应用真空负压 ABC 法显示 HPV-1抗原比常规 ABC 法效果好,敏感性高,时间短,整个过程只需一个多小时,阳性物明显突出,颗粒均匀,色泽鲜艳,呈黄棕色,背景清晰等优点。还可提高抗体的稀释度,降低成本。与微波技术相比,具有设备简单,易于操作,安全可靠,不受条件限制,经对26例尖锐湿疣的病例进行多层次稀释度的研究,取得了满意的结果,认为该法值得推广应用。 相似文献

We report a modification of the immunogold-silver staining method (IGSS) for localizing hepatic phosphoenolpyruvate carboxykinase (PEPCK) in tissue sections, and we compare the efficacy of localizing the primary antibody with either a 5 nm gold labeled secondary antibody or 5 nm gold labeled secondary and tertiary antibodies. Light microscope examination of 10 μm frozen sections demonstrated that the use of combined secondary and tertiary gold labeled antibodies was superior to using a secondary gold labeled antibody alone. The increased labeling density (number of colloidal gold particles/antigenic site/cell) achieved by combined gold labeled antibodies was confirmed by electron microscopy. The increased labeling density resulted in a two-thirds reduction in the time needed for the IGSS physical development of the silver shells and less background. We achieved intense specific staining of hepatocytes expressing PEPCK while minimizing background staining. The use of combined secondary and tertiary gold labeled antibodies enhances the signal-to-noise ratio, achieves high resolution and is a suitable method for use in both light and electron microscopy. 相似文献

A series of ZnB2O4 phosphors doped with different concentrations of Eu and Dy (0.05 0.1, 0.2, 0.5, 1.0 mol%) and co-doped with Ce (1, 2, 5, 7, 10 mol%) respectively was prepared via the solid-state reaction technique and the thermoluminescence (TL) behaviour of gamma ray-irradiated samples was studied. The synthesized samples were irradiated with γ-rays for the dose range 0.03–1.20 kGy. The TL intensity variations with dose, dopant concentration, and the effect of co-doping were studied. The TL response curves for ZnB2O4:Eu3+ and ZnB2O4:Dy3+, ZnB2O4:Eu3,Ce3+ and ZnB2O4:Dy3+,Ce3+ phosphor were observed. It was revealed that ZnB2O4:Eu3+ showed a linear TL behaviour for the dose 0.03–1.20 kGy and ZnB2O4:Dy3+ showed linearity for the gamma dose range 0.03–0.10 kGy. Furthermore, fading for all the samples was observed to be less than 10% for a storage period of 30 days. In addition to this, the trapping parameters, especially activation energies were evaluated using the Ilich method and the initial rise method. 城市绿色开敞空间是城市公共空间的重要组成部分,合理配置城市绿色开敞空间对城市宜居环境建设至关重要。空间可达性常用于评价公共服务设施空间布局的合理性,两步移动搜索法即是一种直观、运算简便、应用广泛的可达性计算方法,但该方法未考虑需求主体之间的差异性。本研究以南京市中心城区为例,利用手机信令数据提取需求者属性特征以及空间分布,通过分类细化需求主体改进搜索策略,改善两步移动搜索法传统研究未考虑不同群体差异性、统计数据滞后、空间尺度粗等弊端。结果表明,南京市中心城区绿色开敞空间可达性呈现出明显的空间分异特征,江南主城可达性总体比江北新区高,低值区集中在人口密度高、绿色开敞空间资源规模小的中心城区周边;不同人群的绿色开敞空间可达性总体格局相似,但在数值上老年人可达性均值是其他年龄段的14.60%,工作人群可达性均值是居住人群的86.02%,老年人作为弱势群体在享受绿色开敞空间服务中被边缘化。研究结果可为优化南京市绿色开敞空间布局提供科学依据,也可为其他公共服务设施可达性评价提供借鉴。 相似文献

本研究旨在建立一种基于流式细胞术的牛多细胞因子检测方法。对前期制备并筛选出的针对牛细胞因子IFN-γ、IL-2、TNF-α、IP-10和MCP-1的单克隆抗体进行荧光标记,与细胞表面分子抗体组合搭配,进行牛多细胞因子流式检测方法的建立和优化。随后利用建立的方法进行BCG体外感染牛外周血单个核细胞的细胞因子表达规律测定,并结合CFP10-ESAT6蛋白刺激剂评价上述细胞因子作为牛结核诊断标识的潜力。建立的牛多细胞因子流式检测方法可以有效测定BCG感染牛外周血T淋巴细胞的细胞因子表达,其中IFN-γ、IL-2、TNF-α在感染40h后持续上升,而IP-10和MCP-1表达水平呈现下降趋势;对于牛外周血CD4+T淋巴细胞IFN-γ、IL-2、TNF-α的联合检测能有效区分牛结核阳性和阴性样品。这一方法为牛病原菌感染、疫苗注射后细胞免疫应答水平评价以及疫病诊断提供了重要技术手段。 相似文献

Cryoconite, the dark sediment on the surface of glaciers, often aggregates into oval or irregular granules serving as biogeochemical factories. They reduce a glacier's albedo, act as biodiversity hotspots by supporting aerobic and anaerobic microbial communities, constitute one of the organic matter (OM) sources on glaciers, and are a feeder for micrometazoans. Although cryoconite granules have multiple roles on glaciers, their formation is poorly understood. Cyanobacteria are ubiquitous and abundant engineers of cryoconite hole ecosystems. This study tested whether cyanobacteria may be responsible for cryoconite granulation as a sole biotic element. Incubation of Greenlandic, Svalbard, and Scandinavian cyanobacteria in different nutrient availabilities and substrata for growth (distilled water alone and water with quartz powder, furnaced cryoconite without OM, or powdered rocks from glacial catchment) revealed that cyanobacteria bind mineral particles into granules. The structures formed in the experiment resembled those commonly observed in natural cryoconite holes: they contained numerous cyanobacterial filaments protruding from aggregated mineral particles. Moreover, all examined strains were confirmed to produce extracellular polymeric substances (EPS), which suggests that cryoconite granulation is most likely due to EPS secretion by gliding cyanobacteria. In the presence of water as the only substrate for growth, cyanobacteria formed mostly carpet-like mats. Our data empirically prove that EPS-producing oscillatorialean cyanobacteria isolated from the diverse community of cryoconite microorganisms can form granules from mineral substrate and that the presence of the mineral substrate increases the probability of the formation of these important and complex biogeochemical microstructures on glaciers. 相似文献

An acceleration of soil respiration with decreasing CO2 concentration was suggested in the field measurements. The result supporrs that obtained in laboratory experiments in our
previous study. The CO2 concentrations in a chamber of the alkali absorption method (the AA-method) were about 150–250 parts/106 lower than that in the atmosphere (about 350 parts/106), while those observed in the open-flow IRGA method (the OF-method) were nearly equal to the soil surface CO2 levels. The AA-method at such low CO2 levels in the chamber appears to overestimate the soil respiration. Our results showed that the rates obtained by the AA-method
were about twice as large as those by the OF-method in field and laboratory measurements. This finding has important consequences
with respect to the validity of the existing data obtained by the AA-method and the estimation of changes in the terrestrial

通过“微生物学实验”课程多年来的混合式教学实践发现,学生的主动性是学习效果的核心要素。鉴于在以往的教学实践中,无论采用何种教学手段,总有部分学生的学习主动性不能得到体现。我们近年来采用轮流组长责任制下的任务驱动教学法,极大地调动了所有学生的学习动力。每位学生在担任组长工作时从课前任务、课堂执行、课后总结等方面都发挥了重要作用,而不担任组长时也会积极主动地配合组长工作。最终,实验课程学习成绩优秀率占比、体现素质能力培养的成绩分项都显著提升。同时,课程教学也收获了良好的学生认可和反馈。 相似文献

生物脱氮是由微生物主导的地球氮循环中的重要环节之一,主要包括硝化、反硝化和厌氧氨氧化(anaerobic ammonium oxidation,anammox)等过程。在微生物联合作用下,污水中的有机氮及氨氮经一系列作用转化为氮气,这种经济高效、环境友好的处理工艺在世界范围内得到广泛应用。群体感应(quorum sensing,QS)以信号分子为媒介通过改变菌群密度和周围环境变化来调节微生物的各种行为。大量的研究已证实调控QS信号分子在生物脱氮中具有应用潜力。本文介绍了各种信号分子类型,从基因组学、实际应用等方面综述了各类信号分子以及检测方法,同时针对酰基高丝氨酸内酯(acyl homoserine lactones,AHLs)类信号分子在生物脱氮中的作用进行详细介绍。然而不足之处在于信号分子研究只是停留在实验室阶段,仅仅研究了单一信号分子对生物脱氮的影响。未来可将信号分子应用于实际污水,研究多种信号分子共同作用以及多种微生物之间的QS现象。 相似文献
