The rate of acidification of media by Avena stem segments was studied with a titrimeter. GA3 increased this rate by an average of 17% if supplied to the segments 90 min prior to measurement. GA3 inhibited the rate by 15% if supplied 10 min prior to measurement. After 90 min incubation, stimulation of elongation had started; at 10 min, GA3 had not yet started to stimulate elongation in the segments.  相似文献

American [Castanea dentata (Marsh) Borkh.] × Chinese [Castanea mollissima Blume] chestnut (Fagac, ae) hybrids are a novel system in which to study influences of phytopathogenic fungi and woody plant hybridization on herbivore susceptibility, as the hybrids are well characterized with regard to resistance to the chestnut blight fungus [Cryphonectria parasita (Murr) Barr (Endothia) Diaporthales: Valsaceae] and variability is present. We chose two groups of resistance‐rated backcross chestnut that shared an F1 parent and had different American parents. Foliage from both backcross groups and the parent trees was sampled on three dates for use in feeding assays with gypsy moth larvae [Lymantria dispar (L.) [Lepidoptera: Lymantriidae], adult Japanese beetles [Popillia japonica Newman (Coleoptera: Scarabaeidae)], and fall webworm larvae [Hyphantria cunea Drury (Lepidoptera: Arctiidae)], respectively. Foliar analyses were performed concurrently and included carbohydrate, tannin, and nitrogen content, toughness, and density. Blight resistance had almost no effect on herbivore performance or foliar chemistry. When the parent trees and backcross groups were compared, however, significant differences in gypsy moth performance and Japanese beetle consumption were evident. There were no differences in fall webworm consumption. Most foliar characteristics measured differed among chestnut genotypes at some point in the season, and all varied seasonally. No clear pattern emerges with respect to the relationship among blight resistance, herbivore susceptibility, foliar properties, and plant genotype, and more research is needed to separate these effects.  相似文献

Modelling chestnut biogeography for American chestnut restoration   总被引:1,自引:0,他引:1  
Aim Chestnuts (Castanea spp.) are ecologically and economically important species. We studied the general biology, distribution and climatic limits of seven chestnut species from around the world. We provided climatic matching of Asiatic species to North America to assist the range‐wide restoration of American chestnut [C. dentata (Marsh.) Borkh.] by incorporating blight‐resistant genes from Asiatic species. Location North America, Europe and East Asia. Methods General chestnut biology was reviewed on the basis of published literature and field observations. Chestnut distributions were established using published range maps and literature. Climatic constraints were analysed for the northern and southern distribution limits and the entire range for each species using principal component analysis (PCA) of fourteen bioclimatic variables. Climatic envelope matching was performed for three Chinese species using Maxent modelling to predict corresponding suitable climate zones for those species in North America. Results Chestnuts are primarily distributed in the warm‐temperate and subtropical zones in the northern hemisphere. PCA results revealed that thermal gradient was the primary control of chestnut distribution. Climatic spaces of different species overlap with one another to different degrees, but strong similarities are shown especially between Chinese species and American species. Climatic envelope matching suggested that large areas in eastern North America have a favourable climate for Chinese species. Main conclusions The general biological traits and climatic limits of the seven chestnut species are very similar. The predictions of Chinese species climatic range corresponded with most of the historical American chestnut range. Thus, a regionally adapted, blight‐resistant, introgressed hybrid American chestnut appears feasible if a sufficiently diverse array of Chinese chestnut germplasm is used as a source of blight resistance. Our study provided a between‐continent climate matching approach to facilitate the range‐wide species restoration, which can be readily applied in planning the restoration of other threatened or endangered species.  相似文献

Several earlier protocols for extracting plant DNA or RNA do not work well for a variety of plants because contaminating substances coprecipitate with the nucleic acids and, thus, are present even at the last DNA-hydration step. While DNA extraction protocols have been published in which pectinase is employed to break down these contaminating substances, here we present an alternative modified pectinase protocol that potentially uses fewer steps and avoids the use of ethylene glycol monoethyl ether and phenol. DNA analyses results are described for 6 plant species demonstrating that the method works across distantly related plant taxa.  相似文献

Exogenous application of indoleacetic acid results in a significant suppression of the linear growth that is promoted by exogenous gibberellic acid in Avena stem segments in a fashion similar to that previously noted in Avena leaf base segments (van Overbeek and Dowding, 1961, Fourth International Conference Plant Growth Regulation). Treatment with the auxin transport inhibitors, methyl-2-chloro-9-hydroxyfluorene-(9)-carboxylate (CFM) or 2,3,5-triiodobenzoic acid (TIBA), alone promotes elongation growth of the stem segments over that of control growth. This effect is interpreted as being due to the interference in the transport of native indoleacetic acid by CFM and TIBA, thus removing the inhibitory effect of native indoleacetic acid on gibberellin-promoted growth in the internodal intercalary meristem. This results in a greater promotion of internodal growth by native gibberellins. In the presence of (2-chloroethyl) trimethylammonium chloride (CCC), the growth-promoting effects of CFM and TIBA are decreased, and the antiauxin, PCIB (4-chloro-phenoxyisobutyric acid), has no growth-promoting effects whatsoever. These results indicate that the CFM and TIBA-promoted growth require the continuous presence of gibberellins. They further support the view that native indoleacetic acid acts as a growth suppressor hormone in its regulation of gibberellin-promoted internodal extension in Avena shoots.  相似文献

Summary Horizontally-placed segments of Avena sativa L. shoots show a negative geotropic response after a period of 30 min. This response is based on cell elongation on the lower side of the leaf-sheath base (pulvinus). Triticum aestivum L., Hordeum vulgare L. and Secale cereale L. also show geotropic responses that are similar to those in Avena shoots. The pulvinus is a highly specialized organ with radial symmetry and is made up of epidermal, vascular, parenchymatous and collenchymatous tissues. Statoliths, which are confined to parenchyma cells around the vascular bundles, sediment towards the gravitational field within 10–15 min of geotropic stimulation. Collenchymatous cells occur as prominent bundle caps, and in Avena, they occupy about 30% of the volume of the pulvinus. Geotropic stimulation causes a 3- to 5-fold increase in the length of the cells on the side nearest to the center of the gravitational field. Growth can also be initiated in vertically-held pulvini by the application of indole-3-acetic acid, 1-naphthaleneacetic acid or 2.4-dichlorophenoxyacetic acid. 2.3.5.-triiodobenzoic acid interferes with growth response produced by geotropic stimulation as well as with the response caused by auxin application. Gibberellic acid and kinetin have no visible effect on the growth of the pulvinus. Polarization microscopy shows a unique, non-uniform stretching of the elongating collenchymatous cells. Nonelongated collenchymatous cells appear uniformally anisotropic. After geotropic stimulation or auxin application, they appear alternately anisotropic and almost isotropic. The location and some morphological, anatomical, and functional aspects of the gravity-sensitive pulvini of a selected number of grass shoots are examined. There are two distinct gravity-sensitive regions near the nodal regions of Gramineae. One, the leaf sheath pulvinus, is located at the base of the sheathing leaf bases, and is characteristic of the subfamily Festucoideae. The other, the internodal pulvinus, is located at the base of the internode, a little above the nodal joint. Most members of the Panicoideae possess internodal pulvini, in addition to more or less developed leaf sheath pulvini. Three members of the Oryzoideae examined possess leaf sheath pulvini only, while Phragmites australis (Arundinoideae) possesses both leaf sheath and internodal pulvini. Leaf sheath pulvini of some grasses develop hairs, cork-silica cell pairs or stomatal apparatuses over the epidermis while many others are devoid of any such idioblasts. Both the leaf sheath and internodal pulvini of all grasses examined preferentially exclude, or accumulate very little silica, whereas the regions of the shoot immediately above and below the pulvini in these same grasses accumulate large quantities of silica. Pulvini remain unsilicified or poorly silicified throughout their life and even after several days following geotropic bending. Pulvini are also distinguished from the regions above and below them by the lack of lignin in the bundle cap cells. Lignin is found only in the xylem vascular tissue, and this consists of annular and helical vessel elements only. The bundle cap cells are rich in pectin and are described as collenchymatous. All pulvini possess specialized zones of cells containing starch statoliths. In response to horizontal displacement of the shoots, the lower side of the pulvini grows by cell elongation only. The collenchymatous cells stretch in a manner that results in alternately thin and thick regions of cell wall.  相似文献
