The collaborative trials described by Arnold et al., 1984a, b, c provided an extensive range of data on the performance of electrostatically charged rotary atomizers on a variety of crops, at different growth stages, using systemic and non-systemic pesticides. SYNOPSIS. Gametocytes differentiated from ring-stage parasites in microcultures of human blood infected with Plasmodium falciparum. Immature gametocytes could be distinguished morphologically from late asexual trophozoites after ~ 40 h of culture. Differentiation into crescentic forms took several days and the incorporation of [3H]-isoleucine by developing gametocytes was demonstrated. About 1% of red cells contained gametocytes at the maximum densities attained. Differentiation of gametocytes occurred either directly from rings placed in culture or from the progeny of subsequent cycles of schizogony and invasion in vitro. The latter occurrence was confirmed by the development of gametocytes in marker fetal red cells added to cultures, although fetal red cells provide a less favorable environment than those with HbA for growth of the parasites.  相似文献

High levels of DNA polymorphism were detected in 27 Solanum tuberosum cultivars examined. Combinations of at least two c-DNA clones have been identified which in conjunction with EcoRI allow important UK potato cultivars to be characterised by their molecular profiles. The widely grown North American cultivar Russett Burbank was also successfully fingerprinted. Estimates of genetic diversity based on restriction fragment length polymorphism (RFLP) data indicate the important role that wild potato species and exotic germplasm have played in the development of the cultivars studied. A graphical method for simultaneously highlighting similarities and differences between genotypes for individual hybridising fragments is presented. This approach is particularly useful in identifying and recording fragments which are unique to certain genotypes. Two potato cultivars: Fiona and Morag produce unique RFLP profiles when digested with EcoRI and EcoRV and probed with a flax ribosomal DNA sequence. Both Fiona and Morag possess incomplete or partial (quantitative) type resistance to G. pallida which was transferred from S. vernei. The preferential transmission of the r-DN A fragments from S. vernei may indicate that this locus is associated with genetic factors controlling resistance to G. pallida.  相似文献

Non-resistant but tolerant cv. Cara and non-resistant but relatively intolerant cv. Pentland Dell were grown in split plots encompassing a range of population densities of potato cyst nematode, Globodera pallida. Light interception and its efficiency of conversion were estimated by regular ground cover measurements and plant harvests. It was concluded that increasing levels of infestation with G. pallida only slightly decreased the efficiency of utilisation of intercepted radiation. Heavy infestation of G. pallida initially decreased the top growth and light interception of both cultivars by similar proportions, but in later harvests, this adverse effect markedly decreased for Cara whereas it slightly increased for Pentland Dell. This difference was due to the heavily infested Cara eventually achieving and maintaining 100% ground cover whereas the equivalent Pentland Dell never exceeded 75% ground cover. Consequently, final tuber yields were decreased much more for Pentland Dell than for Cara though the decreases in tuber yield were less than those for top growth. The importance of nematode effects on top growth, and hence on light interception, with regard to both yield losses and tolerance differences, were clearly demonstrated. Both linear and logarithmic models were used to describe the relationship between the initial population density of G. pallida and yield, and the implications of differences in tolerance on the parameters in the logarithmic model are discussed.  相似文献

Virus particles, antigenically related to mouse sarcoma virus, can be induced from hamster tumour cells. They are “defective” in their failure to replicate in or transform hamster, mouse or rat cells, although they contain viral-specific RNA and, with “helper” mouse leukaemia virus, show sarcomagenic activity.  相似文献

The migrations and winter distributions of most seabirds, particularly small pelagic species, remain poorly understood despite their potential as indicators of marine ecosystem health. Here we report the use of miniature archival light loggers (geolocators) to track the annual migration of Sabine’s Gull Larus sabini, a small (c. 200 g) Arctic‐breeding larid. We describe their migratory routes and identify previously unknown staging sites in the Atlantic Ocean, as well as their main Atlantic wintering area in the southern hemisphere. Sabine’s Gulls breeding in northeast Greenland displayed an average annual migration of almost 32 000 km (n = 6), with the longest return journey spanning close to 39 000 km (not including local movements at staging sites or within the wintering area). On their southern migration, they spent an average of 45 days in the Bay of Biscay and Iberian Sea, off the coasts of France, Spain and Portugal. They all wintered in close association with the cold waters of the Benguela Upwelling, spending an average of 152 days in that area. On their return north, Sabine’s Gulls staged off the west African coast (Morocco, Mauritania, Senegal), spending on average 19 days at this site. This leg of migration was particularly rapid, birds travelling an average of 813 km/day, assisted by the prevailing winds. Sabine’s Gulls generally followed a similar path on their outbound and return migrations, and did not exhibit the broad figure‐of‐eight pattern (anti clockwise in the southern hemisphere and clockwise in the northern hemisphere) seen in other trans‐equatorial seabirds in the Atlantic and Pacific oceans.  相似文献

A new restriction site was engineered in the cloned gene codingfor the large subunit polypeptide of ribulose 1, 5-bisphosphatecarboxylase (Rubisco) of the cyanobacterium Anacystis nidulans.This change resulted in the mutation of a phenylalanine residueto an isoleucine residue in the encoded polypeptide but hadno effect on the assembly or biochemical properties of Rubiscocontaining the polypeptide. The mutation was in a loop regionlinking highly structured domains at the N and C termini ofthe complete large subunit. Using the new restriction site, and a corresponding EcoRl restrictionsite in the cloned gene for the native large subunit polypeptideof wheat Rubisco, chimaeric genes were made encoding the polypeptidewith either the 140 residues of the N-terminal part of the wheatlarge subunit fused to the 336 residues forming the C-terminalregion of the A. nidulans large subunit, or the alternativeof 136 residues comprising of the N-terminal chains of A. nidulanssubunit and the 338 residue chain at the C-terminus of the wheatlarge subunit polypeptide. The chimaeric proteins expressedin E. coli, together with the small subunit of the A. nidulansRubisco, formed an insoluble inactive aggregate mainly in inclusionbodies.
