Chemical sunscreens were incorporated into oil formulations of conidia of two isolates of the entomopathogenic fungusMetarhizium spp. and the formulations exposed to simulated solar radiation. After 2 h exposure several sunscreens gave protection as demonstrated by conidial germination after 24 h incubation on gelatin plates at 26°C but only the formulations with Eusolex 8021 showed higher germination than the unprotected control after 48 h incubation. During 5h of exposure, Eusolex 8021 failed to offer significant protection as demonstrated by conidial germination after 48 h incubation. Conidial damage was proportional to the duration of radiation received. Allowing periods of darkness between exposures did not result in decreased loss of viability. Storing conidia, after exposure to simulated radiation, for 24 h prior to germination reduced their viability.  相似文献

Our results demonstrate high concentrations of the UV absorbing mycotoxin citrinin in the outer layer of spores from three citrinin-producing strains of Penicillium verrucosum, which is released in an aqueous environment. An important function of the toxin could be to act as a sun protectant in order to create favorable conditions during the initial germination process. When spores from these strains of P. verrucosum were examined by confocal microscopy, a clearly visible fluorescent layer associated with the cell wall was observed. The strains were grown on agar plates, and the mycelial mat was washed with saline. This suspension contained at least 95% of the spores and particulate material, which was removed by filtration after counting the conidia. An aliquot of this filtrate was extracted and citrinin was purified by high pressure liquid chromatography. The absorbance at 319 nm was used to calculate the amount of UV absorbing material released from the spores. Based on the spore numbers in the suspension of the saline extract, we estimated that this material released was 1.4–4.1 pg per spore or 8–24% of the spore weight. Citrinin (and minor amounts of ochratoxin A and some other unidentificable fluorescent compounds) were observed in the filtrate when subjected to thin layer chromatography. 食用真菌草菇[Volvariella volvacea(Bull.ex.Fr.)Sing.]VvLaeA在调控真菌发育方面的功能并不清楚。本研究在对草菇VvLaeA进行生物信息学分析基础上,通过聚合酶链式反应(polymerase chain reaction,PCR)的方法将超量表达启动子Vvgpd和草菇VvlaeA基因的开放阅读框(open reading frame,ORF)进行融合,并将融合片段克隆入pK2(bar)载体。采用农杆菌介导的转化方法将重组载体pK2(bar)-OEVvlaeA转入球孢白僵菌中进行表达,测定转化子的菌株发育情况。结果表明,草菇VvLaeA与其他真菌的蛋白同源性较低;和野生型相比,转化菌株的菌落直径显著增大,色素减少,分生孢子产量和萌发率均显著下降,对胁迫反应的敏感性增强。进一步研究发现,转化菌株分生孢子的细胞壁结构成分发生改变,和分生孢子发育相关基因的表达被显著下调。提示草菇VvLaeA能提高球孢白僵菌菌株的生长速率,而负调控色素沉积和分生孢子发育,这为草菇基因功能鉴定提供了启示。  相似文献

A key was produced to describe 10 stages of development of linseed buds, flowers and capsules. Botrytis cinerea conidia germinated more rapidly and germ tubes grew longer on linseed stigmas, petals and mature senescing capsules than on green leaves, sepals and immature capsules. The proportion of conidia which germinated increased and the germ tubes continued growing for longer in the presence of linseed pollen and flower petal extracts. In controlled environment and field experiments, the response of buds, flowers and capsules to inoculation with B. cinerea changed with stage of development; few pre‐flowering buds developed symptoms (brown lesions, then grey mould), but high proportions of flowering and post‐flowering buds did so. Few immature green capsules developed symptoms and the proportion of capsules which developed symptoms increased as they matured. The presence of linseed pollen decreased the incubation period from inoculation with spore suspensions to appearance of B. cinerea symptoms on buds. A disease cycle was produced to suggest the changes in susceptibility of linseed to infection by B. cinerea conidia during bud, flower and capsule development.  相似文献

Aims: This study aimed at modelling the effect of ethanol vapours, in the range 0·7–7·5 kPa, on the inactivation of dry‐harvested conidia of Penicillium chrysogenum, Penicillium digitatum and Penicillium italicum. Methods and Results: Survival curves were modelled by a Weibull model: log (N/N0) = ?1/2·303 (t/α)β. The shape parameter β was different from one in all cases, indicating that the classical first‐order kinetics approach is the exception rather than the rule. Survival curves exhibited upward concavity (β < 1) with the notable exception of P. chrysogenum at ethanol vapour pressures 0·7 and 1·5 kPa. The scale parameter α (h) varied greatly depending on the ethanol vapour pressure and on the species. Conclusions: For safety reasons, it is recommended not to exceed an ethanol vapour pressure of 3·3 kPa. At 2·8 kPa, more than 4 log10 reductions in viable conidia were achieved for all the species after 24‐h exposure. Significance and Impact of the Study: Ethanol has GRAS status in the USA and represents an interesting alternative to fungicides. The effectiveness of ethanol vapours to inactivate dry‐harvested conidia of some Penicillium was demonstrated in this study.  相似文献

Metarhizium anisopliae conidia were formulated with three granular carriers and nine dust diluents and stored over an 8- to 12-month period at 4° or 20°C. The virulence of formulations, with the exception of two dust preparations, was reduced significantly compared to unformulated conidia against Culex pipiens pipiens larvae. The formulation components most detrimental to conidial virulence were corn cob granules, diatomaceous earth, and two Kaolinite diluents. This was exampled by a decline in virulence from ca. 100% for unformulated conidia to 36% or below for these formulations. LT50 values also increased from 2.4–2.6 days for unformulated conidia to above 6 days. In contrast, a diluent derived from dried castor oil (Thixcin R) significantly enhanced conidial virulence at several doses above that of unformulated conidia against C. pipiens larvae. Enhancement occurred whether conidia were formulated prior to storage or stored separate from the diluent and mixed prior to application. The Thixcin R formulation was more effective against Anopheles stephensi larvae, but virulence was reduced against Aedes aegypti larvae. A bentonite formulation (Bentone-38) also maintained conidial virulence effectively, but Thixcin R was a superior diluent. It was shown that conidial virulence of formulations was not correlated with differences in conidial viability. The preparations that were applied dry by a surface method were more virulent than when an aqueous suspension containing a surfactant was used. The results demonstrate the need to assess efficacy of mycoinsecticidal formulations in a virulence bioassay prior to field testing.  相似文献

This study evaluates osmolality of a submerged conidia-producing medium in relation to the following spore characteristics: yield, morphology (dimensions and cell wall structure), chemical properties of cell wall surfaces (charge, hydrophobicity, and lectin binding), cytoplasmic polyols and trehalose, and performance (drying stability and pathogenicity). Spore production was increased by the addition of up to 150 g l?1 polyethylene glycol 200 (PEG). Spores from high osmolality medium (HOM spores) containing 100 g l?1 PEG had thin cell walls and dimensions more similar to blastospores than submerged conidia or aerial conidia. However, a faint electron-dense layer separating primary and secondary HOM spores’ cell walls was discernable by transmission electron microscopy as found in aerial and submerged conidia but not found in blastospores. HOM spores also appeared to have an outer rodlet layer, unlike blastospores, although it was thinner than those observed in submerged conidia. HOM spores’ surfaces possessed hydrophobic microsites, which was further evidence of the presence of a rodlet layer. In addition, HOM spores had concentrations of exposed N-acetyl-β-d-glucosaminyl residues intermediate between blastospores and submerged conidia potentially indicating a masking of underlying cell wall by a rodlet layer. All spore types had exposed α-d-mannosyl and/or α-d-glucosyl residues, but lacked oligosaccharides. Similar to blastospores, HOM spores were less anionic than submerged conida. Although HOM spores had thin cell walls, they were more stable to drying than blastospores and submerged conidia. Relative drying stability did not appear to be the result of differences in polyol or trehalose concentrations, since trehalose concentrations were lower in HOM spores than submerged conidia and polyol concentrations were similar between the two spore types. HOM spores had faster germination rates than submerged conidia, similar to blastospores, and they were more pathogenic to Schistocerca americana than submerged conidia and aerial conidia.  相似文献

Neozygites tanajoae is a very specialized fungus pathogenic to the cassava green mite (CGM), Mononychellus tanajoa, an important cassava pest introduced to Africa from the Neotropics. Conidial discharge from cadavers of CGM that died from infections with 14 isolates of N. tanajoae collected from diverse climates of Brazil was quantified to help select potential candidate strains for introduction to Africa. Studies aimed to identify isolates with lower requirements for relative humidity for sporulation and isolates that discharged more conidia during short periods of moisture. At 96 ± 0.5% RH, production of conidia was variable and even isolates from the Brazilian semi-arid region, e.g., Petrolina and Itaberaba, produced few conidia. Significant differences in the numbers of conidia produced by diverse Brazilian isolates were observed after 6, 9 and 12 h at 100% RH. At 100% RH, production of primary conidia increased considerably from an average of 57 ± 4 conidia at 6 h to 509 ± 37 conidia at 12 h. The isolate sporulating least (BIN21) discharged only 45.7% of the number of conidia produced by isolate BIN1, one of the isolates producing the most spores. Results from this study demonstrate that differences in production of conidia among isolates should be considered when selecting Neozygites isolates for new biological control introductions.  相似文献
