Agrobacterium-mediated gene transfer, or agroinfiltration, can be a highly efficient method for transforming and inducing transient transgene expression in plant tissue. The technique uses the innate DNA secretion pathway of Agrobacterium tumefaciens to vector a particular plasmid-encoded segment of DNA from the bacteria to plant cells. Vacuum is often applied to plant tissue submerged in a suspension of A. tumefaciens to improve agroinfiltration. However, the effects of vacuum application on agroinfiltration and in planta transient transgene expression have not been well quantified. Here we show that vacuum application and release act to drive A. tumefaciens suspension into the interior of leaf tissue. Moreover, the amount of suspension that enters leaves can be predicted based on the vacuum intensity and duration. Furthermore, we show that transient expression levels of an agroinfiltrated reporter gene vary in response to the amount of A. tumefaciens vacuum infiltrated into leaf tissue, suggesting that vacuum infiltration conditions can be tailored to achieve optimal transient transgene expression levels after agroinfiltration.  相似文献

The purpose of this work was to evaluate two methods (floral dip and vacuum infiltration) of in planta transformation of Arabidopsis thaliana. The key issue of this work is the identification of the developmental stages of A. thaliana flower buds subjected to agroinfection, optimal for the successful transformation. Histological tests performed after agroinfection made it possible to establish the patterns of a GUSPlus reporter gene expression in the examined plants and thus precisely define the range of flower developmental stages most appropriate for efficient transformation. Two plasmids, CAMBIA 1305.1 and CAMBIA 2301, were used. Verification of the transgenic nature of plants was carried out by detection of CaMV::gusA and CaMV::GUSPlus transgenes and their expression in transgenic plants by appropriate molecular and histochemical methods. For the flower dip transformation, three concentrations of Silwet L-77 surfactant and two inoculation times were tested. The most efficient treatment appeared to be 2-min-long flower bud inoculation and 400 μl/l surfactant (pCAMBIA 1305.1 − 1.73%; pCAMBIA 2301 − 2.01%). In the case of vacuum infiltration method, the highest efficiency of the transformation occurred when the inoculation time was 4 min (pCAMBIA 2301 − 1.55%; pCAMBIA 1305.1 − 1.37%). 采用水蒸气蒸馏法提取中草药贯叶连翘(全草)挥发油。通过气相色谱/质谱联用技术测定了挥发油的化学成分,从中鉴定出75种化合物,其中百里香酚、桉油精、丁香油酚等酚类物质为首次检出。结果表明,贯叶连翘因产地不同、部位不同,其挥发油的成分有较大的差别。  相似文献

目的:研究土茯苓中的挥发性成分。方法:利用水蒸气蒸馏法提取土茯苓(Smilax glabraRoxb.)挥发油,用GC-MS进行测定,结合计算机检索技术对分离的化合物进行结构鉴定,应用色谱峰面积归一化法计算各成分的相对百分含量。结果:分离了40个化合物,鉴定出22个化学成分,占总挥发性成分的47.88%,其中相对百分含量大于1%的分别确定为棕榈酸17.87%,萜品烯-4-醇7.533%,亚油酸6.775%,正壬烷4.509%,8,11-十八碳二烯酸甲酯2.215%,α-雪松醇1.81%,甲基棕榈酯1.293%。结论:22个挥发性成分均为首次从该植物中得到。  相似文献

SDE-GC-MS法分析三种虫生真菌菌丝中挥发性成分   总被引:2,自引:0,他引:2  
采用同时蒸馏萃取-气相色谱质谱联用(SDE-GC-MS)的方法分析了蝉拟青霉、拟细羽束梗孢、根足被毛孢菌丝的挥发性成分,从中分别鉴定出44、28和19种化合物,它们主要为萜类化合物、芳香族化合物、醇类、烷烃类、酯类和醛类。成分比较发现,3种虫生真菌挥发性物质中有3种主要共有成分,分别为丁羟基甲苯、1-辛烯-3-醇、苯乙醛。除共有成分外,它们各自都有大量特有成分,其中蝉拟青霉主要有5-甲基-2-呋喃-乙酸酯、反式-2,4-癸二烯醛、长叶烯等;拟细羽束梗孢主要有5-羟基-2-癸烯酸-δ-内酯、2,4-二甲基-恶唑、苯酚、β-榄香烯等;根足被毛孢主要有3,4,5-三甲基-苯甲醛、1,3-二甲基-3,4,5,6-四氢化-2(1H)嘧啶、顺式-2-羟基-1-(羟甲基)-9-十八碳一烯酸乙酯。  相似文献

Hybrid lead halide perovskites are promising materials for future photovoltaics applications. Their spectral response can be readily tuned by controlling the halide composition, while their stability is strongly dependent on the film morphology and on the type of organic cation used. Mixed cation and mixed halide systems have led to the most efficient and stable perovskite solar cells reported, so far they are prepared exclusively by solution‐processing. This might be due to the technical difficulties associated with the vacuum deposition from multiple thermal sources, requiring a high level of control over the deposition rate of each precursor during the film formation. In this report, thermal vacuum deposition with multiple sources (3 and 4) is used to prepare for the first time, multications/anions perovskite compounds. These thin‐film absorbers are implemented into fully vacuum deposited solar cells using doped organic semiconductors. A maximum power conversion efficiency of 16% is obtained, with promising device stability. The importance of the control over the film morphology is highlighted, which differs substantially when these compounds are vacuum processed. Avenues to improve the morphology and hence the performance of fully vacuum processed multications/anions perovskite solar cells are proposed.  相似文献

An integrated soil vacuum extraction/ biofiltration system has been designed and installed at a gasoline‐contaminated leaking underground storage tank (LUST) site in southern Delaware. The system remediates contaminated moisture entrained in the air stream, employs automatic water level controls in the filters, and achieves maximum vapor extraction and VOC destruction efficiency with an optimum power input. In addition, the valving and piping layout allows the sequence of air flow through the filters to be reversed at given time intervals, which minimizes biofouling, thereby increasing efficiency by decreasing system backpressure. This integrated system achieves VOC destruction rates of up to 69%. The modular design allows for easy mobilization, setup, and demobilization at LUST sites throughout Delaware.  相似文献

TiO2 has been well studied as an ultraviolet (UV) photocatalyst and electrode material for lithium‐ion rechargeable batteries. Recent studies have shown that hydrogenated TiO2 displayed better photocatalytic and lithium ion battery performances. Here it is demonstrated that the photocatalytic and battery performances of TiO2 nanocrystals can be successfully improved with a facile low‐temperature vacuum process. These TiO2 nanocrystals extend their optical absorption far into the visible‐light region, display nanometer‐scale surface atomic rearrangement, possess superoxide ion characteristics at room temperature without light irradiation, show a 4‐fold improvement in photocatalytic activity, and has 30% better performance in capacity and charge/discharge rates for lithium ion battery. This facile method could provide an alternative and effective approach to improve the performance of TiO2 and other materials towards their practical applications.  相似文献

Ethylenediaminetetraacetic acid (EDTA) solution is used to decalcify bone specimens for histological examination. Sodium hydroxide (NaOH) has been used to dissolve EDTA and to bring EDTA solutions to neutral pH. This solution, however, requires several weeks to decalcify bone specimens. We investigated a new de-calcification fluid using concentrated ammonium hydroxide (NH4OH) to dissolve EDTA and to adjust the pH to neutral. Decalcification was performed using a magnetic stirrer with and without vacuum, or with a sonic cleaner. Decalcification end point was confirmed using both the weight loss and X-ray methods. After decalcification, specimens were processed through paraffin and sections were stained with hematoxylin and eosin. Decalcification employing NH4OH required an average of six days. Light microscopy indicated good retention of cellular detail.  相似文献
