目的:由于长期服用左旋多巴治疗帕金森病,其药物浓度波动刺激易引起异动症,本实验旨在制备突释小,药物释放浓度稳定的左旋多巴甲酯微球制剂。方法:将左旋多巴甲酯用复乳法包裹于PLGA微球内,采用C18反相色谱研究药物包封率和体外释放行为。结果:通过调节药物浓度和不同高分子组合筛选出突释小,包封率高且缓慢释放的处方。结论:左旋多巴甲酯包裹于PLGA能实现理想的缓释效果,降低药物浓度波动,为后期药效学实验提供基础。  相似文献

目的:降钙素(一个由32个氨基酸组成的多肽)是治疗骨质疏松的首选药之一。降钙索的劣势是其半衰期过短,需要一天一次注射给药,本实验旨在制备突释小,药物释放浓度稳定的降钙素微球制剂。方法:制备降钙素羧酸葡聚糖颗粒和降钙素硫酸葡聚糖颗粒组合物,分别将其包裹于PLGA微球内,制备成降钙素组合微球,采用C18反相色谱柱研究药物的包封率和体外释放行为。结果:所制得的降钙素葡聚糖颗粒缓释微球体外释放一个月,释放曲线比较完美,接近零级释放。结论:本研究制得的降钙素葡聚糖颗粒缓释组合微球能实现理想的体外缓释效果,为后期药动学实验提供基础。  相似文献

目的:研究含蛋白的不对称膜高分子囊泡包封进PLGA微球后对其体外释放动力学的改善作用.方法:将包封有BSA蛋白的不对称膜高分子囊泡采用S/O/W法包裹进PLGA微球中,制备复合微球,对微球表征后,以包封葡聚糖颗粒的微球做对照品,于37℃测定微球的体外释放,比较两者的释放曲线,考察不对称膜高分子囊泡时微球中蛋白释放的改善作用.结果:①经扫描电镜(SEM)观察,包裹高分子囊泡的复合微球形态圆整,表面光滑,平均粒径为75.20μm,粒径较为均匀,复合微球制备成功.②比较复合微球和对照微球的释放曲线,发现对照微球有较小的突释,而复合微球的几乎没有突释效应.结论:不对称膜高分子囊泡包封进PLGA微球后可以很好的改善蛋白的释放行为,获得更为理想的释放曲线.  相似文献

目的:考察左旋多巴甲酯在PLGA微球中的稳定性并探讨其稳定方法。方法:利用HPLC的方法考察了左旋多巴甲酯在不同pH值和光照的环境中和微球里的稳定性。结果:左旋多巴甲酯在pH3中稳定,在微球中也可以稳定一周。结论:包封左旋多巴甲酯在PLGA微球中,是一种有效地保护了左旋多巴甲酯在微球中的活性,可以实现长效缓释,是一种可行的方案。  相似文献

目的:探索氢氧化镁对BSA微球体外释放的影响,优化BSA微球的制备工艺。方法:通过水包油包固复乳法制备BSA-PLGA微球。先将BSA与葡聚糖制备成玻璃体颗粒,再将玻璃体颗粒与氢氧化镁包裹进PLGA中,制备成缓释微球。在扫描电镜下观察其形态。然后用Micro BCA法测定其包封率和载药量,并考察其体外释放行为。结果:所制得的微球粒径约60μm,呈较好的球形。添加氢氧化镁后,BSA微球的包封率和载药量都有显著提高。不同含量的氢氧化镁对BSA微球的包封率和载药量影响也不同。在体外释放过程中,载有氢氧化镁的微球14天累积释放量为(85.10±2.67)%,而对照组不到80%。结论:通过调整氢氧化镁的量,可以制得形态完整,大小均匀,突释较小的BSA微球。  相似文献

目的:研究PLGA微球复合明胶支架对蛋白药物的释放影响。方法:将模型蛋白BSA通过复乳法制备成缓释PLGA微球,然后将微球埋置于明胶支架中,形成担载蛋白的PLGA微球复合明胶组织工程支架。考察复合支架体外蛋白释放行为,并用MicroBCA法定量测定释放的BSA量,采用β-半乳糖苷酶催化ONPG的方法检测制备前后蛋白的活性,并与不含PLGA微球直接担载蛋白的支架做对照。结果:PLGA微球复合支架蛋白的包封率能达到73.2%,其中第一天释放20%,对蛋白活性的保持达到70%以上。结论:微球复合明胶支架可以改善一般组织工程支架蛋白药物的突释,提高蛋白药物在制剂,贮存,释放过程中的稳定性。  相似文献

目的:研究包裹在PLGA微球中的多糖纳米颗粒在保护蛋白稳定性和改善药物体外释放行为方面的作用。方法:将模型药物BSA用低温诱导相分离方法担载于多糖纳米颗粒之中,后将其用水包油包固复乳法包裹于PLGA微球内。应用体积排阻色谱(SEC-HPLC)和红外色谱(FTIR)表征蛋白的稳定性,而且也研究了样品的体外释放行为。结果:这种方法能够很好的保护蛋白的稳定性,保持蛋白结构在制备过程中不会改变,而且改善了体外释放行为,减少了突释。结论:多糖纳米颗粒结合PLGA微球能够提供一种有效的解决蛋白控释的途径。  相似文献

聚乳酸乙醇酸共聚物(PLGA)是一种可生物降解的高分子聚合物,具有良好的生物相容性,其降解产物为乳酸和乙醇酸,是机体正常代谢的中间产物,最终可分解为二氧化碳和水,并分别经肺和肾脏排出体外,对人体不产生危害,所以PLGA在微球制剂的制备中常作为首选载体。近年来PLGA微球制剂在医药领域有着飞跃发展,尤其是在抗肿瘤、免疫疫苗、蛋白给药、基因治疗、诊断试剂和细胞支架等方面显现出很大优势。而且已有许多PLGA微球获得美国FDA批准上市,临床应用也有令人满意的效果,未见报道有严重的不良反应。但现阶段国内生产的PLGA缓释微球的质量还有很多不足之处如微球粒径偏大、包封率和载药量偏低、突释过大等,有待进一步提高和改进。本文将综述在制备包裹水溶性药物的PLGA微球过程中相关因素如药物本身理化性质、制备方法、PLGA结构特点、有机溶剂等对微球粒径、包封率的影响,以期为提高以PLGA为药物载体的药效奠定良好的理论基础。  相似文献

In the present study, we aimed to assess the potential anti-amblyopic effects of l-dopa methyl ester (LDME) on visual cortex area 17 in an amblyopic feline model induced by monocular vision deprivation. After LDME administration, pathophysiologic and ultrastructural observations were utilized to examine the morphological changes of nerve cells in visual cortex area 17. Dopamine (DA) and its metabolite contents in visual cortex area 17 were investigated through HPLC analysis. Apoptotic cells in visual cortex area 17 were evaluated by TUNEL assay. Additionally, the c-fos expression both at gene and protein levels was assessed using RT-PCR and immunohistochemistry analyses, respectively. The contents of DA and its metabolites were elevated in visual cortex area 17. Neuronal rejuvenation which occurred in visual cortex area 17 was observed through anatomical and physiological assessments. Similarly, TUNEL results showed that neuronal apoptosis was inhibited in the visual cortex of amblyopic cats by both l-dopa and LDME therapies. Meanwhile, the c-fos expression was notably up-regulated at both the mRNA and protein levels by the treatments. These findings suggested that LDME treatment could effectively increase DA and its metabolite contents, and restrain the apoptotic process, as well as elevate the c-fos expression in nerve cells of visual cortex area 17. Taken together, LDME might ameliorate the functional cytoarchitecture in visual cortex area 17 through mechanisms that elevate DA content and increase endogenous c-fos expression, as well as inhibit neuronal lesion in visual cortex tissue.  相似文献

目的:本实验旨在开发一种胶原酶缓释微球制剂,用以治疗手掌腱膜挛缩症,以减小现有水针剂的不足。方法:利用水相-水相乳化法和低温冷冻相分离法两种方法制备载药颗粒,分别将其包裹于PLGA微球内,制备成胶原酶微球,并用扫描电镜考察其表面形态,对其粒径进行统计学分析,测定体外释放行为并比较。结果:两种方法制备的微球表面光滑圆整,都可以达到缓释的效果,一个星期内释药完全。水相-水相乳化法制备的微球比低温冷冻相分离制备的微球粒径大,且具有统计学差异(P0.05)。水相-水相乳化法制备的微球粒径较均一,其体外释放更加平缓,突释较小。结论:本研究制得的胶原酶微球能实现理想的体外缓释效果,解决了现有技术中胶原酶粉针剂型快速释放并分散的问题。  相似文献

目的:在体外研究京尼平-壳聚糖微球可控性释放具有生物活性的神经生长因子的可行性。方法:采用"乳化-化学交联"技术制备包埋神经生长因子的京尼平-壳聚糖微球,京尼平为化学交联剂;应用扫描电镜、粒径分布、体外缓释动力学及细胞生物活性分别对微球的性能进行研究。结果:京尼平-壳聚糖微球表面光滑,平均粒径在5.1~50.5μm之间;京尼平的浓度可影响微球在体外释放神经生长因子的速度,经高浓度京尼平交联的微球能减缓并持续释放神经生长因子;此外,从京尼平-壳聚糖微球释放的神经生长因子可维持PC12细胞的生物活性,提高NGF生物利用率。结论:京尼平-壳聚糖微球能有效缓释具有生物活性的NGF超过14天,为神经退行性疾病的治疗提供一种治疗策略。  相似文献

目的:开发一种白细胞介素-12(IL-12)长效缓释微球剂型。方法:采用水包油-油包固(S/O/W)法制备了白介素-12因子多糖微粒的聚乳酸-聚羟基乙酸共聚物(PLGA-PLA)微球,研究了微球的表面形态和粒径大小,并且运用ELISA方法考察了微球的体外释放效果和免疫活性。结果:本方法制备的白介素-12因子微球光滑圆整,体外缓释达45天,累积释放率近90%。结论:本方法制备的白介素-12因子微球,不仅具有有效地保护IL-12蛋白活性,同时实现长效缓释的目标,是一种可行的IL-12缓释方案。  相似文献

An objective of the present investigation was to prepare and evaluate Eudragit-coated pectin microspheres for colon targeting of 5-fluorouracil (FU). Pectin microspheres were prepared by emulsion dehydration method using different ratios of FU and pectin (1:3 to 1:6), stirring speeds (500–2000 rpm) and emulsifier concentrations (0.75%–1.5% wt/vol). The yield of preparation and the encapsulation efficiencies were high for all pectin microspheres. Microspheres prepared by using drug:polymer ratio 1:4, stirring speed 1000 rpm, and 1.25% wt/vol concentration of emulsifying agent were selected as an optimized formulation. Eudragit-coating of pectin microspheres was performed by oil-in-oil solvent evaporation method using coat: core ratio (5:1). Pectin microspheres and Eudragit-coated pectin microspheres were evaluated for surface morphology, particle size and size distribution, swellability, percentage drug entrapment, and in vitro drug release in simulated gastrointestinal fluids (SGF). The in vitro drug release study of optimized formulation was also performed in simulated colonic fluid in the presence of 2% rat cecal content. Organ distribution study in albino rats was performed to establish the targeting potential of optimized formulation in the colon. The release profile of FU from Eudragit-coated pectin microspheres was pH dependent. In acidic medium, the release rate was much slower; however, the drug was released quickly at pH 7.4. It is concluded from the present investigation that Eudragit-coated pectin microspheres are promising controlled release carriers for colon-targeted delivery of FU. O-carboxymethylchitosan (OCMC) microspheres containing an antibiotic drug pazufloxacin mesilate (PM) have been successfully prepared by emulsion cross-linking using glutaraldehyde (GA). Various manufacturing parameters, including amount of cross-linking agent and OCMC:PM ratios were altered to optimize process variables during the microspheres production. The structure and morphology were characterized by Fourier transform infrared (FT-IR), wide-angle X-ray diffraction (WXRD) and scanning electron microscopy (SEM). The swelling and releasing behaviors of the microspheres at pH 1.2 and 7.4 media were investigated. The results revealed that the microspheres had a spherical, rough morphology and with a narrow size distribution. The degree of swelling of microspheres at pH 7.4 media was higher than that at pH 1.2 media. The microspheres proved to be successful in prolonging drug release. The release of PM was found to depend upon the extent of matrix cross-linking and drug loading. The release profiles of PM from OCMC microspheres were found to be biphasic with a burst release followed by a gradual release phase, and followed the Higuchi matrix model.  相似文献

The purpose of this research was to develop the taste-masked microspheres of intensely bitter drug ondansetron hydrochloride (OSH) by spray-drying technique. The bitter taste threshold value of OSH was determined. Three different polymers viz. Chitosan, Methocel E15 LV, and Eudragit E100 were used for microsphere formation, and the effect of different polymers and drug–polymer ratios on the taste masking and release properties of microspheres was investigated. The microspheres were characterized by Fourier transform infrared spectroscopy, scanning electron microscopy, Drug loading, in vitro bitter taste evaluation, and drug-release properties. The taste masking was absent in methocel microspheres at all the drug–polymer ratios. The Eudragit microspheres depicted taste masking at 1:2 drug–polymer ratio whereas with Chitosan microspheres the taste masking was achieved at 1:1 drug–polymer ratio. The drug release was about 96.85% for eudragit microspheres and 40.07% for Chitosan microspheres in 15 min.  相似文献
