Exposure of workers to airborne microorganisms in open-air swine houses   总被引:7,自引:0,他引:7  
This study quantified the levels of airborne microorganisms in six swine farms with more than 10,000 pigs in subtropical Taiwan. We evaluated breeding, growing, and finishing stalls, which were primarily open-air buildings, as well as partially enclosed farrowing and nursery piggeries. Airborne culturable bacteria, gram-negative bacteria, and fungi were placed on appropriate media by using an all-glass impinger or single-stage Andersen microbial sampler. Results showed that mean concentrations of culturable bacteria and gram-negative bacteria were 3.3 x 10(5) and 143.7 CFU/m(3), respectively. The concentration of airborne culturable fungi was about 10(3) CFU/m(3), with Cladosporium the predominant genus. The highest airborne levels of culturable bacteria and gram-negative bacteria were identified in the finishing units. The air of the nursery stalls was the least contaminated with culturable and gram-negative bacteria. Irregular and infrequent cleaning, high pig density, no separation of wastes from pen floors, and accumulation of water as a result of the processes for cleaning and reducing pig temperature possibly compromise the benefits of the open characteristic of the finishing units with respect to airborne bacterial concentration.  相似文献

Indoor and outdoor airborne fungal propagule concentrations in Mexico City   总被引:7,自引:0,他引:7  
Thirty homes of asthmatic adults located in Mexico City were examined to determine the predominant culturable fungi and the changes in their airborne concentrations. Fungi were cultured and identified microscopically from air samples collected in naturally ventilated homes, during both wet (July–August) and cool dry (November–December) seasons, and from settled dust from the same homes. Airborne dust from indoor yielded 99–4950 cfu m−3, and settled dust 102–106 cfu g−1 on DG18 agar. The indoor geometric mean concentration of airborne fungi during the cool dry season was 460 cfu m−3 while in the wet season it was 141 cfu m−3. Similarly, numbers of airborne fungal propagules out of doors decreased 60% between the dry and wet season. In general, the total fungal concentrations in indoor air were less than 103 cfu m−3 and a large proportion of them was collected in Stage-2 of the Andersen sampler. Moreover, the ratio between indoor and outdoor concentrations was <3:1. Five of the 30 sampled homes yielded >500 cfu m−3 of one genus, with up to 1493Cladosporium cfu m−3 or 2549Penicillium cfu m−3. Also, these two genera were predominant in both airborne and settled dust, and their concentrations were greater indoors than out, indicating a possible indoor source of fungal propagules. The predominant species wereCladosporium herbarum, Penicillium aurantiogriseum andP. chrysogenum. These results suggest that exposure to large concentrations of fungi occurs indoors and is associated with both seasons of the year and with particular home characteristics.  相似文献

When microorganisms are used for pest control in vegetable production, the active organisms become part of the microbiota growers are exposed to. The aim of this study was to quantify vegetable growers' exposure to the bacterial strain Bacillus thuringiensis kurstaki strain HD1 (termed HD1) from the biocontrol agent Dipel®, and other airborne mesophilic bacteria. Personal (n=102) and stationary (n=43) measurements of exposure were performed in greenhouses and open fields. Air samples were analysed by plate counts, and total counts with a microscope. Isolates resembling HD1 were identified by PCR analysis. HD1-like bacteria were only detected in environments where Dipel® was used. In a greenhouse with Dipel® treated tomato plants, the growers' exposure to airborne HD1-like bacteria reached 5300 cfu/m3 and 1400 cfu/m3 during harvest and clearing of old plants, respectively. In untreated greenhouses, the highest concentration of total mesophilic bacteria, 1,100,000 cfu/m3, was detected in a cucumber greenhouse. The median concentrations of mesophilic bacteria in tomato greenhouses were significantly lower than the median concentrations in cucumber greenhouses. There was no significant difference in exposure to mesophilic bacteria in tomato greenhouses and in vegetable fields. We found that greenhouse workers, especially in cucumber production, were exposed to high concentrations of total bacteria. Thus, the already present airborne bacteria in greenhouses might have a greater influence on growers' health than applied biocontrol strains. However, further studies are needed to establish an occupational threshold limit for airborne bacteria and to secure a healthy working environment for vegetable growers.  相似文献

Numbers of airborne microorganisms, fungi, Gram-negative bacteria, thermophilic microorganisms, endotoxins and dust have been monitored in resource recovery plants and composting plants. The work is still in progress, so this paper decribes only preliminary results. Only low levels (< 15 ng m?3) of endotoxins were found at all locations. Levels of microorganisms, fungi, Gram-negative bacteria and dust changed with quality of waste, activity in the plant etc. Levels of airborne microorganisms, endotoxins and dust could be considerably decreased in resource recovery plants if only waste of good quality, e.g. presorted materials, is handled. For composting plants the highest levels of airborne microorganisms were found during aeration, especially by indoor composting where levels of 8.3 × 105 CFU of mesophilic microorganisms were found.  相似文献

The waste management occupational environment is recognized by the simultaneous presence of several substances and biologic agents. Therefore, workers are exposed simultaneously to multiple contaminants. Occupational exposure to aflatoxin B1 in one Portuguese waste sorting plant was already reported. However, besides this mycotoxin, data regarding fungal contamination showed that exposure to other mycotoxins could be expected. A study was developed to analyze if exposure to other mycotoxins besides aflatoxin B1 was occurring in the workers from the waste sorting plant previously assessed and to discuss how these findings need to be considered in the risk assessment process. In addition to aflatoxin B1 detected previously by ELISA, two additional mycotoxins and one mycotoxin degradation product were detected and quantified by a multi-mycotoxin HPLC-MS/MS approach: Enniatin B and ochratoxin A as well as 2’R-ochratoxin A. Besides the confirmation of co-exposure to several mycotoxins, results probably indicate different exposure routes for the mycotoxins reported.  相似文献

This study investigates the exposure of workers to biological particles in a poultry litter burning plant in operation. The microorganism concentrations were examined at different workplaces during procedures leading to increased emissions. The concentrations of culturable airborne mesophilic, xerophilic and thermophilic microorganisms in the ambient air were tested inside and outside of the burning plant using two different methods of measuring. The focus of this study was on the quantitative evaluation of culturable bacteria as well as the quantitative and qualitative evaluation of gram-negative bacteria, fungi and thermophilic actinomycetes. The maximum airborne concentrations were found in the delivery hall. Mesophilic bacteria concentrations reached up to 1.7 × 106 CFU/m3; gram-negative bacteria up to 9.1 × 102 CFU/m3. Fungal propagule concentrations for xerophilic fungi were between 1.2 × 103 and 2.9 × 104 CFU/m3 and for mesophilic fungi between 4.4 × 102 and 2.9 × 104 CFU/m3. Among fungi, Aspergillus niger, Eurotium herbariorum and Scopulariopsis brevicaulis species were dominant. Thermophilic actinomycetes reached airborne concentrations of 8.7 × 104 CFU/m3, with increased concentrations of the pathogens causing extrinsic allergic alveolitis. The high concentrations of airborne microorganisms in poultry litter burning plants and the potential hazard of the intake of microorganisms including potential pathogens require the introduction of consistent measures in both technical areas and personnel management.  相似文献

Cultivation-based microbiological methods are a gold standard for monitoring of airborne micro-organisms to determine the occupational exposure levels or transmission paths of a particular infectious agent. Some highly contagious microorganisms are not easily culturable but it is becoming evident that cultivation and molecular methods are complementary and in these cases highly relevant. We report a simple and efficient method for sampling and analyzing airborne bacteria with an impactor-type high-flow-rate portable air sampler, currently used for monitoring culturable bacteria or fungi. A method is reported for extraction of nucleic acids from impacted cells without prior cultivation and using agarose as a sampling matrix. The DNA extraction efficiency was determined in spiked samples and, samples taken from a wastewater treatment plant and an alpine area. The abundance, diversity and quantity of total bacteria were analysed by a quantitative polymerase chain reaction, and by construction and analysis of clone libraries. The method does not interfere with downstream PCR analysis and can cover the gap between traditional culture and molecular techniques of bioaerosol monitoring.  相似文献

采用KC-6120空气综合采样器采集空气微生物样品,通过构建16S/18SrDNA克隆文库方法分析青岛市市区街道秋季空气微生物群落结构特征.结果表明: 空气细菌分布在6大类,分别为变形菌门(78.8%)、厚壁菌门(14.6%)、放线菌门(4.0%)、浮霉菌门(1.3%)、蓝藻门(0.7%)和栖热菌门(0.6%),优势菌属为不动杆菌属(39.7%)、葡萄球菌属(11.3%)、鞘脂单胞菌属(8.6%)和副球菌属(6.0%).空气真菌分布在子囊菌门(97.5%)和担子菌门(2.5%),优势菌属为核腔菌属(76.5%)、炭角菌属(13.6%)和外瓶霉属(2.5%).空气微生物中存在不动杆菌属、鞘脂单胞菌、葡萄球菌等致病菌或条件致病菌,以及引发多种农作物枯萎死亡的麦类核腔菌、团炭角菌和角状平脐疣孢等真菌.  相似文献

In work environments with laboratory animals, the bedding of animals binds the excreta as well as other compounds originating from the animals and their environment. These may be generated into the ambient air when the personnel handle bedding in different procedures. This study compares the dustiness of different types of six clean and four soiled beddings from rat or mouse cages. The dust generation of clean bedding varied from <1 to 25 mg/m(3). When used in the cages of rats or mice for 4 days, the dust concentration of the beddings decreased, increased or stayed the same, depending on the type of bedding and animal species. A decrease in dustiness was, however, more common. The levels in the soiled beddings varied from <1 to 8.6 mg/m(3). In the case of the aspen chip bedding, the contents of bedding used in mouse, rat or rabbit cages were analysed for mesophilic bacteria and fungi, mycobacteria and endotoxins. All of these contaminants were variably found in the bedding samples, the maximal concentrations for bacteria were >6 500 000 colony-forming units (cfu)/g, for fungi 212 000 cfu/g, and for endotoxins 6500 ng/g (81 000 EU/g). The results showed that the bedding of laboratory animals may contain biologically effective compounds, and that these may be distributed into the ambient air depending on the characteristics of the bedding material. The dustiness of different bedding types is an important factor affecting the amount and quality of the occupational exposure of the personnel to airborne contaminants.  相似文献

AIMS: The dynamics of bioaerosol generation in specific occupational environments where mail is manually unpacked and sorted was investigated. METHODS AND RESULTS: Total number of airborne particles was determined in four different size classes (0.3-0.5, 0.5-1, 1-5 and >5 microm) by laser particle counting. Time dependent formation of bioaerosols was monitored by culturing methods and by specific staining followed by flow cytometry. Besides handling of regular mail, specially prepared letters ('spiked letters') were added to the mailbags to deliberately release powdered materials from letters and to simulate high impact loads. These letters contained various dry powdered biological and nonbiological materials such as milk powder, mushrooms, herbs and cat litter. Regarding the four size classes, particulate aerosol composition before mail handling was determined as 83.2 +/- 1.0, 15.2 +/- 0.7, 1.7 +/- 0.4 and 0.04 +/- 0.02%, respectively, whereas the composition changed during sorting to 66.8 +/- 7.9, 22.3 +/- 3.6, 10.4 +/- 4.0 and 0.57 +/- 0.27%, respectively. Mail processing resulted in an increase in culturable airborne bacteria and fungi. Maximum concentrations of bacteria reached 450 CFU m(-3), whereas 270 CFU of fungi were detected. CONCLUSIONS: Indoor particle concentrations steadily increased during mail handling mostly associated with particles of diameters >1 microm. However, it was not possible to distinguish spiked letters from nonspiked by simple particle counting and CFU determinations. SIGNIFICANCE AND IMPACT OF STUDY: The dynamics of bioaerosol generation have to be addressed when monitoring specific occupational environments (such as mail sorting facilities) regarding the occurrence of biological particles.  相似文献

Using three sampler devices (SAS, Andersen Six-Stages and All Glass Impinger), the environmental impact of bacterial and fungal aerosols generated by municipal wastewater treatment plants operating with different methods of sludge oxygenation were evaluated. The highest microbial concentrations were recovered above the tanks (2247 cfu m-3) and in downwind positions (1425 cfu m-3), where a linear correlation (P < 0.05) was found between the quantity of sewage treated and the entities of microbial aerosol dispersion. Moreover, an exponential increase (P < 0.05) in the bacteria recovered from the air occurred at increasing times of treatment. However, after long-term plant operation, high bacterial and fungal concentrations were found in almost all of the sites around the plant. Coliforms, enterococci, Escherichia coli and staphylococci were almost always recovered in downwind positions. Considerable fractions (20-40%) of sampled bacteria were able to penetrate the final stages of the Andersen apparatus and thus, are likely to be able to penetrate the lungs. The plant operating with a fine bubble diffused air system instead was found to generate rather low concentrations of bacteria and fungi; moreover, staphylococci and indicator micro-organisms were almost absent. Finally, salmonellae, Shigella, Pseudomonas aeruginosa and Aeromonas spp. were not detected in either of the plants. The results indicate a remarkable dispersion of airborne bacteria and fungi from tanks in which oxygen is supplied via a mechanical agitation of sludge, and suggest the need to convert them to diffused aeration systems which pose a lesser hazard for human health.  相似文献

用JWL-IIB新型固体撞击式空气微生物采样仪对广州市18个监测点的空气微生物含量及其月和季节变化势态进行了观察和研究。结果表明:广州市区室外空气微生物年平均含量为2,298cfu/m^3,室内空气微生物年平均含量为1,792cfu/m^3。各功能区的室外空气微生物平均含量(cfu/m^3)大小顺序为:垃圾压缩站(4,573)〉商业步行街(3,835)〉交通枢纽(1,580)〉居民住宅小区(1,413)〉工业厂区(1,197)〉中心公园(1,187);室内空气微生物含量(cfu/m^3)大小顺序为:交通枢纽(2,511)〉旅游三星酒店(1,699)〉地铁站(1,167)。一年中,市区各监测点室外空气微生物月平均含量变化范围在1,073~4,096cfu/m^3,其中3月份的空气微生物平均含量最高,可达4,096cfu/m^3,而10月份的空气微生物平均含量最低仅为1,073cfu/m^3。四季中,各监测点室外空气细菌和真菌平均含量(cfu/m^3)变化均表现为春季(3,716)〉夏季(2,300)〉冬季(1,816)〉秋季(1,553)。可为广州市区公共场所的卫生防疫和环境治理提供科学依据。  相似文献

Using sodium dodecyl sulfate-polyacrylamide gel electrophoresis of iron-deficient and replete cell envelopes, 59Fe-siderophore uptake studies, and Western immunoblots and cytofluorimetric analyses with monoclonal antibodies (MAbs), we surveyed a panel of gram-negative bacteria to identify outer membrane proteins that are structurally related to the Escherichia coli K-12 ferric enterobactin receptor, FepA. Antibodies within the panel identified FepA epitopes that are conserved among the majority of the bacteria tested, as well as epitopes present in only a few of the strains. In general, epitopes of FepA that are buried in the outer membrane bilayer were more conserved among gram-negative bacteria than epitopes that are exposed on the bacterial cell surface. The surface topology and tertiary structure of FepA are quite similar in E. coli and Shigella flexneri but differ in Salmonella typhimurium. Of the 18 different genera tested, 94% of the bacteria transported ferric enterobactin, including members of the previously unrecognized genera Citrobacter, Edwardsiella, Enterobacter, Haemophilus, Hafnia, Morganella, Neisseria, Proteus, Providencia, Serratia, and Yersinia. The ferric enterobactin receptor contains at least one buried epitope, recognized by MAb 2 (C. K. Murphy, V. I. Kalve, and P. E. Klebba, J. Bacteriol. 172:2736-2746, 1990), that is conserved within the structure of an iron-regulated cell envelope protein in all the bacteria that we have surveyed. With MAb 2, we identified and determined the Mr of cell envelope antigens that are immunologically related to E. coli FepA in all the gram-negative bacteria tested. Collectively, the library of anti-FepA MAbs showed unique patterns of reactivity with the different bacteria, allowing identification and discrimination of species within the following gram-negative genera: Aeromonas, Citrobacter, Edwardsiella, Enterobacter, Escherichia, Haemophilus, Hafnia, Klebsiella, Morganella, Neisseria, Proteus, Providencia, Pseudomonas, Salmonella, Serratia, Shigella, Vibrio, and Yersinia.  相似文献

Although application of composted municipal waste has its benefits, there are also concerns about a range of adverse effects. Bioaerosol emissions are one cause of concern because of related health problems. To assess the concentration of bioaerosols emitted during application of compost, fungi and bacteria were collected using an Andersen biosampler. Temperature and relative humidity were also recorded at each location. Total bacterial concentrations averaged 2,887 cfu/m3 for background samples and 6,727 cfu/m3 for compost-application samples, which constitutes a significant difference. The mean concentration of fungi during application of compost was three times greater than the background, but the difference was not significant. Concentrations of total bacteria in compost-application samples decreased significantly with increasing humidity. These results indicated that bioaerosol emissions as a result of application of compost could increase levels of exposure to bacteria and pose a potential health risk for exposed individuals.  相似文献

采用撞击法对热带雨林西双版纳大气试验站不同试验场地类型(试验场、试验棚、试验库)按干季和雨季共4次采集大气真菌,用统计学法计算出不同试验场地干季和雨季的真菌数量,采用真菌形态学方法,结合ITS1-5.8S-TIS2或26S rDNA D1/D2区序列分析法鉴定到种,并用SPSS和R软件初步探讨大气真菌与环境因素的关系。结果表明云南西双版纳试验站大气真菌浓度为2 200-4 300cfu/m 3;从西双版纳大气试验站分离得到776株真菌鉴定为34属52个种,优势种群为尖孢枝孢菌Cladosporiumoxysporum、变红镰孢菌Fusariumincarnatum、歧皱青霉菌PenicilliumsteckiiAspergillusminisclerotigenesLeptosphaerulinachartarumClonostachysepichloeParaconiothyriumestuarinum;大气环境中真菌多样性较丰富,真菌群落和浓度随季节性变化;3个试验场地不同季节之间真菌的群落结构差异大,影响大气真菌群落结构的重要环境因素是降雨量和气压。  相似文献

Three samples of silage taken from the surface of a silo and from depths of 20 and 45 cm in the silo were studied for identification of the potential agents causing symptoms of organic dust toxic syndrome. The samples were examined by dilution plating before and after aerosolization in an acoustical dust generator. Aerosol samples were collected by liquid impinger and filter cassettes. The samples were examined for total aerobic bacteria, anaerobic bacteria, gram-negative bacteria, lactobacilli, listeriae, thermophilic actinomycetes, fungi, and endotoxin. Very high levels of total aerobic bacteria and fungi were found in the surface sample (up to 10(9) CFU/g in the bulk sample and up to 10(9) CFU/m3 after aerosolization), whereas the corresponding values from the deepest site were 100 to 50,000 times lower. Aspergillus fumigatus predominated among the fungi, whereas Bacillus and gram-negative organisms (Pseudomonas, Alcaligenes, Citrobacter, and Klebsiella species) prevailed among bacteria. Thermophilic actinomycetes occurred in numbers up to 10(7) CFU/g in the bulk samples, whereas anaerobic bacteria, lactobacilli, and listeriae were only few or absent. The concentration of endotoxin was high in the surface sample (up to 211.4 Endotoxin Units/mg) and about 200-fold lower in the sample from the deepest site. The results show that contact with dust from the surface of silage carries the risk of exposure to high concentrations of microorganisms, of which A. fumigatus and endotoxin-producing bacteria are the most probable disease agents.  相似文献
