Abstract

The aim of this work was to determine the effects of cGnRH I pulse frequencies on FSH and LH release and the changes in features and number of cultured laying hen FSH-cells and LH-cells in vitro. Primary adenohypophyseal cell cultures taken from laying hens were stimulated by four 5 min pulses using 1 or 10 nM cGnRH, administered with interpulses between pulses at 15, 30 or 60 min. Pulse frequencies and dose dependent effects were examined in six separate experiments including two controls. After the last interpulse time, the supernatants were collected and stored at ?70° C until the performance of an indirect enzyme-linked immunosorbent assay (ELISA) using chicken LH and chicken FSH antisera at 1:1000 and 1:2000 dilutions, respectively. Supernatants were coated in duplicate on the inner surface of Immulon 2 plates and later blocked with the optimal solutions. They were incubated with each antiserum and subsequently with isotype-specific peroxidase-labeled anti-rabbit antibodies. Hydrogen peroxide/o-phenylenediamine was added as substrate/chromogen and the optical density (OD) was determined at 492 nm. The ABC immunocytochemical method was performed to characterize and re-count the gonadotropes employing anti-chicken FSH and anti-chicken LH as primary antibodies. The number of FSH-LH cells was obtained using stereological analysis and the data were statistically processed. The ODs obtained for each anti-hormone were compared with the control groups and with each other. Significant differences were found in number of aggregated-positive LH cells, which decreased with 1 nM cGnRH-I, 15 vs. 30 min pulses, increased with 30 vs. 60 min pulses, and also with 10 nM cGnRH-I, 30 vs. 60 min pulses. Aggregated positive FSH cells, however, did not show significant differences in percentage at any GnRH dose or pulse frequencies, but did show activity at low pulse frequencies of 15 and 30 min. The results suggest that LH cells varied in percentage in a dose dependent manner at higher pulse frequency (15 min) and were dose independent at low pulse frequency (60 min) and showed inactive features; while FSH cell numbers were unaffected showing features of activity at low pulse frequencies. High and moderate pulse frequencies of cGnRH-I (15-30 min) increased the FSH release in dose independent manner without changes in features or percentage of FSH cells. Low pulse frequency (60 min) of cGnRH-I increased LH release dose independently disminished LH cell percentage and showed changes in cells’ features. These results in avian cells showed differences in responses to GnRH pulse frequencies from those reported earlier in mammals.  相似文献

Summary The trail-laying behaviour of foragers of the antLasius niger was observed in the laboratory on a 20 cm bridge between the nest and the food source. We measured both the frequency of trail laying, as defined by the proportion of trips during which trail laying occurred, and its intensity, as defined by the number of marks laid during one bridge crossing.Foragers do not exhibit trail-laying behaviour until a food source is discovered. Trail laying then occurs more or less equally both to and from the nest, and both its frequency and intensity decrease as the recruitment proceeds. Foragers from very small colonies less than a year old appear to have quantitatively the same trail laying behaviour as those from older and much larger colonies.Groups of recruiters and recruits were individually marked. Their trail laying intensity was similar, both for trips to and from the nest, and for an ant's first, second, third and fourth trip. The frequency diminished rapidly with the number of trips made by each individual, and was 2–3 times higher for recruiters than for recruits, for trips both to and from the nest. Even though foragers stop marking after a variable number of passages, they continue to move between the nest and the food source, and other ants start marking. Different foragers appear to have widely different levels of trail laying, although we cannot say whether these differences are stable between different recruitments.Trail laying is strongly affected by the foragers' position on the bridge, especially for ants returning to the nest which lay up to five times more on the segment closest to the source than that closest to the nest. Foragers on a weakly marked trail appear to mark more than those on a well-marked trail. However, this effect is weak and could partly be attributed to their lower speed.Finally, a model using the experimental data gathered on the individuals' trail-laying behaviour reproduced satisfactorily the colony's overall trail laying.  相似文献

A population of Acrobeloides nanus in Australia is described and illustrated, based on light and scanning electron microscopy. Embryogenesis from egg laying to hatching is followed over a wide range of temperatures. At 15 C, hatching occurs in about 125 hours and at 35 and 37.5 C after about 40 hours. At 40 C, egg development ceases early in cleavage. The capacity of A. nanus to develop over such a range of temperatures, and its anhydrobiotic capabilities, are discussed in relation to its survival and wide distribution in Australia.  相似文献

Populations of animals with resident and migratory individuals provide an ideal system for addressing questions concerning the evolution of migration. Partially migratory populations may persist because residents and migrants have equal fitness or because migration is based on conditional asymmetries. Studies measuring the costs and benefits of migration provide empirical data to test hypotheses concerning the maintenance of partial migration within a population. In this study, we measured the reproductive differences between resident and migrant females in a pond-breeding amphibian, the red-spotted newt Notophthalmus viridescens . We used large field enclosures to repeatedly sample egg laying over the prolonged breeding season of this species. Resident females did not lay a greater number of eggs or begin laying eggs earlier, despite beginning the breeding season earlier and having a higher mass than migrant newts. The only difference in reproduction we detected was that the eggs of resident females hatched into larger larvae compared with the larvae of migrant females. We discuss this result in the context of other potential trade-offs between residency and migration. This study illustrates the phenology of egg laying of N. viridescens and our results contribute to understanding the population dynamics of partially migratory species.  相似文献
