Conidia of Isaria fumosorosea were submitted to three regimes of temperature and moisture to simulate microclimatic conditions which prevail in temperate (43% RH and 28 °C to 98% RH and 15 °C), subtropical (75% RH and 35 °C to 98% RH and 25 °C), and arid areas (13% RH and 40 °C to 33% RH and 15 °C) with daily fluctuating cycles. Germination, conidial viability, and virulence to Spodoptera frugiperda larvae were less affected after 20 days exposure under temperate cycling conditions than under arid and subtropical conditions. Exposure of conidia for 20 days to constant nocturnal simulated conditions of any tested regime weakly affected conidial persistence, whereas diurnal conditions exerted the most detrimental effects of high temperatures. However, when tested at both 45 °C and 50 °C at 33% RH for 160 h, the persistence of I. fumosorosea conidia was relatively higher than expected. These results emphasize that climatic conditions prevailing in environments and ecological fitness of fungal isolates have to be taken into account for assessing microbial control strategies.  相似文献

Aims:  4',6-diamidino-2-phenylindole (DAPI) staining and fluorescent in-situ hybridization (FISH) show great potential for the detection of fungal conidia, also conserving the spatial architecture of their colonization. These investigations are often greatly hampered by the complicated wall structure of many fungal taxa. The aim of the present study was to develop an efficient permeabilization strategy for both DAPI staining and the FISH technique, applicable to various fungal species and maintaining their relationships with surfaces.
Methods and Results:  We compared different DAPI staining permeabilization strategies based on alcohol dehydration, surfactants and osmotic shock, tested with Aspergillus niger conidia. Among four permeabilization methods leading to a strong DAPI signal, only one, based on Triton X-100, EDTA and β-mercaptoethanol followed by hyperosmotic stress, appeared suitable for FISH investigation and was successfully applied to an additional 10 fungal taxa and three environmental samples.
Conclusions:  The effective permeabilization method, which employed a combination of surfactant and osmotic strategies, was successfully applied as preliminary step in both DAPI staining and the FISH protocol.
Significance and Impact of the Study:  The method described is reproducible, simple and inexpensive and might be attractive for other direct visualization techniques.  相似文献

蓖麻根腐病是茄腐镰孢菌(Fusarium solani)引起的根部病害,严重影响蓖麻产量。抗源缺乏制约了抗病品种的选育。为寻找抗病种质、建立抗性分子标记,该研究对252份蓖麻材料的抗性进行了表型和分子标记鉴定。结果表明:(1)浓度为1×106个·mL-1的孢子悬浮液灌根是一种有效的接种方法; 以接种后枯萎天数为基础的5级评价法,可作为鉴定标准。(2)鉴定出130份抗病材料,其中高抗为105份。(3)野生材料中抗病材料比率(66%)远高于栽培材料(35%),建议将野生材料,尤其是中国华南野生材料的研究利用作为今后抗病育种的重要方向。(4)初步建立了8个与抗性关联的SSR标记。该研究结果提供了有效的根腐病抗性鉴定方法和评价标准,筛选出了一批育种迫切需要的抗病基因资源,初步建立了可用于辅助选择的SSR标记,为蓖麻抗根腐病育种奠定了重要基础。  相似文献

The recent advances in biocontrol of stored-grain insects using formulated and unformulated strains of entomopathogenic fungi (EPF) are reviewed and discussed. Several liquid and dry formulations of EPF strains were developed and used against these insects. However, a literature search revealed lack of any commercially registered product of these formulations. Therefore, in order to achieve an effective control of these insects, the following potential areas of future research have been identified and discussed: (i) screening for new effective strains of EPF, in addition to new effective formulations, (ii) applying the most effective strains and formulations selected in the previous step under storage conditions, (iii) optimising the proportions of ingredients in the selected formulations and (iv) integrating the products of the most effective formulations, after registration and commercialisation, in the integrated pest management programmes of stored-grain insects. Such products would constitute an appropriate alternative control means to synthetic insecticides and fumigants commonly used for control of these insects. The various obstacles pertaining to how the selected EPF products are planned to be used in ‘real world’ in stored-grain protection are also critically discussed.  相似文献

AIMS: To find a supplemental ingredient that can be added to routinely used growth media to increase conidial production and decrease aflatoxin biosynthesis in small sclerotial (S strain) isolates of Aspergillus flavus. METHODS AND RESULTS: Molasses was added to three commonly used culture media: coconut agar (CAM), potato dextrose agar (PDA), and vegetable juice agar (V8) and production of conidia, sclerotia, and aflatoxins by A. flavus isolate CA43 was determined. The effect of nitrogen sources in molasses medium (MM) on production of conidia, sclerotia and aflatoxins was examined. Water activity and medium pH were also measured. Conidia harvested from agar plates were counted using a haemocytometer. Sclerotia were weighed after drying at 45 degrees C for 5 days. Aflatoxins B(1) and B(2) were quantified by high-performance liquid chromatography. Addition of molasses to the media did not change water activity or the pH significantly. Supplementing CAM and PDA with molasses increased conidial production and decreased aflatoxins. Two-fold increased yield of conidia was found on MM, which, like V8, did not support aflatoxin production. Adding ammonium to MM significantly increased the production of sclerotia and aflatoxins, but slightly decreased conidial production. Adding urea to MM significantly increased the production of conidia, sclerotia and aflatoxins. CONCLUSIONS: Molasses stimulated conidial production and inhibited aflatoxin production. Its effect on sclerotial production was medium-dependent. Water activity and medium pH were not related to changes in conidial, sclerotial or aflatoxin production. Medium containing molasses alone or molasses plus V8 juice were ideal for conidial production by S strain A. flavus. SIGNIFICANCE AND IMPACT OF THE STUDY: Insight into molecular events associated with the utilization of molasses may help to elucidate the mechanism(s) that decreases aflatoxin biosynthesis. Targeting genetic parameters in S strain A. flavus isolates may reduce aflatoxin contamination of crops by reducing the survival and toxigenicity of these strains.  相似文献

Aims: To determine the effect of water activity (aw = 0·880–0·960) and temperature (15–35°C) on the percentage of viable conidia and mycelial growth of three biocontrol agents effective against water hyacinth in Mali: Alternaria sp. isolate Mlb684, Fusarium sacchari isolate Mln799 and Cadophora malorum isolate Mln715. Methods and Results: The fungi were grown in vitro on plates containing potato dextrose agar medium at different aw values (glycerol being added to adjust the aw). The percentage of viable conidia and radial growth rate decreased with decreasing water activity. Statistical analysis showed a significant effect of aw, temperature and the aw × temperature interaction on mycelial growth (P < 0·0001). Water activity emerged as the factor exerting the greatest influence. Differences were observed between the fungi tested, the C. malorum appearing more tolerant to low aw and the F. sacchari more tolerant to high temperature (35°C). Growth models predicting the combined effect of aw and temperature were developed and response surfaces generated, showing fairly good agreement with the experimental values. Conclusions: Our results confirm the previous finding that aw has a greater influence than temperature on fungal growth. Under most conditions, variation of environmental factors has a detrimental influence on the percentage of viable conidia and mycelial growth rate of fungal isolates. Significance and Impact of the Study: The developed models may contribute to predicting the best environmental conditions for use of these fungi as effective biocontrol agents against water hyacinth.  相似文献

Immunocytochemical localization of stachylysin in Stachybotrys chartarum   总被引:1,自引:0,他引:1  
Stachylysin is a proteinaceous hemolytic agent that is produced by Stachybotrys chartarum. Stachylysin was found, using immunohistochemical and immunocytochemical methods, to be localized in S. chartarum spores/mycelia primarily in the inner wall suggesting that it is constitutively produced. Spores instilled in mouse or rat lung tissues resulted in granuloma formation, which showed the highest stachylysin concentration in the inner wall of the spore and near the spore, with less at distance indicating that it had diffused out from the spore. The in vitro high stachylysin producing strain (58-06) was also highest in vivo, based on immunohistochemistical staining. More stachylysin was observed in the mouse lung tissue at 72 h than at 24 h indicating that production/release is a relatively slow process. The localization of stachylysin in macrophage phagolysosomes suggests that these cells may be involved with hemolysin inactivation. This would be consistent with what is known about asp-hemolysin produced by Aspergillus fumigatus.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

The conidia produced by the mycelial form of Paracoccidioides brasiliensis were examined by scanning electron microscopy for the first time. Several different conidial types were characterized. These included intercalary arthroconidia, several types of septate conidia that are formed from other conidia, pedunculate conidia, and terminal hyphal conidia. In addition, the ultrastructure of the supporting pedestal of the pedunculate conidium was found to be separated from the mother conidium by a septum in some instances, and at other times it was not.  相似文献

Metarhizium flavouiride conidia formulated in oil or water were exposed to simulated solar radiation. Radiation below 320 nm killed conidia and caused delays in the germination of survivors; germination was greater after 48 h of incubation than after 24 h. UV exposure of conidia formulated in oil for 2 h reduced germination from 99% to 37.5% after incubation for 48 h. Exposure of conidia in water to UV for 1 h resulted in 4.7% germination after 24 h incubation compared with 36.5% for conidia formulated in oil. The addition of 1% oxybenzone resulted in 81.9% conidial germination after 3 h exposure and 48 h incubation compared with 28.1% in oil without the sunscreen.  相似文献
