Actinomycete strain SA198, isolated from a Saharan soil sample of Algeria, exhibited antimicrobial activity against Gram-positive and Gram-negative bacteria, and phytopathogenic and toxinogenic fungi. The morphological and chemotaxonomic characteristics of the strain were consistent with those of the genus Saccharothrix. Analysis of the 16S rRNA gene sequence of strain SA198 showed a similarity level ranging between 97.2 and 98.8% within Saccharothrix species, S. australiensis being the most closely related. Two new active products were isolated by reverse HPLC using a C18 column. The ultraviolet–visible (UV–VIS), infrared (IR), mass, and 1H and 14C nuclear magnetic resonance (NMR) spectra showed that these products were new bioactive compounds. The minimum inhibitory concentrations of these antibiotics showed a strong activity against fungi and moderate activities against Gram-positive and Gram-negative bacteria.  相似文献

An actinomycete strain designated G60 was isolated from a Saharan soil sample in Ghardaïa, Algeria, by a dilution agar plating method using chitin-vitamin agar medium supplemented with penicillin. Morphological and chemical studies indicated that this strain belonged to the genus Streptomyces. Analysis of the 16S rDNA sequence showed an identity level within Streptomyces species, with S. coerulescens ISP 5146T and S. bellus ISP 5185T the most closely related (100 % for each). However, the comparison of the morphological and physiological characteristics of the strain with those of the two nearest species showed significant differences. Strain G60 had a very strong activity against pathogenic staphylococci, including methicillin-resistant Staphylococcus aureus (MRSA) ATCC 43300, other clinical isolates of MRSA and vancomycin resistant S. aureus (VRSA) S1. One antimicrobial compound was extracted by n-hexane from the ISP2 culture medium at 5 days of fermentation culture and purified by HPLC. The chemical structure of the compound was determined after spectroscopic (1H NMR, 13C NMR, 1H-1H COSY and 1H-13C HMBC spectra), and spectrometric (mass spectrum) analyses. The bioactive compound was identified as di-(2-ethylhexyl) phthalate.  相似文献

Apigenin, as a natural flavonoid present in several plants is characterized with potential anticancer, antioxidant, and anti-inflammatory properties. Recent studies proposed that apigenin affects depression disorder through unknown mechanistic pathways. The effects of apigenin’s anti-depressive properties on streptozocin-mediated depression have been investigated through the evaluation of behavioral tests, oxidative stress, cellular energy homeostasis and inflammatory responses. The results demonstrated anti-depressive properties of apigenin in behavioral test including forced swimming and splash tests and oxidative stress biomarkers such as reduced glutathione, lipid peroxidation, total antioxidant power and coenzyme Q10 levels. Apigenin, also, demonstrated its regulatory potency in cellular energy homeostasis and immune system gene expression through inhibiting Nlrp3 and Tlr4 overexpression. Furthermore, failure in energy production as the key factor in various psychiatric disorders was reversed by apigenin modulating effect on AMPK gene expression. Overall, 20 mg/kg of apigenin was recognized as the dose suitable for minimizing the undesirable adverse effects in the STZ-mediated depression model proposed in this study. Our data suggested that apigenin could be able to adjust behavioral dysfunction, biochemical biomarkers and recovered cellular antioxidant level in depressed animals. The surprising results were achieved by raise in COQ10 level, which could regulate the overexpression of the AMPK gene in stressful conditions. The regulatory effect of apigenin in inflammatory signaling pathways such as Nlrp3, and Tlr4 gene expression was studied at the surface part of the hippocampus.

An actinobacteria strain PAL114, isolated from a Saharan soil in Algeria, produces bioactive compounds. Morphological and chemical studies indicated that this strain belongs to the genus Streptomyces. Analysis of the 16S rRNA gene sequence showed a similarity level of 99.8 % with S. griseoflavus LMG 19344T, the most closely related species. Two bioactive compounds, named P44 and P40, were extracted by dichloromethane from the cell-free supernatant broth and were purified by HPLC. Minimum inhibitory concentrations (MIC) of the compounds were determined against pathogenic and toxigenic microorganisms, most of which are multiresistant to antibiotics. The P40 fraction showed a strong activity especially against Candida albicans, Bacillus subtilis, and Staphylococcus aureus and has lower MIC values than those of P44 against most microorganisms tested. Chemical structures of compounds were determined based on spectroscopic and spectrometric analyses (UV-visible, mass, 1H, and 13C NMR spectra). The compounds P44 and P40 were identified as vineomycin A1 and chaetoglobosin A, respectively. Vineomycin A1 is known to be produced by some Streptomyces species. However, chaetoglobosin A is known to be produced only by fungi belonging to the genera Chaetomium, Penicillium, and Calonectria. This is the first time that chaetoglobosin A, known for its antimicrobial, anticancer, and cytotoxic effects, is reported in prokaryotes.  相似文献

An actinomycete strain designated PAL54, producing an antibacterial substance, was isolated from a Saharan soil in Ghardaïa, Algeria. Morphological and chemical studies indicated that this strain belonged to the genus Saccharothrix. Analysis of the 16S rDNA sequence showed a similarity level ranging between 96.9 and 99.2% within Saccharothrix species, with S. longispora DSM 43749T, the most closely related. DNA–DNA hybridization confirmed that strain PAL54 belonged to Saccharothrix longispora. It showed very strong activity against pathogenic Gram-positive and Gram-negative bacteria responsible for nosocomial infections and resistant to multiple antibiotics. Strain PAL54 secreted the antibiotic optimally during mid-stationary and decline phases of growth. One antibacterial compound was isolated from the culture broth and purified by HPLC. The active compound was elucidated by uv-visible and NMR spectroscopy and by mass spectrometry. The results showed that this compound was a d(?)-threo chloramphenicol. This is the first report of chloramphenicol production by a Saccharothrix species.  相似文献
