The constricted ‘waist’ of the metamorphosing larva of the polychaete Arenicola cristata is described, using light and electron microscopy. The constriction is shown to be the consequence of the discharge and collapse of a post-trochal ring of epithelial cells which remain as functional components of the post-metamorphic juvenile. Morphological differentiation of neuro-effector and interneuronal contacts is initiated at this time. Muscular and neural changes are discussed in terms of their role in effecting metamorphosis.  相似文献

Sucrose gradient centrifugation was used to separate the microsomalmembranes and purify the plasma membrane ATPase from Ricinuscotyledons. The pellet from a three-step (30, 34, 38%) sucrosegradient was enriched in plasma membrane as determined by acombination of marker assays. The partially purified plasma membrane ATPase was magnesium-dependentand had a pH optimum of 6.5. It showed high sensitivity to vanadate,erythrosin B, SW 26, DCCD and PCMBS but low sensitivity to azide,nitrate and NEM. Substitution of calcium for magnesium resultedin low activity, and in the presence of magnesium, calcium wasinhibitory. KCl stimulation was low (less than 50%) and of thepotassium salts tested all were stimulatory except which was inhibitory. Specificity for nucleotide triphosphateswas high, greatest activity occurring with ATP. Proton-pumping activity measured using quinacrine fluorescencequenching was inhibited by vanadate and erythrosin B but notby nitrate and oligomycin indicating that activity was mainlydue to the plasma membrane ATPase. Hordeum vulgare cv. California Mariout was established in sandculture at two different NaCl concentrations (0.5 mol m–3‘control’ and 100 mol m–3) in the presenceof 6.5 mol m–3 K +. Between 16 and 31 d after germination,before stem elongation started, xylem sap was collected by useof a pressure chamber. Collections were made at three differentsites on leaves 1 and 3: at the base of the sheath, at the baseof the blade, i.e. above the ligule, and at the tip of the blade.Phloem sap was collected from leaf 3 at similar sites throughaphid stylets. The concentrations of K +, Na+, Mg2+ and Ca2+were measured. Ion concentrations in xylem sap collected at the base of leaves1 and 3 were identical, indicating there was no preferentialdelivery of specific ions to older leaves. All ion concentrationsin the xylem decreased from the base of the leaf towards thetip; these gradients were remarkably steep for young leaves,indicating high rates of ion uptake from the xylem. The gradientsdecreased with leaf age, but did not disappear completely. In phloem sap, concentrations of K+ and total osmolality declinedslightly from the tip to the base of leaves of both controland salt-treated plants. By contrast, Na+ concentrations inphloem sap collected from salt-treated plants decreased drasticallyfrom 21 mol m–3 at the tip to 7.5 mol m–3 at thebase. Data of K/Na ratios in xylem and phloem sap were used to constructan empirical model of Na+ and K+ flows within xylem and phloemduring the life cycle of a leaf, indicating recirculation ofNa+ within the leaf. Flow cytometry was employed as a tool to analyze and characterize batches of oocysts from laboratory and field isolates of Eimeria spp. from chickens and to propagate sub-populations of batches of oocysts. Oocyst batches were cleaned of debris by a combination of salt flotation, washing and treatment with dilute sodium hypochlorite (1.5% aqueous). Oocyst size and shape were registered by forward-angle light scatter with the argon laser excitation set at 488 nm at 300 mW. Sub-populations of oocysts were collected by map gating and used for microscopy or for propagation. The profile of particle size was characteristic for each species. Propagation of sub-populations of oocysts of specified sizes resulted in cultures of coccidia that were pure species or nearly pure species. The small size of E. mills caused difficulty in separation from the remaining fine debris. This technique was useful for studying the variation in oocyst size within populations and characterization of field isolates of mixed species. Propagation of pure species from mixed isolates by bit-map gating had the same limitations as micromanipulation because of the overlapping size of Eimeria spp. Chancaerization is further limited by the lack of suitable size/shape standards for flow cytometry-Key words. AtHMA4 was previously shown to contribute to the control of Zn root‐to‐shoot translocation and tolerance to high Zn. However, heterologous expression of 35S::AtHMA4 in tobacco (Nicotiana tabacum cv. Xanthi) results in enhanced Zn sensitivity. This study provides a better understanding of the development of this Zn‐sensitive phenotype and demonstrates that substantial modifications of Zn homeostasis occur due to AtHMA4 expression. We show that ectopically expressing AtHMA4 in tobacco results in overloading the root and leaf apoplast with Zn. The tissue and cellular distribution of Zn, monitored using Zinpyr‐1, was altered in the AtHMA4‐expressing plants compared with wild type. Increased loading of the leaf apoplast with Zn in AtHMA4 transformants induced necrosis; this appeared at lower levels of Zn supply in the transgenics compared with wild type. This study suggests that Zn concentration may be sensed in the apoplast of leaves, and if concentrations are above a certain threshold then particular groups of cells accumulate Zn and necrosis is initiated. Therefore, this could be considered as a mechanism for protecting the other parts of the photosynthetically active leaf from Zn toxicity.  相似文献

SYNOPSIS. The bongo is one of the least-studied of the Tragelaphini.In a study of five captive bongo calves, two males and threefemales, we measured several behaviors important to parentalinvestment theory (Clutton-Brock et al., 1982), in order todescribe their pattern in a rare species, and add to the databasepertaining to sex allocation theory. Variations in individualpatterns of maternal investment, such as suckling rate and boutlength, were measured and graphed over time, with sex of calf,and age and dominance of dam indicated. Instances of "thiefsuckling" by calves and juveniles were also described in termsof kinship among the animals involved. Only one measure of maternalinvestment was biased in favor of males, which had a somewhathigher birth weight than females. Other measures showed littlesex difference. Among these calves, maternal care appeared tovary more with dominance and age of the dam than with sex ofcalf. Data from these animals generally conform more closelyto those of Byers and Moodie (1990). They found little evidenceof sex-biased investment among pronghorn (Antilocapra americana)fawns, and postulated that many ungulates have such a high rateof development that further differential investment is impossible.Robust biological theories are supported by data obtained froma wide variety of species, many of which are impossible to studyclosely in the wild. Additional studies on captive populationsof sexually dimorphic species would contribute usefully to theaccumulation of data pertinent to parental investment theory.  相似文献

A suitable time temperature process for packaging small potatoes in flexible pouches is described. Spores of Bacillus stearothermophilus were inoculated onto the surface of peeled potatoes in flexible pouches. These were evacuated, heat sealed and heated in a steam retort modified to allow a water cooking process with an air overpressure of ca. 68·95 kPa (10 lbf/in2). The D and z values determined in the retort were confirmed by parallel heat-resistance tests with spores in glass ampoules held in an oil bath. Heat treatment at 121·1 °C for 20 min in the retort satisfactorily killed test inocula of spores without overcooking the potatoes. Uninoculated peeled potatoes with a natural level of contamination ( ca. 4 x 103 spores/sealed pouch) were treated in the retort for different times at 121·1 °C and then incubated at 30 °C for 6 months. No pouches heated for 17–25 min showed microbial growth. This procedure may be applied to any type of food if the numbers of naturally occurring heat-resistant contaminants can be related to the number and heat resistance of a suitable test micro-organism.  相似文献

Samples of pectoral muscle tissue from Zebra Finches Poephila guttata were taken from birds at various stages of egg laying. As egg laying progressed, there was a significant loss of both water-soluble and alkali-soluble proteins. Gel filtration of the water-soluble proteins showed three peaks, only one of which changed significantly during the laving cycle. This was a high-molecular-weight protein that declined in concentration during the period of egg laying. This protein may function as a reserve, enabling female birds to provide amino acids for egg protein production from their muscle tissue without impairing normal muscle function.  相似文献
