The objective of this study was to analyze human fecal Lactobacillus community and its relationship with rheumatoid arthritis. Samples taken from rheumatoid arthritis (RA) patients and healthy individuals were analyzed by quantitative real-time PCR. Bacterial DNA was extracted from feces, and amplicons of the Lactobacillus-specific regions of 16S rRNA were analyzed by denaturing gradient gel electrophoresis. The richness, Shannon-Wiener index, and evenness of gut microbiota of both groups were analyzed to compare fecal Lactobacillus community structures. Results of this study demonstrated that fecal microbiota of RA patients contained significantly more Lactobacillus (10.62 ± 1.72 copies/g) than the control group (8.93 ± 1.60 copies/g). Significant increases were observed in RA patients in terms of the richness, Shannon-Wiener, and evenness measures, indicating more bacterial species, and increased bacterial diversity and abundance. These results suggest a potential relationship between Lactobacillus communities and the development and progression of rheumatoid arthritis.  相似文献

为探明长期施肥与土壤动物群落之间的关系,于2001年6月至2002年10月,在陕西黄土区对不同施肥条件下的农田土壤动物类群的群落组成和结构进行了研究.在6种不同施肥处理的小区内,即对照组(不施肥,简称CK)、撂荒(不施肥、不耕作、不种植,简称ABAND)、施氮磷钾(简称NPK)、施氮磷钾+秸秆(简称SNPK)、施氮磷钾+有机肥(简称MNPK)和施1.5倍MNPK(简称1.5MNPK),两年4次共采集了72个定点土壤样品.采用手捡法和Cobb过筛法共获得农田土壤动物标本5495个,隶属6门11纲22目61科2亚科35属.结果显示6种施肥处理中,大型农田土壤动物的个体总数从多到少依次为SNPK>1.5MNPK>NPK>ABAND>MNPK>CK,类群数依次是1.5MNPK>NPK>SNPK>CK>ABAND=MNPK.中小型农田土壤动物个体总数由多到少依次为1.5MNPK>MNPK>ABAND>SNPK>NPK>CK,类群数依次是SNPK=MNPK>CK=NPK=1.5MNPK>ABAND.大型农田土壤动物个体数和类群数分布最多的分别是SNPK和1.5MNPK处理,而中小型农田土壤动物则分别是1.5MNPK和SNPK处理.表明农田土壤动物类群分布与施肥处理有关.农田土壤动物优势类群分布以施氮磷钾(NPK)小区最多,常见类群以对照组(CK)最多,极稀有类群以施1.5倍MNPK小区最多.群落相似性指数分析结果表明,在不同施肥处理之间,农田土壤动物的相似性系数一般较低,而其群落组成的异质性较高如大型土壤动物群落在撂荒地与其他施肥处理之间的相似性明显低于其他各施肥处理之间;而中小型土壤动物群落在对照小区与其他施肥之间的相似性指数明显低于其他各施肥处理之间,反映出不同施肥处理对土壤生态系统内部环境,进而对土壤动物群落产生的影响.  相似文献

27-Hydroxycholesterol (27-HC) has been implicated in the pathological process of estrogen receptor positive breast cancer. However, the role of 27-HC in lung adenocarcinoma is still unclear. Because bone metastasis is a main reason for the high mortality of lung adenocarcinoma, this study aimed to investigate the effect of 27-HC on osteoclastogenesis in lung adenocarcinoma microenvironment. The results showed that the conditioned media (CM) from lung adenocarcinoma cells cocultured with macrophages promoted osteoclast differentiation, which was enhanced by 27-HC. Further investigation showed that CM inhibited miR-139 expression and promoted c-Fos expression. Luciferase reporter assay identified c-Fos as a direct target of miR-139. CM also induced the expression and nuclear translocation of NFATc1 and STAT3 phosphorylation, which was enlarged by 27-HC but was attenuated by miR-139. Coimmunoprecipitation assay demonstrated that 27-HC increased the interaction between NFATc1 and phosphorylated STAT3, which was restricted by miR-139. Chromatin immunoprecipitation assay showed that pSTAT3 could bind to the promoter of c-Fos, c-Fos could bind to the promoter of NFATc1, and both pSTAT3 and NFATc1 could bind to the promoter of Oscar, which were enlarged by 27-HC but were blocked by miR-139. Knockdown of c-Fos mimicked the effect of miR-139. These results suggested that CM, especially containing 27-HC, promoted osteoclastogenesis by inhibiting miR-139 expression and activating the STAT3/c-Fos/NFATc1 pathway.  相似文献

The Kunming (KM) mouse is a closed colony mouse strain widely used in Chinese pharmacology, toxicology, and microbiology research laboratories. However, few studies have examined human flora-associated (HFA) microbial communities in KM mice. In this study, HFA models were built from germ-free KM and C57BL/6J mouse strains, and gut microbial diversity was analyzed by denaturing gradient gel electrophoresis (DGGE) and DNA sequencing. We found that the two strains of HFA mice were significantly different based on the UPGMA dendrogram and the Richness index, but dice similarity coefficients of mouse replicates were not significantly different between HFA-KM and HFA-C57BL/6J. Most of the dominant phyla of human gut microflora could be transferred into the guts of the two mouse strains. However, the predominant genus that formed in HFA-KM was Clostridium sp. and that in HFA-C57BL/6J was Blautia sp. These results imply that genotypes difference between the two mice strains is a critical factor in shaping the intestinal microflora. However, genetic differences of individuals within KM mouse populations failed to lead to individual difference in microflora. Successful generation of HFA-KM mice will facilitate studies examining how diet affects gut microbial structure, and will enable comparative studies for uncovering genetic factors that shape gut microbial communities.  相似文献

模拟氮沉降下南方针叶林红壤的养分淋溶和酸化   总被引:8,自引:0,他引:8  
以中国科学院红壤生态实验站林草生态试验区针叶林红壤为研究对象,在恒温(20 ℃)条件下,通过大土柱(直径10 cm、高60 cm),8个月间隙性淋溶试验模拟研究了不同氮输入量(0、7.8、26和52 mg N/月/柱)对针叶林红壤NO3-、NH4+、H+和土壤盐基离子(Ca2+、Mg2+、K+、Na+)淋溶以及土壤酸化的影响.结果表明,土壤交换态盐基总量、Ca2+和Mg2+淋溶量随氮输入量的增加而增加,土壤交换态Na+和K+则无明显影响.4种N输入处理的土壤交换态盐基总量净淋溶(淋溶出的盐基与淋洗液累计输入的盐基之差)分别占土壤交换性盐基总量的13.9%、18.6%、31.8% 和57.9%,土壤交换态Ca2+净淋溶分别占土壤交换性Ca2+总量的19.6%、25.8%、45.3%和84.8%,土壤交换态Mg2+净淋溶分别占土壤交换性Mg2+总量的4.4%、6.1%、10.9%和17.1%.随氮输入量增加,表层土壤pH值逐渐下降,4种N输入处理的表层土壤pH(KCl)分别为3.85、3.84、3.80和3.75;随氮输入量增加,淋溶液中无机氮、NO3-和H+逐渐增加.氮沉降可促进针叶林红壤的有机氮矿化,加速养分淋失和土壤酸化.  相似文献

With japonica rice 98-08, indica hybrids Shanyou 63, Gangyou 881, and X07S/Zihui 100, and sub-species hybrid Peiai 64S/9311 as materials, chlorophyll (Chl) content, Chl a fluorescence parameters, and membrane lipid peroxidation in flag leaf were measured at late developmental stages under natural conditions. Fv/Fm, qP, PS2, and electron transport rate gradually decreased while qN increased conversely. Excessive photon energy led to the accumulation of active oxygen (O2 ), H2O, malonyldialdehyde, and products of membrane lipid peroxidation, and resulted in reduced Chl content and early ageing subsequent to the photooxidation during flag leaf senescence. There was obvious diversification of these parameters among rice cultivars. In comparison with japonica cv. 98-08 (tolerant to photooxidation), Fv/Fm decreased in indica cv. Shanyou 63 (susceptible to photooxidation) with greater accumulation of active oxygen and a sharp drop in Chl content, which resulted in yellowish early ageing, and affected the filling and setting of rice grains. The mechanism for premature ageing in indica rice was related to irradiance and temperature at filling stages. On a sunny day at above 25 °C, the reaction centre of photosystem 2 (PS2) exhibited a dynamic change on reversible inactivation. Under the intense irradiance at noon, PS2 function in indica rice exhibited obvious down-regulation and photoinhibition. Under intense irradiance with lowered temperatures, PS2 resulted in photo-damage and early ageing, related to the degradation of PS2-D1 protein and the inhibition of endogenous protection systems such as the xanthophyll cycle and enzymes scavenging active oxygen. Hence for high-yield breeding, based on a good plant-type and utilising heterosis and tolerance of photooxidation, the selection of japonica rice or a sterile line with the japonica genotype as female is a strategy worthy of consideration.  相似文献

Cilia of higher animals sense various environmental stimuli. Proper ciliary signaling requires appropriate extent of BBSome‐mediated export of membrane receptors across ciliary barrier transition zone (TZ) through retrograde intraflagellar transport (IFT) machinery. How the barrier passage is controlled, however, remains unknown. Here, we show that small GTPase Rabl2 functions as a molecular switch for the outward TZ passage. Rabl2‐GTP enters cilia by binding to IFT‐B complex. Its GTP hydrolysis enables the outward TZ passage of the BBSome and its cargos with retrograde IFT machinery, whereas its persistent association leads to their shedding from IFT‐B during the passing process and consequently ciliary retention. Rabl2 deficiency or expression of a GTP‐locked mutant impairs the ciliary hedgehog signaling without interfering with ciliation and respectively results in different spectrums of mouse developmental disorders. We propose that the switch role of Rabl2 ensures proper turnover of the BBSome and ciliary membrane receptors to fine‐tune cilia‐dependent signaling for normal embryonic development and organismic homeostasis.  相似文献

Theoretical studies of a new ion-pair receptor, meso-octamethylcalix[4]pyrrole (OMCP), and its interactions with the halide anions F(-), Cl(-), and Br(-) and the cesium halides CsF, CsCl, and CsBr have been performed. Geometries, binding energies, and binding enthalpies were evaluated with the restricted hybrid Becke three-parameter exchange functional (B3LYP) method using the 6-31+G(d) basis set and relativistic effective core potentials. The optimized geometric structures were used to perform natural bond orbital (NBO) analysis. The two typical types of hydrogen bonds, N-H…X(-) and C-H…X(-), were investigated. The results indicate that hydrogen bonding interactions are dominant, and that the halide anions (F(-), Cl(-), and Br(-)) offer lone pair electrons to the σ*(N-H) or σ*(C-H) antibonding orbitals of OMCP. In addition, electrostatic interactions between the lone pair electrons of the halide anion and the LP* orbitals of Cs(+) as well as cation-π interactions between the metal ion and π-orbitals of the pyrrole rings have important roles to play in the Cs(+)?OMCP?X(-) complexes. The current study further demonstrates that this easy-to-make OMCP host compound functions as not only an anion receptor but also an ion-pair receptor.  相似文献

In this study, we investigated the impact of neonatal amoxicillin treatment on the development of the murine intestinal Lactobacillus community. Suckling BALB/c mice received a daily intragastric gavage of amoxicillin or saline from postnatal day 7 (PND 7) to PND 20. Just after the treatment (PND 21) and 5 weeks later (PND 56), the colon digesta samples were analyzed by Lactobacillus-specific quantitative real-time PCR analysis and PCR-denaturing gradient gel electrophoresis (DGGE) technique. Real-time PCR results showed that the levels of lactobacilli in the treatment group were similar to those in the control at PND 56. However, in DGGE analysis the number of DGGE bands and Shannon index were decreased significantly in comparison with control (P < 0.05). The dominant Lactobacillus strain in the murine colon changed from L. johnsonii to L. murinus. These results demonstrated that neonatal amoxicillin treatment led to a significant impact on the biodiversity of the murine intestinal Lactobacillus community within a long-time period.  相似文献
