In the Americas, areas with a high risk of malaria transmission are mainly located in
the Amazon Forest, which extends across nine countries. One keystone step to
understanding the Plasmodium life cycle in Anopheles species from the Amazon Region
is to obtain experimentally infected mosquito vectors. Several attempts to colonise
Ano- pheles species have been conducted, but with only short-lived success or no
success at all. In this review, we review the literature on malaria transmission from
the perspective of its Amazon vectors. Currently, it is possible to develop
experimental Plasmodium vivax infection of the colonised and field-captured vectors
in laboratories located close to Amazonian endemic areas. We are also reviewing
studies related to the immune response to P. vivax infection of Anopheles aquasalis,
a coastal mosquito species. Finally, we discuss the importance of the modulation of
Plasmodium infection by the vector microbiota and also consider the anopheline
genomes. The establishment of experimental mosquito infections with Plasmodium
falciparum, Plasmodium yoelii and Plasmodium berghei parasites that could provide
interesting models for studying malaria in the Amazonian scenario is important.
Understanding the molecular mechanisms involved in the development of the parasites
in New World vectors is crucial in order to better determine the interaction process

Potato late blight, caused by the oomycete pathogen Phytophthora infestans, significantly hampers potato production. Recently, a new Resistance to Phytophthora infestans (Rpi) gene, Rpi-amr1, was cloned from a wild Solanum species, Solanum americanum. Identification of the corresponding recognized effector (Avirulence or Avr) genes from P. infestans is key to elucidating their naturally occurring sequence variation, which in turn informs the potential durability of the cognate late blight resistance. To identify the P. infestans effector recognized by Rpi-amr1, we screened available RXLR effector libraries and used long read and cDNA pathogen-enrichment sequencing (PenSeq) on four P. infestans isolates to explore the untested effectors. Using single-molecule real-time sequencing (SMRT) and cDNA PenSeq, we identified 47 highly expressed effectors from P. infestans, including PITG_07569, which triggers a highly specific cell death response when transiently coexpressed with Rpi-amr1 in Nicotiana benthamiana, suggesting that PITG_07569 is Avramr1. Here we demonstrate that long read and cDNA PenSeq enables the identification of full-length RXLR effector families and their expression profile. This study has revealed key insights into the evolution and polymorphism of a complex RXLR effector family that is associated with the recognition by Rpi-amr1. 相似文献

Carbon emission and sequestration by agricultural land use: a model study for Europe 总被引:13,自引:0,他引:13
A model was developed to calculate carbon fluxes from agricultural soils. The model includes the effects of crop (species, yield and rotation), climate (temperature, rainfall and evapotranspiration) and soil (carbon content and water retention capacity) on the carbon budget of agricultural land. The changes in quality of crop residues and organic material as a result of changes in CO2 concentration and changed management were not considered in this model. The model was parameterized for several arable crops and grassland. Data from agricultural, meteorological, soil, and land use databases were input to the model, and the model was used to evaluate the effects of different carbon dioxide mitigation measures on soil organic carbon in agricultural areas in Europe. Average carbon fluxes under the business as usual scenario in the 2008–2012 commitment period were estimated at 0.52 tC ha?1 y?1 in grassland and ?0.84 tC ha?1 y?1 in arable land. Conversion of arable land to grassland yielded a flux of 1.44 tC ha?1 y?1. Farm management related activities aiming at carbon sequestration ranged from 0.15 tC ha?1 y?1 for the incorporating of straw to 1.50 tC ha?1 y?1 for the application of farmyard manure. Reduced tillage yields a positive flux of 0.25 tC ha?1 y?1. The indirect effect associated with climate was an order of magnitude lower. A temperature rise of 1 °C resulted in a ?0.05 tC ha?1 y?1 change whereas the rising CO2 concentrations gave a 0.01 tC ha?1 y?1 change. Estimates are rendered on a 0.5 × 0.5° grid for the commitment period 2008–2012. The study reveals considerable regional differences in the effectiveness of carbon dioxide abatement measures, resulting from the interaction between crop, soil and climate. Besides, there are substantial differences between the spatial patterns of carbon fluxes that result from different measures. 相似文献

The molecular basis of Plasmodium vivax chloroquine (CQ) resistance
is still unknown. Elucidating the molecular background of parasites that are
sensitive or resistant to CQ will help to identify and monitor the spread of
resistance. By genotyping a panel of molecular markers, we demonstrate a similar
genetic variability between in vitro CQ-resistant and sensitive phenotypes of
P. vivax parasites. However, our studies identified two
loci (MS8 and MSP1-B10) that could be used to discriminate
between both CQ-susceptible phenotypes among P. vivax isolates in
vitro. These preliminary data suggest that microsatellites may be used to identify
and to monitor the spread of P. vivax-resistance around the

Potato defends against Phytophthora infestans infection by resistance (R)-gene-based qualitative resistance as well as a quantitative field resistance. R genes are renowned to be rapidly overcome by this oomycete, and potato cultivars with a decent and durable resistance to current P. infestans populations are hardly available. However, potato cultivar Sarpo Mira has retained resistance in the field over several years. We dissected the resistance of 'Sarpo Mira' in a segregating population by matching the responses to P. infestans RXLR effectors with race-specific resistance to differential strains. The resistance is based on the combination of four pyramided qualitative R genes and a quantitative R gene that was associated with field resistance. The qualitative R genes include R3a, R3b, R4, and the newly identified Rpi-Smira1. The qualitative resistances matched responses to avirulence (AVR)3a, AVR3b, AVR4, and AVRSmira1 RXLR effectors and were overcome by particular P. infestans strains. The quantitative resistance was determined to be conferred by a novel gene, Rpi-Smira2. It was only detected under field conditions and was associated with responses to the RXLR effector AvrSmira2. We foresee that effector-based resistance breeding will facilitate selecting and combining qualitative and quantitative resistances that may lead to a more durable resistance to late blight. 相似文献

Mapping resistance genes is usually accomplished by phenotyping a segregating population for the resistance trait and genotyping
it using a large number of markers. Most resistance genes are of the NBS-LRR type, of which an increasing number is sequenced.
These genes and their analogs (RGAs) are often organized in clusters. Clusters tend to be rather homogenous, viz. containing
genes that show high sequence similarity with each other. From many of these clusters the map position is known. In this study
we present and test a novel method to quickly identify to which cluster a new resistance gene belongs and to produce markers
that can be used for introgression breeding. We used NBS profiling to identify markers in bulked DNA samples prepared from
resistant and susceptible genotypes of small segregating populations. Markers co-segregating with resistance can be tested
on individual plants and directly used for breeding. To identify the resistance gene cluster a gene belongs to, the fragments
were sequenced and the sequences analyzed using bioinformatics tools. Putative map positions arising from this analysis were
validated using markers mapped in the segregating population. The versatility of the approach is demonstrated with a number
of populations derived from wild Solanum species segregating for P. infestans resistance. Newly identified P. infestans resistance genes originating from S. verrucosum, S. schenckii, and S. capsicibaccatum could be mapped to potato chromosomes 6, 4, and 11, respectively. 相似文献

High mortality among hepatocellular carcinoma (HCC) patients reflects both late diagnosis and low curability, due to pharmacoresistance. Taxol (TAX) is toxic for many human HCC-derived cell lines, yet its clinical efficacy on HCCs is poor. Combining TAX with other drugs appears a promising possibility to overcome such refractoriness. We analyzed whether combining tumor necrosis factor (TNF) with TAX would improve their toxicity. Human HCC-derived cell lines were treated with TAX or TNF, alone or combined. Apoptosis was assessed by morphology and flow-cytometry. Several pro- and anti-apoptotic molecules were evaluated by western blotting and/or enzymatic assay. After a 24 hour treatment, TNF was ineffective and TAX modestly cytotoxic, whereas HCC cells were conditionally sensitized to TNF by TAX. Indeed some relevant parameters were shifted to a prodeath setting: TNF-receptor 1 was increased, SOCS3, c-FLIP and pSTAT3 were markedly downregulated. These observations provide a significant clue to critically improve the drug susceptibility of HCC cells by combining 2 agents, TAX and TNF. The sequential application of TAX at a low dosage followed by TNF for only a short time triggered a strong apoptotic response. Of interest, prior TAX administration could also sensitize to TNF-induced apoptosis in the Yoshida AH-130 hepatoma transplanted in mice. Therefore, scrutinizing the possibility to develop similar combination drug regimens in suitable preclinical models seems highly advisable. 相似文献
