以丽格海棠幼嫩叶柄为外植体,采用培养基④(MS培养基,4.0 mg/L 2,4-D,0.5 mg/L6-BA)为胚性愈伤组织诱导培养基,诱导率达86%;以培养基③(MS培养基,3.0 mg/L 2,4-D,0.2 mg/L 6-BA)为胚性愈伤组织增殖培养基进行扩增;胚状体分化培养基为培养基⑥(MS培养基,2.0 mg/L 6-BA,0.2 mg/L NAA,0.2 mg/L IBA),分化率达100%;对幼根发育不良的胚性植株,在培养基⑩(MS培养基,0.1 mg/L IAA)上进行生根培养,生根率100%,平均根数3~5条,带侧根,长约2~3 cm。试管苗在森林土:蛭石:河沙=1:1:1的基质中生长良好,成活率100%。培养基中均附加3%蔗糖、0.6%琼脂,pH值为5.8~6.0。  相似文献

Introgression lines population was effectively used in mapping quantitative trait loci (QTLs), identifying favorable genes, discovering hidden genetic variation, evaluating the action or interaction of QTLs in multiple conditions and providing the favorable experimental materials for plant breeding and genetic research. In this study, an advanced backcross and consecutive selfing strategy was used to develop introgression lines (ILs), which derived from an accession of Oryza rufipogon Griff. collected from Yuanjiang County, Yunnan Province of China, as the donor, and an elite indica cultivar Teqing (O. sativa L.), as the recipient. Introgression segments from O. rufipogon were screened using 179 polymorphic simple sequence repeats (SSR) markers in the genome of each IL. Introgressed segments carried by the introgression lines population contained 120 ILs covering the whole O. rufipogon genome. The mean number of homozygous O. rufipogon segments per introgression line was about 3.88. The average length of introgressed segments was approximate 25.5 cM, and about 20.8% of these segments had sizes less than 10 cM. The genome of each IL harbored the chromosomal fragments of O. rufipogon ranging from 0.54% to 23.7%, with an overall average of 5.79%. At each locus, the ratio of substitution of O. rufipogon alleles had a range of 1.67-9.33, with an average of 5.50. A wide range of alterations in morphological and yield-related traits were also found in the introgression lines population. Using single-point analysis, a total of 37 putative QTLs for yield and yield components were detected at two sites with 7%-20% explaining the phenotypic variance. Nineteen QTLs (51.4%) were detected at both sites, and the alleles from O. rufipogon at fifteen loci (40.5%) improved the yield and yield components in the Teqing background. These O. rufipogon-O, sativa introgression lines will serve as genetic materials for identifying and using favorable genes from common wild rice.  相似文献

应用外周血淋巴细胞染色体畸变和CB微核分析方法,对2000年河南许昌“3.06”60Co辐射事故1例受照者(A)和2000年河南开封“6.26”辐射事故一例过量放射性核素内污染致γ射线照射受照者(B)的生物剂量进行估算。结果表明,“A”和“B”两例受照者依据双+环率估算的剂量分别为1.44Gy和0.15Gy,CB微核率估算的剂量分别为1.43Gy和0.22Gy,与用物理方法估算的剂量比较接近,亦与放射损伤的临床诊断一致。泊松分布检验证实,“A”偏离泊松分布,受到不均匀照射。提示染色体畸变和CB微核分析是非常可靠的生物计量估算方法。 Abstract:Biological doses were estimated by using the yields of dicentrics plus rings(dic+r) and cytokinesis-block micronuclei (CBMN) for two victims of the accident radiation occurred on Mar 6,2000 in the City of Xuchang(victim A),and Jun 26,2000 in the City of Kaifeng(victim B),Henan Province,respectively.The results indicated that the equvalent whole body doses based on the standard dose-response curves established by the analyses of dic+r and CBMN were estimated to be 1.44Gy and 1.43Gy(victim A),and 0.15Gy and 0.22Gy(victim B).These doses were very consistent with the mean doses calculated from physical measurement and conformable to the clinical dianosis.The dic+r aberrations of victim A did not accord with Poisson distribution.The analyses of chromosomal aberrations and CBMN are an extremely reliable method in biological dosimetry.The radiation which victim A was exposed to was heterogeneous.  相似文献

Genetically modified pigs are valuable models of human disease and donors of xenotransplanted organs.Conventional gene targeting in pig somatic cells is extremely inefficient.Zinc-finger nuclease(ZFN)technology has been shown to be a powerful tool for efficiently inducing mutations in the genome.However,ZFN-mediated targeting in pigs has rarely been achieved.Here,we used ZFNs to knock out the porcineα-1,3-galactosyl-transferase(GGTA1)gene,which generates Gal epitopes that trigger hyperacute immune rejection in pig-to-human transplantation.Primary pig fibroblasts were transfected with ZFNs targeting the coding region of GGTA1.Eighteen mono-allelic and four biallelic knockout cell clones were obtained after drug selection with efficiencies of 23.4%and 5.2%,respectively.The biallelic cells were used to produce cloned pigs via somatic cell nuclear transfer(SCNT).Three GGTA1 null piglets were born,and one knockout primary fibroblast cell line was established from a cloned fetus.Gal epitopes on GGTA1 null pig cells were completely eliminated from the cell membrane.Functionally,GGTA1 knockout cells were protected from complement-mediated immune attacks when incubated with human serum.This study demonstrated that ZFN is an efficient tool in creating gene-modified pigs.GGTA1 null pigs and GGTA1 null fetal fibroblasts would benefit research and pig-to-human transplantation.  相似文献

建立以亚热带湿生、水生植物为主的十二套下流行一上流-上行流人工湿地系统作为处理城镇生活污水的对策。以其中四套研究其在不同的水力负荷及气候条件下对污水中磷的去除效果。人工湿地系统随处理运行时间的推移趋于稳定,对污水中的总磷、无机磷显示较好的净化效率,平均去除率在冬季达到40%以上,夏季达到60%以上,出水达到国家地面水Ⅲ级标准。水生植物在系统中起到明显作用,有植物系统的除磷效率及稳定性均高于无植物对照,其中2号茭白-石菖蒲系统的效果最好,总磷平均去除率为65%。4号9蔗-苔草系统在高水力负荷下的净效果优于2号。水力负荷的增加对系统的净效果没有明显影响。  相似文献

Breast tumors are typically heterogeneous and contain diverse subpopulations of tumor cells with differing phenotypic properties. Planar cultures of cancer cell lines are not viable models of investigation of cell-cell and cell-matrix interactions during tumor development. This article presents an in vitro coculture-based 3-dimensional heterogeneous breast tumor model that can be used in drug resistance and drug delivery investigations. Breast cancer cell lines of different phenotypes (MDAMB231, MCF7, and ZR751) were cocultured in a rotating wall vessel bioreactor to form a large number of heterogeneous tumoroids in a single cell culture experiment. Cells in the rotating vessels were labeled with Cell Tracker fluorescent probes to allow for time course fluorescence microscopy to monitor cell aggregation. Histological sections of tumoroids were stained with hematoxylin and eosin, progesterone receptor, E-cadherin (E-cad), and proliferation marker ki67. In vitro tumoroids developed in this study recapture important features of the temporal-spatial organization of solid tumors, including the presence of necrotic areas at the center and higher levels of cell division at the tumor periphery. E-cad-positive MCF7 cells form larger tumoroids than E-cad-negative MDAMB231 cells. In heterogeneous tumors, the irregular surface roughness was mainly due to the presence of MDAMB231 cells, whereas MCF7 cells formed smooth surfaces. Moreover, when heterogeneous tumoroids were placed onto collagen gels, highly invasive MDAMB231 cell-rich surface regions produced extensions into the matrix, whereas poorly invasive MCF7 cells did not. The fact that one can form a large number of 1-mm tumoroids in 1 coculture attests to the potential use of this system at high-throughput investigations of cancer drug development and drug delivery into the tumor.  相似文献
