A stairway that allows the collection of kinetic data is essential for biomechanical studies on stair climbing. There is a need to validate the measured center of pressure (COP) on the surface of a stair in order to verify the accuracy of the calculation of joint kinetics. The purpose of this study was to validate a new stairway design for kinetic analysis of stair climbing through a calibration and error analysis of the COP obtained from this system. The new stairway design allows the collection of kinetic data for multiple steps without any constraint to foot placement. Known vertical forces were applied to known locations on the surface of each stair and each force plate. Multiple regression analyses were conduced to determine the distribution pattern of the error in the measured COP. It was found that the error in the COP was a function of location on the stair or force plate. The magnitude of the vertical force had no significant effect on the error in the measured COP. The distribution pattern of the error in the measured COP on the force plates used in this study matched the results in the literature. A healthy female subject was used as the subject in a stair climbing test. The error in the measured COP had a significant effect on the calculated joint resultant moments, especially the abduction-adduction and internal-external rotation moments. The correction of these errors should make the kinetic calculation in stair climbing more accurate.  相似文献

Fragmentation is a degradation pathway ubiquitously observed in proteins despite the remarkable stability of peptide bond; proteins differ only by how much and where cleavage occurs. The goal of this review is to summarize reports regarding the non-enzymatic fragmentation of the peptide backbone of monoclonal antibodies (mAbs). The sites in the polypeptide chain susceptible to fragmentation are determined by a multitude of factors. Insights are provided on the intimate chemical mechanisms that can make some bonds prone to cleavage due to the presence of specific side-chains. In addition to primary structure, the secondary, tertiary and quaternary structures have a significant impact in modulating the distribution of cleavage sites by altering local flexibility, accessibility to solvent or bringing in close proximity side chains that are remote in sequence. This review focuses on cleavage sites observed in the constant regions of mAbs, with special emphasis on hinge fragmentation. The mechanisms responsible for backbone cleavage are strongly dependent on pH and can be catalyzed by metals or radicals. The distribution of cleavage sites are different under acidic compared to basic conditions, with fragmentation rates exhibiting a minimum in the pH range 5 to 6; therefore, the overall fragmentation pattern observed for a mAb is a complex result of structural and solvent conditions. A critical review of the techniques used to monitor fragmentation is also presented; usually a compromise has to be made between a highly sensitive method with good fragment separation and the capability to identify the cleavage site. The effect of fragmentation on the function of a mAb must be evaluated on a case-by-case basis depending on whether cleavage sites are observed in the variable or constant regions, and on the mechanism of action of the molecule.  相似文献

The impact of war on the population is vast, especially when it comes to those who were directly affected by war, among other things as concentration camp detainees. Because of the specific war experience of this population it is important to better understand the possible contribution of key socio-demographic variables, war traumatization and acute disturbances in mental health to their subjective assessment of their own well-being, which represents a psychological category and is based on a subjective assessment. The starting point is a theoretical precept according to which individual characteristics, together with war experience, can have repercussions on mental health, and eventually on the general well-being of an individual and their quality of life. The study comprised 184participants who had given their informed consent for participation and filled out complete questionnaires. The participants were a convenience sample of male persons who had survived war captivity in the Homeland War in the period from 1991 to 1995. The study was conducted as part of the physical examinations at the University Hospital "Fran Mihaljevi?" in Zagreb. The data was collected using several self-evaluation measuring instruments one of which served to collect socio-demographic data, two to collect data on the participants' mental health, one for the data on the participants' combat and war experiences and one to assess the participants' well-being. The data obtained suggest that only avoidance and arousal symptoms and psychosomatic difficulties are predictors of the well-being of persons who have experienced war captivity.  相似文献

The action of a low-intensity 632.8 nm-wavelength laser radiation on blood elements was studied. The degree of binding the fluorescence probes 1.8 ANS by blood proteins and cells was taken to be the index of the response to irradiation. The change of fluorescence of the oppositely charged probes indicates that a He-Ne laser radiation causes processes leading to a change of charge in proteins of a blood plasma with the negative charge decreased in most cases. The effect depends on individual characteristics of a biological object, on the exposure dose and the incubation time following the irradiation.  相似文献

The rate of GTP hydrolysis in the active site of transducin and that of the release of the phosphate thus formed have been measured. The former step has been found to be a rate-limiting one. The rate constant for GTP hydrolysis is equal to 0.027 s-1 at 23 degrees C, and 0.07 s-1 at 37 degrees C. Besides, it has been shown that the rate of GTPase reaction on the transducin alpha-subunit does not depend on the concentration of a complex of transducin beta- and gamma-subunits or on the presence of cGMP phosphodiesterase and a 48 kDa protein from rod outer segments. According to the results, GTP hydrolysis on transducin proceeds too slowly to account for the rapid quenching of a phosphodiesterase cascade in rod outer segments.  相似文献

The climatic impact on tree radial growth resulting from an atmospheric CO2 doubling was studied for 24 populations of five tree species in the French Alps and the French Mediterranean area. The Arpege AGCM, which predicts a 3 degrees C increase in mean temperature and a light increase of precipitation, is used to estimate the climatic perturbation. The method is based on the integration of estimated climate in an empirical tree-ring to climate model, involving artificial neural networks. Only a few populations are sensitive to the climatic change; all are located on the boundaries of their ecological area and can be divided in two groups. The first one is composed of high altitude populations which show a growth increase induced by the warmer climate during the growing season. The second one, composed of a single Mediterranean Scots pine population, reacts with a severe growth reduction induced by the stronger water stress in summer.  相似文献

The possibility of curling hair is attributed to the fact that the mechanical properties of hair fibres depend on time, temperature and humidity. In this study the dependence of the viscoelastic behaviour of human hair fibres on humidity is characterised in a bending deformation. An experimental set-up was used to perform bending relaxation measurements at different humidities. The relaxation data were fitted using a so-called "stretched exponential equation". The humidity dependence was incorporated by applying time-humidity superposition. Also, the influence of humidity on the initial E-modulus was found. The influence of humidity on the behaviour of human hair fibres could be modelled by using a general characterisation of the behaviour of human hair fibres.The general characterisation is used to predict the recovery in a human hair fibre after curling. The predicted recovery appeared to agree reasonably well with experimentally determined values.  相似文献

The spreading of the muscle proteins myosin and actomyosin depends both on the salt solution in which the proteins are dissolved and on the solution on which they are spread. The spreading is more complete the lower the concentration of the salt solution in which the proteins are dissolved and the higher the salt concentration of the solutions on which the proteins are spread. This effect seems to be due partially to the difference in density allowing the spread droplets a longer life on the surface, and partially to the effect of salt on the charge of the protein. A change in the pH of the substrate has a smaller effect than a change in the salt concentration. Heavy metals like Cu++ or Zn++, inhibit spreading almost completely. The dependence of spreading on these salts and on time was investigated in detail.The effect of the different salts was also compared with the effect of different phosphate compounds. It was noted that the above substances, including the different salts, phosphate compounds, and heavy metals, affect the mechanism of spreading but not films already spread. The viscosity of these fibrillar proteins was compared with other proteins in the monomolecular film state and in myosin an unusually high viscosity was found.  相似文献

The development and survival of free-living stages of gastro-intestinal nematodes of small ruminants are influenced by several abiotic and biotic factors. Within the abiotic factors, most important are the environmental temperature and humidity. They regulate the development of larvae from eggs dispersed on the pasture by the animals faeces. Each parasite species that infect ruminants requires a different time to development, depending on temperature and humidity. Among trichostrongylids, Ostertagia, Teladorsagia and Nematodirus show a strong adaptation to low temperatures. Nematodirus larvae are able to survive to winter inside the egg shell. Temperature and humidity influence the distribution and survival of larvae on pasture. The larval third stage can migrate from faeces to pasture vegetation and they accumulate at the basis of vegetation where stay during the day or in the soil to avoid the desiccation. The forage species affects the migration of larvae on herbage too. Many biological factors contribute to disperse the larvae on the pasture. Dung burying beetles, coprophagous beetles and earthworms can greatly reduce the larvae of some trichostrongylids on pasture. They contribute to the spread of the faecal material on the pasture and allow the larval death as a consequence of drying.  相似文献

The traction exerted by a cell on a planar deformable substrate can be indirectly obtained on the basis of the displacement field of the underlying layer. The usual methodology used to address this inverse problem is based on the exploitation of the Green tensor of the linear elasticity problem in a half space (Boussinesq problem), coupled with a minimization algorithm under force penalization. A possible alternative strategy is to exploit an adjoint equation, obtained on the basis of a suitable minimization requirement. The resulting system of coupled elliptic partial differential equations is applied here to determine the force field per unit surface generated by T24 tumor cells on a polyacrylamide substrate. The shear stress obtained by numerical integration provides quantitative insight of the traction field and is a promising tool to investigate the spatial pattern of force per unit surface generated in cell motion, particularly in the case of such cancer cells.  相似文献

The mechanical characteristics of the ankle such as its kinematics and load transfer properties are influenced by the geometry of the articulating surfaces. A recent, image-based study found that these surfaces can be approximated by a saddle-shaped, skewed, truncated cone with its apex oriented laterally. The goal of this study was to establish a reliable experimental technique to study the relationship between the geometry of the articular surfaces of the ankle and its mobility and stability characteristics and to use this technique to determine if morphological approximations of the ankle surfaces based on recent discoveries, produce close to normal behavior. The study was performed on ten cadavers. For each specimen, a process based on medical imaging, modeling and 3D printing was used to produce two subject specific artificial implantable sets of the ankle surfaces. One set was a replica of the natural surfaces. The second approximated the ankle surfaces as an original saddle-shaped truncated cone with apex oriented laterally. Testing under cyclic loading conditions was then performed on each specimen following a previously established technique to determine its mobility and stability characteristics under three different conditions: natural surfaces; artificial surfaces replicating the natural surface morphology; and artificial approximation based on the saddle-shaped truncated cone concept. A repeated measure analysis of variance was then used to compare between the three conditions. The results show that (1): the artificial surfaces replicating natural morphology produce close to natural mobility and stability behavior thus establishing the reliability of the technique; and (2): the approximated surfaces based on saddle-shaped truncated cone concept produce mobility and stability behavior close to the ankle with natural surfaces.  相似文献

The present paper reports a study on the equilibria and kinetics of the acid-alkaline transition and the azide binding reaction by ferric Aplysia myoglobin. A single completely reversible spectrophotometric titration curve is found over the pH range from similar to 5 to similar to 9, with an apparent pK equals to 7.5 for the acid-alkaline transition. The kinetics of the process, followed by the temperature-jump method, gives, at pH values close to the pK of the transition, one single, well-resolved, relaxation independent of protein concentration and of type of buffer used. The pattern accords to a simple pH dependent reaction, in buffered medium, between the two forms of the protein. The results of the azide binding reaction show that the process conforms to simple equilibrium as expected for a single site protein. The méasured association constant is reported as a function of pH. The kinetics of the reaction of Aplysia metMb with N3- minus shows, on the other hand, a complex behavior. The relaxation pattern is found to strongly depend on pH and ligand concentration in such a way to suggest a linkage between ligand binding and acid-alkaline transition. The system is discussed on the basis of two simplifying conditions, i.e., at low and higher pH with respect to the pK of the acid-alkaline transition. At acid pH the reaction corresponds to a single bimolecular process as expected for a simple binding reaction; at alkaline pH, the dependence of relaxation time on ligand concentration implies the existence of a rate-limiting monomolecular step. On the basis of a reaction scheme implying that binding of the ligand can only occur through the acid (aquomet) form of the protein via the displacement of the water molecule, the experimental data are quantitatively accounted for.  相似文献

Abstract. 1. Fecundity, growth and development were determined at a range of feeding levels and temperatures and compared with the component models for the predator rate of increase.
2. Fecundity and growth rate show a linear dependence on rate of consumption, while development rate conforms to an alternative non-linear model. The non-linearity of development rate is due to a dependence of weight gain during an instar on rate of consumption.
3. The primary influence of temperature is on the rate of consumption, limiting fecundity and growth rate at temperatures below the optimum for the stage, while higher temperatures have a deleterious effect. Temperature has a more direct influence on development rates, allowing comparable growth despite the variation in rates of consumption.  相似文献

The regulation of gene expression is a basic problem of biology. In some cases, the gene activity is regulated by specific binding of regulatory proteins to DNA. In terms of statistical mechanics, this binding is described as the process of adsorption of ligands on the one-dimensional lattice and has a probability nature. As a random physical process, the adsorption of regulatory proteins on DNA introduces a noise to the regulation of gene activity. We derived equations, which make it possible to estimate this noise in the case of the binding of the lac repressor to the operator and showed that these estimates correspond to experimental data. Many ligands are able to bind nonspecifically to DNA. Nonspecific binding is characterized by a lesser equilibrium constant but a greater number of binding sites on the DNA, as compared with specific binding. Relations are presented, which enable one to estimate the probability of the binding of a ligand on a specific site and on nonspecific sites on DNA. The competition between specific and nonspecific binding of regulatory proteins plays a great role in the regulation of gene activity. Similar to the one-dimensional "lattice gas" of particles, ligands adsorbed on DNA produce "one-dimensional" pressure on proteins located at the termini of free regions of DNA. This pressure, an analog of osmotic pressure, may be of importance in processes leading to changes in chromatin structure and activation of gene expression.  相似文献

1. A simple device called a 'pocometer' (POlar COordinate METER) was developed to measure three-dimensional structure of plants. It consists of a tape-measure to measure distance and two protractors to measure zenith angle and azimuth angle.
2. The pocometer can determine locations of points within a few metres distance with a resolution of less than 1cm. Location of any point on a plant can be measured in 10 to 30s depending on the ease of pulling the tape measure to the point of interest.
3. A system to use data obtained with the pocometer to calculate plant light capture was developed. The degree of shading at any point on a plant is estimated by checking obstruction by other plant parts of the view toward the sky at that point.
4. Photon flux density (PFD) on leaf surfaces was estimated for Aucuba japonica , a broad-leaved evergreen shrub, using the above system. The estimated PFDs for individual leaves of a plant corresponded to the sensor-measured PFDs with correlation coefficients of 0·67 to 0·92.  相似文献

The influence of weather and egg contamination on the dynamics of herbage contamination with infective larave of Cooperia oncophora was investigated on artificially contaminated grass plots and in a grazing experiment with 24 first-year grazing calves from May to October 1987 in Lower Saxony, Germany. On the experimental plots the larval translation was highest at the beginning of July and in the second part of September, following high mean weekly temperatures. Between July and September peak recovery of larvae from herbage occurred 4 weeks after contamination. A seasonal pattern of larval translation similar to that on the experimental plots could be demonstrated on the grazed pastures when the number of larvae per m2 of pasture had been adjusted to the previous egg output by means of a contamination index. The resulting 'relative larval density' is regarded as a good indicator for larval development on pasture. From July to September the larval population on pasture resulted mainly from the egg contamination 2-3 weeks earlier. The short persistence of the infective larvae on herbage was probably due to the frequent and heavy rainfall throughout the season, causing a passive washout of larvae into the soil. On single pastures the larval density started to increase within 1 week after the calves had first contact with these fields. The impact of the calves on the distribution of larvae is discussed.  相似文献
