We have analyzed in transgenic tobacco the expression of a chimeric gene containing 5 sequences of the rice rab-16B gene fused to the -glucuronidase (GUS) reporter gene. This construct, a translational fusion (–482 to +184) including 14 amino acids of the RAB-16B protein, is expressed only in zygotic and pollen-derived embryos. In zygotic embryos, GUS activity begins to accumulate 10 days after flowering (daf), and increases until seed maturation at 25 daf. Immunological measurements of endogenous abscisic acid (ABA) accumulation in these seeds showed a close parallel between hormone levels and GUS activity. However, GUS activity could not be reproducibly induced by treatment of immature embryos with ABA (10 M). Neither GUS activity nor GUS mRNA could be detected in leaves of transgenic tobacco even after ABA treatment. In contrast, GUS activity could be induced to high levels in pollen-derived embryos by treatment with ABA. Our results show that 482 bp of 5 sequences of the rice rab-16B promoter can confer in transgenic tobacco developmentally regulated expression in embryos but not ABA-responsive expression in vegetative tissues.  相似文献

The environmental factors governing the distribution and behaviour of total and methylmercury within mercury contaminated shallow lakes, associated with the River Yare, Norfolk, UK, have been assessed in situ through the use of sediment cores. These were analysed for microbial and chemical changes on both a temporal and spatial scale. The distribution of total mercury proved to be site dependent and related to the hydrology, sediment transport dynamics, the degree of sediment disturbance and distance from the contamination source. The core profiles revealed a subsurface peak in total mercury with maximum concentrations residing at depths of 12 and 36 cm, depending upon location, with enrichment extending down to depths in excess of 88 cm. This vertical distribution was deemed to reflect historical emission in the late 1960s and early 1970s rather than post depositional migration effects. The distribution of methylmercury was distinct from that of its inorganic counterpart since it also displayed temporal variability with highest concentrations occurring in the spring and summer. Maximum concentrations also prevailed in the uppermost 12 cm of sediment, with peak loadings at or just below the sediment/water interface and detectable levels being restricted to the upper 36 cm. The temporal and spatial behaviour of methylmercury appeared to reflect variations in redox potential, the availability of the Hg2+ ion and the nature of microbial populations.  相似文献

The susceptibility of 101 pneumococcal isolates from the respiratory tract during 1991–1994 was examined and compared with the susceptibility of isolates over the period of 1975–1990. A rapid increase of resistance was seen not only to penicillin but also other antimicrobial agents. During 1991–1994, 38% of all the isolates were resistant to penicillin. The rates of resistance during this period were 16–23% for three newer cephalosporins, 18% for imipenem, 69% for tetracycline, 31% for erythromycin, 20% for chloramphenicol and 9% for clindamycin. The use of antibiotics within one month prior to pneumococcal isolation was correlated with penicillin resistance (P < 0.05). Serotyping of the isolates by antiserum revealed differences in predominant types between penicillin-resistant (19F, 23F, 4) and -susceptible isolates (15, 4, 11A). Our data suggests that anti-pneumococcal antibiotics should be carefully chosen on the basis of susceptibility tests.  相似文献

We have developed a reproducible biolistic procedure for the efficient transformation of embryogenic suspension cells of an improved aromatic Indica rice variety, Pusa Basmati 1. The -glucuronidase gene was used to assay transient transformation; other plasmids carrying either a potato protease inhibitor 2 (Pin2) gene, or a late embryogenesis-abundant protein (LEA3) gene from barley, were used for the optimization of biolistic process and transgenic plant production. After optimization of the procedure, over 600 transient transformants and at least five fertile plants showing integrative transformation were obtained per bombarded filter. At least 30% of the plants were derived from independent transformation events. The new improved procedure involves the use of a reporter gene or other useful genes driven by the strong rice actin 1 gene (Act1) promoter, osmotic pre-conditioning of cells for 24 h on medium supplemented with 0.25 M mannitol prior to bombardment, use of gold particles for DNA delivery, and use of plant regeneration medium with high (1.0%) agarose concentration.  相似文献

用制备型聚丙烯酰胺凝胶电泳和制备型等电聚焦纯化了曾报道的光敏核不育水稻 (Oryza sativa)农垦 58S叶绿体的特异性蛋白质 P2 ,得到 SDS- PAGE和等电聚焦 (IEF )纯的 P2。经 SDS- PAGE和 IEF测定 ,该纯蛋白质的分子量是 61 k D,等电点是 5.8。现称 P2为 P61。氨基酸序列分析表明 P61的 N-端氨基酸序列与水稻和大麦叶绿体 ATPaseβ亚基的 N-端氨基酸序列同源。  相似文献

Regenerable embryogenic suspensions of elite Indica (group 1) rice varieties IR24, IR64, IR72 and an advanced Indica rice breeding line IR57311-95-2-3 were established within 6–8 weeks from 3–4 week old calli derived from mature seeds. Transgenic rice plants were obtained by introducing a plasmid carrying genes encoding hygromycin phosphotransferase (hph, conferring resistance to hygromycin B) and ß-glucuronidase (uidA), both driven by the CaMV 35S promoter, via particle bombardment of embryogenic suspensions. The effect of osmotic conditioning on transformation was evaluated. Regenerated plants were resistant to hygromycin B and expressed the uidA (GUS) gene. The growth of mother plants (R0) was normal and seeds were produced. Southern blot analysis of R0 and R1 plants showed that hygromycin resistant plants contained intact hph genes that were inherited in a Mendelian fashion. Cropping on jhum fallows in north-eartern India is predominantly done for one year in a jhum cycle. If second year cropping is done, expanse of the forest land required for slashing and burning could be reduced significantly. We tested this hypothesis in a young (6 yr) and an old (20 yr) jhum fallow. We also evaluated if the productivity during second year cropping could be alleviated by auxiliary measures such as tilling the soil or application of fertilizers (chemical or farm-yard manure or both in combination). The results demonstrate that the ecosystem productivity (total dry matter production) and economic yield (rice grain production) decline with shortening of jhum cycle. Second year cropping causes a further decline in ecosystem productivity in old jhum field, but not in young jhum field. Economic yield from second year cropping in its traditional form (without any fertilizer treatment) is not much lower than that in the first year, and can be improved further by manuring the soil. Tilling of soil improves neither ecosystem productivity nor economic yield. Different fertilization treatments respond differently; while inorganic manuring enhances ecosystem productivity, a combination of inorganic and organic manuring improves economic yield  相似文献

Gravitropism of maize and rice coleoptiles was investigated with respect to its dependence on the angle of displacement or the initial stimulation angle (ISA). Close examination of curvature kinetics and the response to a drop in stimulation angle (SA) indicated that the gravtropic response during an early but substantial part of the curvature development is directly related to the ISA, there being no effect of the reduction of SA resulting from the curvature response itself. On the basis of this finding, the relationship between the steady SA and the curvature rate was determined. In maize, the curvature rate increased linearly with the sines of SAs up to an SA of 90 degrees. Rice coleoptiles, however, showed a saturation curve in the same range of SAs. The saturation profile was nearly identical between coleoptiles grown in air and those submerged in water, although the latter elongated much faster. Rice coleoptiles appeared to be far more sensitive to gravity than maize coleoptiles. It is concluded that the sensitivity to gravity, assessed through dependence on ISA, is a property inherent to a given gravitropic organ. Long-term measurements of curvature indicated that the coleoptiles bend back past the vertical. This overshooting was marked in submerged rice coleoptiles.  相似文献
