A protocol was developed for regeneration and Agrobacterium-mediated genetic transformation of Lesquerella fendleri. Calli were first induced from hypocotyls and cotyledons on MS plus 0.5 mg l−1 BA, 1 mg l−1 NAA and 1 mg l−1 2,4-D, then co-cultivated for 2–3 days in darkness on MS supplemented with 0.5 mg l−1 BA, 0.2 mg l−1 NAA and 100 μmol l−1As together with Agrobacterium tumefaciens strain EHA105/pCAMBIA1301 that harbored genes for uidA (GUS) and hygromycin resistance. Following co-cultivation, calli transfected by A. tumefaciens were transferred to MS with 0.5 mg l−1 BA, 0.2 mg l−1NAA, 500 mg l−1 Cef and 10 mg l−1 hygromycin and cultured for 10 days, then the hygromycin was increased to 20 mg l−1 on the same medium. After 4 weeks the resistant regenerants were transferred to MS with 0.5 mg l−1BA, 0.2 mg l−1 NAA, 500 mg l−1 Cef and 25 mg l−1 hygromycin for further selections. Transgenic plants were confirmed by polymerase chain reaction analysis, GUS histochemical assay and genomic Southern blot hybridization. With this approach, the average regeneration frequency from transfected calli was 22.70%, and the number of regenerated shoots per callus was 6–13. Overall results described in this study demonstrate that Agrobacterium-mediated transformation is a promising approach for improvement of this Lesquerella species.  相似文献

Cymbopogon nardus L. could be propagated via tissue culture using axillary buds as explants. The aseptic bud explants obtained using double sterilization methods produced stunted abnormal multiple shoots when they were cultured on Murashige and Skoog medium (MS) supplemented with 1.0 mg L-1 or 2.0 mg L-1 benzyladenine (BA). Stunted shoots that cultured on MS + 1.0 mg L-1 BA + 1.0 mg L-1 N6-isopentenyl-adenine (2iP) could induce elongation of shoots from about 60% of the stunted shoots. Normal multiple shoots could be induced at the highest (19.7 shoots per bud) from the bud explants within six weeks when cultured on proliferation medium consisted of MS supplemented with 0.3 mg L-1 BA and 0.1 mg L-1 indole-3-butyric acid (IBA). The separated individual shoot produced roots when transferred to basic MS solid medium. The essential oils that were contained in the mature plants namely citronellal, geraniol and citronellol were also found in thein vitro C. nardus plantlets. Citronellal was the main essential oil component in the matured plants while geraniol was the main component in thein vitro plantlets.  相似文献

以章丘大葱为试验材料,从形态组织学及生理生化等方面探讨了大葱试管苗玻璃化发生的可能机理。结果表明,玻璃苗形态组织结构与正常苗有很大差异,玻璃苗组织含水量增加,过氧化物酶(POD)活性增强,叶绿素总量、叶绿素a、叶绿素b、过氧化氢酶(CAT)的活性、可溶性蛋白含量均降低。玻璃苗酯酶同工酶谱带与正常苗相比,增加了一条带,同时缺少了两条带;可溶性蛋白电泳分析表明,玻璃苗与正常苗间也存在明显差异。可见,当外界环境条件不适宜植物生长需要时,植物体在基因表达水平上将会受到影响,表达产物的差异可引起生理代谢功能上的紊乱,从而导致形态结构的畸型,进而产生玻璃化苗。  相似文献

The scope of morphological plasticity of vegetative structures among Podostemoideae (Podostemaceae) is documented for Crenias weddelliana, a neotropical species, Maferria indica, a palaeotropical species, and Sphaerothylax abyssinica, from Kenya, and compared with related taxa. The study highlights intrinsic characters of the widely enigmatic plant body of many species of the subfamily Podostemoideae. These include dorsiventrality of shoots occurring irrespective of gravity, incurvate distichy and one‐sided spirodistichy correlated with shoot dorsiventrality, asymmetric leaves, and several types of positioning of the two prophylls and inflorescence structures. The homogeneity of hairs of the ‘Zeylanidium olivaceum type’ established on the subulate leaves of some Indian species is of taxonomic value. The latter also applies to the stipella (not stipule) on the asymmetric compound leaf in New World species. The stimulating effect of cork pieces on hypericin and pseudohypericin biosyntheses was studied in cells of shoots regenerated from the callus cultures of St. John's wort (Hypericum perforatumL.). The addition of the cork matrix slightly stimulated shoot growth and enhanced pseudohypericin biosynthesis about threefold (to 0.4 mg/g dry wt). Pseudohypericin production increased proportionally with the amount of cork material added (from 1 to 4 mg/ml of growth medium). Further increase in the amount of cork pieces inhibited both pseudohypericin production and shoot growth. Organic and aqueous extracts of cork pieces did not affect the production of these substances.  相似文献

To maintain embryogenic cell lines ofPimpinella brachycarpa, we suspension-cultured friable and rapidly growing yellowish calli in an MS liquid medium containing 0.2 ~ 2,4-D and 0.5pM BAP. Efficient somatic embryogenesis was achieved when selected cells were then transferred to an MS medium (0.2% gelrite) that contained 0.2gM 2,4-D, 0.5 uM BAP, and 10.0 laM TDZ (thidiazuron). These cells were cultured at 27°C under continuous illumination (21.5 I~E m-2 s-l). Embryogenic calli expanded about four-fold, and developed into pale yellow calli. Somatic embryogenesis was initiated only from glossy and nodular-type calli. After two more weeks of culture, globular embryos appeared on the surface of calli grown in the MS medium that contained 10.0 /aM TDZ only, or in combination with 0.5 gM NAA. Experimenting with 2,4-D, an auxin, to promote embryogenic calli resulted in excessive browning and death. We overcame this problem by growing glossy embryogenic and nodular calli on media that contained 10.0 gM TDZ. Calli that were not treated with TDZ turned dark brown and were not viable. Up to 74% of the calli showed somatic embryos when the medium was supplemented with 10.0 uM TDZ and 0.5 uM NAA. Embryos from these TDZ-induced, somatic embryogenic calli grew efficiently, forming multiple shoots and developing into normal plants. Therefore, efficient differentiation of suspension-cultured cell clusters into embryogenic calli, along with treatment of subsequent somatic embryos by TDZ, suggests that TDZ probably helps in establishing the optimum cytokinin-auxin ratio required for induction and expression of somatic embryogenesis.  相似文献

A survey of a range of plant tissues showed that the hydroxycinnamate CoA ligase in crude extracts of pea shoots had a high relative activity towards sinapic and other methoxycinnamic acids, together with high activity with p-coumaric acid. The pea enzyme has been resolved by chromatography on DEAE-cellulose into two peaks which differ in their substrate specificity. The form which elutes at relatively low salt concentrations has a ratio activity towards p-coumaric and sinapic acids of about 1.8:1 while the form eluting at higher salt concentrations, although showing very high activity with p-coumaric acid, is inactive towards sinapic acid. The pattern of elution of these forms following gel filtration on Ultragel AcA 34 and Sephadex G100 suggests that these two isoenzymes which differ in ionic properties and substrate specificity can exist in two or three molecular weight forms and there is evidence that these forms are under certain circumstances interconvertible.  相似文献

 Fossil vegetative and reproductive structures from deposits of the Raritan Formation in New Jersey (Turonian, Upper Cretaceous, ∼90 MYBP) include ferns, gymnosperms, and angiosperms. Gymnosperms collected from this formation have been known since the beginning of the 20th century. Fossil leaves, wood and seed cones have been are identified as belonging to the Cupressaceae, Pinaceae, and Taxodiaceae. In the present contribution, we describe a series of fossil dwarf shoots, leaves and a pollen cone belonging to the family Pinaceae. Fossils are charcoalified with preserved three-dimensional structure and excellent cellular detail. The dwarf shoots are assigned to a new species Prepinus crossmanensis sp. nov. and to the previously described Pinus quinquefolia Jeffrey. The new species Prepinus crossmanensis differs in size, shape, presence of hypodermis, sclerenchyma and stomata in the cataphylls, and number and shape of needle leaves from previously known species. Also, isolated leaves were found that were assigned to the new species, Prepinus raritanensis sp. nov. The new species is differentiated by the size and shape of stomata, the presence of number of layers of the hypodermis; and the cell-shape and number of layers of the mesophyll and transfusion tissue. A previously undescribed male cone, Amboystrobus cretacicum gen. and sp. nov., has an axis with spirally attached microsporophylls, each bearing two abaxial ovoid microsporangia. The pollen grains are monosulcate and bisaccate (eusaccate), with an elliptical corpus, granulate exine sculpture, and honeycomb-like wall structure.
