Dopamine and adrenaline injected into mice produce dose-related mydriasis. The effects of both dopamine and adrenaline are antagonized similarly by the alpha-adrenergic blocking agents, phentolamine and thymoxamine, as well as by haloperidol, but are not prevented by pretreatment with reserpine. These results suggest that in mice dopamine produces mydriasis by direct stimulation of alpha-adrenergic receptors in the dilator iridis.  相似文献

The exquisite immunological specificity of cytotoxic T lymphocytes-target cell (CTL-TC) conjugation and lysis is overridden in the presence of certain plant lectins. The role of concanavalin A (Con A) in lectin-dependent, CTL-mediated cytolysis (LDCC) has been investigated. Papain-treated TC are refractory to LDCC, but regain susceptibility following a 3-hr incubation without the enzyme. Papain-treated TC allowed to recover in the presence of tunicamycin (TM; an inhibitor of N-linked glycosylation), are totally refractory to LDCC. Refractoriness of TM-treated TC to LDCC is not due to an overall resistance to lysis or to lack of Con A binding, as these cells can be lysed by specifically sensitized CTL or by H-2 antibody and complement and display a sufficiently high Con A-binding capacity, indistinguishable from intact TC, probably through O-linked, cell-surface glycosyl residues. The finding that TC (TM-treated) capable of binding normal Con A quantities cannot, however, engage in lectin-dependent CTL-TC conjugation and lysis indicates that Con A must react selectively with a specific TC-surface component(s), thereby rendering the TC recognizable by effector CTL, rather than by simply bridging ("glueing") CTL and TC. Affinity absorption and elution from Sepharose-Con A beads as well as specific immunoprecipitations by antibodies against cell surface determinants, have shown effective Con A binding to TC surface components of molecular weights corresponding to 45-kDa product of the H-2K and D MHC genes and, possibly, to a 30-kDa component. Antibodies against MHC proteins but not against non-MHC surface proteins of the TC have produced effective inhibition of LDCC. This and previous investigations show that in nonspecific LDCC as in specific CTL-mediated lysis, TC-MHC determinants are involved in signaling TC recognition and lysis.  相似文献

Summary The response of the cotton plant (Gossypium hirsutum L.) to 9.0 and 12.5 cm intra- and 75.0 and 96.5 cm inter-row spacing was studied under irrigation with saline water (5.5 dS/m, SAR 18). In general, the dry weight per plant matter, the leaf area, number of bolls and flowers, and yield per plant were all affected significantly by intra-row spacing but, not by inter-row spacing. However, on a unit area basis, they were affected only by the inter-row spacing. This indicates that competition between plants exists when intra-row spacing is reduced, whereas no significant competition occurs due to a decrease in the inter-row spacing. Although the effect of intra-row spacing on yield for a unit area was found to be not significant, the effect of inter-row spacing was significant. Yield in plots with the conventional spacing (96.5 cm between rows and 12.5 cm between plants in the row) was 4863 kg/ha, whereas the yield in plots with 75 cm between rows was about 23% higher (5974 kg/ha). The flowering biology and pollination ecology ofLoranthus acaciae was studied at Hazeva in the northern Arava Valley in Israel. Flowers at anthesis had red anthers, a red stigma and a green corolla which turned red as a postfloral phenomenon. Their flowering period was approximately 10 months long (from mid-June until mid-April) during which time two main flowering patterns were distinguished. Some plants flowered twice a year, with separate summer and winter flowering periods; other plants flowered continuously, with two peaks, one in the summer and one in the winter. Several significant differences between summer and winter flowering and fruiting were found: (1) the summer flowering period was shorter than that of winter, (2) flowering synchrony between individual plants was lower in summer than in winter, (3) in summer the plants produced a larger proportion of female flowers, whereas in winter most of the plants produced a larger proportion of hermaphrodites, (4) in summer a limited number of plants produced smaller flowers while the majority produced normal-sized flowers, whereas in winter the entire population produced only normal-sized flowers, and (5) fruit set percentage was lower in summer than in winter.L. acaciae was found to be self-compatible, but, since it was not spontaneously self-pollinated, it showed high dependence on pollinator activity. In summer the flowers were visited by a wide spectrum of pollinators, both birds and insects, while in winter flowers were visited almost exclusively by the orange-tufted sunbird (Nectarinia osea osea, Nectariniidae). These seasonal changes in flowering characteristics and pollinator activity could explain why reproductive success is higher in winter than in summer.  相似文献

Neuroblastoma (NB) arises from the embryonic neural crest and is the most common extracranial solid tumor in children under 5 years of age. Reduced expression of Dicer1 has recently been shown to be in correlation with poor prognosis in NB patients. This study aimed to investigate the mechanisms that could lead to the down-regulation of Dicer1 in neuroblastoma. We used computational prediction to identify potential miRs down-regulating Dicer1 in neuroblastoma. One of the miRs that were predicted to target Dicer1 was miR-192. We measured the levels of miR-192 in 43 primary tumors using real time PCR. Following the silencing of miR-192, the levels of dicer1 cell viability, cell proliferation and migration capability were analyzed. Multivariate analysis identified miR-192 as an independent prognostic marker for relapse in neuroblastoma patients (p=0.04). We were able to show through a dual luciferase assay and side-directed mutational analysis that miR-192 directly binds the 3'' UTR of Dicer1 on positions 1232-1238 and 2282-2288. An increase in cell viability, proliferation and migration rates were evident in NB cells transfected with miR-192-mimic. Yet, there was a significant decrease in proliferation when NB cells were transfected with an miR-192-inhibitor We suggest that miR-192 might be a key player in NB by regulating Dicer1 expression.  相似文献

Bovine ephemeral fever (BEF) is an economically important viral vector-borne cattle disease. Several live-attenuated, inactivated and recombinant vaccines have been tested, demonstrating varying efficacy. However, to the best of our knowledge, duration of immunity conferred by an inactivated vaccine has never been reported. In the last decade, Israel has faced an increasing number of BEF outbreaks. The need for an effective vaccine compatible with strains circulating in the Middle East region led to the development of a MONTANIDE™ ISA 206 VG (water-in-oil-in-water), inactivated vaccine based on a local strain. We tested the safety, immunogenicity and duration of immunity conferred by this vaccine. The induced neutralizing antibody (NA) response was followed for 493 days in 40 cows vaccinated by different protocols. The vaccine did not cause adverse reactions or a decrease in milk production. All cows [except 2 (6.7%) which did not respond to vaccination] showed a significant rise in NA titer of up to 1:256 following the second, third or fourth booster vaccination. Neutralizing antibody levels declined gradually to 1:16 up to 120 days post vaccination. This decline continued in cows vaccinated only twice, whereas cows vaccinated 3 or 4 times showed stable titers of approximately 1:16 for up to 267 days post vaccination. At least three vaccinations with the inactivated BEF vaccine were needed to confer long-lasting immunity. These results may have significant implications for the choice of vaccination protocol with inactivated BEF vaccines. Complementary challenge data should however be added to the above results in order to determine what is the minimal NA response conferring protection from clinical disease.  相似文献

Peronospora belbahrii is a biotrophic oomycete attacking sweet basil. It propagates asexually by producing spores on dichotomously branched sporophores emerging from leaf stomata. Sporulation occurs when infected plants are incubated for at least 7.5h in the dark in moisture-saturated atmosphere at 10-27°C. Exposure to light suppresses spore formation but allows sporophores to emerge from stomata. Incandescent or CW fluorescent light of 3.5 or 6 µmoles.m2.s-1 respectively, caused 100% inhibition of spore formation on lower leaf surface even when only the upper leaf surface was exposed to light. The inhibitory effect of light failed to translocate from an illuminated part of a leaf to a shaded part of the same leaf. Inhibition of sporulation by light was temperature-dependent. Light was fully inhibitory at 15-27°C but not at 10°C, suggesting that enzyme(s) activity and/or photoreceptor protein re-arrangement induced by light occur at ≥15°C. DCMU or paraquat could not abolish light inhibition, indicating that photosystem I and photosystem II are not involved. Narrow band led illumination showed that red light (λmax 625 nm) was most inhibitory and blue light (λmax 440 nm) was least inhibitory, suggesting that inhibition in P. belbahrii, unlike other oomycetes, operates via a red light photoreceptor. Nocturnal illumination of basil in the field (4-10 µmoles.m2.s-1 from 7pm to 7am) suppressed sporulation of P. belbahrii and reduced epidemics of downy mildew, thus reducing the need for fungicide applications. This is the first report on red light inhibition of sporulation in oomycetes and on the practical application of light for disease control in the field.  相似文献

Education for sustainability (EfS) in higher education is an emerging specialisation within the general field of EfS. EfS encompasses cognitive, affective and behavioural aspects, and aims at enhancing a variety of learning outcomes in these domains and reaching students from all programmes. One of the main challenges for higher education educators is to design courses in a way that will effectively promote the various learning outcomes of EfS. A central question is how sustainability should be integrated into the curriculum; which topics should be taught and which pedagogies ought to be applied to improve students’ knowledge, skills and motivation to promote sustainable living. The present study aimed to contribute to the knowledge about students’ learning outcomes yielded by different designs of EfS courses. This multiple-case study of three courses used a mixed-methods design. For each course, we identified its characteristics and analysed students’ self-reported learning outcomes. We found that: (1) a course with a higher degree of participatory learning, employing a system approach, promoted the highest and most varied learning outcomes; (2) the lecture-based course yielded the fewest learning outcomes; and (3) field trips promoted learning outcomes only when accompanied by more advanced pedagogies.  相似文献
