This study was designed to investigate spatial and temporal variation in Gelidium canariensis populations at two shores in
northern Gran Canaria during two years. Spatial scales ranged from some hundred meters (distance between shores), 10 to 30
m (distance between plots) to less than 3 m (distance between quadrats). Gelidium individuals were defined as distinct Gelidium
clumps. The results show a significant difference in size of clumps between shores, but not on the smaller spatial scales.
No significant temporal variation was found. There was no significant temporal or spatial variation in standing crop or density
(counts made in quadrats where Gelidium was present, rather than counts for the total shore). Sporophytic and gametophytic
clumps were also distinguished by identifying reproductive structures in the field. The total proportion of sporophytes was
larger than the proportion of gametophytes, but at a smaller scale there could be a shift in dominance. The survival rate
of clumps was similar between shores with a mean survival rate of 85%, but there was a significant difference in recruitment
between shores. The results indicate a stable population structure.

A metabolomics approach for prediction of bacteremic sepsis in patients in the emergency room (ER) was investigated. In a prospective study, whole blood samples from 65 patients with bacteremic sepsis and 49 ER controls were compared. The blood samples were analyzed using gas chromatography coupled to time-of-flight mass spectrometry. Multivariate and logistic regression modeling using metabolites identified by chromatography or using conventional laboratory parameters and clinical scores of infection were employed. A predictive model of bacteremic sepsis with 107 metabolites was developed and validated. The number of metabolites was reduced stepwise until identifying a set of 6 predictive metabolites. A 6-metabolite predictive logistic regression model showed a sensitivity of 0.91(95% CI 0.69–0.99) and a specificity 0.84 (95% CI 0.58–0.94) with an AUC of 0.93 (95% CI 0.89–1.01). Myristic acid was the single most predictive metabolite, with a sensitivity of 1.00 (95% CI 0.85–1.00) and specificity of 0.95 (95% CI 0.74–0.99), and performed better than various combinations of conventional laboratory and clinical parameters. We found that a metabolomics approach for analysis of acute blood samples was useful for identification of patients with bacteremic sepsis. We describe and examine methods for estimating spatial correlations used in population ecology. We base our analyses on a hypothetical example of a species that has been censured at 30 different locations for 20 years. We assume that the population fluctuations can be described by a simple linear model on logarithmic scale. Stochastic simulations is utilized to check how seven different ways of resampling perform when the goal is to find nominal 95% confidence intervals for the spatial correlation in growth rates at given distances. It turns out that resampling of locations performs badly, with true coverage level as low as 30–40%, especially for small correlations at long distances. Resampling of timepoints performs much better, with coverage varying from 80 to 90%, depending on the strength of density regulation and whether the spatial correlation is estimated for the response variable or for the error terms in the model. Assuming that the underlying model is known, the best results are achieved for parametric bootstrapping, a result that strongly emphasize the importance of defining and estimating a proper population model when studying spatial processes. 相似文献

A large bioreactor is heterogeneous with respect to concentration gradients of substrates fed to the reactor such as oxygen and growth limiting carbon source. Gradient formation will highly depend on the fluid dynamics and mass transfer capacity of the reactor, especially in the area in which the substrate is added. In this study, some production-scale (12 m3 bioreactor) conditions of a recombinant Escherichia coli process were imitated on a laboratory scale. From the large-scale cultivations, it was shown that locally high concentration of the limiting substrate fed to the process, in this case glucose, existed at the level of the feedpoint. The large-scale process was scaled down from: (i) mixing time experiments performed in the large-scale bioreactor in order to identify and describe the oscillating environment and (ii) identification of two distinct glucose concentration zones in the reactor. An important parameter obtained from mixing time experiments was the residence time in the feed zone of about 10 seconds. The size of the feed zone was estimated to 10%. Based on these observations the scale-down reactor with two compartments was designed. It was composed of one stirred tank reactor and an aerated plug flow reactor, in which the effect of oscillating glucose concentration on biomass yield and acetate formation was studied. Results from these experiments indicated that the lower biomass yield and higher acetate formation obtained on a large scale compared to homogeneous small-scale cultivations were not directly caused by the cell response to the glucose oscillation. This was concluded since no acetate was accumulated during scale-down experiments. An explanation for the differences in results between the two reactor scales may be a secondary effect of high glucose concentration resulting in an increased glucose metabolism causing an oxygen consumption rate locally exceeding the transfer rate. The results from pulse response experiments and glucose concentration measurements, at different locations in the reactor, showed a great consistency for the two feeding/pulse positions used in the large-scale bioreactor. Furthermore, measured periodicity from mixing data agrees well with expected circulation times for each impeller volume. Conclusions are drawn concerning the design of the scale-down reactor. 相似文献
