白激酶,属于磷脂酰肌醇-3激酶相关激酶家族(phosphatidylinositol 3-kinase-related kinases,PIKK),是基因组DNA损伤修复过程中的关键蛋白激酶,参与并决定着非同源末端连接DNA损伤修复通路的整个进程.此外DNA-PK还参与了电离辐射诱导的凋亡信号转导通路,免疫细胞V(D)J重组、免疫细胞分化、胰岛素刺激下的细胞应答等过程,具有维持端粒稳定性的功能.DNA-PK活性的升高会降低肿瘤对放射的敏感性,其活性主要受自身磷酸化调控,此外活性氧、EGFR、MG132抑制剂、PP1γ1和PP5等蛋白磷酸酶也有调控DNA-PK活性的作用.  相似文献

细胞内DNA会受部分外界因素(如紫外辐射,电离辐射和化学毒素)和内部因素(如复制错误)的影响而发生损伤,包括DNA双链断裂、DNA错配和DNA交链等。DNA损伤发生后,损伤部位会被一些蛋白识别,进而招募一系列蛋白至损伤部位,形成一个修复系统。DNA双链断裂是最严重的一种DNA损伤,错误修复往往导致疾病的发生。DNA双链断裂(double strand break, DSB)后,细胞启动RNF8/RNF168信号通路进行修复。RNF8和RNF168是这条通路的枢纽蛋白;53BP和BRCA1是关键的效应蛋白,决定着DSB修复的方式;组蛋白泛素化、磷酸化和甲基化等翻译后修饰是这条通路顺利进行的基本条件;染色质重塑、泛素化酶/去泛素化酶平衡和蛋白稳定性是这条通路的主要调节方式。本综述对RNF8/RNF168信号通路进行了梳理总结,希望其能对相关研究者起到参考作用。  相似文献

DNA双链断裂(double-strand breaks, DSBs)是细胞DNA损伤的主要类型,它的修复通过同源重组(HR)和非同源末端连接(NHEJ)两种机制实现.NHEJ是人和哺乳动物细胞DSBs修复的重要通路,主要由DNA依赖性蛋白激酶(DNA-PK)、X射线修复交叉互补蛋白4(XRCC4)、DNA连接酶Ⅳ、Artemis、XLF/Cernunnos和其它DNA损伤修复辅助因子组成.本文重点介绍了NHEJ机制主要成分的特性及其功能,以及这些组分的基因发生突变或缺失所引起的DSBs修复缺陷与辐射敏感性重症联合免疫缺陷(radiosensitive severe combined immunodeficiencies, RS-SCIDs).  相似文献

许多种癌症是由染色体转位引起的。这是因为转位使原癌基因活化并过量表达,或是转位使基因融合。大部分已确认的转位都与免疫系统(免疫球蛋白和T细胞受体基因)本身固有的V(D)J重组有关。此外,外界因素,如电离辐射和某些化学药物等因素使DNA双链断裂,也可能导致染色体转位和癌变。了解染色体转位和癌变机制,对临床诊断和防治具有重要应用价值 。  相似文献

Rad9是一种重要的细胞周期监控点调控蛋白.越来越多的证据显示,Rad9也可与多种DNA损伤修复通路中的蛋白质相互作用,并调节其功能,在DNA损伤修复中发挥重要作用.非同源末端连接修复是DNA双链断裂的一条重要修复途径.Ku70、Ku80和DNA依赖的蛋白激酶催化亚基(DNA-PKcs)共同组成DNA依赖的蛋白激酶复合物(DNA-PK),在非同源末端修复连接中起重要作用.本研究中,检测到Rad9与Ku70有直接的物理相互作用和功能相互作用.我们在不同的细胞模型中发现,Rad9基因敲除、Rad9蛋白去除或Rad9表达降低会导致非同源末端连接效率明显下降.已有的研究表明,DNA损伤可导致细胞中Ku70与染色质结合增加及DNA-PKcs激酶活性增强.我们的结果显示,与野生小鼠细胞相比,Rad9基因敲除的小鼠细胞中, DNA损伤诱导的上述效应均减弱.综上所述,我们的研究首次报道了Rad9与非同源末端连接修复蛋白Ku70间有相互作用,并提示Rad9可通过调节Ku70/Ku80/DNA-PKcs复合物功能参与非同源末端连接修复.  相似文献

从细胞的克隆形成能力和细胞DNA双链断裂及修复几方面分析了两个人卵巢癌细胞株HOC8和A2780对电离辐射的敏感性并探讨了ADP-核糖基转移酶(ADPR)的特异性抑制剂3-氨基苯甲酰胺(3-AB)对二者的辐射增敏效应,结果表明A2780细胞的辐射敏感性大大高于HOC8细胞,其D0值分别为0.9和2.5Gy;γ射线所致两株细胞的初始DNA双链断裂水平没有显著差异,但A2780细胞对DNA双链断裂的修复能力比HOC8细胞低.3AB能降低受照细胞的克隆形成能力及细胞对双链断裂的修复能力,其中对HOC8细胞的作用更为明显.  相似文献

用DNA合成仪合成寡聚脱氧核苷酸。用T4-DNA连接酶把这些寡聚脱氧核苷酸重组成双链DNA。这两个双链DNA的上游是T7-启动子,下游分别编码酵母丙氨酸tRNA的5′半分子(1-35位核苷酸)和3′半分子(35-76位核苷酸)。再把这两个双链DNA克隆到PUC 12质粒中。经点杂交筛选和DNA顺序测定证明克隆是成功的。  相似文献

电离辐射等多种因素可以引起DNA损伤,表现为碱基改变、DNA双链断裂(DNA double-strand breaks,DSBs)和DNA单链断裂(Single-strand breaks,SSBs)等多种形式。DNA损伤后,细胞发生应答,即引起细胞周期阻滞和/或细胞程序性死亡,以减少损伤引起的染色体畸变和基因组不稳定。在细胞应答过程中,p53蛋白水平和活性均发生变化,介导细胞周期阻滞、程序性死亡,并直接参与DNA损伤修复过程。  相似文献

DNA损伤的发生与积累是造成细胞功能紊乱的根本原因,也是引起衰老与肿瘤等疾病发生的关键事件。为维持机体自身遗传物质的完整性与稳定性,生物体内拥有多种针对不同类型DNA损伤的修复方式。Sirtuin蛋白是一组NAD+依赖的、高度保守的组蛋白去乙酰化酶,可通过去乙酰化作用调节众多底物蛋白质的表达、活性与稳定性。 近来的研究显示,DNA损伤修复途径的多个关键蛋白质是Sirtuin的下游底物。Sirtuin蛋白通过调节同源重组修复、非同源末端修复、核苷酸切除修复等途径中的核心蛋白质参与修复包括双链断裂(double stranded breakes, DSBs)在内的多种DNA损伤类型,从而在维持基因组稳定性、寿命以及细胞能量代谢调节等一系列生物学作用中发挥至关重要的作用。本综述将介绍近年来Sirtuin与DNA损伤修复的研究进展。  相似文献

The solution conformation is presented for representatives of each of the major classes of asparaginyl oligosaccharides. In this report the conformation of alpha(1-3)-, alpha(1-2)-, beta(1-2)-, and beta(1-4)-linked units is described. The conformational properties of these glycopeptides were determined by high-resolution 1H nuclear magnetic resonance in conjunction with potential energy calculations. The NMR parameters that were used in this analysis were chemical shifts and nuclear Overhauser enhancements. Potential energy calculations were used to evaluate the preferred conformers available for the different linkages in glycopeptides and to draw conclusions about the behavior in solution of these molecules. It was found that the linkage conformation of the Man alpha 1-3 residues was not affected by substitution either at the 2-position by alpha Man or beta GlcNAc or at the 4-position by beta GlcNAc or by the presence of a bisecting GlcNAc on the adjacent beta Man residue.  相似文献

Microbial hydroxylation of (+/-)-(2Z,4E)-5-(1',2'-epoxy-2',6',6'-trimethylcyclohexyl)-3-methyl-2,4-pentadienoic acid (3a) with Cercospora cruenta, a fungus producing (+)-abscisic acid, gave a four-stereoisomeric mixture consisting of (+)- and (-)-xanthoxin acid (4a), and (+)- and (-)-epi-xanthoxin acid (5a) by an HPLC analysis with a chiral column. Screening of the microorganisms capable of oxidizing (+/-)-3a showed that Cunninghamella echinulata stereoselectively oxidized (+/-)-3a to xanthoxin acid (4a) with the some degree of enantioselectivity as (-)-3a to (-)-4a.  相似文献

An efficient and facile synthesis of a large series of diverse 6-[2-(dialkylamino)vinyl]-, 6-[2-(dialkylamino)ethyl]-, 6-(2-alkoxyethyl)-, and 6-[2-(alkylsulfanyl)ethyl]purine nucleosides (35 examples of both ribo- and 2'-deoxyribonucleosides) was developed. The key transformations involved conjugate nucleophilic additions of amines, alcoholates, or thiolates to Tol-protected 6-alkylylpurine or 6-vinylpurine nucleosides. 6-[(2-Dialkylamino)vinyl]- and some 6-[(2-dialkylamino)ethyl]purine ribonucleosides exerted significant cytostatic effects and some anti-HCV activity with low selectivity.  相似文献

Both enantiomers of (3S)-(-)- and (3R)-(+)-Neodictyoprolenol [(3S,5Z,8Z)-(-)-1,5,8-undecatrien-3-ol] were successfully converted to the algal sex pheromone, (1S,2R)-(-)-dictyopterene B and (1R,2S)-(+)-dictyopterene B in high enantiomeric purities (e. e. > 99%), respectively, by the biomimetic reaction involving phosphorylation and elimination under a mild condition.  相似文献

Three new coordination polymers [M(Pht)(1-MeIm)2]n (where M=Cu (1), Zn (2), Co (3); Pht2−=dianion of o-phthalic acid; 1-MeIm=1-methylimidazole) and two compounds [M(1-MeIm)6](HPht)2 · 2H2O (M=Co (4), Ni (5)) have been synthesized and characterized by X-ray crystallography. The structures of 1-3 (2 is isostructural to 3) consist of [M(1-MeIm)2] building units connected by 1,6-bridging phthalate ions to form infinite chains. In complex 1, each copper(II) center adopts a square coordination mode of N2O2 type by two O atoms from different phthalate ions and two N atoms of 1-MeIm, whereas in 3 two independent metal atoms are tetrahedrally (N2O2) coordinated to a pair of Pht ligands and a pair of 1-MeIm molecules. There are only van der Waals interactions between the chains in 1, while the three-dimensional network in 3 is assembled by C-H?O contacts. In contrast to polymers 1-3 the structures of 4 and 5 (complexes are also isostructural) are made up of the [M(1-MeIm)6]2+ cation, two hydrogen phthalate anions (HPht) and two H2O solvate molecules. The coordination around each metal(II) atom is octahedral with six nitrogen atoms of 1-MeIm. Extended hydrogen bonding networks embracing the solvate water molecules and a phthalate residue as well as the weak C-H?O interactions stabilize the three-dimensional structures. Magnetic studies clearly show that the magnetic ions do not interact with each other. Furthermore, in compound 4 we have another example of a highly anisotropic Co2+ ion with a rhombic g-tensor and large zero-field-splitting. The complexes were also characterized by IR and 1H NMR spectroscopy, thermogravimetric analysis, and all data are discussed in the terms of known structures.  相似文献

The DNA cleavage activities of (+)-catechin (C), (-)-epicatechin (EC), (-)-epigallocatechin (EGC), and (-)-epigallocatechin gallate (EGCg) were examined with 16 different metal ions. Cu(2+) with all the catechins facilitated DNA cleavage, while Ag+ with EGC and EC showed a strong repressive effect. The other metal ions examined showed little effect.  相似文献

The trans-enantiomers of the commercially important anti-protozoal compound Halofuginone have been prepared and characterized, and the absolute configuration was assigned by X-ray crystallography. The activity of both enantiomers against Cryptosporidium parvum was determined in vitro and related to acute toxicity in vivo. It was shown that both the activity and the toxicity are properties of the (2R,3S)-enantiomer. We conclude that with respect to broadening the therapeutic window there is no advantage in application of one enantiomer over the application of the racemic mixture in the treatment of C. parvum infections.  相似文献
