代谢组学是定量描述生物内源性代谢物质的整体及其对内因和外因变化应答规律的的一门新学科。近年来,代谢组学技术在水产动物疾病中的研究备受关注,特别是为感染性疾病发生机制及防控研究提供了一种新的手段。本文介绍了代谢组学技术及其在水产动物研究中的应用,包括代谢组学技术在水产动物感染性疾病、细菌耐药及环境应激等方面应用进行综述,分析了代谢组学在水产动物疾病研究中面临的问题与挑战,并对未来水产动物代谢组学研究趋势进行了展望,以期为代谢组学技术在水产动物疾病发病机制和药物研发方面更深入的运用提供参考。  相似文献

在后基因组时代, 系统生物学研究成为人们关注的焦点。转录组学、蛋白质组学等功能基因组学研究方法可同时检测药物或其他因素影响下大量基因或蛋白质的表达变化情况, 但这些变化不能与生物学功能的变化建立直接联系。代谢组学方法则可为代谢物含量变化与生物表型变化建立直接相关性。代谢组学研究的目的是定量分析一个生物系统内所有代谢物的含量, 进行全面代谢物分析需要分析化学技术的支撑, 核磁共振和基于质谱的分析技术是代谢组学研究的两种主要技术手段。代谢组学研究可产生大量数据信息, 对这些数据进行分析离不开化学统计学的应用, 比如主成分分析、多维缩放、各种聚类分析技术以及功能差异分析等。文章综述了近年来代谢组学分析技术及数据分析技术的研究进展, 在此基础上, 对代谢组学在临床研究及临床前研究中的应用研究进展进行了综述。对疾病代谢表型图谱的研究有助于人们了解疾病发生、发展以及致死的机制; 在临床条件下, 这些代谢图谱可以作为疾病诊断、预后以及治疗的评判标准。代谢物组成的变化是毒物胁迫对机体造成的最终影响, 利用代谢组技术可以直接反映毒物对机体的影响。质谱技术、核磁共振技术的应用使得药物筛选过程可以快速完成, 并有助于实现个性化用药。此外, 利用代谢组学技术还可以进行已知酶的新活性研究, 也可以研究未知酶。  相似文献

基于稳定同位素示踪的代谢组学不仅能检测疾病发生发展及治疗过程中相关代谢物的变化,还可以对生物体系的代谢物进行定量分析并描述代谢特性。目前,稳定同位素示踪的代谢组学技术可在受试者体内直接追踪到代谢网络的单个原子,更有利于发现与疾病病因和诊断及药物疗效评估相关的生物标志物。现就稳定同位素示踪代谢组学在临床研究中的应用做简要概述,以期为将来更好地开展代谢组学在临床方面的研究提供重要依据。  相似文献

粪菌移植技术(fecal microbiota transplantation,FMT)是利用健康人群的粪便或经过处理的粪便中的微生物来治疗消化、代谢等系统诸多疾病的一项古老而又新兴的技术。宏基因组、培养组等肠道微生物组前沿研究技术的飞速发展,为粪菌移植治疗疾病提供了强有力的研究和临床实践武器。宏基因组技术可全面揭示健康及疾病状态下肠道微生态的组成及功能变化,而培养组学技术则可以用来分离和鉴定人类肠道中的诸多在常规培养条件下未可培养菌,两项技术联用不仅可以使我们更为深入地理解粪菌移植在临床实践中的因果规律,还将有力推动粪菌移植技术在未来的应用与发展。基于此,本文综述了宏基因组及培养组学技术在粪菌移植中的应用及未来发展趋势。  相似文献

代谢组学是后基因时代新兴的一门研究生物体内所有小分子代谢物的组学学科,是系统生物学的有机组成部分。由于代谢组学是基于数据驱动的学科,因此如何有效利用数据预处理、模式识别等信息处理技术从代谢组学复杂的高维样本中挖掘深层次的“知识”是代谢组学乃至整个系统生物学的关键问题。对模式识别技术在代谢组学中的应用作了全面的综述。总结代谢组学数据特性及其对模式识别技术的特殊要求,揭示面向代谢组学的模式识别技术所遇到的困难并在此基础上提出相应的解决办法。  相似文献

代谢组学指某一生物系统中产生的或已存在的代谢物组的研究,以质谱和核磁共振技术为分析平台,以信息建模与系统整合为目标。随着代谢组学中的研究方法与技术成为生态学研究的有力工具,生态代谢组学概念应运而生,即研究某一个生物体对环境变化的代谢物组水平的响应。理清代谢组学与生态代谢组学学科发展的脉络,综述代谢组学研究中的常用技术及其优势与局限性,论述代谢组学技术在生态学研究中的应用现状,展望代谢组学技术与其他系统生物学组学技术的结合在生态学中的应用前景,提出生态代谢组学研究者未来要完成的任务和面对的挑战。  相似文献

代谢组学是"后基因组学"时期新兴的一门学科,也是系统生物学的重要组成部分。代谢组学通过全面、定量检测生物样本中多种类型小分子化合物,来了解在内在和外界因素作用下生物体内源性物质的变化及规律,特别适合于临床上研究机体因受到遗传、生长、生理、环境因素和异物、病源等刺激的影响而产生的变化。借助于代谢组学技术不仅能够描述疾病发生、发展以及治疗过程中机体代谢机能的状态和变化,为临床疾病的诊断、病理机制的探索、新治疗靶点的发现等提供新的途径和思路,还可以揭示外界干扰因素(药物/毒物、环境、饮食、生活方式等)对机体的影响,为药效评价和疾病病因的筛查提供基础数据。近年来,代谢组学在临床研究方面得到了广泛的应用,取得了巨大的进展并展现了鼓舞人心的应用前景。该文分别就代谢组学在描述疾病发展状态、研究疾病诊断方法、探索疾病发病原因和发病机理、药效学评价等几个方面的应用及进展进行回顾和综述。  相似文献

Carbon monoxide, long considered only as a toxic gas, has recently been shown to mediate potent anti-inflammatory and other salutary effects in rodents when it is used at low doses. Carbon monoxide is one of the products of the degradation of heme by heme oxygenase 1. Until recently, these beneficial effects of carbon monoxide were shown only when it was given before a stress stimulus. Hagazi and colleagues have recently shown that this substance is effective even when it is given after a disease process has started. The effects of low doses of carbon monoxide are complemented by the production of biliverdin and probably also by ferritin, which are additional products of heme degradation.  相似文献

Carbon monoxide poisoning in a 37-year-old man was complicated by neurologic damage, skin changes, muscle necrosis and nonoliguric renal failure. The relation between nontraumatic rhabdomyolysis and acute renal failure in carbon monoxide poisoning is reviewed. Recognition of the acute renal failure in such cases is important, for this complication can be fatal; the prognosis is excellent, however, if proper medical management is provided.  相似文献

Carbon monoxide poisoning is a significant cause of illness and death. Its protean symptoms probably lead to a gross underestimation of its true incidence. Low levels of carbon monoxide aggravate chronic cardiopulmonary problems, and high levels are associated with cardiac arrhythmias and cerebral edema. Patients who survive acute poisoning are at risk of delayed neurologic sequelae. The measurement of carboxyhemoglobin levels does not reveal the tissue levels of carbon monoxide but is useful in determining therapy. Treatment includes the monitoring and management of cardiac arrhythmias and oxygenation. Hyperbaric oxygenation is beneficial, but there are currently no definite criteria for its use.  相似文献

A physiological role of carbon monoxide has been suggested for coronary myocytes; however, direct evidence is lacking. The objective of this study was to test the effect of chronic carbon monoxide exposure on the K(+) currents of the coronary myocytes. The effect of 3-wk chronic exposure to carbon monoxide was assessed on K(+) currents in isolated rat left coronary myocytes by the use of the patch-clamp technique in the whole cell configuration. Moreover, membrane potential studies were performed on coronary artery rings using intracellular microelectrodes, and coronary blood flow in isolated heart preparation was recorded. Carbon monoxide did not change the amplitude of global whole cell K(+) current, but it did increase the component sensitive to 1 mM 4-aminopyridine. Carbon monoxide exposure hyperpolarized coronary artery segments by approximately 10 mV and, therefore, increased their sensitivity to 4-aminopyridine. This effect was associated with an enhancement of coronary blood flow. We conclude that chronic carbon monoxide increases a 4-aminopyridine-sensitive current in isolated coronary myocytes. This mechanism could, in part, contribute to hyperpolarization and to increased coronary blood flow observed with carbon monoxide.  相似文献

Unexpected carbon monoxide poisonings The poisonous gas carbon monoxide (CO) is not only produced as a result of combustion processes but can also be inhaled during the operation of barbecues, the smoking of water pipes, or in wood pellet stores. Inhalation of carbon monoxide can lead to life‐threatening or chronic poisoning. 目的:探讨七叶皂苷钠对一氧化碳中毒迟发性脑病大鼠模型细胞凋亡及Caspase-3表达的影响。方法:将30只大鼠随机分为正常对照组、一氧化碳中毒迟发性脑病大鼠模型对照组和七叶皂苷钠治疗组,原位末端标记法(TUNEL)检测细胞凋亡,免疫组化法检测Caspase-3的表达。结果:治疗组大鼠脑细胞凋亡率及Caspase-3表达较迟发性脑病对照组显著降低(P<0.01)。结论:七叶皂苷钠对一氧化碳中毒迟发性脑病的脑保护作用机制可能与干预脑细胞凋亡相关基因表达并减少神经细胞凋亡有关。  相似文献

The stress-inducible protein heme oxygenase-1 provides protection against oxidative stress. The anti-inflammatory properties of heme oxygenase-1 may serve as a basis for this cytoprotection. We demonstrate here that carbon monoxide, a by-product of heme catabolism by heme oxygenase, mediates potent anti-inflammatory effects. Both in vivo and in vitro, carbon monoxide at low concentrations differentially and selectively inhibited the expression of lipopolysaccharide-induced pro-inflammatory cytokines tumor necrosis factor-alpha, interleukin-1beta, and macrophage inflammatory protein-1beta and increased the lipopolysaccharide-induced expression of the anti-inflammatory cytokine interleukin-10. Carbon monoxide mediated these anti-inflammatory effects not through a guanylyl cyclase-cGMP or nitric oxide pathway, but instead through a pathway involving the mitogen-activated protein kinases. These data indicate the possibility that carbon monoxide may have an important protective function in inflammatory disease states and thus has potential therapeutic uses.  相似文献

Carbon monoxide induces delayed neurological and neuropathological alterations, including memory loss and cognitive impairment. The bases for the delay remain unknown. Activation of soluble guanylate cyclase by nitric oxide modulates some forms of learning and memory. Carbon monoxide binds to soluble guanylate cyclase, activating it but interfering with its activation by nitric oxide. The aim of this work was to assess whether exposure of rats to carbon monoxide alters the activity of soluble guanylate cyclase or its modulation by nitric oxide in cerebellum or cerebral cortex. Rats exposed chronically or acutely to carbon monoxide were killed 24 h or 7 days later. Acute carbon monoxide exposure decreased cyclic guanosine monophosphate (cGMP) content and reduced activation of soluble guanylate cyclase by nitric oxide. Cortex was more sensitive than cerebellum to chronic exposure, which reduced activation of soluble guanylate cyclase by nitric oxide in cortex. In cerebellum, chronic exposure induced delayed impairment of soluble guanylate cyclase activation by nitric oxide. Acute exposure effects were also stronger at 7 days than at 24 h after exposure. This delayed impaired modulation of soluble guanylate cyclase by nitric oxide may contribute to delayed memory loss and cognitive impairment in humans exposed to carbon monoxide.  相似文献

The objective of this study was to evaluate the brain function characteristics of carbon monoxide poisoning patients using resting-state functional magnetic resonance imaging (fMRI) method. For this purpose, 12 carbon monoxide poisoning patients and healthy controls were subjected to resting-state fMRI scans separately. A regional homogeneity (ReHo) approach was used to analyze the brain function in carbon monoxide poisoning patients. Compared with control group, the value of ReHo in carbon monoxide poisoning group showed distinct decrease in bilateral superior frontal gyrus, middle frontal gyrus, right cuneus, left middle temporal gyrus, right insula, and cerebellum. Therefore, it was concluded that the brain functions in carbon monoxide poisoning patients were abnormal under the resting-state. The cuneate lobe function may indicate the degree of brain hypoxia and strengthening the cerebellar function training may promote the rehabilitation process.  相似文献

Carbon monoxide is produced in the endothelial cells and has possible vasodilator activity through three different pathways. The aim of this study was to demonstrate circulatory effects after administration of saturated carbon monoxide blood and to describe the pharmacokinetics of carbon monoxide. Six pigs were anesthetized and 150 ml blood was removed. This blood was bubbled with carbon monoxide until the carboxyhemoglobin (COHb) levels were 90-99%. A specific amount of this blood was then injected back to the animal. At predetermined times; arterial and mixed venous blood was drawn and analyzed for carbon monoxide. Systemic and pulmonary vascular resistance index (SVRi and PVRi) were measured and exhaled air was sampled and measured for carbon monoxide. Blood samples were gathered over 300 minutes along with measurements of invasive pressures, heart rate, cardiac output, oxygen saturation (SpO2), Hb, temperature and blood gases. We conclude that this type of exposure to carbon monoxide appears to have little or no effect on general vasomotor tone and, after correcting for basal levels of carbon monoxide, elimination occurs through the lungs as predicted by a single compartment model. The half-life of carbon monoxide was determined to be 60.5 minutes (SEM 4.7).  相似文献
