Summary We have examined DNA restriction fragment length polymorphisms (RFLP) of the Ha-ras-1 gene in DNA from 118 lung cancer patients and 123 unaffected controls. When DNA samples were digested with MspI/ HpaII restriction endonucleases. Southern blot analysis demonstrated 4 common, 4 intermediate and 7 different rare alleles in the combined population after hybridization to the pGDa1 probe. Six of the rare alleles were unique for the lung cancer group and 1 rare allele for the control group. The frequency of rare alleles in lung cancer patients (10/236) was significantly different (P<0.01) from the control group (1/246). The lung cancer group also had a significantly lower frequency of the common 2.57 kb fragment than the controls (P<0.02). The results thus indicate that Ha-ras genotyping may be of value in lung cancer risk assessment.  相似文献

ABSTRACT

The use of film as a tool in anthropological fieldwork is a challenge to traditional ways of thinking about scientific research. Nevertheless, documentary filming of the informants in their own context has proved to be an interesting supplement in the exploration of what the anthropological research field actually covers. Furthermore, film work confronts the researchers with aesthetic aspects of mediation that have not been epistemically well attended to within social science. This article highlights pivotal problems in the professional no-man's-land where visual culture studies take place, as exemplified by the film Cool and Crazy.  相似文献

Climate warming is predicted to affect species and trophic interactions worldwide, and alpine ecosystems are expected to be especially sensitive to changes. In this study, we used two ongoing climate warming (open‐top chambers) experiments at Finse, southern Norway, to examine whether warming had an effect on herbivory by leaf‐chewing insects in an alpine Dryas heath community. We recorded feeding marks on the most common vascular plant species in warmed and control plots at two experimental sites at different elevations and carried out a brief inventory of insect herbivores. Experimental warming increased herbivory on Dryas octopetala and Bistorta vivipara. Dryas octopetala also experienced increased herbivory at the lower and warmer site, indicating an overall positive effect of warming, whereas B. vivipara experienced an increased herbivory at the colder and higher site indicating a mixed effect of warming. The Lepidoptera Zygaena exulans and Sympistis nigrita were the two most common leaf‐chewing insects in the Dryas heath. Based on the observed patterns of herbivory, the insects life cycles and feeding preferences, we argue that Z. exulans is the most important herbivore on B. vivipara, and S. nigrita the most important herbivore on D. octopetala. We conclude that if the degree of insect herbivory increases in a warmer world, as suggested by this study and others, complex interactions between plants, insects, and site‐specific conditions make it hard to predict overall effects on plant communities.  相似文献

Immigrant women are a particularly vulnerable part of the immigrant population. In this paper we analyse negative attitudes towards immigrant women in Norway. We focus on immigrant women’s formal job qualifications, their religious background and wearing of hijab – the headscarf sometimes used by Muslim women. Using survey-embedded experiments we are able to analyse the net effects on attitudes of job qualifications, Islamic religious background and the hijab. The results show that native population frequently has more negative views of Muslim women who wear a hijab. The negative effects of a hijab do not seem to be strongly reduced if a woman wearing it has higher education. With a single exception, the results also show that Muslim background in itself (i.e. without the hijab) does not have any strong effect on attitudes of the native population towards immigrant women.  相似文献

Predicted increases in the frequency and duration of drought are expected to negatively affect tree vitality, but we know little about how water shortage will influence needle anatomy and thereby the trees’ photosynthetic and hydraulic capacity. In this study, we evaluated anatomical changes in sun and shade needles of 20‐year‐old Norway spruce trees exposed to artificial drought stress. Canopy position was found to be important for needle structure, as sun needles had significantly higher values than shade needles for all anatomical traits (i.e., cross‐sectional needle area, number of tracheids in needle, needle hydraulic conductivity, and tracheid lumen area), except proportion of xylem area per cross‐sectional needle area. In sun needles, drought reduced all trait values by 10–40%, whereas in shade needles, only tracheid maximum diameter was reduced by drought. Due to the relatively weaker response of shade needles than sun needles in drought‐stressed trees, the difference between the two needle types was reduced by 25% in the drought‐stressed trees compared to the control trees. The observed changes in needle anatomy provide new understanding of how Norway spruce adapts to drought stress and may improve predictions of how forests will respond to global climate change.  相似文献

Background

Patients with chest pain or palpitations often have poor outcomes following a negative cardiac evaluation, with symptom persistence, limitations in everyday activities, and reduced health-related quality of life. The aims of this study were to evaluate illness perceptions before and after negative cardiac evaluations and measure the ability of a self-report questionnaire to predict outcomes.

Methods

Patients (N?=?138) referred for chest pain or palpitations to a cardiac outpatient clinic were assessed before and six months after a negative cardiac evaluation. In addition to Brief Illness Perception Questionnaire (BIPQ), all patients completed the Beck Depression Inventory and SF-36 Health Survey.

Results

The emotional reactions to and understanding of symptoms had not improved six months after a negative cardiac evaluation. A stronger correlation between illness perceptions and health at follow-up than before the cardiac evaluation might explain the tendency for poor outcomes among these patients. Most of the eight BIPQ item scores before the negative cardiac evaluation were predictive of the outcome six months later. A single question asking about the perceived consequences of the complaints (BIPQ Item 1) rated before the cardiac evaluation was collapsed into a dichotomous variable with a cut-off at ≥4 which yields a sensitivity of 51%, a specificity of 85%, a positive predictive value of 71%, a negative predictive value of 69%, and an odds ratio of 5.7 (r?=?.38, p?<?.001) in predicting poor outcomes.

Conclusions

Assessing illness perceptions is important in patients with negative cardiac tests for understanding and predicting outcomes.

Human cytomegalovirus (HCMV) infection is the most common congenital infection in developed countries and is responsible for a substantial fraction of sensorineural hearing impairment (SNHI) in children. The risk of hearing impairment is associated with viral load in urine and blood collected during the first postnatal month. However, although inner ear abnormalities are observed in some children with HCMV-induced SNHI, the exact mechanism whereby congenital HCMV infection causes hearing impairment is unknown. Earlier studies using standard cytogenetic mapping techniques showed that infection of S-phase human fibroblast cells with HCMV resulted in two specific, site-directed, chromosome breaks at band positions 1q21 and 1q42 which include loci involved in dominantly and recessively inherited hearing impairment, respectively. These findings suggested that cells infected with HCMV might provide a reservoir for genetic damage and, in a clinical perspective, a scenario could be envisioned whereby hearing impairment could result from early DNA damage of dividing fetal cells rather than viral replication and cell lysis. In this work we demonstrate, using fine mapping techniques, that HCMV infection in S-phase fibroblast cells induces genetic damage at 1q23.3, within a maximal region of 37 kb, containing five low copy repeat (LCR) elements. The breakpoint is situated between two hearing impairment (HI) loci, DFNA49 and DFNA7, and in close proximity to the MPZ gene previously shown to be involved in autosomal dominant Charcot-Marie-Tooth syndrome (CMT1B) with auditory neuropathy.  相似文献

Objectives

Afforestation changes soil chemical properties over several decades. In contrast, microbial community structure can be shifted within the first decade and so, the direct effects of tree species can be revealed. The aim of this study was to determine the alteration of soil microbial community composition 10 years after afforestation by trees with contrasting functional traits.

Methods

The study was conducted at the BangorDIVERSE temperate forest experiment. Soil samples were collected under single, two and three species mixtures of alder and birch, beech and oak - early and secondary successional species, respectively, and contiguous agricultural field. Soil was analysed for total carbon (C) and nitrogen (N) contents, and microbial community structure (phospholipid fatty acids (PLFAs) analysis).

Results and conclusions

The total PLFAs content (370–640 nmol g?1 soil) in forest plots increased for 30 to 110 % compared to the agricultural soil (290 nmol g?1 soil). In contrast, soil C, N and C/N ratios were altered over 10 years much less - increased only up to 20 % or even decreased (for beech forest).

Afforestation increased bacterial PLFAs by 20–120 %, whereas it had stronger impact on the development of fungal communities (increased by 50–200 %). These effects were proved for all forests, but were more pronounced under the monocultures compared to mixtures. This indicates that species identity has a stronger effect than species diversity. Principal component analysis of PLFAs revealed that under mono and three species mixtures similar microbial communities were formed. In contrast, gram-positive PLFAs and actinomycete PLFAs contributed mainly to differentiation of two species mixtures from other forests. Thus, at the early afforestation stage: i) soil biological properties are altered more than chemical, and ii) tree species identity affects more than species amount on both processes.

Considerable knowledge exists about the effect of aluminium (Al) on root vitality, but whether elevated levels of Al affect soil microorganisms is largely unknown. We thus compared soils from Al-treated and control plots of a field experiment with respect to microbial and chemical parameters, as well as root growth and vitality. The field experiment was established in a 50-year-old Norway spruce (Picea abies L.) stand where no Al or low concentrations of Al had been added every 7–10 days during the growth season for 7 years. Analysis of soil solutions collected using zero tension lysimeters and porous suction cups showed that Al treatment lead to increased concentrations of Al, Ca and Mg and lower pH and [Ca + Mg + K/Al] molar ratio. Corresponding soil analyses showed that soil pH remained unaffected (pH 3.8), that exchangeable Al increased, while exchangeable Ca and Mg decreased due to the Al treatment. Root in-growth into cores placed in the upper 20 cm of the soil during three growth seasons was not affected by Al additions, neither was nutrient concentration or mortality of these roots. The biomass of some taxonomic groups of soil microorganisms, analyzed using specific membrane components (phospholipid fatty acids; PLFAs), was clearly affected by the imposed Al treatment, both in the organic soil horizon and in the underlying mineral soil. Microbial community structure in both horizons was also clearly modified by the Al treatment. Shifts in PLFA trans/cis ratios indicative of short term physiological stress were not observed. Yet, aluminium stress was indicated both by changes in community structure and in ratios of single PLFAs for treated/untreated plots. Thus, soil microorganisms were more sensitive indicators of subtle chemical changes in soil than chemical composition and vitality of roots.  相似文献

Root cap development in cereals and legumes is self-regulated by a repressor that accumulates in the extracellular environment, and immersing the root tip into water results in renewed cap development. By exploiting this phenomenon, root cap mitosis and differentiation can be synchronously induced among populations. In Pisum sativum L., messenger RNA (mRNA) differential display revealed changes in expression of approximately 1% of the sample mRNA population within minutes of induced cap turnover. This profile changes sequentially over a period of 30 min, then stabilizes. Microarray analysis of Medicago truncatula root caps confirmed changes in expression of approximately 1% of the target population, within minutes. A cell specific marker for cap turnover exhibited the same temporal and spatial expression profile in the gymnosperm species Norway spruce (Picea abies) as in pea. Induced cap development provides a means to profile cell-specific gene expression among phylogenetically diverse species from the early moments of mitosis and cellular differentiation.  相似文献
