Genetically transformed alfalfa (Medicago sativa L., cv. Zajearska 83) plantlets were obtained by inoculating somatic embryos with Agrobacterium tumefaciens strains A281/pGA472 and LBA4404/pBI121. Single somatic embryos, 5–7 mm long, were released from a repetitively embryogenic culture, wounded, and cocultivated with the bacteria. The agar-solidified culture medium contained mineral salts, vitamins, 40 g l–1 sucrose, 1 g l–1 yeast extract and 0.05 mg l–1 BA. Five clones, transformed with A281/pGA472, and 4 clones transformed with LBA4404/pBI121, were selected for proliferation by repetitive somatic embryogenesis, on media containing 100 mg l–1 of kanamycin. The transformation of kanamycin-resistant clones was confirmed by assaying the activity of neomycin phosphotransferase II and/or -glucuronidase enzymes, and by the Southern blot analysis. It is suggested that the transformation/regeneration system based on somatic embryogenesis may be suitable for establishing transgenic alfalfa lines. IR‐783 is a kind of heptamethine cyanine dye that exhibits imaging, cancer targeting and anticancer properties. A previous study reported that its imaging and targeting properties were related to mitochondria. However, the molecular mechanism behind the anticancer activity of IR‐783 has not been well demonstrated. In this study, we showed that IR‐783 inhibits cell viability and induces mitochondrial apoptosis in human breast cancer cells. Exposure of MDA‐MB‐231 cells to IR‐783 resulted in the loss of mitochondrial membrane potential (MMP), adenosine triphosphate (ATP) depletion, mitochondrial permeability transition pore (mPTP) opening and cytochrome c (Cyto C) release. Furthermore, we found that IR‐783 induced dynamin‐related protein 1 (Drp1) translocation from the cytosol to the mitochondria, increased the expression of mitochondrial fission proteins mitochondrial fission factor (MFF) and fission‐1 (Fis1), and decreased the expression of mitochondrial fusion proteins mitofusin1 (Mfn1) and optic atrophy 1 (OPA1). Moreover, knockdown of Drp1 markedly blocked IR‐783‐mediated mitochondrial fission, loss of MMP, ATP depletion, mPTP opening and apoptosis. Our in vivo study confirmed that IR‐783 markedly inhibited tumour growth and induced apoptosis in an MDA‐MB‐231 xenograft model in association with the mitochondrial translocation of Drp1. Taken together, these findings suggest that IR‐783 induces apoptosis in human breast cancer cells by increasing Drp1‐mediated mitochondrial fission. Our study uncovered the molecular mechanism of the anti‐breast cancer effects of IR‐783 and provided novel perspectives for the application of IR‐783 in the treatment of breast cancer.  相似文献

Checkpoint kinase 1 (ChK1) is activated in response to DNA damage, acting to temporarily block cell cycle progression and allow for DNA repair. It is envisaged that inhibition of ChK1 will sensitize tumor cells to treatment with DNA-damaging therapies, and may enhance the therapeutic window. High throughput screening identified carboxylate-containing diarylpyrazines as a prominent hit series, but with limited biochemical potency and no cellular activity. Through a series of SAR investigations and X-ray crystallographic analysis the critical role of polar contacts with conserved waters in the kinase back pocket was established. Structure-based design, guided by in silico modeling, transformed the series to better satisfy these contacts and the novel 1,7-diazacarbazole class of inhibitors was discovered. Here we present the genesis of this novel series and the identification of GNE-783, a potent, selective and orally bioavailable inhibitor of ChK1.  相似文献

Protein phosphorylation can be regulated by changes in kinase activity, phosphatase activity, or both. GABAB receptor R2 subunit (GABABR2) is phosphorylated at S783 by 5′-AMP-activated-protein kinase (AMPK), and this phosphorylation modulates GABAB receptor desensitization. Since the GABAB receptor is an integral membrane protein, solubilizing GABABR2 is difficult. To circumvent this problem and to identify specific phosphatases that dephosphorylate S783, we employed an in vitro assay based on dephosphorylation of proteins on PVDF membranes by purified phosphatases. Our method allowed us to demonstrate that S783 in GABABR2 is directly dephosphorylated by PP2A (but not by PP1, PP2B nor PP2C) in a dose-dependent and okadaic acid-sensitive manner. We also show that the level of phosphorylation of the catalytic subunit of AMPK at T172 is reduced by PP1, PP2A and PP2C. Our data indicate that PP2A dephosphorylates GABABR2(S783) less efficiently than AMPK(T172), and that additional phosphatases might be involved in S783 dephosphorylation.  相似文献

A series of new disubstituted halogenated arylcyanoximes was synthesized using nitrosation reaction of the respective phenylacetonitriles by CH3–ONO at room temperature in isopropanol. Six synthesized colorless arylcyanoximes containing two F and/or Cl atoms at 2-, 4-, 5- and 6-positions were characterized by means of NMR, IR, UV–Vis spectroscopy and pKa studies. Crystal structures were determined for four cyanoximes and revealed the presence of only the syn-isomers for fluorinated compounds in a solid state, while the chlorinated arylcyanoxime exists as anti-isomer in the crystal. However, five out of the six protonated arylcyanoximes HL exist as a mixture of syn- and anti-isomers in solutions. Deprotonation of HL with NaOC2H5 in ether solutions leads to yellow NaL which were used as precursors for the synthesis of a series of monovalent Ag, Tl and bivalent Pd, Pt complexes. Seven palladium and platinum arylcyanoximates of [M(HL)2Cl2] composition were synthesized and characterized. Obtained colored compounds are non-electrolytes in solution. However, in EtOH and DMSO solutions Pt(II) cyanoximates undergo two consecutive solvolysis reactions. First order rate constants were measured at 294 K for complexes in both solvents. Binding modes of the cyanoxime ligands and the possible solid state structures of the obtained coordination compounds are suggested on a basis of their IR spectra and MM-2 calculations. Because of their structural resemblance to the cisplatin family of anticancer drugs, synthesized Pd/Pt arylcyanoximates were tested in vitro against human colon carcinoma WiDr cell line using cis-[Pt(NH3)2Cl2] as positive control. Results showed that two Pd(II) and Pt(II) cyanoximates containing oximino(2,4-dichlorophenyl)acetonitrile exhibit cytotoxicity at 0.25 mM concentrations.  相似文献

目的:观察研究近红外荧光染料IR-783在膀胱癌中的特异性成像。方法:通过染料与膀胱癌细胞及正常细胞共孵育,观察近红外荧光染料IR-783是否能够实现膀胱癌细胞的选择性成像。利用细胞器示踪剂观察近红外荧光染料在膀胱癌细胞内的共定位;使用IR-783检测循环血液中及尿液中的膀胱癌细胞。结果:近红外荧光染料IR-783可被膀胱癌细胞选择性摄取。IR-783可选择性聚集在膜性细胞器如线粒体和溶酶体内,这种选择性聚集作用使IR-783可以保持较长的染色效果。近红外染料可以检测到血液或尿液极少量的膀胱癌细胞。结论:近红外荧光染料IR-783能够被膀胱癌细胞特异性吸收,可用于血液和尿液中膀胱肿瘤细胞的特异性诊断,具有重要的临床应用前景。  相似文献

Activation of Checkpoint kinase 1 (Chk1) following DNA damage mediates cell cycle arrest to prevent cells with damaged DNA from entering mitosis. Here we provide a high-resolution analysis of cells as they undergo S- and G₂-checkpoint bypass in response to Chk1 inhibition with the selective Chk1 inhibitor GNE-783. Within 4–8 h of Chk1 inhibition following gemcitabine induced DNA damage, cells with both sub-4N and 4N DNA content prematurely enter mitosis. Coincident with premature transition into mitosis, levels of DNA damage dramatically increase and chromosomes condense and attempt to align along the metaphase plate. Despite an attempt to congress at the metaphase plate, chromosomes rapidly fragment and lose connection to the spindle microtubules. Gemcitabine mediated DNA damage promotes the formation of Rad51 foci; however, while Chk1 inhibition does not disrupt Rad51 foci that are formed in response to gemcitabine, these foci are lost as cells progress into mitosis. Premature entry into mitosis requires the Aurora, Cdk1/2 and Plk1 kinases and even though caspase-2 and -3 are activated upon mitotic exit, they are not required for cell death. Interestingly, p53, but not p21, deficiency enables checkpoint bypass and chemo-potentiation. Finally, we uncover a differential role for the Wee-1 checkpoint kinase in response to DNA damage, as Wee-1, but not Chk1, plays a more prominent role in the maintenance of S- and G₂-checkpoints in p53 proficient cells.  相似文献

Photothermal therapy (PTT) has attracted wide attention due to its noninvasiveness and its thermal ablation ability. As photothermal agents are crucial factor in PTT, those with the characteristics of biocompatibility, non-toxicity and high photothermal stability have attracted great interest. In this work, new indocyanine green (IR-820) was utilized as a photothermal agent and near-infrared (NIR) fluorescence imaging nanoprobe. To improve the biocompatibility, poly(styrene-co-maleic anhydride) (PSMA) was utilized to encapsulate the IR-820 molecules to form novel IR-820@PSMA nanoparticles (NPs). Then, the optical and thermal properties of IR-820@PSMA NPs were studied in detail. The IR-820@PSMA NPs showed excellent photothermal stability and biocompatibility. The cellular uptaking ability of the IR-820@PSMA NPs was further confirmed in HeLa cells by the NIR fluorescent confocal microscopic imaging technique. The IR-820@PSMA NPs assisted PTT of living HeLa cells was conducted under 793 nm laser excitation, and a high PTT efficiency of 73.3% was obtained.  相似文献
