Molecular Characterization of a Barley Gene Induced by Cold Treatment   总被引:4,自引:1,他引:3  
A cDNA library was made from low positive temperature (6 ?C/2?C) grown barley shoot meristems. Several genes which are differentiallyexpressed, as measured by mRNA abundance, were selected fromthe library using a differential screen. This paper reportsan analysis of in vivo expression in several cultivars, theDNA sequence, copy number and chromosomal location of one gene(BLT14). In addition, genomic restriction fragment length polymorphismfor this gene in the 10 most widely UK-grown spring and 9 mostwidely UK-grown winter barley cultivars is analysed. Understanding the relative magnitudes of inbreeding and outbreeding depression in rare plant populations is increasingly important for effective management strategies. There may be positive and negative effects of crossing individuals in fragmented populations. Conservation strategies may include introducing new genetic material into rare plant populations, which may be beneficial or detrimental based on whether hybrid offspring are of increased or decreased quality. Thus, it is important to determine the effects of pollen source on offspring fitness in rare plants. We established pollen crosses (i.e. geitonogamous‐self, autonomous‐self, intrasite‐outcross, intersite‐outcross and open‐pollinated controls) to determine the effects of pollen source on fitness (seeds/fruit and seed mass) and early offspring traits (probability of germination, number of leaves, leaf area and seedling height) in the rare plant Polemonium vanbruntiae. Open‐pollinated, intrasite‐outcross and geitonogamous‐self treatments did not differ in fitness. However, plants receiving autonomous‐self pollen had the lowest fitness and the lowest probability of seed germination. Intersite‐outcross plants contained fewer seeds/fruit, but seeds germinated at higher frequencies and seedlings were more vigorous. We also detected heterosis at the seed germination stage. These data may imply that natural populations of P. vanbruntiae exhibit low genetic variation and little gene flow. Evidence suggests that deleterious alleles were not responsible for reduced germination; rather environmental factors, dichogamy, herkogamy and/or lack of competition among pollen grains may have caused low germinability in selfed offspring. Although self‐pollination may provide some reproductive assurance in P. vanbruntiae, the result is a reduction in germination and size‐related early traits for selfed offspring.  相似文献

Energy crops offer an opportunity to substantially increase bioenergy resources which can replace rapidly depleting fossil fuel reserves and mitigate the effect of climate change. Energy crops are typically established within traditional agricultural systems such as tillage land or grassland. Associated land use conversion has environmental implications. The aim of this paper is to propose a framework to examine how such environmental implications can be assessed, based on (a) a Strategic Environmental Assessment (SEA) approach which considers potential impacts at different stages of a plan across a wide range of environmental receptors and (b) a literature review. The example we used was that of Miscanthus replacing grassland farming. This scenario is particularly relevant to Ireland, where over 90% of the agricultural land is permanent pasture, but is also applicable to grassland conversion throughout Europe and the United States. Two consecutive phases of land‐use change were identified for assessment, each with a distinct set of environmental impacts. The first was a transition phase, lasting from initial livestock clearance and grassland ploughing until the Miscanthus crop became established (2–3 years). The second phase was the mature crop phase, lasting up to 25 years. Miscanthus cultivation was more likely to impact negatively on the environment during the transition phase than the mature phase, primarily due to abrupt disturbance and the time required for a new equilibrium to establish. However, a literature review of the impact on the environmental receptors revealed that replacing Irish agricultural grassland with Miscanthus had the potential to improve biodiversity, water, air and soil quality, and climatic factors once the crop became established and reached maturity. In order to confirm these findings an appropriate monitoring programme involving objectives and indicators associated with each environmental receptor would need to be developed.  相似文献

The introduction of exotic species to ecosystems can have severe consequences for populations of native organisms, but logistical limitations and shortage of historical data often hinder attempts to quantify the ecological implications of such relationships. The establishment and rapid expansion of Ring‐necked Parakeets Psittacula krameri in England therefore presents a rare opportunity to apply novel analytical methods to existing extensive national bird monitoring data from the UK Breeding Bird Survey for an invasive species. A previous study from Belgium suggests that Ring‐necked Parakeets have the potential to reduce the abundance of Eurasian Nuthatch Sitta europaea through competition for nesting cavities. Our analysis provides no evidence for a significant impact through competition on Nuthatch populations or those of any other cavity‐nesting species within the Parakeet’s current range in the UK. However, we cannot exclude the possibility that competitive exclusion could be occurring at a minority of sites at which availability of nest cavities is limiting. This may yet have significant implications for future conflict if Parakeets continue to increase in numbers and range.  相似文献

Most agricultural land has been compacted to some degree by heavy machinery or livestock trampling. This legacy is expected to influence the success of tree seedling recruits in farmland areas where natural regeneration is being encouraged. We investigated the impact of soil compaction on seedlings of a woodland eucalypt (Eucalyptus albens) and an annual grass competitor (Vulpia myuros) in a laboratory experiment. Replicate soil cores were created at five bulk density levels; 1.0, 1.1, 1.2, 1.3 or 1.4 Mg m?3 with a soil water content of 20%. The depth of root penetration declined linearly with increasing bulk density, resulting in a decrease in root depth of around 75% in the most compacted soil compared with the least compacted soil for both species. Shoot length and primary root length did not vary between soil bulk density levels for either species, but seedlings responded to increasing levels of compaction with oblique (non‐vertical) root growth. Results suggest that young seedlings of both E. albens and V. myuros will be more susceptible to surface drying in compacted than uncompacted soils and therefore face a greater risk of desiccation during the critical months following germination. Any competitive advantage that V. myuros may have over E. albens is not evident in differential response to soil compaction.  相似文献

The selection of a sampling protocol is critical to study amphibian and reptile communities and in many instances researchers have combined the use of visual encounter surveys (VES) conducted on trails and off trails. The effect of human‐made trails on relative abundance estimates of amphibians and reptiles has been assessed in a few temperate locations, but data are lacking for tropical sites. Our study was designed to address this issue by comparing abundance estimates of frogs and lizards on and off trails in a lowland rainforest in south‐eastern Perú. We used nocturnal VES to sample frogs and lizards along transects established on trails and off trails in two different forest types. We found that the observed relative abundance estimates of frogs and lizards were affected by the location of transects (on trail vs. off trail) and the type of forest (floodplain forest vs. terra firme forest). We also found an interaction between the two main effects, indicating that the effect of transect location with respect to trails varies as a function of habitat. Observed frog abundances were higher on trails than off trails, indicating that studies that include VES on trails will bias relative abundance estimates in contrast to studies that include only VES off trails. We suggest that transects should be established only off trails, especially for monitoring studies because trail use by humans can have a strong influence on observed animal abundance.  相似文献

The availability of expressed sequence data derived from gene discovery programs enables mining for simple sequence repeats (SSR), providing useful genetic markers for crop improvement. These markers are inexpensive, require minimal labour to produce and can frequently be associated with functionally annotated genes. This study presents the development and characterization of 16 expressed sequence tags (EST)‐SSR markers from Brassica juncea and their cross‐amplification across Brassica species. Sixteen primer pairs were assessed for polymorphism in all genomes of the diploid and amphidiploid Brassica species. The markers show reliable amplification, considerable polymorphism and high transferability across species, demonstrating the utility of EST‐SSRs for genetic analysis of brassicas.  相似文献

Abstract Female viviparous lizards from temperate locations in the Southern Hemisphere (New Zealand, Tasmania (Australia), South Africa and South America) often have reproductive activity spanning many months of the year. In contrast, vitellogenesis and pregnancy are often confined to the spring/summer months in viviparous species from temperate zones of the Northern Hemisphere. An extreme Southern Hemisphere example is the nocturnal common gecko from New Zealand, Hoplodactylus maculatus (Gray 1845), in which females exhibit biennial reproduction with pregnancy lasting up to 14 months in a cool‐climate population. Here, we examined whether such an extended reproductive cycle also occurs in a diurnal species, the jewelled gecko Naultinus gemmeus (McCann 1955), at a similar latitude. Palpation was used to assess reproductive condition non‐invasively. In contrast to the nearby higher‐altitude population of H. maculatus, N. gemmeus reproduces annually. Vitellogenesis occurs from autumn to spring in both species, but pregnancy ends after about 7 months in N. gemmeus. Birth occurs in the seemingly unpropitious season of mid‐ to late autumn, a pattern that may be unique for lizards from cool‐temperate zones. We hypothesize that there are major differences between populations of N. gemmeus and H. maculatus with respect to survival of autumn‐born neonates and/or costs to females from remaining pregnant over winter. Museum specimens of N. gemmeus support anatomical inferences from palpation; they also suggest that vitellogenesis may begin before the end of pregnancy (which may be essential to completing each reproductive cycle within a year) and that some populations may show gestation in utero over winter, as in H. maculatus. Extended gestation appears to be a common response to cool climates for Southern Hemisphere lizards that have independently evolved viviparity.  相似文献
