Specific strains of fluorescent Pseudomonas spp. inhabit the environment surrounding plant roots and some even the root interior. Introducing such bacterial strains to plant roots can lead to increased plant growth, usually due to suppression of plant pathogenic microorganisms. We review the modes of action and traits of these beneficial Pseudomonas bacteria involved in disease suppression. The complex regulation of biological control traits in relation to the functioning in the root environment is discussed. Understanding the complexity of the interactions is instrumental in the exploitation of beneficial Pseudomonas spp. in controlling plant diseases.  相似文献

Fungi that occur inside asymptomatic plant tissues are known as fungal endophytes. Different genera of fungal entomopathogens have been reported as naturally occurring fungal endophytes, and it has been shown that it is possible to inoculate plants with fungal entomopathogens, making them endophytic. Their mode of action against insects appears to be due to antibiosis or feeding deterrence. Research aimed at understanding the fungal ecology of entomopathogenic fungi, and their role as fungal endophytes, could lead to a new paradigm on how to successfully use these organisms in biological control programs.  相似文献

Old trees are rare in the landscape, as are many of their associated species. Veteranisation is a method by which attempts are made to create microhabitats, otherwise found only in old trees, in younger trees at an earlier stage than would occur naturally. Here, we analysed the early fungal succession in 6 y-old veteranisation wounds in ca. 100 y old living oak trunks by DNA-barcoding of the wood at eight sites in Sweden and Norway. We hypothesised basidiomycetes would be most abundant, and exposed sapwood and heartwood would select for different communities. We identified 686 fungal taxa, mainly ascomycetes, with a large overlap in species composition and surprisingly similar species richness, i.e. 325 vs. 308–360, between intact and different types of damaged wood, respectively. Endophytes continued to be present and common in damaged wood. The results demonstrate that damage to sapwood and heartwood partly select for different fungi and that 6 y is too early to evaluate if veteranisation can positively favour fungi of conservation interest.  相似文献

The basidiomycete fungus Crinipellis perniciosa (Stahel) Singer is the causal agent of Witches' Broom Disease of Cacao (Theobromacacao L.) which is the main factor limiting cacao production in the Americas. Pod losses of up to 90% are experienced in affected areas as evidenced by the 50% drop in production in Bahia province, Brazil following the arrival of the C. perniciosa in the area in 1989. The disease has proven particularly difficult to control and many farmers in affected areas have given up cacao cultivation. In order to evaluate the potential of endophytes as a biological control agent of this phytopathogen, the endophytic fungal community of resistant and susceptible cacao plants as well as affected branches was studied between 2001 and 2002. The fungal community was identified by morphological traits and rDNA sequencing as belonging to the genera Acremonium, Blastomyces, Botryosphaeria, Cladosporium, Colletotrichum, Cordyceps, Diaporthe, Fusarium, Geotrichum, Gibberella, Gliocladium, Lasiodiplodia, Monilochoetes, Nectria, Pestalotiopsis, Phomopsis, Pleurotus, Pseudofusarium, Rhizopycnis, Syncephalastrum, Trichoderma, Verticillium and Xylaria. These fungi were evaluated both in vitro and in vivo by their ability to inhibit C. perniciosa. Among these, some were identified as potential antagonists, but only one fungus (Gliocladium catenulatum) reduced the incidence of Witches' Broom Disease in cacao seedlings to 70%.  相似文献

Various pink-pigmented facultative methylotrophic (PPFM) bacteria (strains iEII3, iEIV1, iEI6, iEII1, iEIII3 iEIII4, iEIII5, iRII1, iRII2, iRIII1, iRIV1 and iRIV2) were obtained from the rhizosphere and endosphere of hyperaccumulating plant Thlaspi goesingense grown in Redschlag, Austria [R. Idris, R. Trifonova, M. Puschenreiter, W.W. Wenzel, A. Sessitsch, Bacterial communities associated with flowering plants of the Ni hyperaccumulator Thlaspi goesingense, Appl. Environ. Microbiol. 70 (2004) 2667–2677]. Due to their unexpected diversity, abundance and nickel tolerance they were further characterized by detailed 16S rRNA gene analysis, DNA–DNA hybridization, fatty acid analysis, heavy metal tolerance, screening for known Ni resistance genes and phenotypic analysis. These strains were found to exhibit different multiple heavy metal resistance characteristics to Ni, Cd, Co, Zn and Cr. On the basis of their physiological and genotypic properties, strains could be grouped with Methylobacterium extorquens and M. mesophilicum. One endophyte, strain iEII3, was found to belong to a novel species for which the name M. goesingense is proposed.  相似文献

We investigated the endophytic fungal community associated with the Amazonian medicinal plant Carapa guianensis and its potential for providing bioactive compounds. A total of 162 endophytic fungal isolates were obtained and identified by molecular methods. These isolates were classified into 35 different taxa in the genera Aspergilllus, Beltrania, Botryosphaeria, Colletotrichum, Diaporthe, Endomelanconiopsis, Fusarium, Guignardia, Pestalotiopsis, Phomopsis, Pilidiella, Trichoderma, and Xylaria. The most frequent colonisers recovered of C. guianensis were Colletotrichum sp. 1, Diaporthe cf. mayteni, and Pestalotiopsis sp. 1. The fungal community had a moderate richness but high diversity and evenness indices. Colletotrichum sp. and Pilidiella wangiensis displayed selective antibacterial activity; Diaporthe cf. mayteni and Endomelanconiopsis endophytica showed high activity against amastigote forms of Trypanosoma cruzi; and Colletrotrichum sp. Guignardia mangiferae, Pestalotiopsis sp., and Diaporthe melonis were able to inhibit yellow fever virus proliferation. Our results suggest that the plants living in the tropical forest, such as the Amazonian hotspot region, can live in symbiosis with hidden and underestimated rich communities of endophytic fungi, which deserve protocols and/or specific laws to keep its future conservation. The capability of these endophytic fungi to produce bioactive compounds may be part of their chemical defense and adaptive response to survive and colonizing the plant host in wild environment. Consequently, these fungal communities may provide a source of bioactive molecules, including those able to inhibit or control neglected tropical diseases.  相似文献

Despite the importance of bryophyte-associated microorganisms in various ecological aspects including their crucial roles in the soil-enrichment of organic mass and N2 fixation, nonetheless, little is known about the microbial diversity of the bryophyte phyllospheres (epi-/endophytes). To get insights into bacterial community structures and their dynamics on the bryophyte habitats in different ecosystems and their potential biological roles, we utilized the 16S rRNA gene PCR-DGGE and subsequent phylogenetic analyses to investigate the bacterial community of eight bryophyte species collected from three distinct ecosystems from western Japan. Forty-two bacterial species belonging to γ-proteobacteria and Firmicutes with 71.4% and 28.6%, respectively, were identified among 90 DGGE gel band population. These DGGE-bands were assigned to 13 different genera with obvious predomination the genus Clostridium with 21.4% from the total bacterial community. These analyses provide new insights into bryophyte-associated bacteria and their relations to the ecosystems.  相似文献

  • Endophytic microbes isolated from plants growing in nutrient‐deficient environments often possess properties that improve nutrition of agriculturally important plants. A consortium of non‐rhizobial endophytic microbes isolated from a macrophyte Typha angustifolia growing in the marginal wetlands associated with a Uranium mine was characterized for their beneficial effect on rice and the mechanisms of growth promotion were investigated.
  • The microbes were identified and characterized for their potential plant growth promoting (PGP) properties. Effect of these microbes on nitrogen (N)‐metabolism of rice was tested as Typha endophytes were predominantly (N)‐fixing. Relative N‐use efficiency and expression of genes involved in N‐uptake and assimilation were investigated in treated plants.
  • Evidence of horizontal gene transfer (HGT) of dinitrogen reductase gene was observed within the consortium from a Pseudomonas stutzeri strain. The consortium behaved as plant probiotic and showed substantial growth benefits to Typha, their natural host as well as to rice. Typha endophytes colonized rice endosphere significantly increasing biomass, shoot length and chlorophyll content in rice plants both under N‐sufficient and N‐deficient conditions. N‐uptake and assimilation genes were upregulated in plants treated with the endophytes even after three weeks post infection.
  • Our results suggested, HGT of nitrogen‐fixation trait to be highly prevalent among endophytes isolated from nutrient‐poor habitats of the uranium mine. A long‐term nitrogen deficiency response in the treated plants was elicited by the consortium improving N‐uptake, assimilation and relative N‐use efficiency of rice plants. This appeared to be at least one of the main strategies of plant growth promotion.

Entophytic bacteria (EBs) are very diverse and found in virtually all plant species studied. These natural EBs live insides the host plant and can be used to maximize crop and fruit yield by exploiting their potential. In this paper, EBs characterization from various citrus genotypes and their influence on the morphological and physiological functioning of sour orange (Citrus aurantium) seedlings are described. To assess the influence of 10 distinct EBs, three different techniques (injection, soil mix, and spray) were applied for single and mixed inoculation on sour orange (C. aurantium) seedlings. The selected strains were identified as firmicutes (Enterococcus faecalis, Bacillus safensis, Bacillus cereus, Bacillus megaterium, Brevibacillus borstelensis & Staphylococcus haemolyticus), and gamma Proteobacteria (Enterobacter hormachaei, Proteus mirabilis, Pseudomonas aeruginosa, & Pseudomonas sp.) by 16S rRNA gene sequencing. To investigate the influence of these EBs on host plant morphology, different parameters (morphometric) were recorded after five WOI (weeks of inoculation), including shoot/root length, shoot/root fresh and dry biomass, and biophysical analyses i.e., relative water content (RLWC). Physiological markers such as chlorophyll & carotenoid content, protein content, proline content, phenolics, and flavonoids were also analyzed to determine the influence of endophytes on sour orange seedlings. Five strains such as SM-34, SM-20, SM-36, SM-68, and SM-56 significantly improved the development and physiology of sour orange seedlings. Bacillus cereus and Pseudomonas aeruginosa produced the best outcomes in terms of plant growth. The relative quantification of bacterial inoculums was determined using real-time PCR. A rise in the number of bacterial cells in inoculated treatment suggests that bacterial strains survived and colonized successfully, and also shown their competitiveness with native bacterial community structure. As per the results of inoculation methods, soil mixing, and injection methods were determined to be effective for bacterial inoculation to plants but a variable trend was found for different parameters with test bacterial strains. After testing their impact on field conditions, these strains can be applied as fertilizers as an alternative to conventional chemical fertilizer, although in the context of mixed inoculation of bacterial strains, 5 M and 6 M performed best and enhanced plant growth-promoting activity.  相似文献

A Xylaria sp. was isolated as an endophyte from a surface-sterilized Concord grape leaf (Vitis labrusca). Dereplication and LC–MS analysis of the filtrate extracts identified two new structures, compound 1, 13-O-methyl-(5R) diplosporin, and 2 a new agistatine. Additionally, five known compounds 37 including an agistatine, coriloxin, (5R)-diplosporin, and two mellein derivatives were also isolated. This is the first report of these structurally-related metabolites isolated from the same fungus, suggesting they share a related biosynthesis. Metabolites were isolated using liquid chromatography-solid phase extraction (LC-SPE) and semi-preparative HPLC. Structures were elucidated on the basis of 1D and 2D NMR analysis, HRMS measurements and X-ray structural analysis for compound 1.  相似文献
