Region 20 of the polytene X chromosome of Drosophila melanogaster was studied in salivary glands (SG) and pseudonurse cells (PNC) of otu mutants. In SG chromosomes the morphology of the region strongly depends on two modifiers of position effect variegation: temperature and amount of heterochromatin. It is banded in XYY males at 25° C and β-heterochromatic in X0 males at 14° C, i.e. it shows dynamic transitions. In PNC chromosomes region 20 is not heterochromatic, but demonstrates a clear banding pattern. Some molecular markers of mitotic heterochromatin were localized by means of in situ hybridization on PNC chromosomes: DNA of the gene su(f) in section 20C, the nucleolar organizer and 359-bp satellite in 20F. The 359-bp satellite, which has been considered to be specific for heterochromatin of the mitotic X chromosome, was found at two additional sites on chromosome 3L, proximally to 80C. The right arm of the X chromosome in SG chromosomes was localized in the inversion In(1LR)pn2b: the telomeric HeT-A DNA and AAGAG satellite from the right arm are polytenized, having been relocated from heterochromatin to euchromatin. The in vitro effect of four isolates of the nematophagous fungi Duddingtonia flagrans (AC 001), Monacrosporium sinense (SF 53) and Pochonia chlamydosporia (VC 1 and VC 4) on eggs of Schistosoma mansoni was examined. One thousand S. mansoni eggs were plated on 2% water–agar with the grown isolates and control without fungus. After 7, 14 and 21 days, the eggs were removed and classified according to the following parameters: type 1, lytic effect without morphological damage to eggshell; type 2, lytic effect with morphological alteration of embryo and eggshell; and type 3, lytic effect with morphological alteration of embryo and eggshell, besides hyphal penetration and internal egg colonization. Significant differences (P < 0.01) were found among the studied fungal isolates for ovicidal activity, confirming type 3 effect for the isolates VC 1 and VC 4, which characterizes the ovicidal activity of a fungus. Type 3 effect was only found for P. chlamydosporia (VC 1 and VC 4) with 26.6 and 17.2%, 25.6 and 22.6%, 27.4 and 23.9% in the 7, 14 and 21 days respectively (P < 0.01). P. chlamydosporia can thus be a potential biological control agent for S. mansoni eggs.  相似文献

MYB类转录因子在植物生长发育、代谢、应答生物胁迫和非生物胁迫的响应等生物过程发挥重要作用。为探究马尾松R2R3-MYB基因结构及功能,该研究以转录组数据为研究区域,从中筛选获得了17个马尾松R2R3-MYB基因,利用生物信息学对基因进行理化性质、系统进化树等分析,同时利用荧光定量PCR技术分析基因的组织特异性以及在花发育时期和非生物胁迫下的表达模式。结果表明:(1)17个PmMYBs亚细胞定位于细胞核,均无跨膜结构,且均含有Motif1、Motif2保守基序。系统发育进化树将马尾松PmMYBs划分为9个亚家族,且与火炬松、白云杉等裸子针叶植物关系较近。(2)17个基因均属于组成型表达,但在不同组织的表达量不同;所有基因均参与了花发育和非生物胁迫,不同基因在花发育不同时期的表达存在差异,有7个基因可能参与了雌雄性状转变;大部分基因响应非生物胁迫上调表达,但响应胁迫的时间存在差异;少数基因在胁迫中下调表达,尤其是PmMYB11基因在所有胁迫中均明显下调表达。该研究较系统地分析了马尾松R2R3-MYB基因的结构特征、系统进化及其在花发育时期和非生物胁迫下的表达模式,为深入探究马尾松R2R3...  相似文献

香豆酸-3-羟化酶属于植物中最大的蛋白酶细胞色素P450家族之一,在植物生命活动中发挥着重要作用。为了解地黄香豆酸-3-羟化酶基因RgC3H合成毛蕊花糖苷的功能,该研究基于地黄代谢组学分析获得KEGG途径中的C3H,采用多重比对在NCBI中获得同源基因的一个保守序列,并基于该保守序列和地黄SRA数据库,采用电子克隆和RT-PCR克隆技术获得地黄C3H基因全长CDS(RgC3H),对其进行生物信息学分析。结果表明:RgC3H基因全长为1 530 bp,且编码一个含509个氨基酸、分子量为57.91 kD、无信号肽的蛋白质; 基于氨基酸序列的结构分析显示,RgC3H有一个保守区域-P450结构域; 系统进化分析结果显示,RgC3H与芝麻和猴面花的C3H基因具有很高的同源性。上述结果为进一步研究RgC3H基因在地黄毛蕊花糖苷生物合成途径中的作用奠定了基础。  相似文献

MYB 是一类常见的转录因子,广泛参与植物花青素生物合成的调控。为探究 MYB转录因子在甜荞花青素生物合成中的调控作用,该研究从红花甜荞和白花甜荞转录组学数据中筛选并克隆出一个和花青素生物合成相关的MYB基因,将其命名为 FeR2R3-MYB,GenBank 登录号为 MT151381.1,并对该序列进行生物信息学分析,以及利用 qRT-PCR 分析FeR2R3-MYB基因在白花甜荞和红花甜荞中的表达特征。结果表明:(1)FeR2R3-MYB基因全长 831 bp,编码 276 个氨基酸,蛋白的相对分子质量为 30.95 kD,理论等电点(pI)为 8.73,蛋白的不稳定指数为 69.64,属于不稳定蛋白,总疏水值为-0.679,整条肽链呈现亲水特性。(2)FeR2R3-MYB 具有典型的 R2R3-MYB 结构域,属于 R2R3-MYB 亚家族。(3)FeR2R3-MYB 与同属蓼科的苦荞和虎杖亲缘关系比较近。(4)FeR2R3-MYB 的启动子序列共含有 9 个光照响应元件、17 个转录因子结合位点、4 个非生物响应元件和 2 个激素响应元件。(5)亚细胞定位发现 FeR2R3-MYB 只在细胞核中表达。(6)FeR2R3-MYB 基因的表达量在叶片和花序中红花甜荞均高于白花甜荞,推测 FeR2R3-MYB 基因可以正向调节甜荞花青素生物合成。综上所述,该研究结果为进一步深化 FeR2R3-MYB 基因在甜荞花青素生物合成途径中的功能及表达调控方面的研究提供了基础。  相似文献

分别构建了含cry3Acry3A+vhb基因的植物表达载体pBCry3A和pBC3Vhb,并通过根癌农杆菌介导转化了马铃薯. 对转化再生植株进行PCR和DNA印迹分析表明,外源基因已整合到马铃薯基因组中, 且连续三代无性繁殖后转基因仍存在. ELISA分析表明cry3A基因在转基因植株中得到了高效表达, 在单转cry3A植株中最高表达量达0.1%, 转cry3Avhb双基因株系中为0.065%. 水涝试验显示,转双基因且vhb mRNA的RT-PCR呈阳性的马铃薯植株,对低氧胁迫有较好的耐受性, 表明获得的上述转双基因马铃薯株系可能会具有很好的抗虫和耐涝性能.  相似文献

Six geographic samples of Cyclina sinensis were collected from the coast along China and analyzed to reveal morphological and genetic variation by using nine allozyme loci and 11 morphological variables. The discriminant function analysis (DFA) of morphology suggested a clear separation between the southern and northern populations. Polymorphism was detected at nine loci across all six populations. The mean allele number ranged from 2.44 to 2.78, and the mean observed heterozygosity ranged from 0.218 to 0.296. High level of genetic differentiation was found between three northern populations and three southern populations. The marked genetic differentiation can be explained by the upwelling of the Zhejiang province and the freshwater outflow of Yangtze River. The results obtained in this study indicate that the northern and southern populations of C. sinensis should be treated as separate units for conservation management.  相似文献

Populus serves as a model tree for biotechnology and molecular biology research due to the availability of the reference genome sequence of Populus trichocarpa (Torr. & Gray) genotype ‘Nisqually-1’. However, ‘Nisqually-1’ has been shown to be very recalcitrant to micropropagation, regeneration and transformation. In this study, a highly efficient micropropagation protocol from greenhouse-grown shoot tips of ‘Nisqually-1’ was established. The optimal micropropagation protocol involves growing in vitro shoots in plant growth regulator-free Murashige and Skoog (MS) basal medium supplemented with 3% sucrose, 0.3% Gelrite? and 5–10 g L−1 of activated charcoal. Plants grown on this medium were significantly longer, and contained significantly higher concentrations of chlorophyll. This highly effective protocol provides a consistent supply of quality leaf and stem materials throughout the year for transformation experiments and other in vitro manipulations, therefore eliminating inconsistency due to seasonal and greenhouse environmental variations and the need for repetitive tissue sterilization.  相似文献

该研究以华丽龙胆(Gentiana sino ornata)5个不同开放阶段(H1~H5)的蓝色花冠为试材,利用RT PCR技术克隆GsF3′5′HGsFNS全长序列,并进行生物信息学分析,比较GsF3′5′HGsFNS在不同组织和不同开放阶段的基因表达模式。结果显示:(1)所克隆的GsF3′5′HGsFNS基因分别包含1 560 bp和1 590 bp开放阅读框(OFR),并分别编码520和529个氨基酸。(2)结构分析显示,GsF3′5′H和GsFNS均具有典型的F3′5′H和FNSⅡ蛋白保守结构域。(3)系统进化树分析表明,GsF3′5′H和GsFNS亲缘关系最近的物种是三花龙胆(Gentiana triflora)。(4)qRT PCR结果显示,GsF3′5′HGsFNS基因在根、茎、叶和花冠中均表达,其中GsF3′5′H基因在花冠H3阶段表达量最高。GsFNS在根中表达量最高,其次在花冠H4阶段两基因的表达均较高。研究推测,GsF3′5′H基因表达产生的飞燕草素苷和GsFNS表达产生的黄酮共着色作用可能使华丽龙胆的花冠呈更稳定艳丽的蓝色,为蓝色花分子育种提供重要的基因资源。  相似文献

We have attempted to investigate the correlation between the detergent-perturbed structural integrity of the Cyt b 6 f complex from the marine green alga Bryopsis corticulans and its photo-protective properties, for which the nonionic detergents n-octyl-β-d-glucopyranoside (β-OG) and n-dodecyl-β-d-maltoside (β-DM), respectively, were used for the preparation of Cyt b 6 f, and the singlet oxygen (1O2*) production as well as the triplet excited-state chlorophyll a (3Chl a*) formation and deactivation were examined by spectroscopic means. Near-infrared luminescence of 1O2 * (~1,270 nm) on photo-irradiation was detected for the β-OG preparation where the complex is mainly in oligomeric state, but not for the β-DM one in which the complex exists in dimeric form. Under anaerobic condition, photo-excitation of Chl a in the β-DM preparation generated 3Chl a* with a lower quantum yield of ΦT ~ 0.02 and a longer lifetime of ~600 μs with respect to those as in the case of β-OG preparation, ΦT ~ 0.12 and 200–300 μs. These results prove that the enzymatically active and intact Cyt b 6 f complex on photo-excitation tends to produce little 3Chl a* or 1O2 *, which implies that the pigment–protein assembly of Cyt b 6 f complex per se is crucial for photo-protection. The problems of delimitation of species of Prosopis originate from the few morphological discontinuities which exist among some of them; some, however, originated as a result of wide distribution of germplasm without proper knowledge of the species, in particular, much material catalogued as P. juliflora, but being of other species, was distributed for reforestation projects worldwide. This work tests the morphological results obtained for P. pallida and P. limensis of the Peruvian–Ecuadorian coast and for P. juliflora of the Caribbean Basin of Colombia and Venezuela utilizing a study of AFLPs and a study of the morphology of plantlets developed in a conventional garden study. The phenogram obtained for the AFLPs demonstrates each of the three species to be a well differentiated cluster and the molecular variance between them is significantly greater than the variance within each species. Study of the plantlets also indicates statistically significant differences for four morphological characters between P. juliflora and the other two species (P. pallida and P. limensis). These results, in addition to the morphological differentiation evident between adult plants of P. pallida and P. limensis and the clear separation of these two species from P. juliflora, corroborate the genetic identity of the three taxa analyzed.  相似文献

Subunit 3 (Rpb3) of eukaryotic RNA polymerase II is a homologue of the α subunit of prokaryotic RNA polymerase, which plays a key role in subunit assembly of this complex enzyme by providing the contact surfaces for both β and β′ subunits. Previously we demonstrated that the Schizosaccharomyces pombe Rpb3 protein forms a core subassembly together with Rpb2 (the β homologue) and Rpb11 (the second α homologue) subunits, as in the case of the prokaryotic α2β complex. In order to obtain further insight into the physiological role(s) of Rpb3, we subjected the S. pombe rpb3 gene to mutagenesis. A total of nine temperature-sensitive (Ts) and three cold-sensitive (Cs) S. pombe mutants have been isolated, each (with the exception of one double mutant) carrying a single mutation in the rpb3 gene in one of the four regions (A–D) that are conserved between the homologues of eukaryotic subunit 3. The three Cs mutations were all located in region A, in agreement with the central role of the corresponding region in the assembly of prokaryotic RNA polymerase; the Ts mutations, in contrast, were found in all four regions. Growth of the Ts mutants was reduced to various extents at non-permissive temperatures. Since the metabolic stability of most Ts mutant Rpb3 proteins was markedly reduced at non-permissive temperature, we predict that these mutant Rpb3 proteins are defective in polymerase assembly or the mutant RNA polymerases containing mutant Rpb3 subunits are unstable. In accordance with this prediction, the Ts phenotype of all the mutants was suppressed to varying extents by over-expression of Rpb11, the pairing partner of Rpb3 in the core subassembly. We conclude that the majority of rpb3 mutations affect the assembly of Rpb3, even though their effects on subunit assembly vary depending on the location of the mutation considered. Genetic transformation of androgenic-derived amphidiploid Festulolium plants (Lolium perenne L. × Festuca pratensis Huds., 2n = 4x = 28) by Agrobacterium tumefaciens has been achieved. Anther culture-induced calli of Festulolium “Bx351” were inoculated with Agrobacterium tumefaciens strain LBA4404 carrying pIG121-Hm encoding the hygromycin resistance (hph) and β-glucuronidase (uidA) genes under the control of a CaMV 35S promoter. Twenty-three putative transformants were obtained from the hygromycin selection, 19 of which (82.6%) showed GUS activity. The integration of transgene was detected by using genomic DNA PCR analysis, RT-PCR analysis and Southern blot hybridization, respectively, which revealed that foreign gene was integrated into the genomes of dihaploid transformants (2n = 2x = 14). The haploid embryogenic system offers a stable means of transformation, as the introduced trait can be readily fixed through chromosome doubling. An erratum to this article can be found at  相似文献

该研究构建植物表达载体pBin438 Vip3A,通过农杆菌介导法转化棉花品种‘冀合713’,将新型抗虫基因Vip3A导入到棉花植株,创制对棉铃虫抗性的转基因棉花新种质。结果表明:(1)PCR检测Vip3A基因已经导入到棉花基因组中且能够稳定遗传。(2)室内抗虫性鉴定表明,与对照相比转基因植株对棉铃虫的抗性显著提高,并获得2株高抗和3株抗棉铃虫的转Vip3A基因株系。(3)Southern blotting结果显示,转基因株系BV01为单拷贝。(4)Elisa检测表明,外源Vip3A基因在BV01的根、茎、叶、花、种子中都有表达,在叶片中的Vip3A蛋白表达为苗期>蕾期>花期>铃期>吐絮期。该研究创制了新型抗虫转基因棉花材料,为培育棉花新型抗虫品种提供了种质资源。  相似文献
