对番茄内生细菌数量动态及其对青枯病的生物防治研究结果表明:番茄内生细菌可来源于种子内部。番茄不同生育期,内生细菌数量最多在成株期,其中抗病品种根、茎分别为24.3×104CFU/g鲜重和22.9×104CFU/g鲜重,感病品种根、茎分别为9.8×104CFU/g鲜重和13.4×104CFU/g鲜重。抗病品种中具有拮抗青枯菌的内生细菌菌株为17个,感病品种中7个。部分内生细菌具促进番茄种子萌发和防治番茄青枯病的作用,其中5R和3R内生菌株的防病效果分别达91.7%和81.3%。  相似文献

Ralstonia solanacearum is an economically important, bacterial plant pathogen which affects a wide range of crop plants. R. solanacearum survives in the soil for many years and weeds serve as symptomless carrier. One of the important aspects in controlling R. solanacearum is its early detection. In this study, detection threshold of R. solanacearum in the soil was standardised using polymerase chain reaction (PCR) method. The minimum threshold limit ranged between 6.8 × 10 and 3.6 × 102 CFU g?1 of soil. Using this standardised protocol R. solanacearum was detected from the rhizosphere soil of eggplants showing varying degrees of wilt. PCR method was quite sensitive to detect R. solanacearum from the xylem fluid of eggplant. Presence of R. solanacearum in the soil infected with capsicum wilt was also demonstrated successfully and the minimum detection limit was 4 × 102 CFU g?1 of soil. The bacterium was not detected from the eggplant seeds collected during 2006 and 2007 seasons. However, the bacterium was detected from the weed (Alternanthera sessilis) grown in the eggplant field indicating the possibility of weeds serving as symptomless carrier. Using our method, it is possible to detect R. solanacearum from soil, plant and weeds grown in the field at an early stage so that proper management strategies could be taken to prevent the infection and further spread of the pathogen.  相似文献

为了考查应用电解水消除细菌污染的可行性,对氧化电解水的杀菌效果及对食品加工表面材料的消毒效果进行了研究。结果表明,含0.1%NaCl的自来水经7min的电解后所获得的氧化电解水,能在2min内将菌液浓度分别为4.20×106CFU/mL,2.18×106CFU/mL,1.44×106CFU/mL,2.10×106CFU/mL,1.94×106CFU/mL的埃希氏大肠杆菌(Escherichia coliO157:H7)、沙门氏菌(Salmonella enteritidis)、单核细胞增生李斯特菌(Listeria monocytogenes)、摩化摩根菌(Morganella morganii)、副溶血性弧菌(Vibrio parahaemolyticus)几乎全部杀死。另外,对食品加工表面接触材料中的地板砖、不锈钢板、瓷砖进行染菌消毒试验结果表明,含0.1%NaCl的电解水同样能将上述浓度的菌液感染到食品表面接触材料后在5min之内几乎全部将其杀死,是一种理想的食品表面材料消毒剂。  相似文献

Treatments with conidia of Penicillium oxalicum produced in a solid‐state fermentation system were applied at similar densities (6 × 106 spores/g seedbed substrate) to tomato seedbeds in water suspensions (T1: 5 days before sowing, or T2: 7 days before transplanting; 15 days after sowing), or in mixture with the production substrate (T3: 7 days before transplanting; 15 days after sowing). Treatments T2 and T3 significantly (P = 0.05) reduced fusarium wilt of tomato in both greenhouse (artificial inoculation) (33 and 28%, respectively) and field conditions (naturally infested soils) (51 and 72%, respectively), while treatment T1 was efficient only in greenhouse (52%). Verticillium wilt disease reduction was obtained with T3 in two field experiments (56 and 46%, respectively), while T1 and T2 reduced disease only in one field experiment (52% for both T1 and T2). Treatment with conidia of P. oxalicum plus fermentation substrate (T3) resulted in better establishment of a stable and effective population of P. oxalicum in seedbed soil and rhizosphere providing populations of approx. 107 CFU/g soil before transplanting. Results indicate that it will be necessary to apply P. oxalicum at a rate of approx. 106–107 CFU/g in seedbed substrate and rhizosphere before transplanting for effective control of fusarium and verticillium wilt of tomato, and that formulation of P. oxalicum has a substantial influence on its efficacy.  相似文献

The current study aimed at evaluating the possibility of native Bacillus pumilus species to control Fusarium wilt in tomato and examine its effect on plant growth. Biocontrol traits of B. pumilus strains, biofilm assay, root colonisation and in vivo studies under pot conditions were determined. Strain ToIrMA-KC806242 formed biofilm efficiently and could colonise and survive on tomato rhizosphere (3.1 × 104 CFU/g of root). The amount of auxin production was recorded 29.7 μg/ml at the 96th hour of incubation. Siderophore production was determined positive, while ToIrMA was not able to solubilise phosphate compounds or produce cyanide hydrogen. Statistical analysis of data revealed that the increase in root and shoot length was recorded 60 and 84%, respectively, over control. In addition, about 73% reduction in disease incidence was determined in vivo experiments. In conclusion, this study suggests B. pumilus ToIrMA strain as a possible biocontrol agent in the field experiments.  相似文献

Starch industry wastewater was efficiently employed for the production of Sinorhizobium meliloti and the concentrated culture was used for the development of a biofertilizer formulation. Tween‐80 (0.02 g/L) acted as the best emulsifier for a Sinorhizobium–canola oil emulsion. The stability of the emulsion and survival of the organism was enhanced by supplementation of xanthan gum at pH 8. The refrigerated condition was most favorable for stability and survival of the microorganism. The survival of microorganism at 4±1°C was 2.78×1010 and 2.01×1010 CFU (colony forming unit)/mL on storage for 1 and 2 months, respectively. The values were higher than the prescribed cell count (×103 CFU/mL) for field application. At 40°C, the survival of bacteria reduced from 3×1010 CFU/mL to 8.1×109 and 8.8×106 CFU/mL in 1 and 2 months, respectively. Emulsion‐coated seed was incubated at different temperatures and a cell count of 105 CFU/seed was observed after 2 months of storage at 4°C, which was equal to the highest level of the described requirement (103–105 CFU/seed). Emulsion supplemented with xanthan gum improved the shelf‐life under optimized conditions (Sinorhizobium concentrate – canola oil (1:1) emulsion with 0.02 g/L Tween‐80; storage at pH 8 and temperature 4±1°C) and this emulsion with the required cell count and prolonged viability was used for the pre‐inoculation of seed or for in situ soil application.  相似文献

A novel flow‐injection chemiluminescence method was developed for the selective determination of human immunoglobulin G (IgG) in the presence of thiomersal by changing the flow rates of peristaltic pump. The study was based on the independence and additivity of the CL signals of human IgG and thiomersal in the galangin–potassium permanganate–polyphosphoric acid system. In meantime, two equations relating to the concentrations of mixing solutions of human IgG and thiomersal vs the CL intensity were established and solved, on the basis of which the content of thiomersal included in samples was simultaneously determined too. The enhanced CL intensity was in proportion to concerntrations in the range 8.0 × 10?7 to 8.0 × 10?5 g/mL for human IgG and 1.0 × 10?7 to 2.0 × 10?6 g/mL for thiomersal with the detection limits of 5.0 × 10?7 g/mL for human IgG and 6.0 × 10?8 g/mL for thiomersal, respectively. The relative standard deviation for 1.0 × 10?5 g/mL human IgG was 0.8% and for 2.0 × 10?7 g/mL thiomersal it was 2.0% (n = 10). The proposed method was applied to determine three synthetic samples with recoveries of 91.5–109.5%. In addition, the possible chemiluminescence mechanisms are discussed as well. We assessed the seasonal abundance and distribution of Vibrio species as well as some selected environmental parameters in the treated effluents of two wastewater treatment plants (WWTP), one each located in a suburban and urban community of Eastern Cape Province, South Africa. Vibrio population density ranged from 2.1×105 to 4.36×104 CFU/ml in the suburban community and from 2.80×105 to 1.80×105 CFU/ml in the urban community. Vibrio species associated with 180 μ, 60 μ, and 20 μ plankton sizes were observed at densities of 0–136×103 CFU/ml, 0–8.40×102 CFU/ml, and 0–6.80×102 CFU/ml, respectively at the suburban community’s WWTP. In the urban community, observed densities of culturable Vibrio were 0–2.80×102 CFU/ml (180 μ), 0–6.60×102 CFU/ml (60 μm), and 0–1.80× 103 CFU/ml (20 μm). The abundance of free-living Vibrio species ranged from 0 to 1.0×102 and 1.0×103 CFU/ml in the suburban and urban communities’ WWTPs, respectively. Molecular confirmation of the presumptive Vibrio isolates revealed the presence of V. fluvialis (41.38%), V. vulnificus (34.48%), and V. parahaemolyticus (24.14%) in the suburban community effluents. In the urban community molecular confirmation revealed that the same species were present at slightly different percentages, V. fluvialis (40%), V. vulnificus (36%), and V. parahaemolyticus (24%). There was no significant correlation between Vibrio abundance and season, either as free-living or plankton-associated entities, but Vibrio species abundance was positively correlated with temperature (r=0.565; p<0.01), salinity, and dissolved oxygen (p<0.05). Turbidity and pH showed significant seasonal variation (p<0.05) across the seasons in both locations. This study underscores the potential of WWTPs to be sources of Vibrio pathogens in the watershed of suburban and urban communities in South Africa.  相似文献

Bacillus sp. strain TAT105 is a thermophilic, ammonium-tolerant bacterium that grows assimilating ammonium nitrogen and reduces ammonia emission during composting of swine feces. To develop a practical use of TAT105, a dried solid culture of TAT105 (5.3 × 109 CFU/g of dry matter) was prepared as an additive. It could be stored for one year without significant reduction of TAT105. Laboratory-scale composting of swine feces was conducted by mixing the additive. When the additive, mixed with an equal weight of water one day before use, was added to obtain a TAT105 concentration of above 107 CFU/g of dry matter in the initial material, the ammonia concentration emitted was lower and nitrogen loss was approximately 22% lower in the treatment with the additive than in the control treatment without the additive. The colony formation on an agar medium containing high ammonium could be used for enumeration of TAT105 in the composted materials.  相似文献

Employing known susceptible and resistant genotypes and pure bacterial inoculum (0.1 OD; 108 CFU/ml?1), five different inoculation methods were tried to assess the response of tomato genotypes to Ralstonia solanacearum. This included seed‐soaking inoculation, seed‐sowing followed by inoculum drenching, or at 2‐week stage through petiole‐excision inoculation, soaking of planting medium with inoculum either directly or after imparting seedling root‐injury. Seed‐based inoculations or mere inoculum drenching at 2 weeks did not induce much disease in seedlings. Petiole inoculation induced 90–100% mortality in susceptible checks but also 50–60% mortality in normally resistant genotypes within 7–10 days. Root‐injury inoculation at 2‐week seedling stage appeared the best for early and clearer distinction between resistant and susceptible lines. The observations suggest a role played by the root system in governing genotypic resistance to the pathogen. Direct shoot inoculation is to be adopted only for selecting highly resistant lines or to thin down segregating populations during resistance breeding.  相似文献

Aims: To isolate the rhizosphere competence‐defective transposon Tn5 mutant of Pantoea agglomerans NBRISRM (SRM) and to identify the gene causing defect in its root colonization ability. Methods and Results: From over 5000 clones containing Tn5, one mutant P. agglomerans NBRISRMT (SRMT) showing 6 log units less colonization when compared with SRM, after 30 days in sand‐nonsterilized soil assay system was selected for further work to determine the effects of the mutation on rhizosphere competence. Southern hybridization analysis of restricted genomic DNA of SRMT demonstrated that the mutant had a single Tn5 insert. SRM increased in titre to about 2 × 108 CFU g?1 root, compared with the indigenous bacterial population of heterotrophs of about 5 × 107 CFU g?1 root. In contrast, 30 days later, the titre value of SRMT was almost undetectable at 1 × 102 CFU g?1 root, demonstrating its inability to survive and colonize the rhizosphere. Sequencing of the flanking region of the Tn5 mutant revealed that Tn5 disrupted the purB gene. Conclusions: A defect in the colonization phenotype of the SRMT was attributed to the disruption in adenylosuccinate lyase (EC 4.3.2.2) which is encoded by the pur B gene and is required for rhizosphere colonization in P. agglomerans. Significantly less exopolysaccharide and biofilm was formed by SRMT when compared to SRM, because of the disruption of the purB gene. Significance and Impact of the Study: This work provides the first evidence for a functional role of purB gene in rhizosphere competence and root colonization by any rhizobacteria.  相似文献

从深圳大鹏湾南澳赤潮爆发海域的表层海水中分离得到1株对海洋原甲藻(Prorocentrum micans)具有溶藻活性的海洋细菌,菌株编号为N10。利用液相感染法研究了该溶藻细菌的溶藻效果和溶藻作用方式。结果表明,菌株N10能使藻细胞失去运动活性,并膨胀变形,细胞膜内物质聚集于一端,藻细胞最终破裂死亡。菌悬液接种到藻液中的量越大,初始细菌密度越高,其溶藻效果越强。菌悬液以1∶10的体积比接种到藻液中时,藻细胞在24 h的死亡率为83%,至72 h全部溶解死亡;体积比为1∶20的藻细胞在24 h的死亡率为71%,之后藻细胞密度略有波动,120 h时死亡率达77%;而体积比为1∶100的藻细胞密度在前24 h有所下降,死亡率达39%,之后藻细胞密度又开始明显上升;对照组的藻细胞密度均呈明显上升趋势。菌悬液过滤液和高温加热处理后的菌悬液过滤液对海洋原甲藻均无溶藻活性,表明菌株N10的溶藻方式为直接溶藻。通过16S rRNA序列分析并与GenBank数据进行同源性检索,并结合细菌形态及生理生化特征,菌株N10隶属于黄杆菌科(Flavobacteriaceae)中的Muricauda sp.。  相似文献

As part of a program to develop starter cultures aiding in the spoilage control and sanitation of African fermented foods, a cereal-based food (‘ogi’ and its solid form ‘agidi’ or ‘eko’) was prepared using a bacteriocin-producingLactobacillus strain as the starter culture. The survival of an enterotoxigenicEscherichia coli strain was investigated in the naturally fermented food and in food fermented with the starter bacteriocin-producingLactobacillus strain. An inhibition ofE. coli was observed within 2 h of incubation in ‘ogi’ fermented with the bacteriocin producing strain. After 6 h, the viable count ofE. coli in locally fermented ‘ogi’ was log 6.41 (2.54×106CFU/mL), whereas in ‘ogi’ fermented with the bacteriocin producer it was reduced to log 1.70 (0.5×102 CFU/mL). Comparison of the shelf life of ‘agidi’ prepared from the naturally fermented food with that fermented with the bacteriocin-producing starter culture showed that the latter had a better shelf life (kept for 11 d before spoilage occurred as compared with 7 d for the natural one). The results are discussed in terms of the potential of bacteriocin-producing cultures in the control and retardation of spoilage and food-forne infections in some African fermented foods.  相似文献

A new antagonist/pathogen combination is reported, with Bacillus pumilus showing strong antagonistic activity against Penicillium digitatum. B. pumilus (1.6 × 1010 to 1.6 × 1012*cfu ml-1) gave significant control of P. digitatum infection in Valencia orange, which was as effective as imazalil (500 μg ml-1) and was significantly better than benomyl treatment (500 μg ml-1). When lower concentrations of B. pumilus (1.9 × 107 to 1.9 × 109 cfu ml-1) were tested on Washington Navel orange and Lisbon lemon fruit, the antagonist caused significant control of P. digitatum infection at two inoculum levels (6.5 × 104 and 6.5 × 105 spores ml-5). Concentrations of both the pathogen and the antagonist affected the biocontrol effect.  相似文献

A novel chemiluminescence method for the determination of 6‐mercaptopurine was established based on 6‐mercaptopurine inhibition of the chemiluminescence emission of potassium permanganate–thioacetamide–sodium hexametaphosphate system. The peak height was proportional to log 6‐mercaptopurine concentration in the range 7.0 × 10?10 to 1.0 × 10?7 g/mL and the detection limit was 1.9 × 10?11 g/mL (S/N = 3). The relative standard deviation was 1.5% for the determination of 8.0 × 10?8 g/mL 6‐mercaptopurine (n = 11). Quantitative and qualitative studies of the bacterial flora of farmed freshwater prawn (Macrobrachium rosenbergii) larvae in Saudi Arabia were performed, and isolates identified where possible. Physico‐chemical characteristics, bacterial counts, and the nature of the bacterial flora of larvae rearing tank water, sediment, tank wall surfaces, larval surface, supplied water, and feed were investigated. Bacterial counts ranged from 2.1 ± 1.3 × 105 to 2.2 ± 0.8 × 107 colony forming units (CFU) ml?1 in tank water; 4.4 ± 0.9 × 107 to 8.3 ± 1.7 ×109 CFU g?1 in tank sediment; 8.6 ± 1.0 × 102 to 9.8 ±0.7 × 104 CFU cm?2 on the tank wall surface; 1.3 ± 1.1 × 104 to 7.7 ± 1.6 × 106 CFU per larva surface, 7.9 ± 1.2 × 105 to 5.0 ± 1.5 × 107 CFU g?1 in washed larval tissue slurries, 9.1 ± 0.7 × 103 CFU ml?1 in supplied water, and 2.4 ± 1.9 ×1010 CFU g?1 in mixed feed. Fourteen bacterial genera were identified, including Chryseomonas sp., Vibrio spp., Cellulomonas sp., Aeromonas hydrophila, and Pasteurella sp. The tank water and sediment had similar bacteria to those on the prawn larvae. Chryseomonas sp., Cellulomonas sp. and Vibrio sp. were the most dominant species (prevalence >10%) in tank water; Chryseomonas sp., Pseudomonas alcaligenes and Shewanella putrefaciens in the sediment; Ps. alcaligenes and Cellulomonas sp. on the tank wall surface; Chryseomonas sp., and Cellulomonas sp. on the larval surface; and Chryseomonas sp., Vibrio vulnificus, Sh. putrefaciens and V. alginolyticus in the washed larval tissue slurries (prevalence 10%). Pseudomonas alcaligenes, Moraxella sp., Serratia liquefaciens, Gordona sp. and Burkholderia glumae were absent in larvae but identified in the culture water, tank sediment, and tank wall surface. Pseudomonas sp., Chryseomonas sp., Pasteurella sp. and V. alginolyticus were the prevalent bacteria (>12%) in supplied water. The feed contained V. alginolyticus, A. hydrophila and Cellulomonas sp. as the dominant bacteria (>13%). In the culture water and larvae samples, 83% of the feed and supplied water bacteria were identified.  相似文献

Phytophthora foot rot of black pepper caused by Phytophthora capsici is a major disease of black pepper throughout production areas in Vietnam. The disease causes collar, foot and tap root rots and eventual death of the infected vine. Potassium phosphonate was evaluated for the control of this disease in greenhouse and field trials. In greenhouse trials three-month-old vines treated with phosphonate by soil drenching (10–20 g a.i./l) and then inoculated with P. capsici mycelium (2% v/v soil) had significantly less foot rot compared to vines grown in non-treated soil. In field trials mature vines were treated with phosphonate at 50–100 g a.i/pole soil drenching or 10 g a.i./l by root infusion. After 10 days root, stem and leaf specimens were removed for bioassay by inoculation with 5 ml of P. capsici zoospores suspension (106–108 spores/ml). Soil drenching with phosphonate inhibited the colonisation of pathogen on excised leaf, stem and root tissues, significantly more than phosphonate root infusion. Our study provides further evidence supporting the efficacy of potassium phosphonate in the management of black pepper foot rot caused by P. capsici. The excised leaf and stem bioassay used in this study is a rapid and useful technique for testing the efficacy of systemic fungicides in controlling this disease.  相似文献

Bioaerosol concentrations inside one naturally ventilated and one mechanically ventilated swine finishing barn were assessed by sampling air using membrane filtration and impaction (six-stage Andersen sampler), and assayed by culture method. The barns, located on the same commercial farm in northeast Kansas, did not show any significant difference (p > 0.05) in concentrations of total and respirable airborne microorganisms. The overall mean total concentrations inside the two barns were 6.6 × 104 colony forming units (CFU)/m3 (SD = 3.8 × 104 CFU/m3) as measured by filtration and 8.6 × 104 CFU/m3 (SD = 5.1 × 104 CFU/m3) by impaction. The overall mean respirable concentrations were 9.0 × 103 CFU/m3 (SD = 4.1 × 103 CFU/m3) measured by filtration and 2.8 × 104 CFU/m3 (SD = 2.2 × 104 CFU/m3) by impaction. Total and respirable CFU concentrations measured by impaction were significantly (p < 0.05) higher than that by filtration. The persistent strains of microorganisms were various species of the following genera: Staphylococcus, Pseudomonas, Bacillus, Listeria, Enterococcus, Nocardia, Lactobacillus, and Penicillium. It appears that filtration sampling can be used for a qualitative survey of bioaerosols in swine barns while the Andersen sampler is suitable for both quantitative and qualitative assessments. A simple and sensitive flow injection chemiluminescence method has been developed for the determination of ferulic acid (FA) based on the significant enhancement effect of FA on the CL signal of the N‐bromobutanimide (NBS)–eosin–CrCl3 system in alkaline solution. Under optimum conditions, the enhanced CL intensity is linearly related to the concentration of FA in its pharmaceutical preparations and human plasma samples. The corresponding linear regression equations were established over the 4.0 × 10–10–1.0 × 10–7 g/mL for FA tablets and 2.0 × 10–10–1.0 × 10–7 g/mL for plasma samples. The limit of detection for FA tablets and limit of quantification for plasma samples were 2.8 × 10–10 g/mL (3 σ) and 3.04 × 10–10 g/mL (10 σ), respectively. A complete analysis could be performed within 40 s, including washing and sampling, giving a throughput of ≈90/h. The proposed method was successfully applied to the determination of FA in pharmaceutical preparations and human plasma samples with satisfactory results. The recoveries of pharmaceutical preparations and human plasma samples at three different concentrations were 97.8–102.6% and 96.7–104.0%, respectively. In the present study, the endophytic bacterium Bacillus subtilis strain Em7 (GU258545.1) was evaluated as a biological control agent for Sclerotinia sclerotiorum on oilseed rape. In petri dish, strain Em7 not only strongly inhibited pathogen mycelium growth but also germination of sclerotia at concentrations between 109 and 1011 colony forming unit (CFU)·ml?1. Scanning electron microscopy and transmission electron microscopy studies revealed that in the presence of strain Em7, hyphae of S. sclerotiorum showed leakage and disintegration of hyphal cytoplasm. Furthermore, the strain Em7 showed a broad antifungal spectrum on mycelium growth of numerous important plant pathogenic fungi. Light microscopic observations revealed that strain Em7 caused morphological alterations including increased branching, swelling and collapse of cytoplasm. In the greenhouse, spray treatments of cell suspensions of strain Em7 (1×109 CFU·ml?1) reduced leaf and stem rot incidence and severity in the seedling and blossom stage. The control efficacy was higher when strain Em7 cell suspension was applied one day prior to inoculation of the pathogen than after inoculation. Three-year field trials showed that two applications of strain Em7 cell suspension at blossom stage significantly reduced disease incidence and severity by 50–70%. There was no significant difference in control efficacy among treatments with strain Em7 cell suspension and the fungicides containing carbendazim or tebuconazole (P = 0.05). Thus, our results strongly suggest that B. subtilis strain Em7 is a promising biological control agent for control of oilseed rape Sclerotinia stem rot.  相似文献
