Objective

A study was conducted to recover carbapenem-resistant bacteria from the faeces of dairy cattle and identify the underlying genetic mechanisms associated with reduced phenotypic susceptibility to carbapenems.

Methods

One hundred and fifty-nine faecal samples from dairy cattle were screened for carbapenem-resistant bacteria. Phenotypic screening was conducted on two media containing ertapenem. The isolates from the screening step were characterised via disk diffusion, Modified Hodge, and Carba NP assays. Carbapenem-resistant bacteria and carbapenemase-producing isolates were subjected to Gram staining and biochemical testing to include Gram-negative bacilli. Whole genome sequencing was performed on bacteria that exhibited either a carbapenemase-producing phenotype or were not susceptible to ertapenem and were presumptively Enterobacteriaceae.

Results

Of 323 isolates collected from the screening media, 28 were selected for WGS; 21 of which were based on a carbapenemase-producing phenotype and 7 were presumptively Enterobacteriaceae and not susceptible to ertapenem. Based on analysis of WGS data, isolates included: 3 Escherichia coli harbouring blaCMY-2 and truncated ompF genes; 8 Aeromonas harbouring blacphA-like genes; 1 Acinetobacter baumannii harbouring a novel blaOXA gene (blaOXA-497); and 6 Pseudomonas with conserved domains of various carbapenemase-producing genes.

Conclusions

Carbapenem resistant bacteria appear to be rare in cattle. Nonetheless, carbapenem-resistant bacteria were detected across various genera and were found to harbour a variety of mechanisms conferring reduced susceptibility. The development and dissemination of carbapenem-resistant bacteria in livestock would have grave implications for therapeutic treatment options in human medicine; thus, continued monitoring of carbapenem susceptibility among enteric bacteria of livestock is warranted.  相似文献

The identification of species and population boundaries is important in both evolutionary and conservation biology. In recent years, new population genetic and computational methods for estimating population parameters and testing hypotheses in a quantitative manner have emerged. Using a Bayesian framework and a quantitative model‐testing approach, we evaluated the species status and genetic connectedness of bottlenose dolphin (Tursiops spp.) populations off remote northwestern Australia, with a focus on pelagic ‘offshore’ dolphins subject to incidental capture in a trawl fishery. We analysed 71 dolphin samples from three sites beyond the 50 m depth contour (the inshore boundary of the fishery) and up to 170 km offshore, including incidentally caught and free‐ranging individuals associating with trawl vessels, and 273 dolphins sampled at 12 coastal sites inshore of the 50 m depth contour and within 10 km of the coast. Results from 19 nuclear microsatellite markers showed significant population structure between dolphins from within the fishery and coastal sites, but also among dolphins from coastal sites, identifying three coastal populations. Moreover, we found no current or historic gene flow into the offshore population in the region of the fishery, indicating a complete lack of recruitment from coastal sites. Mitochondrial DNA corroborated our findings of genetic isolation between dolphins from the offshore population and coastal sites. Most offshore individuals formed a monophyletic clade with common bottlenose dolphins (T. truncatus), while all 273 individuals sampled coastally formed a well‐supported clade of Indo‐Pacific bottlenose dolphins (T. aduncus). By including a quantitative modelling approach, our study explicitly took evolutionary processes into account for informing the conservation and management of protected species. As such, it may serve as a template for other, similarly inaccessible study populations.  相似文献

In addition to crustaceans, remains from 17 individual squid were found in the stomach of a 58 cm slender sunfish Ranzania laevis from Australia, adding a new prey item to their little studied diet. Taken together with existing data from the literature, crustaceans appear to be a common prey item, with larger R. laevis (26–65 cm) also taking small fish and squid. Along with new documentation on breaching, the unexpected finding of squid in the stomach confirms that these fish are fast and agile predators.  相似文献

During October to December 1997, we trawled estuarine habitats in the Joseph Bonaparte Gulf (JBG) to determine the distribution of juvenile red-legged banana prawns, Penaeus indicus (H. Milne Edwards, 1837) and white banana prawns, Penaeus merguiensis (de Man, 1888). We made 229 beam-trawls at 185 sites, mostly over a 100-m path (3-min duration). A Global Positioning System (GPS) receiver was used to verify our location. During October to December 1998, we intensively resampled three of the rivers that were sampled in 1997 to confirm the gulf-wide distribution of P. indicus and P. merguiensis and to investigate the microhabitat use of P. indicus. We chose previously sampled and new sites in Forsyth Creek (eastern JBG), the Lyne River (Cambridge Gulf), and the Berkeley River (western JBG). We made 249 trawls at 21 sites, mostly over 100 m.Juvenile banana prawns were abundant in eastern JBG, Cambridge Gulf and western JBG. They were not abundant in southern JBG, although fewer trawls were made there, due to its inaccessibility. In eastern JBG and Cambridge Gulf, over 96% and 73% (respectively) of juvenile banana prawns were P. indicus and they were more abundant there than in the western JBG. Conversely, in the western JBG over 93% of the juvenile banana prawns were P. merguiensis and they were more abundant than in the eastern JBG and Cambridge Gulf. The Lyne River in the northwestern Cambridge Gulf seems to be the transition zone; both P. indicus and P. merguiensis are equally abundant.P. indicus are most abundant on the mangrove-lined muddy banks of waterways within mangrove forests, similar habitats to P. merguiensis. Within these habitats, they were most abundant in gutters and small creeks, rather than rivers and large creeks. Few P. indicus or P. merguiensis were caught in 100 m2 trawls undertaken midriver (on the channel bottom and on emergent banks), although these habitats may be only 100 m from the mangrove-lined habitats. In all creek and river habitats, both species are most catchable at low tide (irrespective of daylight or darkness) when they move out of the mangrove forests and accumulate in the remnant water bodies.The offshore fishery for P. indicus is in northwestern JBG in waters 50-80 m deep, about 300 and 200 km, respectively, from where juveniles are abundant in their extensive inshore habitats in east JBG and in Cambridge Gulf, demonstrating a geographical separation of the juvenile and adult phases. Postlarval P. indicus, spawned offshore, must use tides and currents to travel south and east to reach nursery habitats. Emigrant subadults must migrate north and west, across relatively shallow inshore sand substrates (30-40 m deep) to reach their offshore habitats.  相似文献

Beach seines, gill nets and otter trawls were used during 1979–81 to sample extensively the fish fauna of the Peel-Harvey estuarine system. Approximately 145,000 individuals were caught, representing 29 familes and 55 species. The Clupeidae, Teraponidae, Mugilidae, Apogonidae, Atherinidae and Gerreidae were the dominant families, each contributing at least 8.9% to the total catch and together contributing 86.2%. Seasonal catch data and length-frequency distributions were used to help clarify the way in which the estuary was utilised by fish. Nine of the 15 most abundant fish species were marine species which entered the estuary for variable periods while the other six were represented by populations in which the individuals were capable of passing through the whole of their life cycle within the estuary. In order of abundance, the nine marine species were Hyperlophus vittatus, Pelates sexlineatus, Aldrichetta forsteri, Mugil cephalus, Torquigener pleurogramma, Favonigobius lateralis, Pranesus ogilbyi, Gymnapistes marmoratus and Sillago schomburgkii , while the six species with estuarine populations were Apogon rueppellii, Atherinosoma elongata, Nematalosa vlaminghi. Atherinosoma wallacei, Cnidoglanis macrocephalus and Amniataba caudavittatus. The extent and seasonality of the distribution of the species within the estuary and associated river systems varied considerably. Evidence is also presented to show that fish tend to move further away from the shallow banks near the shore with increasing age and size. The fish fauna of the Peel-Harvey is compared with those of Cockburn Sound and the estuaries of the Swan-Avon and Blackwood rivers in Western Australia and with Botany Bay in eastern Australia.  相似文献
