Cooling and lubrication agents like triethanolamine (TEA) are essential for many purposes in industry. Due to biodegradation, they need continuous replacement, and byproducts of degradation may be toxic. This study investigates an industrial (1,200 m³) cooling-lubrication circuit (CLC) that has been in operation for 20 years and is supposedly in an ecological equilibrium, thus offering a unique habitat. Next-generation (Illumina Miseq 16S rRNA amplicon) sequencing was used to profile the CLC-based microbiota and relate it to TEA and bicine dynamics at the sampling sites, influent, machine rooms, biofilms and effluent. Pseudomonas pseudoalcaligenes dominated the effluent and influent sites, while Alcaligenes faecalis dominated biofilms, and both species were identified as the major TEA degrading bacteria. It was shown that a 15 min heat treatment at 50°C was able to slow down the growth of both species, a promising option to control TEA degradation at large scale.  相似文献

The importance of assembly processes in shaping biological communities is poorly understood, especially for microbes. Here, we report on the forces that structure soil bacterial communities along a 2000 m elevational gradient. We characterized the relative importance of habitat filtering and competition on phylogenetic structure and turnover in bacterial communities. Bacterial communities exhibited a phylogenetically clustered pattern and were more clustered with increasing elevation. Biotic factors (i.e., relative abundance of dominant bacterial lineages) appeared to be most important to the degree of clustering, evidencing the role of the competitive ability of entire clades in shaping the communities. Phylogenetic turnover showed the greatest correlation to elevation. After controlling the elevation, biotic factors showed greater correlation to phylogenetic turnover than all the habitat variables (i.e., climate, soil and vegetation). Structural equation modelling also identified that elevation and soil organic matter exerted indirect effects on phylogenetic diversity and turnover by determining the dominance of microbial competitors. Our results suggest that competition among bacterial taxa induced by soil carbon contributes to the phylogenetic pattern across elevational gradient in the Tibetan Plateau. This highlights the importance of considering not only abiotic filtering but also biotic interactions in soil bacterial communities across stressful elevational gradients.  相似文献

Large-scale composting of source-separated household waste has expanded in recent years in the Nordic countries. One problem can be low pH at the start of the process. Incoming biowaste at four composting plants was characterised chemically, physically and microbiologically. The pH of food waste ranged from 4.7 to 6.1 and organic acid concentration from 24 to 81 mmol kg(-1). The bacterial diversity in the waste samples was high, with all samples dominated by Gammaproteobacteria, particularly Pseudomonas and Enterobacteria (Escherichia coli, Klebsiella, Enterobacter). Lactic acid bacteria were also numerically important and are known to negatively affect the composting process because the lactic acid they produce lowers the pH, inhibiting other bacteria. The bacterial groups needed for efficient composting, i.e. Bacillales and Actinobacteria, were present in appreciable amounts. The results indicated that start-up problems in the composting process can be prevented by recycling bulk material and compost.  相似文献

Prokaryotic diversity was investigated near the inlet and outlet of a plug-flow reactor. After analyzing 800 clones, 50 bacterial and 3 archaeal phylogenetic groups were defined. Clostridia (>92%) dominated among bacteria and Methanoculleus (>90%) among archaea. Significant changes in pH and volatile fatty acids did not invoke a major shift in the phylogenetic groups. We suggest that the environmental filter imposed by the saline conditions (20 g liter−1) selected a stable community of halotolerant and halophilic prokaryotes.The anaerobic digestion of organic wastes constitutes a major research focus due to the global needs for waste recycling and renewable energy production. Currently, the linkage between digester performance and the diversity and dynamics of anaerobic prokaryotes is still not fully understood (2). Bacterial diversity in anaerobic reactors has always been judged to be greater than archaeal diversity (9, 13, 30). This probably reflects the metabolic flexibility of bacteria and the range of available substrates in complex input materials. However, several recent discoveries pose the question as to whether archaeal diversity and physiological versatility are greater than currently thought: that is, the huge diversity of yet-to-be cultured archaea (4, 6), the detection of energy metabolisms not known previously in archaea (e.g., chemoorganotrophy [1]), and the unexpected predominance of archaeal groups among prokaryotes in unstressed environments, such as ammonia oxidizers in soils (19).Several surveys have investigated the shifts in prokaryotic diversity occurring with waste maturation or under different reactor operating conditions. Some evidence demonstrates bacterial phylogenetic stability under constant operation conditions (18). Generally, however, the dominant bacterial communities are very dynamic, showing chaotic shifts even with stable reactor performance (9, 32). Hypothetically, this is due to the functional redundancy among diverse phylogenetic groups allowing oscillations of their populations with no effects on the reactor function (2). Archaeal communities are less dynamic than bacterial communities (32), their shifts being related to changes in reactor performance (6) and correlated with important process parameters such as volatile fatty acids (VFAs) (13, 16).We aimed to analyze the change in prokaryotic diversity in a plug-flow reactor associated with the maturation of biowastes. In a previous study, stable bacterial and archaeal denaturing gradient gel electrophoresis patterns were found in the sludge collected close to the outlet over a year of unstable reactor performance (23). This temporal pattern contradicts the general idea of extremely dynamic bacterial communities proliferating in bioreactors. Here, we investigated the phylogenetic identity of the organisms in sludge samples collected near the inlet and outlet pipes after a period of stable operation and performance in terms of pH and biogas production.  相似文献

An automated system for continuous soil respiration and microbial biomass measurements based on Infra Red Gas Analysis was constructed. The switching device is computer controlled and allows hourly measurements of up to 24 samples when switching intervals of 2.5 min are selected. This allows the use of the substrate-induced respiration method for biomass determination. A software package to run the system was developed.  相似文献

The qualitative and quantitative changes in the bacterial community composition in two mesophilic, commercially used biogas plants were monitored by denaturing gradient gel electrophoresis (DGGE) and real-time PCR. The main objective was to evaluate the influence of the co-substrate maize silage on total bacteria and some selected bacterial groups by comparing full-scale reactors fed solely with pig manure or additionally with maize silage. DGGE fingerprints reflected shifts in the bacterial community structure associated with maize silage as co-substrate and the real-time PCR results showed clear changes in the quantitative composition of the bacterial consortia of each fermenter. A clear dominance of Clostridia in all surveyed fermenters and considerably lower abundance of Bacteroidetes in the biogas plant fed with maize silage was shown.  相似文献

The existence of anaerobic ammonia-oxidizing (anammox) bacteria was postulated in the late 1970s. Approximately 20 years later, these lithotrophic members of the nitrogen cycle were identified as deep-branching members of the planctomycetes. Recently, full-scale implementation of biological deammonification was successfully achieved in the DEMON reactor at the wastewater treatment plant in Strass, Austria. The sludge of this reactor contains red granules and brownish flocs that can be physically separated. The two fractions yielded different banding patterns in denaturing gradient gel electrophoresis of PCR products obtained with primer sets targeting the 16S rRNA genes of planctomycetes. Comparative analysis of partial sequences of almost full-length 16S rRNA gene clones obtained from the granules and flocs confirms the differences in the community composition of the two fractions. The sequences retrieved from the red granules were 93% similar to those of Candidatus Brocadia anammoxidans, a bacterium known to catalyze the anaerobic ammonia oxidation.  相似文献

Several boreal and alpine forests are depleted in nutrients due to acidification. Fertilization may be a remedy, but rapidly-soluble salts (N, P, K, Mg) may pose nitrate problems for the groundwater or decrease microbial activity.With the aim to investigate potential nitrogen leaching after fertilization we set up an experiment employing intact soil cores (11 cm diameter, 20–40 cm long) from a mixed forest and a Picea abies stand (soil type Rendsina) in the Northern Calcareous Alps of Austria. The cores were fertilized with a commercial NPK fertilizer or a methylene-urea-apatite-biotite (MuAB) fertilizer at a rate corresponding to 300 kg N ha-1 and incubated for 28 weeks together with unfertilized controls. Both soil water (retrieved 5 cm below the soil surface) and leachate were analyzed for nitrate and ammonium in regular intervals. After the incubation, soil microbial biomass and basal repiration were determined and a nitrogen mineralization assay was performed.For the control, in the soil water and leachate maximum NH4 + and NO3 - concentrations of 5 and 11 mg N L-1, respectively, were found. Compared to that, MuAB fertilizer resulted in a slow increase of NH4 + and NO3 - in the soil water (up to 11 and 35 mg N L-1 respectively) and in the leachate (4 mg NH4 +-N L-1 and 44 mg NO3 --N L-1). Highest nitrogen loads were found for the fast release NPK fertilizer, with NH4 + and NO3 - concentrations up to 170 and 270 mg N L-1, respectively, in the soil water. NH4 +-N levels in the leachate remained below 5, while NO3-N levels were up to 190 mg L-1. Fast- release NPK caused a significant decrease of microbial biomass and basal respiration. These parameters were not affected by MuAB fertilizer.The results suggest that the MuAB fertilizer may be an ecologically appropriate alternative to fast-release mineral fertilizers for improving forest soils.  相似文献
