An off-line filtering ditch–pond system was designed and constructed to control the small point and runoff pollution at the Wuhan City Zoo, Hubei Province, China. The quantity and quality of wastewater discharge and runoff from 16 rainfall events were measured to test the effectiveness of the off-line treatment train. The results showed that the water quality was improved and high retention rates for water and pollutants were also achieved by the off-line treatment train. In the outflows, the event mean concentrations (EMCs) of TSS, COD, TN, TDN, TP and TDP were reduced by 75%, 50%, 50%, 57%, 74% and 80% compared to the inflows. In 2005, the annual inflow volume in the catchment was 6783 m3 and the water retention rate was 80.1%. The retention rates in the annual loads of TSS, COD, TN and TP came to 86.4%, 85.5%, 83.9% and 82.9%, respectively. Therefore, the off-line filtering ditch–pond system was shown to be an effective and economical measure to control diffuse pollution. It would be worthwhile to extend the off-line treatment train to regions with limited land resources, especially in urban areas.  相似文献

A novel label-free electrogenerated chemiluminescence (ECL) aptasensor for the determination of lysozyme is designed employing lysozyme binding aptamer (LBA) as molecular recognition element for lysozyme as a model analyte and Ru(bpy)(3)(2+) as an ECL signal compound. This ECL aptasensor was fabricated by self-assembling the thiolated LBA onto the surface of a gold electrode. Using this aptasensor, sensitive quantitative detection of lysozyme is realized on basis of the competition of lysozyme with Ru(bpy)(3)(2+) cation for the binding sites of LBA. In the presence of lysozyme, the aptamer sequence prefers to form the LBA-lysozyme complex, the less negative environment allows Ru(bpy)(3)(2+) cations to be less bound electrostatically to the LBAs on the electrode surface, in conjunction with the generation of a decreased ECL signal. The integrated ECL intensity versus the concentration of lysozyme was linear in the range from 6.4×10(-10) M to 6.4×10(-7) M. The detection limit was 1.2×10(-10) M. This work demonstrates that using the competition of target protein with an ECL signal compound Ru(bpy)(3)(2+) for binding sites of special aptamer confined on the electrode is promising approach for the design of label-free ECL aptasensors for the determination of proteins.  相似文献

A simple and highly sensitive electrochemical impedance spectroscopy (EIS) biosensor based on a thrombin-binding aptamer as molecular recognition element was developed for the determination of thrombin. The signal enhancement was achieved by using gold nanoparticles (GNPs), which was electrodeposited onto a glassy carbon electrode (GCE), as a platform for the immobilization of the thiolated aptamer. In the measurement of thrombin, the change in interfacial electron transfer resistance of the biosensor using a redox couple of [Fe(CN)6]3−/4− as the probe was monitored. The increase of the electron transfer resistance of the biosensor is linear with the concentration of thrombin in the range from 0.12 nM to 30 nM. The association and dissociation rate constants of the immobilized aptamer–thrombin complex were 6.7 × 103 M−1 s−1 and 1.0 × 10−4 s−1, respectively. The association and dissociation constants of three different immobilized aptamers binding with thrombin were measured and the difference of the dissociation constants obtained was discussed. This work demonstrates that GNPs electrodeposited on GCE used as a platform for the immobilization of the thiolated aptamer can improve the sensitivity of an EIS biosensor for the determination of protein. This work also demonstrates that EIS method is an efficient method for the determination of association and dissociation constants on GNPs modified GCE.  相似文献

氮肥施用量对设施番茄氮素利用及土壤NO3--N累积的影响   总被引:2,自引:0,他引:2  
在宁夏引黄灌区滴灌条件下,以温棚番茄为研究对象,采用田间试验与室内分析的方法,研究不同施氮量对番茄产量、氮素利用率及土壤NO3--N残留的影响,并对土壤-番茄体系的氮平衡进行了表观评估。试验结果表明:2004年秋冬茬番茄,施用氮肥显著增加了当茬番茄果实、植株产量 (增产幅度19.9%~29.6%)及地上部总吸氮量(204~232.6 kg/hm2)。2005年冬春茬番茄,与空白处理产量相比(106 kg/hm2)只有N200处理的果实增产达显著水平(120 kg/hm2)。两季番茄的氮肥利用率都随氮肥施用量的增加而明显降低(3.3%~10.9%)。2004年秋冬茬番茄收获后,表层0~30 cm的NO3--N大量累积(200~650 kg/hm2),其累积量随施氮量增加呈增加的趋势,经过2005年冬春茬番茄种植后,上茬0~30 cm土层NO3--N向下有淋失的趋势,淋失层次主要在90 cm以上的土体(250~380 kg/hm2)。综合考虑番茄果实产量、氮肥利用率及土壤NO3--N残留等因素,秋冬茬番茄推荐氮肥用量在100~200 kg/hm2和适量的磷钾肥配施为当茬氮肥优化管理处理。而冬春茬番茄氮肥推荐在200~400kg/ hm2 范围可以满足当茬番茄对氮肥的需求。  相似文献

Electrogenerated chemiluminescence (ECL) of a ruthenium complex polymer modified carbon paste electrode and its analytical applications were investigated. The ruthenium complex polymer was prepared using bis(2,2‐bipyridine) (4,4‐dicarboxy‐2,2‐bipyridine) ruthenium(II). The ECL behaviours of ruthenium complex polymer modified carbon paste electrode were investigated in the absence and presence of tripropylamine (TPA). The modified carbon paste electrode exhibited long‐term stability and fine reproducibility. The ECL intensity of the modified carbon paste electrode was linear with the concentration of TPA in the range 2.0 × 10–6–3.8 × 10–3 mol/L, with a detection limit (S:N = 3) of 6 × 10–7 mol/L. It was also found that raceanisodamine could enhance the ECL intensity of the modified electrode. The ECL intensity of the modified carbon paste electrode was linear with the concentration of raceanisodamine in the range 1.1 × 10–5–6.0 × 10–4 mol/L, with a detection limit (S:N = 3) of 6 × 10–6 mol/L. This work demonstrates that the entrapment of ruthenium complex in a highly cross‐linked polymer is a promising approach to construct an ECL modified electrode with long‐term stability and fine reproducibility. 碱性螺旋-环-螺旋(Basic helix-loop-helix protein,bHLH)家族成员Twist2对间质细胞系的发生和发育起转录调节作用,经直接或间接机制发挥分子开关功能,从而激活或抑制靶基因。Twist2能直接结合DNA上 E-box保守序列,招募共激活物或抑制剂;能与E蛋白调节因子发生蛋白-蛋白相互作用,干扰激活或抑制功能。Twist2无义突变导致Setleis综合征。对Twist2的早期研究多集中在骨骼发育,随后在多种肿瘤中发现其有表达差异,研究表明Twist2在肿瘤的上皮-间质转化(Epithelial-mesenchymal transition,EMT)中发挥着重要作用。Twist2参与了多条通路的调控,其调控作用的发挥受到时空表达、磷酸化、二聚化和细胞定位的调节,在机体的正常发育、体内平衡和疾病发生机制中研究Twist2的作用显得尤为重要。文章对Twist2在成骨分化、肿瘤形成和EMT中的作用及其分子机制进行综述,以便帮助了解Twist2的生物学功能,为进一步在疾病的诊断、发展、以及治疗等方面的转化应用研究提供依据。  相似文献

A sulfite oxidase (SO(X)) (EC 1.8.3.1) purified from Syzygium cumini leaves was immobilized onto carboxylated gold coated magnetic nanoparticles (Fe(3)O(4)@GNPs) electrodeposited onto the surface of a gold (Au) electrode through N-ethyl-N'-(3-dimethylaminopropyl) carbodiimide (EDC)-N-hydroxy succinimide (NHS) chemistry. An amperometric sulfite biosensor was fabricated using SO(X)/Fe(3)O(4)@GNPs/Au electrode as working electrode, Ag/AgCl as standard and Pt wire as auxiliary electrode. The working electrode was characterized by Fourier Transform Infrared (FTIR) Spectroscopy, Cyclic Voltammetry (CV), Scanning Electron Microscopy (SEM) and Electrochemical Impedance Spectroscopy (EIS) before and after immobilization of SO(X). The biosensor showed optimum response within 2s when operated at 0.2V (vs. Ag/AgCl) in 0.1 M Tris-HCl buffer, pH 8.5 and at 35 °C. Linear range and detection limit were 0.50-1000 μM and 0.15 μM (S/N=3) respectively. Biosensor was evaluated with 96.46% recovery of added sulfite in red wine and 1.7% and 3.3% within and between batch coefficients of variation respectively. Biosensor measured sulfite level in red and white wines. There was good correlation (r=0.99) between red wines sulfite value by standard DTNB (5,5'-dithio-bis-(2-nitrobenzoic acid)) method and the present method. Enzyme electrode was used 300 times over a period of 4 months, when stored at 4 °C. Biosensor has advantages over earlier biosensors that it has excellent electrocatalysis towards sulfite, lower detection limit, higher storage stability and no interference by ascorbate, cysteine, fructose and ethanol.  相似文献

A pot experiment with acid yellow–brown soil was conducted to investigate the interactive effects of molybdenum (Mo) and phosphorus (P) fertilizers on the photosynthetic characteristics of seedlings and grain yield of Brassica napus which is sensitive to soil P and Mo deficiency. Both Mo and P fertilizers were applied at three levels (0 mg Mo kg?1, 0.15 mg Mo kg?1, 0.30 mg Mo kg?1 soil; 0 mg P kg?1, 80 mg P kg?1, 160 mg P kg?1 soil). The results showed that P fertilizer application increased grain yield, soluble sugar concentrations of seedling leaves, DM and P accumulation of seedling shoots of Brassica napus in the absence or presence of Mo fertilizer. In contrast, Mo fertilizer increased these parameters only in the presence of P fertilizer. Mo accumulation in shoots, chlorophyll concentrations and net photosynthesis rate (P n) of seedling leaves were increased by both Mo and P fertilizers, particularly with the combination of the two fertilizers. The results also showed that the Mo and P fertilizers increased photosynthetic rate through two different mechanisms, with Mo increasing photosynthetic activity of mesophyll cells, and P increasing stomatal conductance. The results demonstrate that there was a synergetic effect on photosynthesis and grain yield between Mo and P fertilizers and it is conducive for Brassica napus growth to co-apply the two fertilizers.  相似文献

A highly sensitive homogeneous electrogenerated chemiluminescence (ECL) immunoassay for the determination of anti-digoxin antibody and digoxin hapten was developed employing Ru(bpy)(2)(dcbpy)NHS (bpy = 2,2'-bipyridyl; dcbpy = 2,2'-bipyridine-4,4'-dicarboxylic acid; NHS = N-hydroxysuccinimide ester) as an electrochemiluminescent label and bovine serum albumin (BSA) as a carrier protein. A digoxin hapten was indirectly heavily labelled with Ru(bpy)(2)(dcbpy)NHS through BSA to form Ru(bpy)(2)(dcbpy)NHS-BSA-digoxin conjugate. The ECL intensity of the immunocomplex of the conjugate with anti-digoxin antibody markedly decreased when the immunoreaction between Ru(bpy)(2)(dcbpy)NHS-BSA-digoxin conjugate and anti-digoxin antibody took place. Two formats, direct homogeneous immunoassay for anti-digoxin antibody and competitive immunoassay for digoxin, were developed to determine anti-digoxin antibody and digoxin, respectively. The anti-digoxin antibody concentration in the range 7.6 x 10(-8)-7.6 x 10(-6) g/mL was determined by direct homogeneous format. Digoxin hapten was determined throughout the range 4.0 x 10(-10)-1.0 x 10(-7) g/mL with a detection limit of 1.0 x 10(-10) g/mL by competitive format. The relative standard derivation for 6.0 x 10(-9) g/mL was 4.3%. The method has been applied to assaying digoxin in control human serum.  相似文献
