The preparation of highly purified medium chain acyl-CoA synthetase (Acid: CoA ligase, AMP-forming (EC 6.2.1.2)) from the cell extracts of is described. The enzyme is inducibly formed in the cells of the microorganism, when it is grown with butyrate as a major carbon source. The purified enzyme is homogeneous on disc gel electrophoresis. Its molecular weight is approximately 142,000, and it is possibly composed of 4 identical subunits of approximately 37,000 molecular weight and has isoelectric point of 4.3. The enzyme catalyzes the stoichiometric conversion of butyrate and CoA to butyryl-CoA in the presence of ATP and Mg2+. It also activates fatty acids with carbon chain lengths of 3 to 5 well, but is inactive toward fatty acids with carbon chain lengths of more than 6. The enzyme is sulfhydryl dependent and inactivated by silver and mercury compounds. 相似文献

A new phosphorylating agent, S-(N-monomethoxytritylaminoethyl)-O-(O-chlorophenyl) phosphorothioate, was prepared and reacted with a 5'-hydroxyl group of an oligonucleotide using 1-mesitylene-sulfonyl-3-nitrotriazole (MSNT) as a condensing agent. After base labile protecting groups were removed, the partially deprotected oligonucleotide was separated on a reversed phase column and converted to the oligonucleotide with an aminoethyl or a phosphoryl group at the 5'-end by treatment with 80% acetic acid or iodine-water, respectively. The syntheses of ppT, pppT, A5'pp5'T and A5'ppp5'T were also performed by treatment of 5'-O-(N-monomethoxytritylaminoethylthiophosphoryl) thymidine with tri-n-octylammonium salt of phosphoric acid, pyrophosphoric acid, pA and ppA, respectively. 相似文献

Winged bean Kunitz chymotrypsin inhibitor (WCI) is encoded by a multigene family and accumulation of its mRNA is restricted in mid-maturation stage seeds and tuberous roots. In this paper, we analyzed the accumulation of mRNA derived from each WCI gene using a novel method: sequence-specific termination analysis. The velocity of the α-β transition was measured in two cases, i.e., when α-crystals stayed in the saturated aqueous solution, and when they were left to stand in an air-bath at various temperatures ranging from 20° to 100°C. The effects of Ca2+ in the external medium on intact mung beanroots under high NaCl stress were investigated. With increasingexternal concentrations of NaCl, mung bean roots showed suppressionof elongation and a decrease in the intracellular concentrationof K+. Addition of Ca2+ to the external medium alleviated theinhibition of root elongation under the high NaCl stress andmaintained a high intracellular concentration of K+ in the elongatingregion of the roots. This counter effect of Ca2+ against theNaCl stress on roots was correlated with the ratio of [Ca2+]/[Na+]2in the external medium. A value above 5.0 ? 104 mM1resulted in almost complete recovery of root elongation undervarious high concentrations of NaCl. Root elongation for 24h under NaCl stress was correlated with the extent to whichthe intracellular concentration of K+ was in excess of 10 mM.Maintenance of an adequate concentration of K+ in root cellsis essential for root elongation under salt stress.
