Gene flow from genetically modified (GM) crops to the same species or wild relatives is a major concern in risk assessment. Transgenic rice with insect and/or disease resistance, herbicide, salt and/or drought tolerance and improved quality has been successfully developed. However, data on rice gene flow from environmental risk assessment studies are currently insufficient for the large-scale commercialization of GM rice. We have provided data on the gene flow frequency at 17 distances between a GM japonica line containing the bar gene as a pollen donor and two indica hybrid rice varieties and four male-sterile (ms) lines. The GM line was planted in a 640m2 in an isolated experimental plot (2.4 ha), which simulates actual conditions of rice production with pollen competition. Results showed that: (1) under parallel plantation at the 0-m zone, the transgene flow frequency to the ms lines ranged from 3.145 to 36.116% and was significantly higher than that to hybrid rice cultivars (0.037–0.045%). (2) Gene flow frequency decreased as the distance increased, with a sharp cutoff point at about 1–2 m; (3) The maximum distance of transgene flow was 30–40 m to rice cultivars and 40–150 m to ms lines. We believe that these data will be useful for the risk assessment and management of transgenic rice lines, especially in Asia where 90% of world's rice is produced and hybrid rice varieties are extensively used. We have taken a mutational approach to identify genes important for male fertility in Arabidopsis thaliana and have isolated a number of nuclear male/ sterile mutants in which vegetative growth and female fertility are not altered. Here we describe detailed developmental analyses of four mutants, each of which defines a complementation group and has a distinct developmental end point. All four mutants represent premeiotic developmental lesions. In ms3, tapetum and middle layer hypertrophy result in the degeneration of microsporocytes. In ms4, microspore dyads persist for most of anther development as a result of impaired meiotic division. In ms5, degeneration occurs in all anther cells at an early stage of development. In ms15, both the tapetum and microsporocytes degenerate early in anther development. Each of these mutants had shorter filaments and a greater number of inflorescences than congenic male-fertile plants. The differences in the developmental phenotypes of these mutants, together with the non-allelic nature of the mutations indicate that four different genes important for pollen development, have been identified.  相似文献

1992年3月至11月,我们在海南和长沙两地,先后使用38种化学调控剂,对水稻两用不育系,三系不育系及普通核不育系进行了喷施和注射试验,筛选出7种较为有效的调控剂,代号为CR1—CR7,其中CR5和CR7表现较为突出。这些调控剂对两用不育系和三系不育系均表现出一定程度的恢复效应,且CR1和CR2还能在可育条件下提高培矮64S的结实率,但所有调控剂对普通核不育系均无恢复作用。  相似文献

油菜细胞质雄性不育系叶绿体DNA特异片段的分子克隆   总被引:7,自引:0,他引:7  
采用高离子浓度缓冲液法分别提取油菜不育系及保持系的叶绿体DNA。经Sepharcse 4B柱层析纯化后,得到具有较高纯度的叶绿体DNA样品。将其分别用Eco RI、Bam HI、HimdHI、PstI和XhoI 5种限制性内切酶酶解,得到5种限制性内切酶图谱。其中除PstI图谱外,其它4种酶谱均显示出明显的两系间叶绿体DNA结构差异。以pBR 322为载体,分别克隆了不育系Bam HI图谱上的3个特异片段。得到的3个克隆,经克隆杂交及电泳分析后,证实分别带有上述3个目的片段。这些特异片段的特性及其与花粉育性的关系尚在研究中。  相似文献

Summary In order to identify factors necessary for the establishment of the reticulate pollen wall pattern, we have characterized a T-DNA tagged mutant ofArabidopsis thaliana that is defective in pattern formation. This study reports the results of an ultrastructural comparison of pollen wall formation in the mutant to wall development in wild-type plants. Pollen wall development in the mutant parallels that of wild-type until the early tetrad stage. At this point in wild-type plants, the microspore plasma membrane assumes a regular pattern of ridges and valleys. Initial sporopollenin deposition occurs on the ridges marking the beginning of probacula formation. In contrast, the plasma membrane in the mutant appears irregular with flattened protuberances and rare invaginations. As a result, the wild-type regular pattern of ridges and valleys is not formed. Sporopollenin is randomly deposited on the plasma membrane and aggregates on the locule wall; it is not anchored to the membrane. Our finding that the mutation blocks the normal invagination of the plasma membrane and disrupts the proper deposition of sporopollenin during wall formation suggests that the mutation could be in a gene responsible for pattern formation. These results also provide direct evidence that the plasma membrane plays a critical role in the establishment of the pollen wall pattern.  相似文献

Summary Restriction endonuclease patterns of chloroplast DNA (cpDNA) were consistently distinguishable between fertile and male-sterile cytoplasms of sorghum [Sorghum bicolor (L.) Moench], whereas no differences in restriction patterns of cpDNA among male-sterile (A1) lines, including six isocytoplasmic strains, were revealed in this study. It is suggested that chloroplast DNA may contribute to the male sterility of A1 lines used currently in hybrid sorghum production.This research was supported by a research grant from Kansas Grain Sorghum Commission, Kansas Board of Agriculture. 采用焦锑酸钾沉淀法研究了温敏雄性不育水稻(Oryza sativa L.)培矮64S在高温引起雄性不育与正常可育花药发育过种中Ca2+的分布变化.结果表明,当培矮64S生长在较高温度条件下引起雄性不育,与可育花药相比,不育花粉母细胞中有较多的液泡、较多的Ca2+沉积和较少的线粒体,并且有较多的Ca2+沉积在不育花药的中间层、表皮层和绒毡层中.到四分体与单细胞花粉时期,不育花药的木质部细胞的次生加厚壁上有较多的Ca2+沉淀,连接组织中的Ca2+沉淀也大大增加,所有不育花粉外壁较厚而发育都不正常.在单核细胞早期,不育花粉的四分体细胞中有较明显的大液泡出现.不育花药中的Ca2+在花药发育的各时期均比可育花药要多.这些结果说明在高温生长条件下,花粉母细胞发育的异常、花药中Ca2+沉积的增加、绒毡层与花粉外壁发育的异常可能与培矮64S花粉败育相关.  相似文献

Summary Restriction fragment length polymorphisms (RFLPs) were used to assess chloroplast DNA (cpDNA) variation in a population of soybeans subjected to continuous cycles of forced outcrossing. This population was derived by crossing 39 female lines with four male-sterile (Ms2ms2) maintainer lines and advancing each generation by selecting only outcrossed seed borne on male-sterile (ms2ms2) plants. Analysis of the original 39 female lines revealed three groups based on cpDNA RFLPs. These three groups had been previously documented in soybeans, and the distribution of these groups among the female parents of this population was similar to that observed in germ plasm surveys of soybean. Thirty-four of the female parents had group I cpDNA, 3 had group II, and 2 had group III. Plants collected from this population after seven cycles of outcrossing were scored for four morphological traits (flower color, pubescence color, seed color, and pubescence type) known to be controlled by alleles at single nuclear loci. The frequencies of the phenotypes observed in this study indicated that the population underwent random mating with respect to flower and pubescence color, but deviated from random mating at the other two loci. Analysis of 158 of these same plants collected from the population after seven cycles of outcrossing revealed no individuals with group II or group III cpDNAs. The fixation of the group I cpDNA marker in this outcrossing population was judged to result primarily from selection against individuals in the population with the rare cpDNAs.Published as Paper no. 9640, Journal Series, Nebraska Agric Res Div Project no. 12-187. Seed germination in a male-sterile 7B-1 mutant in tomato is reletively more resistant to the inhibitory effects of a high osmoticum induced by mannitol and polyethylene glycol, to various salts, including NaCl, Na2SO4, KCl and K2SO4, and to low-temperature stress, compared to the wild-type (WT) seeds. The inhibitory effects of various stresses could be partly or completely overcome by fluridone (FLU), an inhibitor of abscisic acid (ABA) biosynthesis. However, lower concentration of fluridone was required for the 7B-1 mutant than for WT seeds, and the mutant seeds were more sensitive to the inhibitory effects of exogenous ABA. The data suggest that 7B-1 seed has a pre-existing level of elevated ABA which imparts resistance to the various stresses. The ability to regulate male sterility in the 7B-1 mutant by photoperiod, as previously reported by Sawhney (1997), and its resistance to abiotic stresses, as reported here, makes this a useful system for tomato breeding and in hybrid programs.
